Different Sites Of Alcohol Action In The Nmda Receptor Glun2a And Glun2b Subunits, Yulin Zhao, Hong Ren, Donard S. Dwyer, Robert W. Peoples

The NMDA receptor is a major target of alcohol action in the CNS, and recent behavioral and cellular studies have pointed to the importance of the GluN2B subunit in alcohol action. We and others have previously characterized four amino acid positions in the third and fourth membrane-associated (M) domains of the NMDA receptor GluN2A subunit that influence both ion channel gating and alcohol sensitivity. In this study, we found that substitution mutations at two of the four corresponding positions in the GluN2B subunit, F637 and G826, influence ethanol sensitivity and ion channel gating. Tumor Necrosis Factor-a Converting Enzyme (TACE) induces active TNFa and may contribute to the development of colitis in humans. I hypothesized that pharmacological blockade of TNFa production would improve colitis scoring through decreased expression of inflammatory biomarkers. Acute colitis was induced in wild type BALB/c mice using 5% dextran sulfate sodium (DSS) in drinking water for 7 days. TACE inhibition was accomplished through twice daily intraperitoneal injection of DPC- 333 (10mg/kg; BSM Inc.) To determine the effects of TACE blockade during colitis, the following experimental groups (n=6-7/group) were tested: 1) vehicle; 2) DPC-333; 3) 5% DSS and vehicle; and 4) …

The objective of this study is to identify genetic factors associated with chronic kidney disease (CKD) and related cardiometabolic phenotypes among participants of the Genetics of Kidney Disease in Zuni Indians study. The study was conducted as a community-based participatory research project in the Zuni Indians, a small endogamous tribe in rural New Mexico. We recruited 998 members from 28 extended multigenerational families, ascertained through probands with CKD who had at least one sibling with CKD. With the global rise in frequency of harmful algal blooms in estuarine environments comes an increase in prevalence of toxic metabolites, such as microcystin (MC), that some of the cyanobacteria involved will produce. At high concentrations, MC may accumulate in consumer tissues and have deleterious effects on organisms; however impacts of the toxin on aquatic living resources at ecologically relevant concentrations have not been widely documented. We analyzed the effects of MC on juveniles of five fish species from the James River, Virginia to determine if MC has the potential to impede growth. TGFβ is involved in many pathological conditions, including autoimmune disorders, cancer, and cardiovascular and allergic diseases. We have previously found that TGFβ can suppress IgE-mediated mast cell activation in human and mouse mast cells in vitro. IL-33 is a recently discovered member of the IL-1 family capable of inducing mast cell responses and enhancing IgE-mediated activation. In this study, we investigated the effects of TGFβ on IL-33-mediated mast cell activation. Bone marrow-derived mast cells cultured in TGFβ -1, -2, or -3 showed reduced IL-33-mediated production of TNF, IL-6, IL-13 and MCP-1, in a concentration-dependent manner. Race & Non-Racial Characteristics In Sentencing Length And Sentencing Type Disparity, Davis A. Estes

Utilizing data from the United States Department of Justice Bureau of Justice Statistics Survey of Inmates in State and Federal Correctional Facilities (SISCF) 2004, this research investigates the possibility that African American drug offenders receive lengthier prison sentences and are imposed more range of time or indeterminate sentences as opposed to flat sentences based on race and/or non-racial characteristics; specifically parole status prior to sentencing, plea agreement status, prior criminal history, education status prior to arrest, employment status prior to arrest, and parental incarceration. While regression analysis revealed racial sentencing disparity for length of sentence and type of sentence (p< .05 and p< .001 respectively), among non-racial characteristics, only education status prior to arrest proved a significant predictor for length of sentence (p≤ .001). African American drug offenders were more likely sentenced to indeterminate sentences as opposed to flat sentences and were less likely to receive short sentences of 0 to 4 years or medium sentences of 4 to 10 years as compared to long sentences of 10 or more years. Potential research implications include the necessity for additional research regarding racial sentencing disparity as length or type of sentences as opposed to disparity as a numerical or a percentage difference between racial groups as well as the utilizing of inmate data that encompasses recent changes in drug sentencing laws, e.g. crack cocaine versus powder cocaine. Future research might also consider the evolution of marijuana laws in the United States and the potential impact on racial sentencing disparity.

Lignocellulosic biomass can be saccharified to produce reducing sugars that can be converted into various valuable products. This research aimed to produce reducing sugars from sago waste via sequential ionic liquid dissolution-solid acid saccharification process. The study included determining the best ionic liquid and solid acid catalyst combination, process optimisation, and kinetic study of the process, as well as product separation and catalyst recyclability. The generation of important and useful products (e.g. ethanol, lactic acid etc.) through microbial fermentation often involves the breakdown of complex polymeric feedstock such as starch and cellulose through enzymatic scissions followed by subsequent metabolic conversion. This thesis presents an effective method for synthesis and processing of polymethylmethacrylate-co-methacrylic acid (PMMA-co-MAA) biochips with controlled surface properties aimed for successful detection of the Dengue virus (DV). By variation of synthesis reaction parameters and using spin-coating technique, different compositions of the PMMA-co-MAA coated biochips were prepared. Madu, pemanis semulajadi, dianggap oleh sains sebagai makanan yang berkhasiat kerana mengandungi gula dan pelbagai bahan fitokimia.
