Real-Time Rt-Pcr Assays To Detect West Nile Virus In Mosquito Pools, Nicole Marie Herman Jan 2015

Real-Time Rt-Pcr Assays To Detect West Nile Virus In Mosquito Pools, Nicole Marie Herman

Public Health Theses

Introduction: West Nile virus (WNV) is a mosquito-borne flavivirus that was first reported in North America in 1999. Human infections with West Nile are often mild or asymptomatic, but some infections progress to life-threatening encephalitis. The Connecticut Agricultural Experiment Station performs viral cell culture screening on mosquito pools collected throughout the state each summer to detect viruses circulating in mosquito populations. Three different real-time RT-PCR assays are used to identify WNV in positive cell cultures. Many surveillance programs do not use cell culture and only use real-time PCR to screen pools. Although this can provide for faster analyses, some positive …

Pirna Sequence Variants Associated With Prostate Cancer In African Americans And Caucasians, Michael Cameron Lerro Jan 2015

Pirna Sequence Variants Associated With Prostate Cancer In African Americans And Caucasians, Michael Cameron Lerro

Public Health Theses

Background: Prostate cancer is the most common cancer among men, and the incidence is 1.65 times higher in African Americans than Caucasians. The newly discovered PIWI/piRNA pathway, which regulates transposon and gene expression via small non-coding RNAs, has been implicated in certain aspects of cancer etiology. We hypothesize a novel involvement of the pathway via aberrant gene regulation due to sequence variants within piRNAs and test this hypothesis in relation to prostate cancer in both an African American and Caucasian sample. Methods: To interrogate SNPs embedded in piRNA sequences, we utilized genome-wide genotype data to impute 1,000 Genomes SNPs falling …

Effects Of Cobalt On Dna Double Strand Break Repair-Deficient Cells, Sean Raph Ms Jan 2015

Effects Of Cobalt On Dna Double Strand Break Repair-Deficient Cells, Sean Raph Ms

All Student Scholarship

Epidemiology studies of human workers exposed to particulate cobalt have found increased incidence of lung cancer. A hallmark of lung cancer, chromosome instability(CIN) manifests as either numerical or structural abnormalities, which are thought to result from unrepaired or miss-repaired DNA double strand breaks, homologous recombination (HR) and non homologous end joining (NHEJ). Previous studies have shown that HR protects cells against metal induced CIN while NHEJ does not. We investigated whether this outcome occurs for cobalt.

Anaerobic Digestion Of Feedlot Manure, Andrea K. Watson, Adam L. Shreck, Amy M. Schmidt Schmidt, Terry J. Klopfenstein, Galen E. Erickson Jan 2015

Anaerobic Digestion Of Feedlot Manure, Andrea K. Watson, Adam L. Shreck, Amy M. Schmidt Schmidt, Terry J. Klopfenstein, Galen E. Erickson

Nebraska Beef Cattle Reports

Cattle diet can impact manure quality and quantity but has minimal impacts on methane production from anaerobic digestion of manure. Quality of manure, measured as OM, does affectmethane production and is largely impacted by the environment cattle are housed in and methods used to collect manure. As the amount of ash contamination of manure was increased, or OM content of the manure was decreased, organic matter degradation and methaneproduction were decreased. With adequate daily cleanout of ash from digesters, open-lot beef cattle manurecan be used for anaerobic digestion.

Effects Of Calf Age At Weaning On Cow And Calf Performance And Feed Utilization In An Intensive Production System, Jason M. Warner, Curtis J. Bittner, Karla H. Jenkins Jenkins, Rick J. Rasby, Matt K. Luebbe, Galen E. Erickson, Terry J. Klopfenstein

Nebraska Beef Cattle Reports

The effects of calf weaning age on cow and calf performance, reproduction, and feed utilization were investigated in a two-year study. Early weaning increasedcow BW in January. Pregnancy rates were not impacted by calf age at weaning. Dry matter intake (DMI) was similar between normal-weaned cow-calf pairs and early-weaned cows and calves. Feed requirements and utilization were comparable between early- and normal-weaned pairs when fed high energy diets, implying weaning decisions should be made on the basis of management rather than feed efficiency.

Effect Of Corn Residue Removal On Subsequent Crop Yields, Mary E. Drewnoski, L. Aaron Stalker, James C. Macdonald Donald, Galen E. Erickson, Kathy J. Hanford, Terry J. Klopfenstein Jan 2015

Effect Of Corn Residue Removal On Subsequent Crop Yields, Mary E. Drewnoski, L. Aaron Stalker, James C. Macdonald Donald, Galen E. Erickson, Kathy J. Hanford, Terry J. Klopfenstein

Nebraska Beef Cattle Reports

Two studies were conducted to evaluate the effects of corn residue harvest on subsequent crop yields. In a long-term study (16 years), cattle grazing corn residue in the spring (February to the middle of April) or the fall (November through January) slightly improved subsequent soybean yields and had no effect on corn yields in an irrigated field maintained in an annual corn-soybean rotation at Mead, Neb. In a five-year study, fall grazing (December through January) or baling of corn residue had no effect on subsequent corn grain yields in a field maintained in continuous corn production at Brule, Neb. These …

A Basic Mechanism Of Beef Tenderization: Feeding Wet Distillers Grains Plus Solubles Contributes To Sarcoplasmic Reticulum Membrane Instability, Michael D. Chao, Katherine I. Domenech, Chris R. Calkins Calkins Jan 2015

A Basic Mechanism Of Beef Tenderization: Feeding Wet Distillers Grains Plus Solubles Contributes To Sarcoplasmic Reticulum Membrane Instability, Michael D. Chao, Katherine I. Domenech, Chris R. Calkins Calkins

Nebraska Beef Cattle Reports

Feeding wet distillers grains plus solubles (WDGS) could increase polyunsaturated fatty acid (PUFA) concentration in the sarcoplasmic reticulum (SR) membrane, thereby altering membrane integrity, resulting in more rapid post-rigor calcium leakage, greater enzyme activity and improved tenderness. Steers were finished on either 0% WDGS or 50% WDGS. Steaks from steers fed WDGS were more tender and had greater free calcium concentrations. Feeding WDGS also increased proportions of PUFA in SR membrane and altered SR lipid and phospholipid profiles. These findings suggest that feeding increased concentrations of WDGS in the finishing diet can possibly increase meat tenderness through the proposed mechanism.

Prevalence Of Obesity And Overweight, Its Clinical Markers And Associated Factors In A High Risk South-Asian Population, Faridah Amin, Syeda Sadia Fatima, Najmul Islam, Anwar H. Gilani

Department of Biological & Biomedical Sciences

Background: Obesity is a global epidemic, which is a risk factor for cardiovascular diseases and metabolic abnormalities. It is measured by body mass index (BMI), waist circumference (WC), waist-hip ratio (WHR), body fat (BF) distribution and abdominal fat mass, each having its own merits and limitations. Variability in body composition between ethnic groups in South-Asians is significant and may not be truly reflected by BMI alone, which may result in misclassification. This study therefore, aims to determine the frequency of obesity, body fat composition and distribution, in a high risk population of an urban slum of Karachi, Pakistan. This survey …

Computational Characterization Of Carboxyphosphate, Traci M. Clymer Jan 2015

Computational Characterization Of Carboxyphosphate, Traci M. Clymer

Electronic Theses and Dissertations

Carboxyphosphate (CP) is an important intermediate involved in reactions catalyzed by acetyl-CoA carboxylase, pyruvate carboxylase, N5-Carboxyaminoimidazole ribonucleotide synthetase, propionyl-CoA carboxylase, urea amidolyase, and carbamoyl phosphate synthetase. Despite its important role, properties for CP have never been reported due to its short estimated half-life (t1/2 70 ms). Thus, the high level ab initio methods, MP2 and CCSD(T), along with the DFT functionals: B3LYP, BB1K, M05-2X, M06-2X, and MPW1K were used to investigate the structure and energetics of CP in both vacuum and the PCM continuum solvation model of water. It was found that CP adopts a novel pseudo-cyclic structure featuring an …

Topographical Differences In Stress Vulnerability In Experimental Parkinson's Disease, Jessica M. Posimo Jan 2015

Topographical Differences In Stress Vulnerability In Experimental Parkinson's Disease, Jessica M. Posimo

Electronic Theses and Dissertations

Parkinson's disease is characterized by the progressive spread of protein misfolding stress, or proteotoxicity, across the brain. During this protracted process, the allocortex of the temporal lobe develops protein inclusions before the neocortex in the frontal and parietal lobes. In the present study we tested the hypothesis that the staged appearance of proteotoxicity in allocortex followed by neocortex is the result of intrinsic vulnerability differences. We microdissected the neocortex and multiple subregions of the allocortex from rat brains and plated the primary neo- and allocortical neurons for parallel in vitro studies. Cells were then exposed to a number of Parkinson's …

Comparison Of A Video Game Based Learning Environment And A Traditional Learning Environment, Jessica Barron Jan 2015

Comparison Of A Video Game Based Learning Environment And A Traditional Learning Environment, Jessica Barron

Electronic Theses and Dissertations

There are many ways to teach within a classroom environment. Traditional teaching methods have been successful because they can be applied in almost any classroom and are useful for imparting knowledge to a learner (Chang & Fisher, 2001). However, understanding and being able to apply that knowledge can be a challenge within a normal classroom environment. Simulations and games have been used in both the past and the present by the military and the government to teach and fine tune desired skills (Balci, Bertelrud, Esterbrook, & Nance, 1997). Video games and simulations give the learner a chance to apply their …

Analytical Pyrolysis Principles And Applications To Environmental Science, Michael A. Kruge

Department of Earth and Environmental Studies Faculty Scholarship and Creative Works

Over the past half century, analytical pyrolysis has proven itself to be an effective means for the semiquantitative characterization of complex macromolecular organic substances. It has been demonstrated that instruments such as Py-FID, Py-MS, and in particular, Py-GC/MS can provide valuable geochemical insights when applied to a wide variety of problems in environmental science. The more widespread use of analytical pyrolysis methods in the evaluation of environmental pollution is recommended, because of their relatively low cost and information-rich results.

Pyrolysis is the heating of organic substances in an inert, oxygen-free atmosphere, thereby avoiding combustion. When performed on a large scale, …

Department of Earth and Environmental Studies Faculty Scholarship and Creative Works

The inner shelf of the East China Sea is a river-dominated margin characterized by fine-grained mud deposits and a rapid sedimentation rate. Three short sediment cores (similar to 2.7m in length) were examined to characterize spatial variations in magnetic mineral diagenesis. The sediment cores were analyzed for sedimentation rates, magnetic properties, particle size distribution, organic carbon, and total sulfur content. The two more proximal cores with higher sedimentation rates (similar to 2.2cm/yr and similar to 0.96cm/yr) do not exhibit obvious effects of reductive dissolution of magnetite with increasing depth, which is consistent with their lower total sulfur content. The offshore …

Effect Of Particles On Evaporation Of Droplet Containing Particles, Yan Wei Jan 2015

Effect Of Particles On Evaporation Of Droplet Containing Particles, Yan Wei

Electronic Theses and Dissertations

The evaporation of droplet containing insoluble particles has grown into an active area of research due to the needs for nanofluids for applications in heat transfer, combustion, and manufacturing desired micro/nano particles in the pharmaceutical industry. The evaporation of droplets containing particles involves complicated multiphase heat and mass transport. The evaporation process consists of two stages: the first stage consists of evaporation until a shell of particle forms or when the solid to liquid ratio is sufficiently large and the second stage, where the droplet size is commonly assumed to be unchanged. The dissertation investigates the evaporation kinetics in the …

Trihalomethane Removal And Re-Formation In Spray Aeration Processes Treating Disinfected Groundwater, Cassandra Smith Jan 2015

Trihalomethane Removal And Re-Formation In Spray Aeration Processes Treating Disinfected Groundwater, Cassandra Smith

Electronic Theses and Dissertations

Historically, chlorination has been widely utilized as a primary and secondary disinfectant in municipal water supplies. Although chlorine disinfection is effective in inactivating pathogenic microbes, the use of chlorine creates the unintentional formation of regulated chemicals. On January 4, 2006, the United States Environmental Protection Agency (EPA) promulgated the Stage 2 Disinfectants/Disinfection by-product rule (DBPR) that focuses on public health protection by limiting exposure to four trihalomethanes (THM) and five haloacetic acids (HAA5), formed when chlorine is used for microbial pathogen control. This thesis examines post-aeration TTHM formation when employing spray-aeration processes to remove semi-volatile TTHMs from chlorinated potable water …

The polymer substrate is laser patterned to control the droplet's contact line dynamics and the droplet's contact angle is maintained using a computer controlled syringe pump. A room temperature silicon wafer is translated through the …

Tidal Hydrodynamic Response To Sea Level Rise And Coastal Geomorphology In The Northern Gulf Of Mexico, Davina Passeri Jan 2015

Tidal Hydrodynamic Response To Sea Level Rise And Coastal Geomorphology In The Northern Gulf Of Mexico, Davina Passeri

Electronic Theses and Dissertations

Sea level rise (SLR) has the potential to affect coastal environments in a multitude of ways, including submergence, increased flooding, and increased shoreline erosion. Low-lying coastal environments such as the Northern Gulf of Mexico (NGOM) are particularly vulnerable to the effects of SLR, which may have serious consequences for coastal communities as well as ecologically and economically significant estuaries. Evaluating potential changes in tidal hydrodynamics under SLR is essential for understanding impacts to navigation, ecological habitats, infrastructure and the morphologic evolution of the coastline. The intent of this research is to evaluate the dynamic effects of SLR and coastal geomorphology …
