High Throughput Screening Of Priming Candidates For Impact On Nonviral Gene Delivery, Albert L. Nguyen Dec 2015

High Throughput Screening Of Priming Candidates For Impact On Nonviral Gene Delivery, Albert L. Nguyen

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Priming, in the context of nonviral gene delivery, is the treatment of cells with a compound prior to gene transfer that enhances transfection efficiency and/or transgene expression. Essentially, it is the application of an adjuvant approach to gene delivery. Effective transfection strategies may require priming to compete with the efficiency of viral transduction in order to achieve clinically relevant efficiency and expression in vivo. To search for priming compounds, a high throughput screen of the NIH Clinical Collection was performed using 25kDa b-PEI, an EGFP/luciferase plasmid, and HEK293T cells. The EGFP reporter was multiplexed with Hoechst 33342 and Resazurin fluorescence …

The Experience Of Influence Among Youth Leaders, Kelly Moguel Dec 2015

The Experience Of Influence Among Youth Leaders, Kelly Moguel

Department of Agricultural Leadership, Education, and Communication: Dissertations, Theses, and Student Research

While the term “leadership” can be difficult to define, one term that is commonly seen in its association is “influence”. While the field of leadership has empirically identified several outcomes associated with adult influence (i.e., greater organizational performance, higher subordinate satisfaction, higher subordinate engagement), little is known about influence in youth leaders. The purpose of this phenomenological study was to gain a better understanding of how youth leaders experience influence. Twenty-nine in-depth, semi-structured interviews from fifth- 12th-grade students in a leadership mentoring program called Nebraska Human Resources Institute (NHRI) at the University of Nebraska-Lincoln were conducted to ascertain …

Effect Of Simulated Canopy Cover On Created Vernal Pools In The Northeastern Us, Elizabeth Anne Bruen Dec 2015

Effect Of Simulated Canopy Cover On Created Vernal Pools In The Northeastern Us, Elizabeth Anne Bruen

Theses

Vernal pools in the Northeastern United States are small, forested wetlands characterized by ephemeral flooding. These unique ecosystems provide critical habitat for both terrestrial and aquatic organisms, abate seasonal flooding, and are biogeochemical hotspots. Vernal pools may be created to maintain key ecosystem services associated with loss of natural wetlands. In many instances, however, created pools do not mimic the ecological functions and services of extant vernal pools due to shortcomings associated with the surrounding land use and canopy cover, hydrology, or invasion by opportunistic plants. Because of the importance of these systems to regional biodiversity, understanding the conditions required …

Efficient Implementations Of Pairing-Based Cryptography On Embedded Systems, Rajeev Verma Dec 2015

Efficient Implementations Of Pairing-Based Cryptography On Embedded Systems, Rajeev Verma

Theses

Many cryptographic applications use bilinear pairing such as identity based signature, instance identity-based key agreement, searchable public-key encryption, short signature scheme, certificate less encryption and blind signature. Elliptic curves over finite field are the most secure and efficient way to implement bilinear pairings for the these applications. Pairing based cryptosystems are being implemented on different platforms such as low-power and mobile devices. Recently, hardware capabilities of embedded devices have been emerging which can support efficient and faster implementations of pairings on hand-held devices. In this thesis, the main focus is optimization of Optimal Ate-pairing using special class of ordinary curves, …

Spectroscopic And Theoretical Characterization Of Retinylidene Proteins And Biological Chromophores For Use In Biophotonic Applications, Jordan A. Greco Dec 2015

Spectroscopic And Theoretical Characterization Of Retinylidene Proteins And Biological Chromophores For Use In Biophotonic Applications, Jordan A. Greco

Doctoral Dissertations

Retinylidene proteins are found in all domains of life and perform diverse biological functions, including light-induced ion transport and phototransduction in visual systems. One such protein, bacteriorhodopsin (BR), is the light-activated proton pump that produces chemical energy for the archaeon, Halobacterium salinarum. In recent years, a branching reaction from the photocycle of BR has been discovered to produce a stable photoproduct, the Q state, using a sequential multiphoton process. The Q state has optical properties that enable the use of BR in bioelectronic technologies, however, the native protein has limited access to this photoproduct. A review is …

Extracellular Regulation Of Glial Cell Functions, Kasey Marie Johnson Dec 2015

Extracellular Regulation Of Glial Cell Functions, Kasey Marie Johnson

Doctoral Dissertations

The extracellular environment plays an important and crucial role in orchestrating the behaviors of the cells in the central nervous system (CNS). In my thesis work, I studied how the extracellular environment regulates a subset of glial cells, Oligodendrocytes (OLs) and astrocytes. Moreover, I studied the relationship of the astrocyte and the OL, particularly for astrocytes to promote a favorable environment for OL maturation. The extracellular matric (ECM) is composed of a variety of factors, including glycoproteins and secreted extracellular proteins.

In my thesis work, I focused on a subset of glycoproteins: Laminin (Ln), Tenascin-C (TnC), Fibronectin (Fn), and Vitronectin …

Executive Summary: The Effects Of High Fidelity Simulation On Hesi Grades, Monica Taylor Dec 2015

Executive Summary: The Effects Of High Fidelity Simulation On Hesi Grades, Monica Taylor

DNP and Student Works

The nursing program at a community college in Texas had a steady decrease in National Council Licensure Examination for Registered Nurses (NCLEX-RN®) and Health Education Systems, Incorporated (HESI) scores for the past five years. After making several changes in the program, HESI scores continued to be low. In 2012, the school built a new state of the art high fidelity simulation lab. The faculty considered high fidelity simulation (HFS) as a possible solution to improve the scores. The purpose of this project was to examine whether the integration of HFS with traditional clinical learning experiences would increase the students' performance …

Officiating Removal, Leah Litman Dec 2015

Officiating Removal, Leah Litman

Articles

For the last several years, the Commonwealth of Pennsylvania has quietly attempted to curtail capital defendants' representation in state postconviction proceedings. In 2011, various justices on the Pennsylvania Supreme Court began to call for federally funded community defender organizations to stop representing capital defendants in state postconviction proceedings. The justices argued, among other things, that the organizations' representation of capital defendants constituted impermissible federal interference with state governmental processes and burdened state judicial resources. The court also alleged the community defender organizations were in violation of federal statutes, which only authorized the organizations to assist state prisoners in federal, but …

Converted Measurement Trackers For Systems With Nonlinear Measurement Functions, Steven V. Bordonaro Dec 2015

Converted Measurement Trackers For Systems With Nonlinear Measurement Functions, Steven V. Bordonaro

Doctoral Dissertations

Converted measurement tracking is a technique that filters in the coordinate system where the underlying process of interest is linear and Gaussian, and requires the measurements to be nonlinearly transformed to fit. The goal of the transformation is to allow for tracking in the coordinate system that is most natural for describing system dynamics. There are two potential issues that arise when performing converted measurement tracking. The first is conversion bias that occurs when the measurement transformation introduces a bias in the expected value of the converted measurement. The second is estimation bias that occurs because the estimate of the …

Development Of Semi-Circular Bending (Scb) Fracture Test For Bituminous Mixtures, Gabriel Nsengiyumva

Department of Civil and Environmental Engineering: Dissertations, Theses, and Student Research

Granted that most distresses in asphalt (flexible) concrete (AC) pavements are directly related to fracture, it becomes clear that identifying and characterizing fracture properties of AC mixtures is a critical step towards a better pavement design. This thesis examines the testing variables of a reliable and practical semicircular bending (SCB) test for evaluating the fracture characteristics of asphalt concrete mixtures at intermediate service temperature conditions. The first part of this thesis investigates the repeatability of the SCB fracture test method by integrating a statistical-experimental approach to identify testing variables of the SCB test that result in repeatable test results. Toward …
