The theory of intersectionality posits that inclusion in multiple stigmatized identity groups results in intersectional rather than additive oppression, making it impossible to examine any one form of oppression in isolation. Black lesbians experience multiple forms of oppression—based on their sex, gender, race, and sexual orientation—making their experiences an ideal opportunity for analyzing the impact of intersectionality. Drawing from feminist theory and research, this thesis uses multiple Black lesbian narratives selected from two texts, interwoven with research from Black scholars, to present an innovative method of inquiry designed to uncover the intersections in their lives. In this thesis, I focus on questions of fan power, resistance, and producer interaction, particularly around the issue of sexuality. I examine the fan cultures surrounding Supernatural and Sherlock, constructing two case studies. Each case study examines the text of the shows, the producer-fan relationship and the fan re-workings of the text, specifically fanfiction. This approach allows for an analysis situated firmly within the world of the fan. By situating my work against the larger conversation regarding resistance and power in fan studies, I work to further trouble the characterization of fan spaces as resistant. Improved Oncolytic Virotherapy By Increasing Virus Spread Within Tumors., Stephen Wechman

Oncolytic adenoviruses (Ads) have great therapeutic potential for lung cancer. Cancer selective E1b-deleted Ads are safe, however their efficacy remains limited clinically. To improve E1b-deleted Ads, Adhz60 was selected for greater anti-cancer efficacy by a process called bioselection, producing AdUV. AdUV preferentially lysed A549 and H1299 lung cancer cells more effectively than both the cancer selective Adhz60 and non-selective Ad5. AdUV induced greater LC3-II expression, relative to LC3-I, indicating that AdUV (30.9-fold) induced autophagy more effectively than Ad5 (12.2-fold) and Adhz60 (7.8-fold) in A549 cells. Mice treated with AdUV had significantly smaller tumors (p-value < 0.001) and prolonged survival (p-value = 0.0005) than mice treated with the negative control AdGFP. These results suggest that AdUV efficiently lyses lung cancer cells in vitro and in vivo.

Receptor Tyrosine Kinases (RTK’s) like EGFR, Insulin Receptor, IGF1R and others mediate growth, proliferation and balance between apoptosis and survival. Dysregulation of RTK activity has been identified in a wide range of cancers and contributes to initiation, progression and metastasis of tumors. Ubiquilins (UBQLN) are fairly novel proteins and are studied mostly in the field of neurodegenerative disorders. Our laboratory recently discovered a link between UBQLN1 function and tumorigenesis. UBQLN1 is lost in about 50% of non-small cell lung cancer (NSCLC) cases. We have identified an interaction between UBQLN1 and IGF1R. Furthermore, UBQLN1 regulates activity of IGF1R and EGFR. Hepatocellular Carcinoma (HCC) is a challenging condition to treat in the clinic due to its distinct tumor phenotype, aggressive growth, and chemotherapy resistance. Cancer Stem Cells (CSCs) are a poorly differentiated subpopulation of cells within the tumor microenvironment, and role of CSCs in tumor initiation and chemotherapy resistance is not well understood in HCC. In this study, we have successfully identified HCC cells with CSC-like properties that showed resistance to Doxorubicin. Using the established serum free culture techniques, we have enriched CSC-like cells among hepatocellular cancer cells. A double-blind placebo-controlled trial determined the long term effects of Protandim® supplementation on 5-km running performance, as well as on acute and long-term oxidative damage assessed by serum lipid peroxides (Thiobarbituric acid-reacting substances: TBARS). Thirty-eight subjects were placed into one of two groups: an experimental (675 mg/day Protandim®) or control group (675 mg/day corn starch). 5-km runs were performed at baseline and 88 (SD 4) days post-supplementation with samples taken immediately pre- and 10-min post 5-km run, as well as at rest ∼30 and ∼60 days during the supplementation period. Zeolite and sand mixture is an ideal reactive filling material for the Permeable Reactive barrier (PRB) due to its higher hydraulic conductivity and sorption capacity. This study applied three ASTM standard tests to examine the geotechnical engineering properties of ASTM 20-30 sand and zeolite (clinoptilolite) mixtures with varying zeolite mass percentages (25%, 50%and 75%). Conducted lab tests including: Proctor compaction, hydraulic conductivity in rigid wall permeameter and one-dimensional consolidation. The goals of this study were to quantify the impact of zeolite content and compactive effort on the density, conduction and compressibility of the sand-zeolite mixtures.

Human arylamine N-acetyltransferase 1 (NAT1) is a phase II xenobiotic metabolizing enzyme that is found in almost all tissues of the body. Expression of NAT1 is commonly elevated in several cancers including breast cancer. However, the exact mechanism by which NAT1 expression affects cancer risk and progression remains unclear. Three MDA-MB-231 breast adenocarcinoma cell lines that stably express wild-type, increased, and decreased levels of human NAT1 have been constructed and characterized for NAT1 mRNA, NAT1 acetylation activity, cell doubling rates, and endogenous acetyl-CoA levels. In rodent models, suramin improves recovery following acute kidney injury (AKI). We hypothesized that suramin would be useful in protection against cisplatin-induced AKI. This hypothesis was tested by pre-treating C57BL/6j mice with suramin prior to cisplatin. Our data indicates that suramin protects the kidney from injury by decreasing cisplatininduced decreases in kidney function. Renal histology indicated that suramin significantly protected from cisplatin-induced AKI. Data indicate that suramin pretreatment attenuated mRNA expression of pro-inflammatory cytokines and chemokines following cisplatin treatment, while also decreasing markers of cell stress and cell death. Background: The Epidermal Growth Factor Receptor (EGFR) is a 170- kilodalton transmembrane protein that belongs to the ErbB family of receptor tyrosine kinases. Upon ligand-mediated activation, the EGFR responsible for cell growth, proliferation, and tissue homeostasis in epithelial cells; however, the EGFR is overexpressed in many human malignancies including MDA-MB-468 cells, a metastatic breast epithelial cell line. Previous studies have indicated that within the MDA-MB-468 cell line, receptors at the cell surface promote cell growth when activated with Epidermal Growth Factor (EGF) ligand. The first purpose of the present dissertation was to apply a high throughput assay to systematically screen a library of food and drug administration (FDA)-approved drugs as potential ligands for the cannabinoid receptor 2 (CB2). A cell-based, homogenous time resolved fluorescence (HTRF) method for measuring changes in intracellular cAMP levels was validated and found to be suitable for testing ligands that may act on CB2. Among the 640 FDA-approved drugs screened, raloxifene, a drug used to treat/prevent post-menopausal osteoporosis, was identified for the first time to be a novel CB2 inverse agonist. Although increasing numbers of adults are entering college, their attrition rates are higher than those of their traditional student counterparts (ACT, 2010; Bergman, 2012). Additionally African American women are entering college in larger numbers than their African American male counterparts, and more so than within other minorities or underrepresented students populations (Mangino, 2010; NCES, 2012; 2014; Ntiri, 2001; Rosales & Person, 2003). According to the CDC, the most common type of heart disease is coronary artery disease, which frequently leads to myocardial infarction (MI). Therapeutic approaches to lessen the resulting cardiovascular injury associated with MI are limited. Recently, the management paradigm for cardiac injury has entered the molecular era and microRNAs (miRNAs) have been shown to act as negative regulators of gene expression by inhibiting mRNA translation and/or stimulating mRNA degradation. A single miRNA can modulate physiological or disease phenotypes by regulating whole functional systems. Individuals who provide emotional, social, and physical care to a care recipient with cancer, without receiving financial compensation, are considered informal cancer caregivers. Research has identified that depression is a significant concern for cancer caregivers and likely impacts the care recipient’s health as well. However, limited literature has evaluated constructs that may affect depression in cancer caregivers. The current study explored how intrinsic and extrinsic cultural motivations, caregiver burden, and repetitive thinking relate to depression in a sample of 46 current cancer caregivers. c-kit+ cardiac progenitor cells (CPCs) have recently gained much attention due to the therapeutic effects they exert on cardiac function following their administration into the infarcted heart as evidenced by animal studies and by a recent clinical trial (SCIPIO). Romantic attraction is commonly studied in the scientific literature, with countless theories providing hypotheses for why we find particular characteristics attractive. This study focused on psychodynamic theories to investigate the attraction process, particularly regarding the influence of parental figures on romantic attraction through the psychoanalytic theory of partner selection (i.e., template matching hypothesis used interchangeably), attachment theory, and Core Conflictual Relational Themes theory. Ras is the most frequently activated oncogene found in human cancer, but its mechanisms of action remain only partially understood. Ras activates multiple signaling pathways in order to promote transformation, but can also exhibit a potent ability to induce growth arrest and death. NORE1A (RASSF5) is a direct Ras effector that acts as a tumor suppressor promoting senescence and apoptosis. Its expression is frequently lost in human tumors and its mechanism of action remains unclear. Here I show that NORE1A forms a direct, Ras regulated, complex with β-TrCP, the substrate recognition component of the SCF-β-TrCP ubiquitin ligase complex. NORE1A also …

Contemporary American Jews individually define their own Jewish identity developed through both religious and cultural foundations (Cohen & Eisen, 2000), yet there is limited empirical research on the experience of Jewish identity and its impact on mental and spiritual well-being. Beyond denominational affiliation, each Jewish individual builds a Jewish identity based on personally desired levels of participation in one’s Jewish community (Ideal Jewish Identity) as well as perceived expectations and obligations of a Jewish life (Ought Jewish Identity), although the internalized desires and expectations may not be fulfilled. Three dynamic models are proposed to study the mechanism of glucose-insulin regulatory system and the possible causes of diabetes mellitus. The progression of diabetes comes along with the apoptosis of pancreatic beta-cells. A dynamical system model is formulated based on physiology and studied by geometric singular perturbation theory. The analytical studies reveal rich analytical features, such as persistence of solutions, Hopf bifurcation and backward bifurcation, while numerical studies successfully fit available longitudinal T2DM data of Pima Indian tribe. This study examined the utility of older adults’ awareness of their executive functioning abilities to predict future cognitive decline. The recently revised Cognitive Awareness Model (CAM) forms the conceptual background of this approach, and suggests that executive dysfunction disrupts awareness in a manner distinct from episodic memory dysfunction. The study design examined how awareness of executive functioning ability may predict both continuous decline on neuropsychological testing and qualitative change in diagnostic status. This form of prediction was tested using a longitudinal sample (n = 661) of older adults with either normal cognition, mild cognitive impairment (MCI), or Alzheimer’s disease (AD). …

In the face of a changing climate, questions regarding sub-lethal effects of elevated habitat temperature on the physiology of ectotherms remain unanswered. In particular, long-term responses of ectotherms to the warming trend in tropical regions are unknown and significantly understudied due primarily to the difficulties in specimen and community traceability. In freshwater lakes employed as cooling reservoirs for power plants, increased physiological stress from high water temperature can lead to an increase in mortality, reduce growth, and potentially alter the community structure of fishes. Interest in reducing methane (CH4) emissions by cattle has increased recently. Loss of feed energy as CH4 represents both an environmental concern and an energetic loss to the animal. Manipulation of dietary composition has proven to be an effective mitigation strategy. Experiments were conducted to evaluate the effect of several dietary characteristics on CH4 production in growing and finishing cattle, and to develop a novel, non-invasive method for quantifying emissions from animals in a production setting. Bypassing tyrosine kinases responsible for Stat5a/b phosphorylation would be advantageous for therapy development for Stat5a/b-regulated cancers. Here, we sought to identify small molecule inhibitors of Stat5a/b for lead optimization and therapy development for prostate cancer and Bcr-Abl-driven leukemias. In silico screening of chemical structure databases combined with medicinal chemistry was used for identification of a panel of small molecule inhibitors to block SH2 domain-mediated docking of Stat5a/b to the receptor-kinase complex and subsequent phosphorylation and dimerization. Rickettsiales-like organisms (RLOs) are thought to be related to bacteria in the order Rickettsiales. They have been reported to occur in the staghorn coral (Acropora cervicornis), and this study investigated trends of infection over time, and in relation to the health of infected corals. This study focuses on tissue samples taken mostly from visibly healthy A. cervicornis thickets in Broward County, Florida, and processed for histological examination. Samples were originally collected and analyzed to document reproduction during years 2001 through 2012, and tissue loss diseases (white-band disease [WBD] types I and II, and rapid tissue loss). …

Wetland resources in South Florida are regulated at three redundant jurisdictional levels: local or municipal regulations set forth by many independent jurisdictions, State regulations derived directly from Florida Statutes, and Federal regulations promulgated primarily under the Clean Water Act. All three levels of government can have jurisdiction over projects that affect regulated wetland resources, yet inconsistent policies and standards remain and continue to confound regulators despite decades of intensive coordination efforts and a rapidly growing scientific research base. Ecologists, particularly those engaged in biogeomorphic studies, often seek to connect data from three or more domains. Using three-block partial least squares regression, we present a procedure to quantify and define bi-variance and tri-variance of data blocks related to plant communities, their soil parameters, and topography. Bi-variance indicates the total amount of covariation between these three domains taken in pairs, whereas tri-variance refers to the common variance shared by all domains. We characterized relationships among three domains (plant communities, soil properties, topography) for a salt marsh, four coastal dunes, and two temperate forests spanning several regions in the world.
