Synthesis, Characterization, In Vitro Evalution, And Preclinical Profiling Of Β-Cyclodextrin Polyrotaxane Families For Use As Potential Niemann-Pick Type C Therapeutics, Christopher James Collins

Niemann-Pick Disease Type C (NPC) is a rare, autosomal recessive genetic disorder featuring a loss of proteins responsible for unesterified cholesterol (UC) trafficking through the late endosomes/lysosomes (LE/LY) of every cell of the body. Disruption of this pathway leads to abnormal accumulation and storage of UC and other lipids. A broad range of visceral and neurological symptoms result from this accumulation exhibiting a variable age of onset and a disease progression that is ultimately fatal. The disease has an incidence of approximately 1 in 120,000 live births and has no known effective treatment.

Habit formation is a well-characterized process involving a shift from response-outcome as the leading driving force underlying action to a stimulus-response mechanistic process. It is hypothesized that a transition from goal-directed behavior to habitual behavior is driven by a transition of the bulk of cognitive load from neocortical toward lower neurological mechanisms. While many neurotransmitter systems have a demonstrated involvement in this process, the dopamine system is particularly implicated. Habit formation is assessed through the maintenance of extinction responding for an undesired, or devalued, outcome.

High resolution (1-10 km) numerical weather prediction (NWP) models face major challenges trying to improve representation of moist processes. In particular, simulating the interaction between the land surface and regional convection and rainfall is a source of uncertainties and presents three main barriers: (i) NWP models generally have simple land surface schemes, (ii) land-atmosphere coupling is not properly represented in models, and (iii) many assumptions made in deriving the theory of convective parameterizations are no longer valid at “gray scales” (e.g., 1-10 km). This mixed methods dissertation is guided by a sociocultural framework and a pragmatist stance to contribute to early literacy scholarship about minority children. In particular, I explore the relationship between low socioeconomic status Puerto Rican children’s reading experience and their social cognition by: (1) assessing them through quantitative and qualitative measures that reveal their capacities in these areas at the end of their first grade year, and (2) documenting their reading experiences at home and at school through ethnographic qualitative methods. A main challenge in chemotherapy is to deliver an anti-cancer drug selectively to tumor and avoid off-target exposure to other body tissues and organs. Nanoparticles (NPs) have been considered a promising approach for tumor drug delivery, with popularity attributable to the famous “Enhanced Permeability and Retention effect”, where small particles enter tumor tissues through leaky vasculature and be retained there. Currently, the phagocytic clearance of NPs is avoided by coating NP surface with Polyethylene glycol (PEG). My dissertation explores the ecology of aphid-vectored viruses in Midwestern cucurbits; in particular, it focuses on identifying source populations of vectors and virus reservoirs within fields and interactions of vector aphids with land cover surrounding and within fields. Initially, I identified the most commonly occurring viruses and aphid species associated with virus infections in pumpkin fields located across Indiana. This was done by assaying cucurbit leaf tissue with multiplex-rt-PCR targeting all four aphid-vectored, nonpersistent viruses found in cucurbits (papaya ringspot virus type-W, watermelon mosaic virus type-2, zucchini yellow mosaic virus, cucumber mosaic virus) and concurrently monitoring aphid species alightment in …

Relationships Between Behavioral Measures And Productivity In Growing Beef Cattle, Kelsey Bruno

The relationships between behavioral measures, growth performance, and immunocompetence in receiving beef steers were explored in three experiments. Specifically, exit velocity (EV) and objective chute score (OCS) were examined as temperament measures. In experiment 1, no main effects or interactions with degradable intake protein (DIP) or interactions between temperament measures were observed (P ≥ 0.11); however, high OCS steers had greater intake and gain:feed (P < 0.10) and slow EV steers had higher intake and gain (P < 0.10) than their counterparts. In experiment 2, during week two of social observations, dominance hierarchy rankings were dependent on OCS and EV (P < 0.05); slow EV steers also had increased antibody responses and gain (P < 0.10) and high OCS steers had increased gain (P < 0.10). In experiment 3, subjective chute scores and OCS were positively correlated (P < 0.01) and both EV and OCS treatments changed over time (P < 0.10); intake, vaccine titer response, and gain:feed responses to monensin were dependent on OCS treatment (P < 0.10). The studies suggest that temperament, measured by EV and OCS, affects growth performance and health related measures and is related to social dominance behavior in receiving beef steers.

Precision dairy farming technologies provide a variety of functions to dairy farmers. Little is known about dairy producer perception of these technologies. A study was performed to understand dairy producer perception of parameters monitored by precision dairy farming technologies. Calving has potential to be predicted using these same parameters and technologies. A second study was performed using two commercially marketed technologies in calving prediction. Threonine has been reported to be the second limiting amino acid in typical equine diets, but its actual requirement has not been determined in horses. The indicator amino acid oxidation (IAAO) method has been successfully used for evaluating amino acid metabolism and requirements in animals and humans. The objective of this research was to use the IAAO method to estimate threonine requirements in mature horses fed two different feed compositions. Dynamic morphing of marine species to match with environment changes in color and texture is an advanced means for surviving, self-defense, and reproduction. Here we use colloids that are placed inside a multilayered structure to demonstrate color and texture morphing. The multilayer is composed of a thermal insulating base layer, a light absorbing mid layer, and a liquid top layer. When external light of moderate intensity (∼0.2 W cm−2) strikes the structure, colloids inside the liquid layer will be assembled to locations with an optimal absorption. In this review, we discuss new emerging medical applications of the rapidly evolving field of mammalian synthetic biology.We start with simple mammalian synthetic biological components and move towards more complex and therapy-oriented gene circuits. A comprehensive list of ON–OFF switches, categorized into transcriptional, post-transcriptional, translational and posttranslational, is presented in the first sections. Subsequently, Boolean logic gates, synthetic mammalian oscillators and toggle switches will be described. Several synthetic gene networks are further reviewed in the medical applications section, including cancer therapy gene circuits, immuno-regulatory networks, among others. As college populations are becoming increasingly diverse, institutions must find ways to meet the needs of their nontraditional students. Nontraditional adult learners are self-directed, ready to learn, and are internally motivated to engage in problem-centered learning. The purpose of this study was to expand and refine an active learning seminar in a higher education setting to improve the quality of teaching, student engagement, and retention rates. Minority students are lagging behind their non-minority peers in academic achievement. Compounding this problem is the lack of research on minority students' perceptions on their connections to school, their feelings of autonomy, and their relationship with their parents. These variables are important considerations in this problem, as Ryan and Deci's self-determination theory suggests a strong relationship between student performance in school and students' perceptions of their intrinsic and extrinsic motivators. The 5 factor model of personality, including the traits of openness, conscientiousness, extraversion, agreeableness, and neuroticism, is a well-established theoretical model for describing how personality is structured. Hirsh (2010) demonstrated the big 5 personality traits, excluding extraversion, were correlated with pro-environmental attitudes. The purpose of this quantitative study was to replicate previous findings, and discover if there was a correlation with a person's pro-environmental behaviors and the big 5 personality traits. Research with geriatric populations suggests high levels of clinical depression and greater financial and psychological costs of treatment in long-term care facilities with more restrictive care. Research on learned helplessness, a construct separate from depression, suggests learned helplessness and perceived control are useful theories for the study of elder depression, but the relationship between depression and learned helplessness in this population is not clear. This cross-sectional quantitative study examined the relationship between depression and learned helplessness by comparing residents over age 65 in less restrictive assisted living (n =42) versus those in more restrictive skilled nursing facilities (n =63). Postpartum Depression (PPD) affects 15% of women after childbirth. Its etiology includes psychoneuroimmunologic factors with long-lasting postpartum stressors that lead to allostatic overload. Using mindfulness-based stress reduction (MBSR) for PPD has not yet been studied. Addressing this literature gap, the potential benefits of including an 8-week MBSR component based on Beck's theory of PPD at support groups were examined in this phenomenological study based on a sample of 10 women and 2 group facilitators. Recent scholarship and media attention has focused on the prevalence of sexually harassing behavior directed at working teenagers, and the emergence of sexual harassment lawsuits by these minors against their employers. Although many of the legal issues concerning workplace sexual harassment and adult workers (and the various state and federal jurisprudence prohibiting it) have been widely discussed, there is surprisingly little discourse, research, and precedent addressing the problem of workplace sexual harassment and teen workers.

There is an increasing demand in higher education institutions for training in complex environmental problems. Such training requires a careful mix of conventional methods and innovative solutions, a task not always easy to accomplish. In this paper we review literature on this theme, highlight relevant advances in the pedagogical literature, and report on some examples resulting from our recent efforts to teach complex environmental issues. The examples range from full credit courses in sustainable development and research methods to project-based and in-class activity units.
