The Biological And Conservational Implications Of Hatchling Biology In The Kemp's Ridley (Lepidochelys Kempii) Sea Turtle, Amy Nicole Bonka
All ETDs from UAB
The Kemp’s ridley sea turtle is currently listed as critically endangered, and has a very limited range, often found along the Atlantic coast though primarily inhabiting the Gulf of Mexico, with the majority of all nesting occurring near Rancho Nuevo, Mexico. Historically, large numbers of turtles have been reported to nest at Rancho Nuevo; though numbers dropped due to egg take and by-catch in the fishing and shrimping industries. Extensive conservation efforts over the last several decades significantly increased the number of nests; however the recovery status of the Kemp’s ridley is still in question, and the continued research and …

Hypersaline environments (salt concentrations greater than two molar) are found on all continents and represent a woefully understudied ecosystem. To date, most of the hypersaline environments studied have been either lakes (e.g. The Great Salt Lake, Utah) or deep ocean/sea brine pools (e.g. Red Sea), both of which have an abundance of accessible water for the organisms living there. However, more extreme hypersaline environments have not been characterized as thoroughly. One of these environments is the Bonneville Salt Flats (BSF) in Western Utah. The BSF is 80 square miles in size and consists of a salt crust of varying thickness …

The diversity of enzymes and the reactions which they catalyze is enormous. Careful study of enzymatic reaction mechanisms has taught us a great deal about chemical reactivity and has given us insight into the chemical and physical basis of life. One particularly ubiquitous class of enzymes, the RNA polymerases, are expressed in all forms of life. RNA polymerases are central components of nucleic acid metabolism and display a variety of distinctive enzymatic features; two points which solicit their thorough study. The central theme of this dissertation is the use of kinetic techniques to describe enzymatic reaction mechanisms. Although the main …

Freshwater ecosystems play a central role in the environment. Through various processes such as nutrient recycling, groundwater recharging, and the attenuation of many pollutants these biological systems help to maintain environmental health. Unfortunately, many recent studies have identified emerging threats to these special environments and the life they sustain. One source of mounting concern is a group of compounds that interact with the endocrine systems of humans and wildlife. Known as endocrine disrupting chemicals (EDCs), these natural and synthetic chemicals may mimic or interfere with the action of natural hormones—thus disrupting the endocrine system. Multiple studies have reported negative effects …

Transcriptional Regulation Of The Human Ribosomal Rna Gene, Blake L. Atwood
All ETDs from UAB
The ribosome has the major role of synthesizing almost all the proteins of the cell. One of the main structural components of the ribosome are the ribosomal RNAs 18S, 28S, 5.8S and the 5S which are bound by numerous proteins to make up a fully functional unit. In the human genome there are approximately 400 copies of the rRNA gene, which contains all but the 5S rRNA in one long transcription unit. This gene is found in tandem repeats on the acrocentric arm of chromosomes 13, 14, 15, 21 and 22. It is critical that this gene is tightly regulated …

Synonymous codon changes (SCCs) do not alter the amino acid sequence of proteins, but may significantly change the phenotype. The frequency of synonymous codons is species- and gene-specific, described as codon usage bias (CUB). Over the past 50 years, research has shown that SCCs affect protein biogenesis, development of human disorders and the function of recombinant proteins developed for therapeutic applications. However, the molecular mechanisms by which SCCs alter the function of gene products are not completely understood. In Chapter 2 we summarize present knowledge in this field. The most common cause of cystic fibrosis (CF) is the deletion of …

The studies presented herein were born through the observations of the challenges that are faced by forensic toxicology laboratories. While these challenges were first observed in the forensic toxicology field, they are common across clinical, workplace drug testing, and pain management laboratories alike. The challenge is how to increase efficiency without sacrificing quality. Current drug screening protocols such as immunoassays (IA), gas chromatography mass spectrometry (GC/MS), and liquid-liquid chromatography tandem mass spectrometry (LC/MS/MS) are robust but inefficient and limited, with some analyses providing only class information. These analyses when coupled require up to four days to complete for a confirmed, …

Retinitis pigmentosa (RP) is a group of retinal degenerative diseases characterized by the loss of rod photoreceptor cells that can be followed by cone degeneration eventually culminating in total photoreceptor cell loss. This disease affects approximately 1.5 million people worldwide and can be transmitted in an autosomal dominant (ADRP) manner accounting for nearly 30% of all RP cases. Dominant rhodopsin mutations have been divided into class I and class II based on the folding of rhodopsin protein, thermal stability, and chromophore regeneration. A threonine-17-methionine (T17M) mutation in rhodopsin is a class II mutation, characterized by a thermal instability/folding defect and …

Materials and Methods: Theoretical calculations of microstrains of bone surrounding 10 different miniscrews were done. These miniscrews were considered under loading forces of 500 gm., and the microstains of both trabecular and compact bone were calculated separately when the thickness of the compact bone was at 1mm or 2mm. The microstrains were calculated at 4 different …
Presence And Mobilization Of Staphylococcus Aureus Pathogenicity Islands (Sapis) Among Bovine Mastitis Isolates, Laura Kate Parker
Presence And Mobilization Of Staphylococcus Aureus Pathogenicity Islands (Sapis) Among Bovine Mastitis Isolates, Laura Kate Parker
All ETDs from UAB
Bovine mastitis is inflammation of the mammary gland that occurs in cattle, as a result of invading bacteria. Staphylococcus aureus, a causative agent of bovine mastitis, produces toxins such as the toxic shock syndrome toxin (TSST-1) and the staphylococcal enterotoxins (SEs). The genes for these toxins are coded on S. aureus pathogenicity islands (SaPIs), which are mobile genetic elements associated with virulence factors. SaPIs can be mobilized by bacteriophages, resulting in the production of transducing particles that can spread virulence factors throughout a bacterial population. This research assessed the role and prevalence of SaPIs among bovine mastitis samples. In this …

Circadian rhythms are approximately 24-hour cycles in behavioral and physiological processes, such as sleep/wake cycles and cognition. In mammals, these rhythms are endogenously generated and entrained to the external light cycle by the primary circadian pacemaker, the suprachiasmatic nucleus (SCN). A hallmark characteristic of SCN neurons is the daily rhythm in the spontaneous action potential frequency, with high activity during the day and low activity at night. These SCN firing rate patterns are critical for maintaining robust, consolidated behavioral rhythms, and persistent changes in SCN neuronal activity are involved light-induced behavioral shifts. Many of the ionic components underlying daily and …

Breast cancer remains the most frequently diagnosed cancer amongst women worldwide. Despite advances in treatment, patients with more aggressive basal-like and HER2-positive cancer experience high rates of recurrence and necessitate novel therapeutic approaches. A class of inhibitors targeted against poly (ADP-ribose) polymerases (PARP), important DNA repair proteins, are effective in cancers with a defective DNA repair response. As such, extensive preclinical and clinical research has examined their efficacy in these cancers. Although this data is promising, it is only applicable to a lim-ited patient population. The main goal of this dissertation is to expand the utility of these well-tolerated drugs …

C-reactive protein (CRP) is an acute phase reactant whose blood concentration rises in response all types of inflammation. And while CRP is widely used as a clinical marker of inflammation, it has several biological functions, e.g. activation of the classical complement cascade and binding Fcγ receptors in order to mediate multiple immune processes. The full extent of CRP’s physiological activities has not yet been fully elucidated, but there is significant evidence to establish that CRP plays an important role in the onset and progression of autoimmune disease. Seminal work from our lab showed that human CRP, hepatically expressed in transgenic …
Examination Of The Dynamic Assembly Equilibrium For E. Coli Clpb, Jiabei Lin
Examination Of The Dynamic Assembly Equilibrium For E. Coli Clpb, Jiabei Lin
All ETDs from UAB
As a member of the Clp/Hsp 100 chaperone family, Escherichia coli ClpB is able to disaggregate denatured proteins with assistance of DnaKJE co-chaperones to help cell survive under stress. However, the working mechanism of ClpB disaggregation activity remains unclear. The active structure of ClpB is shown to be a hexameric ring and ATP binding and hydrolysis are required for ClpB to perform its chaperone activity. Thus, studying the energetics and kinetics of the ATP linked ClpB assembly equilibrium is essential for the quantitative examination of ClpB - protein substrate interaction to fully reveal its disaggregation mechanism. ATPgS (a slowly hydrolysable …

This article is about a successful adoption of course redesign model of mathematics courses at the Department of Mathematics in the University of Texas at Brownsville (UTB), a Hispanic serve university in the south Texas. Around 2007 National Center for Academic Transformation (NCAT) recommended six models for using of information technology to redesign courses in mathematics: the supplemental model, the replacement model, the emporium model, the fully online model, the buffet model, and the linked workshop model. Because of the local educational environment and resources none of the six models could be implemented exactly like it was described by NCAT. …

Troxel Revisited: A New Approach To Third Party Childcare, Jeffrey A. Parness
Faculty Articles, Papers, and Other Scholarship
In 2000 in Troxel v. Granville, four United States Supreme Court justices determined that the “liberty interests of parents in the care, custody, and control of their children” generally foreclose states from compelling requested grandparent childcare over current parental objections. Yet, these four justices recognized that “special factors” might justify judicial interference as long as the contrary contemporary wishes of parents were accorded “at least some special weight.” The plurality, and one concurring justice, reserved the question of whether any “nonparental” visitation, presumedly also encompassing stepparents, siblings and others, must “include a showing of harm or potential harm to the …

The ascarid nematodes Toxocara canis (Werner, 1782) and Toxocara cati (Schrank, 1788) may infect humans resulting in toxocariasis. A prior study associated species of Toxocara Stiles, 1905 with cognitive deficits in children. To determine if a similar association between toxocariasis and cognition exists in adults, we analysed a large dataset from the United States’ Center for Disease Control’s National Health and Nutrition Examination Survey. We used linear-regression and multivariate models to examine the association between toxocariasis as assessed by the presence of anti-Toxocara IgG antibodies and three measures of cognitive function – simple reaction time (SRT), symbol-digit substitution (SDS) and …

Recent detailed chemical structure analyses of three demineralized kerogens from Green River oil shale samples were used to generate input parameters for the chemical percolation devolatilization (CPD) model. This model uses a lattice network to describe pyrolysis of solid hydrocarbons, such as coal and biomass. It was necessary to modify the formulation of the CPD model to account for the long aliphatic carbon chains found in oil shale, because gases formed from these long chains condense at room temperature and are counted as tar. It was initially assumed that 20% of the aliphatic material was released as light gas during …

Epidemiological research suggests that different indicators of socioeconomic status (SES) such as income and education may have independent and/or interactive effects on health outcomes. In this study, we examined both simple and more complex associations (i.e., interactions) between different indicators of SES and ambulatory blood pressure (ABP) during daily life. Our sample consisted of 94 married couples who completed a one-day ABP protocol. Both income and education were independently related to systolic blood pressure and only income was significantly related to diastolic blood pressure. There were also statistical interactions such that individuals with high levels of both income and education …

Psychometric studies of the ERN, CRN, Pe, and Pc ERPs are increasing. Coherent integration of these results is difficultwith classical test theory because the definition of error depends on the measure of reliability. This study usedgeneralizability theory, which extends the ideas of classical test theory, as a framework for evaluating the influence ofpsychopathology and number of trials on dependability of measurement. Participants included 34 people meeting criteriafor major depression, 29 meeting criteria for an anxiety disorder, and 319 controls. For all ERPs, within-person variancewas larger than between-person variance across groups, indicating many trials are needed for adequate dependability (atleast 13). …

We determine the regularized van der Waals contribution to pressure within a spherical cavity of vapor in a homogeneous, isotropic, infinite medium. The spherical Hamaker function, , has been defined, for the first time, in contrast to the conventional Hamaker function for planar surfaces, . For the materials under consideration, the pressure inside the cavity varies as , where a is the radius of the cavity. For radii below a transition radius, the surface energy (or surface tension) becomes size dependent and could have important implications for homogeneous nucleation of nanosized bubbles in liquids, as well as cavitation of bubbles.

The demand for ecosystem services and the ability of natural ecosystems to provide those services evolve over time as population, land use, and management practices change. Regionalization of ecosystem service activity, or the expansion of the area providing ecosystem services to a population, is a common response in densely populated coastal regions, with important consequences for watershed water and nitrogen (N) fluxes to the coastal zone. We link biophysical and historical information to explore the causes and consequences of change in ecosystem service activity—focusing on water provisioning and N regulation—from 1850 to 2010 in a coastal suburban watershed, the Ipswich …

Long-Term Characterization Of The Chronic Dopamine Microelectrode And Effect Of Electrical Conditioning, Eric P. Marr
EWU Masters Thesis Collection
Background: Dopamine (DA) is a neurotransmitter involved in movement, reward learning and addiction. Fast-scan cyclic voltammetry (FSCV) has long been an indispensable tool for monitoring real-time DA signaling. Development of polyimide fused silica-encased FSCV microelectrodes have made the technique more suitable for chronic DA monitoring (months) in vivo. Methods: In this study, electrically stimulated DA signals were evoked weekly in an effort to characterize the recovery time and stability of DA signals recorded long-term with silica-encased chronic DA microelectrodes. Additionally, electrical conditioning (etching), previously shown to improve microelectrode sensitivity in vitro, was performed to investigate the long-term impact on DA …
