Leaky Intestine And Impaired Microbiome In An Amyotrophic Lateral Sclerosis Mouse Model, Shaoping Wu, Jianxun Yi, Yong-Guo Zhang, Jingsong Zhou, Jun Sun

Faculty and Staff Publications

Emerging evidence has demonstrated that intestinal homeostasis and the microbiome play essential roles in neurological diseases, such as Parkinson's disease. Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by a progressive loss of motor neurons and muscle atrophy. Currently, there is no effective treatment. Most patients die within 3-5 years due to respiratory paralysis. Although the death of motor neurons is a hallmark of ALS, other organs may also contribute to the disease progression. We examined the gut of an ALS mouse model, G93A, which expresses mutant superoxide dismutase (SOD1(G93A)), and discovered a damaged tight junction structure and …

Characterizing Hiv-Associated Neurocognitive Disorder In Two Underserved Sociodemographic Groups, Julia S. Seay Mar 2015

Characterizing Hiv-Associated Neurocognitive Disorder In Two Underserved Sociodemographic Groups, Julia S. Seay

Open Access Dissertations

The purpose of this project is to characterize neurocognitive functioning and correlates of neurocognitive functioning, as well as to examine the diagnostic accuracy of relatively promising neurocognitive tests in two underserved sociodemographic groups living with HIV: monolingual Spanish-speaking Hispanics and English-speaking African-Americans, in order to better understand HIV-Associated Neurocognitive Disorder (HAND) within these groups. Examining HAND in underserved groups is paramount to the development of effective HAND screening and treatment algorithms in clinics that serve these groups, such as the AIDS Clinical Research Unit (ACRU) at the University of Miami. The current project was drawn from an initiative to develop …

Modern biological research requires rapid, complex, and reproducible integration of multiple experimental results generated both internally and externally (e.g., from public repositories). Although large systematic meta-analyses are among the most effective approaches both for clinical biomarker discovery and for computational inference of biomolecular mechanisms, identifying, acquiring, and integrating relevant experimental results from multiple sources for a given study can be time-consuming and error-prone. To enable efficient and reproducible integration of diverse experimental results, we developed a novel approach for standardized acquisition and analysis of high-throughput and heterogeneous biological data. This allowed, first, novel biomolecular network reconstruction in human prostate cancer, …

Varying Influences Of Selection And Demography In Host-Adapted Populations Of The Tick-Transmitted Bacterium, Anaplasma Phagocytophilum Speciation And Evolutionary Genetics, Matthew Aardema, Friederike D. Von Loewenich

Department of Biology Faculty Scholarship and Creative Works

Background: The host range of a pathogenic bacterial strain likely influences its effective population size, which in turn affects the efficacy of selection. Transmission between competent hosts may occur more frequently for host generalists than for specialists. This could allow higher bacterial population densities to persist within an ecological community and increase the efficacy of selection in these populations. Conversely, specialist strains may be better adapted to their hosts and consequently achieve greater within-host population densities, with corresponding increases in selection efficacy. To assess these different hypotheses, we examined the effective population sizes of three strains of the bacterium Anaplasma …

Dennis, Usha Ponnappan, Ralph L. Kodell, Kimberly K. Garner, Christopher M. Parkes, Melinda M. Bopp, Kalpana P. Padala, Charlotte A. Peterson, Prasad R. Padala, Dennis H. Sullivan

Physical Therapy Faculty Publications

BACKGROUND: Immune function may influence the ability of older adults to maintain or improve muscle mass, strength, and function during aging. Thus, nutritional supplementation that supports the immune system could complement resistance exercise as an intervention for age-associated muscle loss. The current study will determine the relationship between immune function and exercise training outcomes for older adults who consume a nutritional supplement or placebo during resistance training and post-training follow-up. The supplement was chosen due to evidence suggesting its ingredients [arginine (Arg), glutamine (Gln), and β-hydroxy β-methylbutyrate (HMB)] can improve immune function, promote muscle growth, and counteract muscle loss. …

Multi-Isotope Analysis To Reconstruct Dietary And Migration Patterns Of An Avar Population From Sajópetri, Hungary, Ad 568-895, Liotta Desiree Noche-Dowdy

USF Tampa Graduate Theses and Dissertations

The Avar were nomadic people from Central Asia who migrated into the Carpathian Basin in Central-Eastern Europe during the mid to late Migration Period (AD 568 - 895). Archaeological evaluation of grave goods and documentation of mortuary practices have been the primary means of understanding the Avar. However, this approach has largely neglected skeletal and biochemical analysis, in particular as these approaches relate to the biological variation, ancestry, and dietary patterns of the Avar.

There remains debate as to whether disparities existed among the socially stratified Avar population of ancient Hungary. It is argued by some that these disparities existed …

Molecular Modeling And Insilico Characterization Of Mutations Associated With Multiple Drug Resistant Tuberculosis (Mdr-Tb) And Extensive Drug Resistant Tuberculosis (Xdr-Tb), Sravanthi Joginipelli

Theses and Dissertations

The increasing emergence of multiple and extensive drug resistant tuberculosis poses a significant threat to effective control of tuberculosis (TB) globally. Resistance is a serious threat in the battle against the treatment and eradication of Tuberculosis. The continuing rise in tuberculosis incidence and the problem of drug resistance strains have prompted the research on developing new drug candidates and understanding the mechanism of drug resistance. Highly frequent mutations in gene sequence of mycobacterium tuberculosis were identified and were used to understand the mechanism of resistance and were computationally modeled to identify potential drug candidates for the treatment of MDR-TB and …

© Streva et al. Background: There are over a half a million copies of L1 retroelements in the human genome which are responsible for as much as 0.5% of new human genetic diseases. Most new L1 inserts arise from young source elements that are polymorphic in the human genome. Highly active polymorphic "hot" L1 source elements have been shown to be capable of extremely high levels of mobilization and result in numerous instances of disease. Additionally, hot polymorphic L1s have been described to be highly active within numerous cancer genomes. These hot L1s result in mutagenesis by insertion of new …

Probabilistic Records Linkage Using Historical Canadian Censuses And Vital Statistics, Stacey A. Hallman Mar 2015

Probabilistic Records Linkage Using Historical Canadian Censuses And Vital Statistics, Stacey A. Hallman

Population Change and Lifecourse Strategic Knowledge Cluster Conferences

Poster Presentation

This poster presents the results of a records linkage project to analyze potential mortality risks for young adults in Ontario during the 1918 influenza epidemic. With a team of research assistants at Western University, McMaster University, and the Université de Montréal, death records were linked to birth records in order to determine exact date of birth for the calculation of exact age at death. This date of birth is then compared to age as listed on other historical documents, such as the 1901 and 1911 Canadian censuses. The family environment (or living situation) was transcribed for each individual …

Melding Data Collection Methodology With Community Assistance: Benefits To Both Researchers And The Indigenous Groups They Study, Douglas S. London

Journal of Ecological Anthropology

I present a description of a model of melding data collection with community aid in the form of health educator training that emerged in the process of research collaboration during 2009-2011 with the Kawymeno Waorani foragers of Amazonian Ecuador. Some guidelines are suggested as to how benefits to both parties might be achieved when collecting data with indigenous populations. In this article I describe some of the advantages and pitfalls of melding data collection and community aid with research when collaborating with vulnerable indigenous groups.

Informal Child Care And Adolescent Psychological Well-Being: Hong Kong’S “Children Of 1997” Birth Cohort, Cherry Y. Leung, Gabriel M. Leung, Mary Schooling

Publications and Research

Background
Informal child care (child care by untrained family members, relatives or employees in the home) in Western populations is often associated with poorer psychological well-being, which may be confounded by socioeconomic position. We examined the association of informal child care, common in non-Western settings, with adolescent psychological wellbeing, using Hong Kong’s Chinese “Children of 1997” birth cohort.

Methods
Multivariable linear regression was used to examine the adjusted associations of informal child care (at 0.5, 3, 5 and 11 years) with parent-reported Rutter score for child behavior at 11 years, self-reported Culture-Free Self-Esteem Inventories score at 11 years and selfreported …

Direct Detection Of Fungal Siderophores On Bats With White-Nose Syndrome Via Fluorescence Microscopy-Guided Ambient Ionization Mass Spectrometry, Samantha J. Mascuch, Wilna J. Moree, Cheng-Chih Hsu

KIP Articles

White-nose syndrome (WNS) caused by the pathogenic fungus Pseudogymnoascus destructans is decimating the populations of several hibernating North American bat species. Little is known about the molecular interplay between pathogen and host in this disease. Fluorescence microscopy ambient ionization mass spectrometry was used to generate metabolic profiles from the wings of both healthy and diseased bats of the genus Myotis. Fungal siderophores, molecules that scavenge iron from the environment, were detected on the wings of bats with WNS, but not on healthy bats. This work is among the first examples in which microbial molecules are directly detected from an infected …

Best Practice Guidelines For Uncomplicated Urinary Tract Infections To Reduce Rates Of Antibiotic Resistance: A Ce Module For Clinicians, Romina Lo-Montano

Dissertations

Uncomplicated urinary tract infections (UTIs) accounted for over 8 million visits to ambulatory clinics. The annual cost of evaluation and treatment for uncomplicated UTIs on the health care system is estimated to be approximately $2.14 billion (Barber, et al., 2013). The improper treatment of patients with uncomplicated UTIs will result in increased healthcare expenditures, increase exposure of patients to potential side effects and an increase in antibiotic resistance. This capstone projected addressed the problem of non-adherence to evidence-based guidelines to treat uncomplicated UTI in women, among primary care providers. The purpose of this capstone project was to develop an evidence-based …

Household Demography And Early Childhood Mortality In A Rice-Farming Village In Northern Laos, Shinsuke Tomita, Daniel M. Parker, Julia Jennings, James Wood

Anthropology Faculty Scholarship

This paper extends Alexandr Chayanov’s model of changing household demography (specifically the ratio of food consumers to food producers) and its influence on agricultural behavior so that it includes possible adverse effects of a rising ratio on nutritional status and early childhood mortality within the household. We apply the model to 35 years’ worth of longitudinal demographic and economic data collected in the irrigated-rice growing village of Na Savang in northern Laos. When appropriate controls are included for other household variables, unobserved inter-household heterogeneity, and changes in local conditions and national policy over the study period, the analysis suggests that …

Solids Accumulation Rates Of Latrines At Rural Schools In Nimba County, Liberia, Caraline M. Murphy

USF Tampa Graduate Theses and Dissertations

Access to appropriate sanitation facilities as well as access to clean drinking water are considered fundamental human rights (Carr, 2001; Bjorklund & Sjodin, 2010), yet roughly 2.5 billion people on the planet lack access to an improved form of sanitation (WHO/UNICEF, 2014). Additionally, many entities responsible for emergency excreta management and sanitation management design guidelines, specifically solids accumulation rates in latrine pits, use rates that are 30-60 years old and may be established from dated knowledge on diet and lifestyle trends (Franceys et al., 1992; Harvey et al., 2002; Harvey, 2007). Using solids accumulation rates that are dated as well …
