Long-Acting Antiretroviral Nanoformulation Development And Subcellular Trafficking, Dongwei Guo
Long-Acting Antiretroviral Nanoformulation Development And Subcellular Trafficking, Dongwei Guo
Theses & Dissertations
The introduction of nanoformulated antiretroviral therapeutic regimens is a promising alternative to standard once a day oral treatment of HIV infection. Our lab has pioneered this effort and was successful in harnessing mononuclear phagocytes (monocytes, dendritic cells and macrophages) as nanoformulated drug carriers. The approach was developed as Trojan horses for drug transport, delivery and distribution to sites of viral replication in order to facilitate microbial elimination in HIV sanctuaries. However, the remaining challenges for current antiretroviral nanoformulations include to formulate a broad range of hydrophilic short-acting drugs, and to elucidate the mechanism of sequestered nanoparticles in macrophages at the …
Role Of B Cell And Hematopoietic Cell Intrinsic Actions Of Erα In Lupus Pathogenesis, Dana E. Tabor
Role Of B Cell And Hematopoietic Cell Intrinsic Actions Of Erα In Lupus Pathogenesis, Dana E. Tabor
Theses & Dissertations
Lupus is a chronic autoimmune disease characterized by the presence of autoimmune B and T cells and the production of pathogenic antibodies against nuclear antigens. Lupus predominately affects women between menarche and menopause. There are both genetic and environmental risk factors which affect an individuals’ risk of developing lupus. Estrogens are a risk factor for developing lupus and are thought to contribute significantly to the initiation and progression of disease. In lupus-prone mice, genetic knockout of a receptor for estrogen, estrogen receptor alpha (ERα), causes significant attenuation of lupus. Previous studies have not identified the cell type or …
Assessment And Mechanisms Of Autonomic Function In Health And Disease, Peter Ricci Pellegrino
Assessment And Mechanisms Of Autonomic Function In Health And Disease, Peter Ricci Pellegrino
Theses & Dissertations
The autonomic nervous system is a master regulator of homeostasis, and the conviction that autonomic outflow is important on a patient-by-patient, minute-to-minute basis in both health and disease is the motivation for this thesis. The dissertation explores three aims that advance our understanding of the autonomic nervous system by elucidating the molecular mechanisms of autonomic regulation, validating widely used techniques for autonomic assessment, and developing and applying a new method to assess sympathetic vascular control.
The first aim of the dissertation was to investigate the role of the Rho kinase pathway as a mediator of the autonomic effects of central …
Disease-Modifying Drug Possibly Linked To Placental Insufficiency : Severe Placental Complications In A Pregnant Woman With Multiple Sclerosis, Sultan M. Salahudheen, Muzibunnisa A. Begam
Disease-Modifying Drug Possibly Linked To Placental Insufficiency : Severe Placental Complications In A Pregnant Woman With Multiple Sclerosis, Sultan M. Salahudheen, Muzibunnisa A. Begam
Sultan Qaboos University Medical Journal
Disease-modifying drugs (DMDs) such as interferon (IFN)-β and glatiramer acetate are often prescribed to slow disability progression in patients with multiple sclerosis (MS). However, adverse pregnancy outcomes have been reported with these medications. We report the rare occurrence of severe placental complications in a 30-yearold pregnant woman with MS who continued to take IFN-β during her first trimester. She presented at the Tawam Hospital, Al Ain, United Arab Emirates, in 2013 with early-onset fetal growth restriction. At 30 gestational weeks, she developed severe pre-eclampsia. The baby was delivered via emergency Caesarean section and was discharged at the age of two …

Chemical Reaction Between Boric Acid And Phosphine Indicates Boric Acid As An Antidote For Aluminium Phosphide Poisoning, Motahareh Soltani, Seyed F. Shetab-Boushehri, Seyed V.
Sultan Qaboos University Medical Journal
Objectives: Aluminium phosphide (AlP) is a fumigant pesticide which protects stored grains from insects and rodents. When it comes into contact with moisture, AlP releases phosphine (PH3), a highly toxic gas. No efficient antidote has been found for AlP poisoning so far and most people who are poisoned do not survive. Boric acid is a Lewis acid with an empty p orbital which accepts electrons. This study aimed to investigate the neutralisation of PH3 gas with boric acid. Methods: This study was carried out at the Baharlou Hospital, Tehran University of Medical Sciences, Tehran, Iran, between December 2013 and February …

The Use Of Platelet Rich Fibrin In Pulpal And Periodontal Regeneration, Elisabeth L. Easley
Theses & Dissertations
Purpose: Platelet rich fibrin (PRF) has been postulated to aid in regeneration. Therefore, the purpose of this study is to evaluate the inflammatory response of PRF in vitro, and analyze clinically and histologically the effectiveness of PRF on pulpal regeneration after reimplantation.
Materials and Methods: The in vivo experiment included 18 patients (69 teeth). Teeth were extracted and apicoectomy performed. The tooth was reimplanted and splinted after PRF was condensed into the apex. Control teeth received the same treatment with the exception of the PRF. After 3-11 months, vitality was checked, teeth were extracted and analyzed histologically. For the in …
The Role Of Glutaminase And Extracellular Vesicles In Macrophages And Microglia, Beiqing Wu
The Role Of Glutaminase And Extracellular Vesicles In Macrophages And Microglia, Beiqing Wu
Theses & Dissertations
Glutamate serves as a crucial excitatory neurotransmitter that is essential for the proper functioning of the brain. However, excess levels of glutamate are neurotoxic and contribute to the pathogenesis of various neurodegenerative diseases, inducing HIV-1 associated neurocognitive disorders (HAND). Glutaminase 1 (GLS1) is an important mitochondrial enzyme responsible for producing glutamate from glutamine. GLS1 is upregulated during HAND and released from mitochondria to cytosol and extracellular space. However, why and how GLS1 is released remains unknown. In chapter II, we demonstrated that extracellular vesicles (EVs) carry GLS1 as cargos from cytosol to extracellular space during HIV infection and innate immune …
The Development Of Transforming Growth Factor-Beta 1 Associated Fibrosis In The Gastrocnemius Of Peripheral Artery Disease Patients And Response To Revascularization And Supervised Exercise Therapies, Duy M. Ha
Theses & Dissertations
Peripheral Artery Disease (PAD) affects over 200 million people worldwide with a 5-year mortality rate as high as 30%. Atherosclerotic blockages impair blood flow to the lower limbs, causing ischemic leg pain and dysfunction upon exercise that later progresses to ulcerations and gangrene and often requires amputation. The standard therapies, revascularization and exercise, allow patients to walk further and longer but are at best maintained short-term. One explanation may be the development of myopathic changes in the limb that progress despite improved hemodynamics—a paradigm shift in the classic view of PAD pathophysiology as solely a mismatch between blood supply and …
Mutations Affecting Epigenetic Regulators And Their Role In Peripheral T-Cell Lymphomas, Chao Wang
Mutations Affecting Epigenetic Regulators And Their Role In Peripheral T-Cell Lymphomas, Chao Wang
Theses & Dissertations
Peripheral T-cell lymphoma (PTCL) is a heterogeneous group of generally aggressive lymphoid malignancies, accounting for 10-15% of all non-Hodgkin lymphomas. Angioimmunoblastic T-cell lymphoma (AITL) represents approximately 20% of all PTCLs and is recognized as a distinct entity. Accurate diagnosis and classification of PTCL remain challenging. With the exception of ALK+ ALCL, patients with PTCL generally have a poor prognosis with standard chemotherapy and even with the availability of many novel drugs, including HDAC inhibitor (romidepsin and belinostat), gemcitabine, and bortezomib. Therefore, deciphering the pathogenesis of this group of diseases is needed to identify novel treatable targets for better therapeutic intervention. …
Aldehyde Adducts And Lung Injury, Muna Sapkota
Aldehyde Adducts And Lung Injury, Muna Sapkota
Theses & Dissertations
From the time of discovery, people have used both alcohol and tobacco in various forms all around the world. Alcohol abuse and cigarette smoking causes thousands of preventable deaths in the US each year. Several lung diseases such as COPD, asthma, respiratory infections, pneumonia, acute respiratory distress syndrome, cancer of neck, throat, mouth and lung are associated with smoking and drinking. Even though some studies have examined the role of alcohol and cigarette smoke in lung diseases, only a few studies have focused on the role of reactive aldehydes generated in the lungs of those who abuse alcohol and smoke …
Changes To Echocardiography-Derived Left Ventricular Filling Pressures And Cardiac Output In Response To Fluid Boluses In Elderly Patients With Left Ventricular Diastolic Dysfunction Undergoing Vascular Surgery, Sasha K. Shillcutt
Theses & Dissertations
Left ventricular diastolic dysfunction (LVDD) of the heart is a condition where the heart does not relax properly. This condition is important during times of stress, as LVDD is associated with significant morbidity of elderly surgical patients. LVDD is often asymptomatic and unrecognized as many of these patients have normal ejection fractions. However, LVDD may lead to heart failure in patients with preserved systolic function, with the incidence being as high as 50% in hospitalized elderly patients. The diagnosis of LVDD is an independent risk factor for postoperative major adverse cardiac events (MACE) and negatively impacts post-surgery readmission rates. Anesthesiologists …
Evaluation Of A Modified Incidental Teaching Procedure To Increase Child Compliance, Corey M. Cohrs
Evaluation Of A Modified Incidental Teaching Procedure To Increase Child Compliance, Corey M. Cohrs
Theses & Dissertations
Teaching noncompliant children to engage in compliant behavior has long been a goal for many parents, teachers, and therapists (Patterson, Shaw, & Ebner, 1969). This goal has driven the development of behavioral technology, including entire manualized treatment programs, specifically intended for the treatment of noncompliance (Forehand & McMahon, 1981; Hembree-Kigin & McNeil, 1995). These programs have typically included time-out based interventions that are associated with resistance to instruction (Roberts, 1982;Roberts, 1984). Given children’s frequent resistance to traditional approaches (e.g., timeout; Ducharme & Popynick, 1993), alternative interventions for the treatment of noncompliance are warranted.
A modified version of incidental teaching (Hart …
The Role Of Inflammatory Monocytes In Post-Influenza Methicillin-Resistant Staphylococcus Aureus Pneumonia, Karl J. Fischer
The Role Of Inflammatory Monocytes In Post-Influenza Methicillin-Resistant Staphylococcus Aureus Pneumonia, Karl J. Fischer
Theses & Dissertations
Secondary bacterial infections following influenza can lead to poor clinical outcomes and mortality. It is widely accepted that susceptibility to secondary bacterial infections is attributable to a suppressed innate antibacterial immunity. In contrast, a dysregulated host inflammatory response may also contribute to disease severity. Inflammation induced by viral infection alone significantly affects lung pathology, potentially exacerbating the destruction of the respiratory tract. Interestingly, the role of inflammatory mediators such as inflammatory monocytes have been extensively studied during influenza infection alone; however, their role during secondary bacterial infection are still not fully established. The objective of this study was to analyze …
Synthesis And Development Of Long-Acting Abacavir Prodrug Nanoformulations, Dhirender Singh
Synthesis And Development Of Long-Acting Abacavir Prodrug Nanoformulations, Dhirender Singh
Theses & Dissertations
Over the past decade, work from our laboratory has demonstrated the potential of targeted nanoformulated antiretroviral therapy (nanoART) to produce sustained high plasma and tissue drug concentrations for weeks following a single intramuscular (IM) administration that can suppress ongoing viral replication and mitigate dose associated viral resistance. While progress has occurred towards developing long-acting nanoformulations for protease and nonnucleoside reverse transcriptase (RT) inhibitors, development of nanoformulations of hydrophilic nucleoside RT inhibitor drugs have remained elusive. Abacavir (ABC); a hydrophilic molecule exhibited limited utilities to develop into long-acting nanoformulation platform. Furthermore, inefficient conversion of ABC to its biological active metabolites; carbovir …
The Pathogenesis Of Natural Killer/T Cell Lymphoma, Bei Jiang
The Pathogenesis Of Natural Killer/T Cell Lymphoma, Bei Jiang
Theses & Dissertations
Extranodal natural killer/T-cell lymphomas (NKTCL) are uncommon lymphomas with poor prognosis. In order to gain a better understanding of the molecular pathogenesis of NKTCL, we obtained and compared the global gene expression profile, global somatic mutation profile, and global methylation profile of normal NK cells, malignant NK cell lines, and NKTCL patient samples. We showed that the JAK-STAT pathway was constitutively activated in NKTCL through the cooperation between activation of signal transducer and activator of transcription proteins (STATs) via activating mutations and inactivation of STAT suppressor protein suppressor of cytokine signaling 6 (SOCS6) through promoter hypermethylation. Activating STAT3 and STAT5B …
Surveillance Of Non-Fatal Agricultural Injuries Among Farm Operators In The Central States Region Of The United States, Ketki Patel
Theses & Dissertations
Agriculture is a major industry in the U.S. with high rates of fatal and non-fatal occupational injuries. The dynamic nature of the U.S. agriculture industry, regional variations in farming practices, and the diverse workforce make surveillance of injuries challenging. A recent National Academies (U.S.) evaluation reported that data for non-fatal agricultural injury are scarce, and mainly available through national surveys. Limited data are available for employees in the agriculture sector, especially farm owners and operators. The objectives of this study were to- 1) review and evaluate existing survey-based systems for surveillance of non-fatal agricultural injuries on U.S. farms, and 2) …
Voltage-Gated K+ Channels And Hiv-1-Induced Neural Injury: Implications For Pathogenesis Of Hiv-1-Associated Neurocognitive Disorders, Han Liu
Theses & Dissertations
Human immunodeficiency virus-1 (HIV-1)-associated neurocognitive disorder (HAND) is a subcortical disease involving neuronal loss and myelin damage. Myelin is deposited by oligodendrocytes through a complex process including oligodendrocyte progenitor cell (OPC) proliferation and maturation. Oligodendrocytes/OPCs are susceptible to viral proteins such as Tat and that myelin damage is associated with oligodendrocyte number decrease. It has been shown that activation of voltage-gated K+ (KV) channels mediates apoptosis in various cell types. KV1.3 is the most predominant KV channel expressed in OPCs/oligodendrocytes and potentially involved in OPC developmental regulation. We studied the involvement of KV …

The Moral Dimension Of Pre-Reflective Self-Awareness, Susana Monsó
The Moral Dimension Of Pre-Reflective Self-Awareness, Susana Monsó
Animal Sentience
Rowlands offers a de-intellectualised account of personhood that is meant to secure the unity of a mental life. I argue that his characterisation also singles out a morally relevant feature of individuals. Along the same lines that the orthodox understanding of personhood reflects a fundamental precondition for moral agency, Rowlands’s notion provides a fundamental precondition for moral patienthood.
What Makes Us Conscious Is Not What Makes Us Human, Ezequiel Morsella, Erica B. Walker
What Makes Us Conscious Is Not What Makes Us Human, Ezequiel Morsella, Erica B. Walker
Animal Sentience
Consistent with the promising proposal of Klein & Barron (K & B), we discuss how what makes us conscious appears to be distinct from and more widespread in the animal kingdom than what distinguishes us from other species. Many of the abilities that do distinguish humans from other species (e.g., syntax and co-articulation in speech production) can be mediated unconsciously. The kind of functional architecture proposed by K & B may engender an “action selection bottleneck” in both humans and nonhuman species. As noted by K & B, this bottleneck is intimately related to conscious processing.

Introspection And Anecdotes Won’T Prove What Animals Are Thinking And Feeling, Michael Colombo, Melissa Johnston
Animal Sentience
We review Part Three, “Whines and Pet Peeves,” of Safina’s book Beyond Words: What Animals Think and Feel. Part Three is concerned mainly with the evidence for Theory of Mind in nonhuman animals. Like Safina, we believe that animals have a Theory of Mind, and like Safina, we question the utility of such concepts. There are many studies that Safina could have used to support his argument, and although some are mentioned, he prefers to make his point mainly by relying on anecdotes and introspection. The end result, we believe, is to devalue the scientific process.
