Activation And Inhibition Of Pannexin 1 Channels, David George Jackson Jun 2015

Activation And Inhibition Of Pannexin 1 Channels, David George Jackson

Open Access Dissertations

Pannexin 1 is one of three pannexins, a vertebrate protein family related to the invertebrate innexins. Unlike its innexin relatives, Pannexin 1 does not form gap junctions in vivo, but it does form plasma membrane channels that act as ATP release channels. ATP release through Pannexin 1 serves multiple physiological roles throughout the body, including in the central nervous system, or CNS. Pannexin 1 also plays a pathophysiological role in CNS injury as part of the cell death-inducing P2X7-Pannexin 1 complex. During CNS injury, increased extracellular K+ activates Pannexin 1 by an unknown mechanism. Since Pannexin 1 block might ameliorate …

Studies in human populations have shown a significant correlation between procollagen C-endopeptidase enhancer protein 2 (PCPE2) single nucleotide polymorphisms and plasma HDL cholesterol concentrations. PCPE2, a 52-kDa glycoprotein located in the extracellular matrix, enhances the cleavage of C-terminal procollagen by bone morphogenetic protein 1 (BMP1). Our studies here focused on investigating the basis for the elevated concentration of enlarged plasma HDL in PCPE2-deficient mice to determine whether they protected against diet-induced atherosclerosis. PCPE2-deficient mice were crossed with LDL receptor-deficient mice to obtain LDLr-/-, PCPE2-/- mice, which had elevated HDL levels compared with LDLr-/- mice with similar …

Ocean Sampling Day was initiated by the EU-funded Micro B3 (Marine Microbial Biodiversity, Bioinformatics, Biotechnology) project to obtain a snapshot of the marine microbial biodiversity and function of the world’s oceans. It is a simultaneous global mega-sequencing campaign aiming to generate the largest standardized microbial data set in a single day. This will be achievable only through the coordinated efforts of an Ocean Sampling Day Consortium, supportive partnerships and networks between sites. This commentary outlines the establishment, function and aims of the Consortium and describes our vision for a sustainable study of marine microbial communities and their embedded functional traits.

Psychosocial factors play a role in HIV progression. Expressive writing (EW) interventions have demonstrated positive effects on psychosocial and disease-related outcomes in HIV. Linguistic properties of written essays have been related to psychological outcomes, although such research is sparse in the HIV literature. This project examines the relationship of word usage to psychosocial and disease-related factors in HIV. As part of a larger randomized, controlled trial of EW, this project examined the subset of subjects (n = 121) randomized to write about trauma. Linguistic Inquiry and Word Count (LIWC) software was used to analyze word usage (pronouns, positive, negative, social, …

Numerical Modeling Of Onshore Plankton Transport, Atsushi Fujimura

Open Access Dissertations

Cross-shore exchange of plankton plays an important role in marine ecosystems and coastal communities. Larvae of many intertidal invertebrate species grow offshore and come back to the shore for settlement by crossing the energetic surf zone: however, shoreward transport mechanisms are not well understood. To test the possible onshore transport mechanisms, numerical simulations are performed based on the field campaigns conducted at Sand City beach (a mildly sloping rip-channeled beach) and Carmel River State Beach (CRSB, a steep pocket beach). The model consists of the 3D hydrodynamic model Delft3D and an individual based model for larval tracking. As weak swimmers, …

The aging brain is often characterized by the presence of multiple comorbidities resulting in synergistic damaging effects in the brain as demonstrated through the interaction of Alzheimer's disease (AD) and stroke. Gangliosides, a family of membrane lipids enriched in the central nervous system, may have a mechanistic role in mediating the brain's response to injury as their expression is altered in a number of disease and injury states. Matrix-Assisted Laser Desorption Ionization (MALDI) Imaging Mass Spectrometry (IMS) was used to study the expression of A-series ganglioside species GD1a, GM1, GM2, and GM3 to determine alteration of their expression profiles in …

Chromatin To Clinic: The Molecular Rationale For Parp1 Inhibitor Function., Felix Y. Feng, Johann S. De Bono, Mark A. Rubin, Karen E Knudsen

Department of Cancer Biology Faculty Papers

Poly(ADP-ribose) polymerase 1 (PARP1) inhibitors were recently shown to have potential clinical impact in a number of disease settings, particularly as related to cancer therapy, treatment for cardiovascular dysfunction, and suppression of inflammation. The molecular basis for PARP1 inhibitor function is complex, and appears to depend on the dual roles of PARP1 in DNA damage repair and transcriptional regulation. Here, the mechanisms by which PARP-1 inhibitors elicit clinical response are discussed, and strategies for translating the preclinical elucidation of PARP-1 function into advances in disease management are reviewed.

A System Dynamics Approach To The Efficacy Of Oxime Therapy In Sub Lethal Exposure To Sarin Gas, Daniel J. Droste

Theses and Dissertations

The 2010 National Security Strategy states, The effective dissemination of a lethal agent would endanger the lives of thousands of people and have unprecedented economic, societal, and political consequences. We must continue to work at home with first responders and health officials to reduce the risk associated with high-consequence threats . Nerve agents, such as Sarin gas, are considered high consequence threats. The threat of use of agents such as Sarin is as much a threat today as any other time in our history. However, the suggested treatment protocol is not as precise as it could be. Debate exists over …

Fishing and pollution are chronic stressors that can prolong recovery of coral reefs and contribute to ecosystem decline. While this premise is generally accepted, management interventions are complicated because the contributions from individual stressors are difficult to distinguish. The present study examined the extent to which fishing pressure and pollution predicted progress towards the Micronesia Challenge, an international conservation strategy initiated by the political leaders of 6 nations to conserve at least 30% of marine resources by 2020. The analyses were rooted in a defined measure of coral-reef-ecosystem condition, comprised of biological metrics that described functional processes on coral reefs. …

Induced Ankylosis Of A Primary Molar For Skeletal Anchorage In The Mandible As Alternative To Mini-Implants, Matina V. Angelopoulou, Despina Koletsi, George Vadiakas, Demetrios J. Halazonetis

School of Dentistry Faculty Research and Publications

Background

Mesial protraction of mandibular posterior teeth requires increased anchorage to avoid undesired tooth movements. Orthodontic mini-implants have become a popular and successful way to increase skeletal anchorage in such cases. However, mini-implants may cause injury to adjacent teeth or anatomical structures and may lead to tissue inflammation. Induced ankylosed primary teeth have been used in the past as abutments for the protraction of the maxilla in cases of maxillary retrognathism. However, this technique has not been described in the literature for the protraction of mandibular molars. The aim of this paper is to present, through a case report, an …

Quantitative Hermeneutics: Counting Forestructures On A Path From W.M. Davis To The Concept Of Multiple-Permeability Karst Aquifers, H. L. Vacher, Lee J. Florea

International Journal of Speleology

Hermeneutics is the theory of interpretation. One of its major components is recognizing prejudgments, or forestructures, that we bring to our objects of study. In this paper, we construct a historical narrative of the evolution of thinking about the role of caves in relation to groundwater flow in limestone, and we tabulate forestructures as they appear in the story. This account consists of three overlapping time periods: the before and after of an incident that repelled hydrogeologists and students of karst from each other in the middle of the 20th century; a period, up to around the turn of …

Relationship Of Phenotypic Variation And Genetic Admixture In The Saltmarsh–Nelson's Sparrow Hybrid Zone, Jennifer Walsh, W. Gregory Shriver, Brian J. Olsen, Kathleen M. O'Brien, Adrienne I. Kovach

New Hampshire Agricultural Experiment Station Publications

Hybridization is influential in shaping species' dynamics and has many evolutionary and conservation implications. Identification of hybrid individuals typically relies on morphological data, but the assumption that hybrids express intermediate traits is not always valid, because of complex patterns of introgression and selection. We characterized phenotypic and genotypic variation across a hybrid zone between 2 tidal-marsh birds, the Saltmarsh Sparrow (Ammodramus caudacutus) and Nelson's Sparrow (A. nelsoni) (n = 290), and we sought to identify morphological traits that could be used to classify admixed individuals. Sparrows were sampled from a total of 34 marshes, including 23 sympatric and 11 putatively …

Staphylococcus aureus infections exert a tremendous burden on the health-care system, and the threat of drug-resistant strains continues to grow. The bacteriolytic enzyme lysostaphin is a potent antistaphylococcal agent with proven efficacy against both drug-sensitive and drug-resistant strains; however, the enzyme’s own bacterial origins cause undesirable immunogenicity and pose a barrier to clinical translation. Here, we deimmunized lysostaphin using a computationally guided process that optimizes sets of mutations to delete immunogenic T cell epitopes without disrupting protein function. In vitro analyses showed the methods to be both efficient and effective, producing seven different deimmunized designs exhibiting high function and reduced …

Within minutes after emission, complex photochemistry in biomass burning smoke plumes can cause large changes in the concentrations of ozone (O3) and organic aerosol (OA). Being able to understand and simulate this rapid chemical evolution under a wide variety of conditions is a critical part of forecasting the impact of these fires on air quality, atmospheric composition, and climate. Here we use version 2.1 of the Aerosol Simulation Program (ASP) to simulate the evolution of O3 and secondary organic aerosol (SOA) within a young biomass burning smoke plume from the Williams prescribed fire in chaparral, which was sampled over …

Research Publications (2011 to 2015)

Despite the vital role of paper publication and citation in higher education institutions (HEIs), literature on publication exercises is relatively scarce. There are a number of factors which influence the rate of university publications and citations. Accordingly, with a focus on policy perspectives, this paper discusses publication exercises by addressing the factors that can increase or decrease the rate of publication and citation in HEIs. The investigated zones are divided into two macro and micro levels, in which macro level deals with global policy and micro level is related to local and university policies. The effective factors and their relevant …

Species distributions are limited by a complex array of abiotic and biotic factors. In general, abiotic (climatic) factors are thought to explain species' broad geographic distributions, while biotic factors regulate species' abundance patterns at local scales. We used species distribution models to test the hypothesis that a biotic interaction with a tree, the Colombian oak (Quercus humboldtii), limits the broad-scale distribution of …
