Electrochemical Monitoring Of Cell Wall-Regulated Transient Extracellular Oxidative Burst From Single Plant Cells, Jun-Tao Liu, Yan-Ling Liu, Zhi Cheng, Shi-Jing Chen, Wei-Hua Huang Feb 2015

Electrochemical Monitoring Of Cell Wall-Regulated Transient Extracellular Oxidative Burst From Single Plant Cells, Jun-Tao Liu, Yan-Ling Liu, Zhi Cheng, Shi-Jing Chen, Wei-Hua Huang

Journal of Electrochemistry

Oxidative burst by rapid, transient generation of reactive oxygen species (ROS) is a fundamental process for plant disease defense and signal transduction. ROS can be protective or toxic depending on their concentrations, and critical balance between ROS production and scavenging is regulated by complex and perfect mechanisms in plant cells. However, the process of rapid and transient ROS burst from plant cells and their regulatory mechanisms are far from being completely understood, owing to the methodological obstacles in real-time monitoring such a fast process. In this work, we showed the application of microelectrode electrochemistry with high spatiotemporal resolution to monitor …

Fabrication And Application Of Cytochrome P450 Electrochemcial Biosensor In Drug Metabolism, Xuan Xu, Ju-Sheng Lu, Song-Qin Liu

Journal of Electrochemistry

The process of drug metabolism plays an important role in drug efficacy and toxicity in vivo. Thus, the development of cheap, continent, rapid and high-throughput method for drug metabolism studies has great guiding significance for the design of new drugs, the determination of drug dosage and the detection of clinical drugs. Since the key role played by Cytochrome P450 (CYP450) in phase-I drug reaction, the constructed CYP450 enzyme biosensor can be used for the initial screening of drugs. It is found that the replacement of coenzyme NADPH with an electrode is to provide two electrons demanded in the catalytic reaction. …

Session F-2: Is It All Just Random?, Nicole Ross

Professional Learning Day

Students in middle school often learn basic genetics and gain experience using a Punnett Square analysis to determine probable genotype and phenotype outcomes for a family. What students may not learn is how these probabilities relate to the overall population. Probability is an integral part of genetics and heredity. Understanding what is meant by “probability” and how it applies to genetics is an important focus in the CCSS. One way to teach these concepts is to tap into popular literature. In this session, we will explore the genetics and probability behind the character, Auggie, in the book, Wonder, by R.J. …

The Entrance As A Complex Ecotone In A Neotropical Cave, Xavier Prous, Rodrigo Lopes Ferreira, Claudia Maria Jacobi Feb 2015

The Entrance As A Complex Ecotone In A Neotropical Cave, Xavier Prous, Rodrigo Lopes Ferreira, Claudia Maria Jacobi

International Journal of Speleology

Cave entrances are transition zones with intermediate characteristics between epigean environments, which bear lower environmental stability, and hypogean environments, with lower food resources. Associated to these interfaces there is a specific community, capable of exploiting its unique and intermediate characteristics. This work investigated this community in a Brazilian limnestone cave, identifying its arthropod species composition and spatial distribution, and exploring its relationships with climatic and structural environment characteristics and trophic relations. An ecotone zone was identified, with 55 species found only in the ecotone, 49 species were shared with the epigean community, 37 species with the hypogean community, and 14 …

Proteolytic Regulation Of Epithelial Sodium Channels By Urokinase Plasminogen Activator: Cutting Edge And Cleavage Sites, Hong Long Ji, Runzhen Zhao, Andrey A. Komissarov, Yongchang Chang, Yongfeng Liu, Michael A. Matthay

Translational Neuroscience

Plasminogen activator inhibitor 1 (PAI-1) level is extremely elevated in the edematous fluid of acutely injured lungs and pleurae. Elevated PAI-1 specifically inactivates pulmonary urokinase-type (uPA) and tissue-type plasminogen activators (tPA). We hypothesized that plasminogen activation and fibrinolysis may alter epithelial sodium channel (ENaC) activity, a key player in clearing edematous fluid. Two-chain urokinase (tcuPA) has been found to strongly stimulate heterologous human αβγ ENaC activity in a dose- and time-dependent manner. This activity of tcuPA was completely ablated by PAI-1. Furthermore, a mutation (S195A) of the active site of the enzyme also prevented ENaC activation. By comparison, three truncation …

Dendritic Distributions Of LH Channels In Experimentally-Derived Multi-Compartment Models Of Oriens-Lacunosum/Moleculare (O-Lm) Hippocampal Interneurons, Vladislav Sekulic, Tse-Chiang Chen, J. Josh Lawrence, Frances K. Skinner Feb 2015

Dendritic Distributions Of LH Channels In Experimentally-Derived Multi-Compartment Models Of Oriens-Lacunosum/Moleculare (O-Lm) Hippocampal Interneurons, Vladislav Sekulic, Tse-Chiang Chen, J. Josh Lawrence, Frances K. Skinner

Biomedical and Pharmaceutical Sciences Faculty Publications

The O-LM cell type mediates feedback inhibition onto hippocampal pyramidal cells and gates information flow in the CA1. Its functions depend on the presence of voltage-gated channels (VGCs), which affect its integrative properties and response to synaptic input. Given the challenges associated with determining densities and distributions of VGCs on interneuron dendrites, we take advantage of computational modeling to consider different possibilities. In this work, we focus on hyperpolarization-activated channels (h-channels) in O-LM cells. While h-channels are known to be present in O-LM cells, it is unknown whether they are present on their dendrites. In previous work, we used ensemble …

The Effect Of Light Intensity On Prey Detection Behavior In Two Lake Malawi Cichlids, Aulonocara Stuartgranti And Tramitichromis Sp., Margot A.B. Schwalbe, Jacqueline F. Webb

Biological Sciences Faculty Publications

Two sand-dwelling cichlids from Lake Malawi (Aulonocara stuartgranti, Tramitichromis sp.) that feed on benthic invertebrates, but have different lateral line phenotypes, use lateral line and/or visual cues to detect prey under light versus dark conditions. The current study examined how ecologically relevant variation in light intensity [0–800 lux (lx)] influences detection of prey (mobile, immobile) in each species by analyzing six behavioral parameters. Both species fed at light intensities ≥1 lx and trends in behavior among light intensities were informative. However, prey type and/or time of day (but not light intensity) predicted all four parameters analyzed with generalized …

Regulation Of Mammalian Nucleotide Metabolism And Biosynthesis, Andrew N. Lane, Teresa W-M Fan Feb 2015

Regulation Of Mammalian Nucleotide Metabolism And Biosynthesis, Andrew N. Lane, Teresa W-M Fan

Toxicology and Cancer Biology Faculty Publications

Nucleotides are required for a wide variety of biological processes and are constantly synthesized de novo in all cells. When cells proliferate, increased nucleotide synthesis is necessary for DNA replication and for RNA production to support protein synthesis at different stages of the cell cycle, during which these events are regulated at multiple levels. Therefore the synthesis of the precursor nucleotides is also strongly regulated at multiple levels. Nucleotide synthesis is an energy intensive process that uses multiple metabolic pathways across different cell compartments and several sources of carbon and nitrogen. The processes are regulated at the transcription level by …

Protein Sectors: Statistical Coupling Analysis Versus Conservation, Tiberiu Teşileanu, Lucy J. Colwell, Stanislas Leibler

Publications and Research

Statistical coupling analysis (SCA) is a method for analyzing multiple sequence alignments that was used to identify groups of coevolving residues termed “sectors”. The method applies spectral analysis to a matrix obtained by combining correlation information with sequence conservation. It has been asserted that the protein sectors identified by SCA are functionally significant, with different sectors controlling different biochemical properties of the protein. Here we reconsider the available experimental data and note that it involves almost exclusively proteins with a single sector. We show that in this case sequence conservation is the dominating factor in SCA, and can alone be …

Winter Rye As A Bioenergy Feedstock: Impact Of Crop Maturity On Composition, Biological Solubilization And Potential Revenue, Xiongjun Shao, Kay Dimarco, Tom L. Richard, Lee R. Lynd

Dartmouth Scholarship

Background:

Winter annual crops such as winter rye (Secale cereale L) can produce biomass feedstock on seasonally fallow land that continues to provide high-value food and feed from summer annuals such as corn and soybeans. As energy double crops, winter grasses are likely to be harvested while still immature and thus structurally different from the fully senesced plant material typically used for biofuels. This study investigates the dynamic trends in biomass yield, composition, and biological solubilization over the course of a spring harvest season.

Results:

The water soluble fraction decreased with increasing maturity while total carbohydrate content stayed …

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

Femtosecond x-ray laser pulses are used to probe the CO oxidation reaction on Ru initiated by an optical laser pulse. On a timescale of a few hundred femtoseconds, the optical laser pulse excites motions of CO and O on the surface allowing the reactants to collide and, with a transient close to a picosecond (ps), new electronic states appear in the O K-edge x-ray absorption spectrum. Density functional theory calculations indicate that these result from changes in the adsorption site and bond-formation between CO and O with a distribution of OC—O bond lengths close to the transition state (TS). After …

Stoichiometric Controls On Denitrification In High Nitrate Watersheds, Brian Grebliunas Feb 2015

Stoichiometric Controls On Denitrification In High Nitrate Watersheds, Brian Grebliunas

Theses and Dissertations

Watershed biogeochemistry throughout Midwestern agroecosystems have been altered through hydrologic manipulation and over-application of nitrogenous fertilizers. As a result, nitrate (NO3-N) export from subsurface drainage has negative impacts on local and downstream ecosystem health. Wetland installation has proven to be a viable option for targeted management where a large proportion of NO3-N is removed through the bacterially mediated process denitrification. For denitrification to maintain high rates under prolonged NO3-N saturation, a stable supply of labile dissolved organic carbon (DOC) is required. The focus of this dissertation was to study how the stoichiometry of agricultural wetlands limits denitrification within a controlled …

School of Integrative Biological & Chemical Sciences (Formerly Dept. of Biology)

In a recent Letter to the Editor, Norris et al. questioned the validity of some of our data reported by Feria-Arroyo et al. The main issue investigated by us was the potential impact of climate change on the probable distribution of the tick vector Ixodes scapularis in the Texas-Mexico transboundary region. As an ancillary issue, an analysis of sequence data for the intergenic spacer of Borrelia burgdorferi was conducted. In the present letter, we provide further evidence supporting our original results, and advocate that extensive study of the population genetics of B. burgdorferi is needed in the Texas-Mexico transboundary …
