Meta-Analysis Of Genome-Wide Association Studies With Correlated Individuals: Application To The Hispanic Community Health Study/Study Of Latinos (Hchs/Sol), Tamar Sofer, John R. Shaffer, Misa Graff, Qibin Qi, Adrienne M. Stilp, Stephanie M. Gogarten, Kari E. North, Carmen R. Isasi, Cathy C. Laurie, Adam A. Szpiro

UW Biostatistics Working Paper Series

Investigators often meta-analyze multiple genome-wide association studies (GWASs) to increase the power to detect associations of single nucleotide polymorphisms (SNPs) with a trait. Meta-analysis is also performed within a single cohort that is stratified by, e.g., sex or ancestry group. Having correlated individuals among the strata may complicate meta-analyses, limit power, and inflate Type 1 error. For example, in the Hispanic Community Health Study/Study of Latinos (HCHS/SOL), sources of correlation include genetic relatedness, shared household, and shared community. We propose a novel mixed-effect model for meta-analysis, “MetaCor", which accounts for correlation between stratum-specific effect estimates. Simulations show that MetaCor controls …

Fitting In: A Study Of Lesbian Mothers In Rural Southeastern Michigan, Allison Amadea Ranusch Nov 2015

Fitting In: A Study Of Lesbian Mothers In Rural Southeastern Michigan, Allison Amadea Ranusch

Master's Theses and Doctoral Dissertations

Lesbian parents residing in rural and small communities have received little scholarly attention. The primary focus of the existing research has highlighted the isolation experienced by lesbian parents within such locations, but little examination has been made of other issues or their coping mechanisms. This study was designed to address these deficits in knowledge and understanding through a series of eleven in-depth, qualitative interviews. Several stigma management strategies were discovered. For example, it was reported that engaging in farming and trade helped lesbian mothers "fit in" enough to gain some level of acceptance from their immediate neighbors. Stigma was also …

Development Of 11-Plex Assay For The Rapid Screening Of Samples For Detection Of Shiga Toxin-Producing E. Coli, Travis Woods

Biomedical Engineering ETDs

Shiga toxin-producing Escherichia coli (STEC) have been identified by the USDA as a serious threat to the nations health stemming from contaminations in the food supply, specifically, the beef chain. I have developed an assay that is able to screen samples for STEC in a rapid and multiplexed format. Multiplex oligonucleotide ligation-PCR (MOL- PCR) is a nucleic acid based assay patented at Los Alamos National Laboratory (LANL) that uses flow cytometry and multiplex microsphere arrays for detection of nucleic acid based signatures. By using MOL-PCR for detecting unique STEC DNA signatures in samples from the beef supply chain (farm to …

Cut From The Same Cloth: The Us Textile And Apparel Industry And Post-Disaster Designs For Haiti, Ransford Edwards Jr. Nov 2015

Cut From The Same Cloth: The Us Textile And Apparel Industry And Post-Disaster Designs For Haiti, Ransford Edwards Jr.

Humanities and Politics Faculty Articles

In the aftermath of the 2010 Haitian earthquake, various neoliberal strategies have been advanced to help in short-term disaster mitigation and reconstruction, as well as more long-term improvements in the country’s overall economic integration and growth. One such strategy has been focused on revitalizing the country’s apparel assembly industries through an aggressive expansion of export processing zones (EPZs). The disaster, it appears, represented an important opportunity to improve economic conditions by reorganizing the country’s role in the global apparel commodity chain. However, this reorganization conflicts with the preferences of US textile and apparel producers who have used trade preference programs …

Conserved Molecular Underpinnings And Characterization Of A Role For Caveolin-1 In The Tumor Microenvironment Of Mature T-Cell Lymphomas, Tyler A. Herek, Timothy D. Shew, Heather N. Spurgin, Christine E. Cutucache

Biology Faculty Publications

Neoplasms of extra-thymic T-cell origin represent a rare and difficult population characterized by poor clinical outcome, aggressive presentation, and poorly defined molecular characteristics. Much work has been done to gain greater insights into distinguishing features among malignant subtypes, but there also exists a need to identify unifying characteristics to assist in rapid diagnosis and subsequent potential treatment. Herein, we investigated gene expression data of five different mature T-cell lymphoma subtypes (n = 187) and found 21 genes to be up- and down-regulated across all malignancies in comparison to healthy CD4+ and CD8+ T-cell controls (n = 52). From these results, …

Species Specific Microcalcification In Reef Building Caribbean Corals In Ocean Acidification Conditions, Ashley M. Dungan Nov 2015

Species Specific Microcalcification In Reef Building Caribbean Corals In Ocean Acidification Conditions, Ashley M. Dungan

HCNSO Student Theses and Dissertations

Coral reefs are one of the most economically important ecosystems on the planet. Despite their great contribution to the world economy, anthropogenic influence via carbon dioxide emissions is leading to unprecedented changes with concerns about subsequent negative impacts on reefs. Surface ocean pH has dropped 0.1 units in the past century; in spite of this rapid shift in oceanic chemistry, it is unclear if individual species or life stages of Caribbean stony corals will be more sensitive to ocean acidification (OA). Examined is the relationship between CO2-induced seawater acidification, net calcification, photosynthesis, and respiration in three model Caribbean …

Beyond Fruit: Examining Community In A Community Orchard, Emily Jane Becker

Dissertations and Theses

The Fruits of Diversity Community Orchard, located in Portland, Oregon in an affordable housing neighborhood, is a site of alternative food provisioning in which a group of people, organized by two nonprofits, work together to manage fruit and nut producing plants. Through conversations with volunteers who participate regularly and participant observation, this study explores the questions: What does community mean in the context of a community orchard? In what ways does partnering with a nonprofit from outside the neighborhood influence community and the way the project is operationalized?

This thesis situates community orchards within the literature on alternative food networks …

Early Shifts Of Brain Metabolism By Caloric Restriction Preserve White Matter Integrity And Long-Term Memory In Aging Mice, Janet Guo, Vikas Bakshi, Ai-Ling Lin Nov 2015

Early Shifts Of Brain Metabolism By Caloric Restriction Preserve White Matter Integrity And Long-Term Memory In Aging Mice, Janet Guo, Vikas Bakshi, Ai-Ling Lin

Sanders-Brown Center on Aging Faculty Publications

Preservation of brain integrity with age is highly associated with lifespan determination. Caloric restriction (CR) has been shown to increase longevity and healthspan in various species; however, its effects on preserving living brain functions in aging remain largely unexplored. In the study, we used multimodal, non-invasive neuroimaging (PET/MRI/MRS) to determine in vivo brain glucose metabolism, energy metabolites, and white matter structural integrity in young and old mice fed with either control or 40% CR diet. In addition, we determined the animals’ memory and learning ability with behavioral assessments. Blood glucose, blood ketone bodies, and body weight were also measured. We …
