Turkish Journal of Biology
A gene encoding ß-mannanase from Aspergillus niger GIM3.452 was amplified and inserted into a pPIC9K vector. The resulting recombinant plasmid, pPIC9K-MAN, was transformed into Pichia pastoris GS115. One strain (GSKM-1) having the highest ß-mannanase activity of 26.6 U/mL was obtained. In order to increase the secretion of ß-mannanase in P. pastoris, we constructed a double recombinant yeast and made it coexpress protein disulfide isomerase. One strain (GSKZ\alphaM2) with the highest ß-mannanase activity of 40 U/mL was then obtained and used to optimize expression conditions. When the GSKZ\alphaM2 strain was induced under the optimized conditions (methanol concentration 1.5%, induction time 7 …

Turkish Journal of Biology
Cynarin (CYN) is the main derivative of caffeoylquinic acid, found in leaves and heads of artichoke. It may have hepatoprotective, antiatherosclerotic, antioxidative, choleretic, and cholesterol-lowering effects. We tested the effects of various doses of CYN on the proliferative potential, survival, morphology, and stress response (SR) markers heme oxygenase-1 (HO-1) and heat shock protein-70 (HSP70) in normal human skin fibroblasts (FSF-1), telomerase-immortalized mesenchymal stem cells (hTERT-MSC), and cervical cancer cells (HeLa). The effects of CYN on cell proliferation and morphology were dose- and cell type-dependent, with 500 µM CYN as the upper limit for all cell types. While the growth and …

Turkish Journal of Biology
Our earlier studies found that in vitro estradiol modulates mitochondrial Ca2+ transport in discrete brain regions. The present study examined the role of estradiol receptors (ERs) in estradiol-induced inhibition of Ca^{2+} efflux from synaptosomal mitochondria isolated from rat caudate nuclei and brain stems. Radioactively labeled CaCl_2 (0.6?0.75 µCi ^45CaCl_{2}) was used for Ca^{2+} transport monitoring. The results revealed that in the presence of ER antagonist 7\alpha,17ß-[9[(4,4,5,5,5-pentafluoropentyl)sulfinyl]nonyl]estra-1,3,5(10)-triene-3,17-diol (ICI 182,780) (1 µmol/L), the inhibitory effect of estradiol on mitochondrial Ca^{2+} efflux was more than 60% decreased, suggesting the involvement of ER in this mode of estradiol neuromodulatory action. When particular contributions of …

Single Nucleotide Polymorphism Discovery Through Illumina-Based Transcriptome Sequencing And Mapping In Lentil, Hülya Yilmaz Temel, Deni̇z Göl, Hi̇lal Betül Kaya Akkale, Abdullah Kahri̇man, Muhammed Bahatti̇n Tanyolaç
Turkish Journal of Agriculture and Forestry
Lentil, which belongs to the family Leguminosae (Fabaceae), is a diploid (2n = 2x = 14 chromosomes) self-pollinating crop with a genome size of 4063 Mbp. Because of its nutritional importance and role in the fixation of nitrogen from the atmosphere, lentil is a widely used crop species in molecular genetic studies. By using DNA markers, to date, a limited number of polymorphic bands have been generated. Therefore, it is necessary to develop additional markers to saturate the genome at high density. Single nucleotide polymorphism (SNP) markers are promising for this purpose because of their abundance, stability, and heredity; they …

Assessing Age, Growth, And Reproduction Of Alburnus Mossulensis And Acanthobrama Marmid (Cyprinidae) Populations In Karakaya Dam Lake (Turkey), Aysel Alkan Uçkun, Di̇dem Gökçe
Turkish Journal of Zoology
In total, 626 individuals of Alburnus mossulensis and 586 individuals of Acanthobrama marmid were collected monthly from Karakaya Dam Lake on the upper Euphrates River. The gonadosomatic index, somatic condition, oocyte size, and fecundity were calculated on a monthly basis. Reading of scales indicated that the maximum age was 4+ years for A. mossulensis and A. marmid. The estimated length-weight relationships were W = 0.206 × FL^{2.065} for females and W = 0.119 × FL^{2.138} for males in A. mossulensis, and W = 0.029 × FL^{2.678} for females and W = 0.030 × FL^{2.631} for males in A. marmid. Growth …

Life History Traits In Bufotes Variabilis (Pallas, 1769) From 2 Different Altitudes In Turkey, Abdullah Altunişik, Nurhayat Özdemi̇r
Turkish Journal of Zoology
Life history characteristics (e.g., longevity, age at maturity, body size, and body mass) of the green toad, Bufotes variabilis, were investigated from 2 populations inhabiting different altitudes (Çankırı, 960 m a.s.l., n = 57; Adana, 13 m a.s.l., n = 52) in Turkey. Age was determined using skeletochronology. The oldest individual was 11 years old (range: 4-11 years) in the highland population, while the oldest individual was 6 years old in the lowland population (range: 2-6 years). Sexual maturity was estimated at 3-4 years in the highland population and 2-3 years in the lowland population by the skeletochronological pattern. Mean …
