Single-Use Technologies: Bridging Polymer Science to Biotechnology Applications

During their life-cycle (i.e., manufacture to end-use), single-use products in the biotechnology industry come in contact with processing equipment, packaging components, and delivery systems. Small amounts of chemicals that may transfer to the single-use product at any step in the life-cycle may subsequently leach into the pharmaceutical drug product and be delivered to the patient along with the dose. It is important that this chemical transfer does not occur in quantities that alter the safety, identity, strength or quality of the drug product. The assessment of potential leachable chemicals begins with a detailed analysis of compounds released from the single-use …

Daily Minority Stress And Affect Among Gay And Bisexual Men: A 30-Day Diary Study, Adam I. Eldahan, John E. Pachankis, H. Jonathan Rendina, Ana Ventuneac, Christian Grov, Jeffrey T. Parsons

Publications and Research

Background. This study examined the time-variant association between daily minority stress and daily affect among gay and bisexual men. Tests of time-lagged associations allow for a stronger causal examination of minority stress-affect associations compared with static assessments. Multilevel modeling allows for comparison of associations between minority stress and daily affect when minority stress is modeled as a between-person factor and a within-person time-fluctuating state.

Methods. 371 gay and bisexual men in New York City completed a 30-day daily diary, recording daily experiences of minority stress and positive affect (PA), negative affect (NA), and anxious affect (AA). Multilevel analyses examined …

Loss Of Glycosaminoglycan Receptor Binding After Mosquito Cell Passage Reduces Chikungunya Virus Infectivity, Dhiraj Acharya, Amber M. Paul, John F. Anderson, Faqing Huang, Fengwei Bai

Faculty Publications

Chikungunya virus (CHIKV) is a mosquito-transmitted alphavirus that can cause fever and chronic arthritis in humans. CHIKV that is generated in mosquito or mammalian cells differs in glycosylation patterns of viral proteins, which may affect its replication and virulence. Herein, we compare replication, pathogenicity, and receptor binding of CHIKV generated in Vero cells (mammal) or C6/36 cells (mosquito) through a single passage. We demonstrate that mosquito cell-derived CHIKV (CHIKVmos) has slower replication than mammalian cell-derived CHIKV (CHIKVvero), when tested in both human and murine cell lines. Consistent with this, CHIKVmos infection in both cell lines …

Articles

The purpose of this paper is to recommend a framework and methodology for incorporating hydrologic data and ecohydrologic relationships in Ecological Site Descriptions (ESDs) and thereby enhance the utility of ESDs for assessing rangelands and guiding resilience-based management strategies. Resilience-based strategies assess and manage ecological state dynamics that affect state vulnerability and, therefore, provide opportunities to adapt management. Many rangelands are spatially heterogeneous or sparsely vegetated where the vegetation structure strongly influences infiltration and soil retention. Infiltration and soil retention further influence soil water recharge, nutrient availability, and overall plant productivity. These key ecohydrologic relationships govern the ecologic resilience of …

Identification Of The Substrate Access Portal Of 5-Lipoxygenase, Sunayana Mitra, Sue G. Bartlett, Marcia E. Newcomer Oct 2015

Identification Of The Substrate Access Portal Of 5-Lipoxygenase, Sunayana Mitra, Sue G. Bartlett, Marcia E. Newcomer

Faculty Publications

© 2015 American Chemical Society. The overproduction of inflammatory lipid mediators derived from arachidonic acid contributes to asthma and cardiovascular diseases, among other pathologies. Consequently, the enzyme that initiates the synthesis of pro-inflammatory leukotrienes, 5-lipoxygenase (5-LOX), is a target for drug design. The crystal structure of 5-LOX revealed a fully encapsulated active site; thus the point of substrate entry is not known. We asked whether a structural motif, a "cork" present in 5-LOX but absent in other mammalian lipoxygenases, might be ejected to allow substrate access. Our results indicate that reduction of cork volume facilitates access to the active site. …

Sedentary Behavior In Preschoolers: How Many Days Of Accelerometer Monitoring Is Needed?, Wonwoo Byun, Michael W. Beets, Russell R. Pate

Faculty Publications

The reliability of accelerometry for measuring sedentary behavior in preschoolers has not been determined, thus we determined how many days of accelerometry monitoring are necessary to reliably estimate daily time spent in sedentary behavior in preschoolers. In total, 191 and 150 preschoolers (three to five years) wore ActiGraph accelerometers (15-s epoch) during the in-school (≥4 days) and the total-day (≥6 days) period respectively. Accelerometry data were summarized as time spent in sedentary behavior (min/h) using three different cutpoints developed for preschool-age children (

Use Of Cactus Mucilage As A Dispersant And Absorbant For Oil In Oil-Water Mixtures, Norma A. Alcantar, Dawn I. Fox, Sylvia Thomas, Ryan G. Toomey Oct 2015

Use Of Cactus Mucilage As A Dispersant And Absorbant For Oil In Oil-Water Mixtures, Norma A. Alcantar, Dawn I. Fox, Sylvia Thomas, Ryan G. Toomey

USF Patents

Despite advances in off-shore oil exploration and drilling technology, accidental oil spills do occur. Depending on their magnitude and location, irreparable damage may be inflicted on marine and coastal ecosystems. Emulsification of a water/surfactant/oil system results in the production of oil droplets and this depends on the formulation and composition variables, mixing characteristics and system preparation. Thus, research is ongoing on improving methods of cleaning up oil spills. Cactus mucilage was used to disperse and absorb oil from a simulated oil slick in different salt concentrations. Results show correlations between emulsion properties and interfacial characteristics of mucilage as function of …

A Critical Role For Alpha-Synuclein In Development And Function Of T Lymphocytes, Afshin Shameli, Wenbin Xiao, Yan Zheng, Susan Shyu, John Sumodi, Howard J. Meyerson, Clifford V. Harding, Robert W. Maitta Oct 2015

A Critical Role For Alpha-Synuclein In Development And Function Of T Lymphocytes, Afshin Shameli, Wenbin Xiao, Yan Zheng, Susan Shyu, John Sumodi, Howard J. Meyerson, Clifford V. Harding, Robert W. Maitta

Faculty Scholarship

Alpha-synuclein is highly expressed in the central nervous system and plays an important role in pathogenesis of neurodegenerative disorders such as Parkinson's disease and Lewy body dementia. Previous studies have demonstrated the expression of α-synuclein in hematopoietic elements and peripheral blood mononuclear cells, although its roles in hematopoiesis and adaptive immunity are not studied. Using an α-synuclein knock out (KO) mouse model, we have recently shown that α-synuclein deficiency is associated with a mild defect in late stages of hematopoiesis. More importantly, we demonstrated a marked defect in B lymphocyte development and IgG, but not IgM production in these mice. …

Prosecuting Online Threats After Elonis, Michael Pierce Oct 2015

Prosecuting Online Threats After Elonis, Michael Pierce

NULR Online

In the essay, Mr. Pierce discusses what, exactly, the government must prove before it can, consistent with the First Amendment, prosecute someone who posts threatening messages on Facebook. Last Term, a divided Court wrestled with this issue in Elonis v. United States, reversing the defendant's conviction but leaving an important question unanswered: does the government need to prove that a speaker was reckless with his words or, alternatively, that he specifically intended them to be interpreted as threats? The essay suggests that instead of deciding which standard is best for all online threats, lower courts should adopt libel law’s …

Translational Research In Education For Knowledge Mobilisation: A Study Of Use And Teacher Perception In Primary Schools In England, Uk, T Ovenden-Hope, L La Velle

Plymouth Institute of Education

In order to achieve the standard expected in the classroom, teachers benefit from continuing professional development that enables access to resources with which to inform their practice. Smith and Helfenbein's “Translational Research in Education: Collaboration and Commitment in Urban Contexts.” In The Collaborative Turn: Working Together in Qualitative Research (2009), edited by W. S. Gershon. Rotterdam: Sense, identified that translational research ‘creates a space for collaborative, co-constructed inquiry that values and utilises the expertise of all stakeholders involved’ (page 91). This paper presents an overview and investigation of a founding model, MESH (Mapping Education Specialist knowHow guides, www.meshguides.org), for translational …
