Pannexin1 Channels Dominate Atp Release In The Cochlea Ensuring Endocochlear Potential And Auditory Receptor Potential Generation And Hearing, Jin Chen, Yan Zhu, Chun Liang, Jing Chen, Hong-Bo Zhao

Otolaryngology--Head & Neck Surgery Faculty Publications

Pannexin1 (Panx1) is a gap junction gene in vertebrates whose proteins mainly function as non-junctional channels on the cell surface. Panx1 channels can release ATP under physiological conditions and play critical roles in many physiological and pathological processes. Here, we report that Panx1 deficiency can reduce ATP release and endocochlear potential (EP) generation in the cochlea inducing hearing loss. Panx1 extensively expresses in the cochlea, including the cochlear lateral wall. We found that deletion of Panx1 in the cochlear lateral wall almost abolished ATP release under physiological conditions. Positive EP is a driving force for current through hair cells to …

Methods Of Treating Stroke Using Stem Cell-Like Menstrual Blood Cells, Paul R. Sanberg, Cesario V. Borlongan, Julie Allickson

USF Patents

A cell type that is a complete match of the transplant recipient appears as an optimal scenario to open treatment options to a large patient population with minimal complications. The use of autologous bone marrow or umbilical cord blood has been proposed as a good source of stem cells for cell therapy. Menstrual blood is found to be another important source of stem cells. Assays of cultured menstrual blood reveal that they express embryonic like-stem cell phenotypic markers and neuronal phenotypic markers under appropriate conditioned media. Oxygen glucose deprivation stroke models show that OGD-exposed primary rat neurons, co-cultured with menstrual …

Compositions And Methods Of Treatment For Myocilin Glaucoma By Selectively Inhibiting Grp94, Chad Anthony Dickey, Amirthaa Suntharalingam, Brian S. J. Blagg

USF Patents

A compound and method for treating myocilin glaucoma using a selective Grp94 inhibitor is presented. Clearance of mutant myocilin can be promoted by selectively targeting the endoplasmic reticulum (ER) chaperone Grp94 using siRNA knockdown or small molecule inhibitors. Grp94 contributes to the intracellular accumulation of mutant myocilin. Tailored treatments aimed at disrupting the Grp94/mutant myocilin interaction can be used as a new therapeutic strategy for myocilin glaucoma. The inventors developed a compound having a general backbone structure of geldanamycin (GDA) and radicicol (RDC) in which a more hydrophobic surrogate of the quinone in GDA is linked to the resorcinol in …

Nutrition Regulation Of Male Accessory Gland Growth And Maturation In Tribolium Castaneum, Jingjing Xu, Ashlee L. Anciro, Subba R. Palli

Entomology Faculty Publications

Insulin/IGF-1 signaling (IIS) pathway is known to control growth, development and reproduction. Insulin-like peptide mediated body size plasticity in Drosophila melanogaster has been reported. Here, our studies showed that IIS pathway and nutrition regulate growth and maturation of the male accessory gland (MAG) in the red flour beetle, Tribolium castaneum. The size of MAG increased from day 1 to day 5 post-adult emergence (PAE). This increase in the size of MAG is contributed by an increase in cell size, but not cell number. The growth of MAG was impaired after double-stranded RNA (dsRNA)-mediated knockdown in the expression of genes …

Micro-Motion Controller Ii, Bergren Antell, Michael Mcnaul, Steve Shushnar

Bioengineering Senior Theses

The purpose of this project was to improve upon the functionality of a micro-motion controller designed by another senior design group at this university. The original controller design facilitated motion in only two dimensions, and by modifying the platform of the design to accommodate another axis of rotation, we were able to achieve a full range of 3-D motion in our own product. Additionally, we designed a new system in which a motor could be mounted on the base plate of the device which would rotate the upper platform on its own through a simple belt-and-pulley system. And lastly, we …

Analysis Of Neurite Outgrowth On Electrospun Fiber Scaffolds In The Presence Of Blebbistatin And Paclitaxel, A. R. D'Amato, N. J. Schaub, C. D.L. Johnson, J. Cardenas, R. J. Gilbert

Chemical and Biochemical Engineering Faculty Research & Creative Works

In an attempt to increase the rate of neurite extension from E9 chick embryo dorsal root ganglion (DRG) on electro spun fiber scaffolds we experimented with two drugs that have been used previously to affect neuronal growth. Blebbistatin and paclitaxel were added to DRG cultured on electro spun fibers in an attempt to increase the rate of neurite extension along fibrous scaffolds. Blebbistatin increased the length of neurites extending from the DRG explant in comparison to the drug-free control group. In contrast, paclitaxel completely eliminated all neurite extension from the DRG explant when compared to the control. A third treatment …

God's Eye News: The Use Of Drones In Journalism, A Documentary Film, Robert Lance Carroll Jun 2015

God's Eye News: The Use Of Drones In Journalism, A Documentary Film, Robert Lance Carroll

Theses and Dissertations

This thesis uses the format of documentary film to investigate the aesthetic, legal, and ethical issues surrounding the use of Unmanned Aircraft Systems, commonly known as drones, in journalism. Particularly important are the topics of public perception, safety, freedom of speech, and privacy. Do journalists' First Amendment protections extend to the right to gather images using drones? How will the privacy of citizens be protected against aerial cameras that can go virtually unnoticed? Can drones be safely integrated into the National Airspace System? The goal of the documentary is not necessarily to answer these questions, but to gather opinions from …

' Cancer Is Like Teenagers': Figurative Comparisons In Surgical Consultations For Early-Stage Breast Cancer, Keith Cox Jun 2015

' Cancer Is Like Teenagers': Figurative Comparisons In Surgical Consultations For Early-Stage Breast Cancer, Keith Cox

Theses and Dissertations

This thesis reports findings from a conversation analytic study that investigate how surgeons describe cancer to early-stage breast cancer patients during preoperative consultations. Drawing upon audio recordings of actual consultations, this study examines one surgeon's use of a type of figurative expression, an analogy, to describe what cancer is and how it behaves. My analysis focuses on the role that this analogy plays in the activities that unfold in one phase of the surgical consultation, and reveals some social organizational dimensions of figurative expressions that have gone unexplored in prior research.

Chemical and Biochemical Engineering Faculty Research & Creative Works

We hypothesized that fiber diameter within an electro spun fiber scaffold would affect astrocyte elongation and maturation. To examine the relationship, astrocytes were seeded on 400nm and 800nm diameter electro spun PLLA fibers. Immunohistochemistry was used to monitor cell morphology, and GLT-1 expression over the first 96 hours after seeding. The results indicate that astrocytes extended along the primary axis of the 800nm fibers more rapidly than astrocytes seeded on fibers of half the size. While GLT-1 was expressed diffusely throughout the surface of the astrocytes, elongated cell processes exhibited dense regions of GLT-1. These results show that 800nm fibers …

The Coloniality Of Language: Race, Expressivity, Power, And The Darker Side Of Modernity, Gabriela A. Veronelli Jun 2015

The Coloniality Of Language: Race, Expressivity, Power, And The Darker Side Of Modernity, Gabriela A. Veronelli

Wagadu: A Journal of Transnational Women's & Gender Studies

This article presents a new framework to analyze linguistic relations of power that examines the linguistic effects of what Aníbal Quijano has theorized as “the coloniality of power.” The argument is organized in two sections. The first section introduces “the coloniality of language,” an expression the author uses to refer to the process of racialization of colonized populations as communicative agents beginning in the sixteenth century and continuing until today. This section includes an account of the language and communication paradigm being developed at the time of the Conquest, which, the author argues, contains the coloniality inside. In the second …

Race-Based Epistemologies: The Role Of Race And Dominance In Knowledge Production, Shana Almeida

Wagadu: A Journal of Transnational Women's & Gender Studies

Mainstream academia in the West reinscribes racial thinking by strategically reducing the theoretical contributions of racialized and Indigenous scholars to “stories.” In my analysis, I describe how this reduction reproduces the ideological, discursive and material racism in knowledge production by centering the white western scholar as the “true” knower.

On The Dynamics Of Chronic Myelogenous Leukemia, The!Immune Response, And Multiple Drugs, John Shamshoian, David Kato, Caleb Miller

Baker/Koob Endowments Awarded Projects

Chronic myelogenous leukemia (CML) is a cancer that begins in the bone marrow and spreads to the bloodstream and other organs if left uncontrolled. Although most patients undergo some sort of remission with the standard drug imatinib, patients will inevitably relapse [1]. Many CML researchers utilize mathematical models to understand leukemia and immune-‐ response interaction and to create novel methods of treatment. We adopt a time-‐delay differential equation model, used by Michor et. al [2], which incorporates biologically significant parameters and leukemia –T-‐cell interactions. Inconveniently, the model is a system of nine nonlinear differential equations encompassing over two dozen parameters. …

Application Of Pyroprinting For Source Tracking Of E. Coli In Pennington Creek, Charles Moritz, Dillon Shapiro, Christopher Pann Jun 2015

Application Of Pyroprinting For Source Tracking Of E. Coli In Pennington Creek, Charles Moritz, Dillon Shapiro, Christopher Pann

Baker/Koob Endowments Awarded Projects

The goal of this project is to determine the source of fecal contamination in Pennington Creek. Dangerously high numbers of E. coli were recorded in the creek by the Morro Bay National Estuary Program (MBNEP) over the last 10 years. This is of particular concern because the creek runs directly through an outdoor school for K-12 education. One potential source of the contamination is the cattle in fields on either side of the creek for several miles, though they are supposedly kept out by fences. Another possibility is local wildlife—specifically wild turkeys, which roam around in flocks of over 100 …
