Proteomic Signature Of The Murine Intervertebral Disc, Matthew R. Mccann, Priya Patel, Agya Frimpong, Yizhi Xiao, Walter L. Siqueira, Cheryle A. Séguin

Bone and Joint Institute

© 2015 McCann et al. Low back pain is the most common musculoskeletal problem and the single most common cause of disability, often attributed to degeneration of the intervertebral disc. Lack of effective treatment is directly related to our limited understanding of the pathways responsible for maintaining disc health. While transcriptional analysis has permitted initial insights into the biology of the intervertebral disc, complete proteomic characterization is required. We therefore employed liquid chromatography electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) protein/peptide separation and mass spectrometric analyses to characterize the protein content of intervertebral discs from skeletally mature wild-type mice. A total …

The role of the primary cilium in key signaling pathways depends on dynamic regulation of ciliary membrane protein composition, yet we know little about the motors or membrane events that regulate ciliary membrane protein trafficking in existing organelles. Recently, we showed that cilium-generated signaling in Chlamydomonas induced rapid, anterograde IFT-independent, cytoplasmic microtubule-dependent redistribution of the membrane polypeptide, SAG1-C65, from the plasma membrane to the periciliary region and the ciliary membrane. Here, we report that the retrograde IFT motor, cytoplasmic dynein 1b, is required in the cytoplasm for this rapid redistribution. Furthermore, signaling-induced trafficking of SAG1-C65 into cilia is unidirectional and …

Escherichia Coli O157:H7 Bacteriophage Φ241 Isolated From An Industrial Cucumber Fermentation At High Acidity And Salinity, Zhongjing Lu, Fred Breidt

Faculty Articles

A novel phage, Φ241, specific for Escherichia coli O157:H7 was isolated from an industrial cucumber fermentation where both acidity (pH ≤ 3.7) and salinity (≥5% NaCl) were high. The phage belongs to the Myoviridae family. Its latent period was 15 min and average burst size was 53 phage particles per infected cell. The phage was able to lyse 48 E. coli O157:H7 strains, but none of the 18 non-O157 strains (including E. coli O104:H7) or the 2 O antigen-negative mutants of O157:H7 strain, 43895Δper (also lacking H7 antigen) and F12 (still expressing H7 antigen). However, the phage was able to …

Mdr1 Sirna Loaded Hyaluronic Acid-Based Cd44 Targeted Nanoparticle Systems Circumvent Paclitaxel Resistance In Ovarian Cancer, Xiaoqian Yang, Arun K. Iyer, Amit Singh, Edwin Choy, Francis J. Hornicek, Mansoor M. Amiji, Zhenfeng Duan

Pharmaceutical Sciences Faculty Publications

Development of multidrug resistance (MDR) is an almost universal phenomenon in patients with ovarian cancer, and this severely limits the ultimate success of chemotherapy in the clinic. Overexpression of the MDR1 gene and corresponding P-glycoprotein (Pgp) is one of the best known MDR mechanisms. MDR1 siRNA based strategies were proposed to circumvent MDR, however, systemic, safe, and effective targeted delivery is still a major challenge. Cluster of differentiation 44 (CD44) targeted hyaluronic acid (HA) based nanoparticle has been shown to successfully deliver chemotherapy agents or siRNAs into tumor cells. The goal of this study is to evaluate the ability of …

Whiteness As Cursed Property: An Interdisciplinary Intervention With Joyce Carol Oates’S Bellefleur And Cheryl Harris’S “Whiteness As Property”, Karen Gaffney Feb 2015

Whiteness As Cursed Property: An Interdisciplinary Intervention With Joyce Carol Oates’S Bellefleur And Cheryl Harris’S “Whiteness As Property”, Karen Gaffney

Bearing Witness: Joyce Carol Oates Studies

This article begins with the assertion that now more than ever, in the aftermath of Ferguson and in a time when many believe our society to be post-racial, we need to bring together scholars and activists who care about racial justice, regardless of discipline, and build interdisciplinary tools for fighting racism. Furthermore, we need to understand and reveal how whiteness has been socially constructed because the power of whiteness lies in its invisibility, and that fuels the perpetuation of systemic racism. In making whiteness visible, we can see how it has been wielded as a weapon, which in turn will …

Oligotyping Reveals Stronger Relationship Of Organic Soil Bacterial Community Structure With N-Amendments And Soil Chemistry In Comparison To That Of Mineral Soil At Harvard Forest, Ma, Usa, Swathi A. Turlapati, Rakesh Minocha, Stephanie Long, Jordan Ramsdell, Subhash C. Minocha

New Hampshire Agricultural Experiment Station Publications

The impact of chronic nitrogen amendments on bacterial communities was evaluated at Harvard Forest, Petersham, MA, USA. Thirty soil samples (3 treatments × 2 soil horizons × 5 subplots) were collected in 2009 from untreated (control), low nitrogen-amended (LN; 50 kg NH4NO3 ha-1 yr-1) and high nitrogen-amended (HN; 150 kg NH4NO3 ha-1 yr-1) plots. PCR-amplified partial 16S rRNA gene sequences made from soil DNA were subjected to pyrosequencing (Turlapati et al., 2013) and analyses using oligotyping. The parameters M (the minimum count of the most abundant unique sequence in an oligotype) and s (the minimum number of samples in which …

Posterior Approach To Kidney Dissection: An Old Surgical Approach For Integrated Medical Curricula, Frank J. Daly, David L. Bolender, Deepali Jain, Sheryl Uyeda, Todd M. Hoagland

Biomedical Sciences Faculty Publications

Integrated medical curricular changes are altering the historical regional anatomy approach to abdominal dissection. The renal system is linked physiologically and biochemically to the cardiovascular and respiratory systems, yet anatomist often approach the urinary system as part of the abdomen and pelvic regions. As part of an integrated curriculum, the renal system must be covered relatively quickly after the thorax in the cadaver lab, often without the opportunity to fully appreciate the rest of the abdominal contents. This paper provides dissection instructions that follow the one of the historical surgical approaches for nephrectomy, including preservation of the posterior abdominal wall …

Macular Pigment Spatial Distribution Effects On Glare Disability/Efectos De La Distribución Espacial Del Pigmento Macular En El Nivel De Discapacidad Por Deslumbramiento, Christopher M. Putnam, Carl J. Bassi

Educator Preparation & Leadership Faculty Works

Purpose

This project explored the relationship of the macular pigment optical density (MPOD) spatial profile with measures of glare disability (GD) across the macula.

Methods

A novel device was used to measure MPOD across the central 16° of retina along four radii using customized heterochromatic flicker photometry (cHFP)at eccentricities of 0°, 2°, 4°, 6° and 8°. MPOD was measured as discrete and integrated values at all measured retinal loci. GD was calculated as a difference in contrast sensitivity (CS) between no glare and glare conditions using identical stimuli presented at the same eccentricities. GD was defined as [(CSNo Glare − …

Objectives: Despite substantial research documenting the relationships between intimate partner violence (IPV) victimization and HIV risks among women worldwide, few studies have examined these relationships among the growing population of migrant women who are disproportionately affected by these co-occurring problems. This cross-sectional study examined associations between lifetime IPV victimization and HIV risks among female migrants in Almaty, Kazakhstan. Methods: Survey interviews and testing for HIV and Syphilis were conducted among a random sample of 225 female migrant vendors who were employed in one of the largest markets in Central Asia. Multivariate regression estimated associations between experiencing any lifetime physical and/or …

Association Of Fungal Secondary Metabolism And Sclerotial Biology, Ana M. Calvo, Jeffrey W. Cary Feb 2015

Association Of Fungal Secondary Metabolism And Sclerotial Biology, Ana M. Calvo, Jeffrey W. Cary

Faculty Articles, Papers, and Other Scholarship

Fungal secondary metabolism and morphological development have been shown to be intimately associated at the genetic level. Much of the literature has focused on the co-regulation of secondary metabolite production (e.g., sterigmatocystin and aflatoxin in Aspergillus nidulans and Aspergillus flavus, respectively) with conidiation or formation of sexual fruiting bodies. However, many of these genetic links also control sclerotial production. Sclerotia are resistant structures produced by a number of fungal genera. They also represent the principal source of primary inoculum for some phytopathogenic fungi. In nature, higher plants often concentrate secondary metabolites in reproductive structures as a means of defense against …

Loop Diuretics For Patients Receiving Blood Transfusions, Michael Sarai, Aaron M. Tejani

Faculty and Staff Publications

Background: Blood transfusions are associated with significant morbidity and mortality. Prophylactic administration of loop diuretics (furosemide, bumetanide, ethacrynic acid, or torsemide) is common practice, especially among people who are at risk for circulatory overload, pulmonary oedema or both.

Objectives: This review aimed to determine if the prophylactic administration of loop diuretics (furosemide, bumetanide, ethacrynic acid, or torsemide) provides a therapeutic advantage (that is, a favourable risk benefit ratio) in adults and children who are recipients of any blood product transfusion versus placebo, no treatment, or general fluid restriction measures.

Search methods: We searched the Cochrane Renal Group's Specialised Register to …
