Functional Impact Of 14 Single Nucleotide Polymorphisms Causing Missense Mutations Of Human α7 Nicotinic Receptor, Qinhui Zhang, Yingjie Du, Jianliang Zhang, Xiaojun Xu, Fenqin Xue, Cong Guo, Yao Huang, Ronald J. Lukas, Yongchang Chang Sep 2015

Functional Impact Of 14 Single Nucleotide Polymorphisms Causing Missense Mutations Of Human α7 Nicotinic Receptor, Qinhui Zhang, Yingjie Du, Jianliang Zhang, Xiaojun Xu, Fenqin Xue, Cong Guo, Yao Huang, Ronald J. Lukas, Yongchang Chang

Translational Neuroscience

The α7nicotinic receptor (nAChR) is a major subtype of the nAChRs in the central nervous system, and the receptor plays an important role in brain function. In the dbSNP database, there are 55 single nucleotide polymorphisms (SNPs) that cause missense mutations of the human α7nAChR in the coding region. In this study, we tested the impact of 14 SNPs that cause missense mutations in the agonist binding site or the coupling region between binding site and channel gate on the receptor function. The wild type or mutant receptors were expressed or co-expressed in Xenopus oocytes, and the agonist-induced currents were …

Life History, Oviposition Characteristics, And Management Of The Ozark Baltimore Checkerspot (Euphydryas Phaeton Ozarkae), Stephen Robertson Sep 2015

Life History, Oviposition Characteristics, And Management Of The Ozark Baltimore Checkerspot (Euphydryas Phaeton Ozarkae), Stephen Robertson

Theses and Dissertations

The Ozark Baltimore Checkerspot (Euphydryas phaeton ozarkae Masters) is an understudied nymphalid butterfly endemic to the Ozarks that specializes on False Foxglove (Aureolaria sp.) as the larval food plant. Few studies have focused on this animal and much of what is known has been derived from studies of the eastern subspecies (Euhpydryas phaeton phaeton Drury). Genetic evidence shows only slight variation between subspecies, but critical ecological differences are found between the two taxa. Here I present an in-depth life history with respect to general ecology and offer evidence towards species identity. Oviposition behavior (search and egg-site selection) is a key …

Seasonal Prevalence Of Shiga-Toxin Producing Escherichia Coli On Beef Cattle Farms In Eastern Arkansas, Tiffani Elizabeth Breckenridge Sep 2015

Seasonal Prevalence Of Shiga-Toxin Producing Escherichia Coli On Beef Cattle Farms In Eastern Arkansas, Tiffani Elizabeth Breckenridge

Student Theses and Dissertations

Sixty-five million U.S. citizens become ill from foodborne bacteria annually; 73,000 of these bacterial illnesses are caused by Shiga-toxin producing Escherichia coli (STEC) O157. Cattle are a reservoir of STEC; therefore, STEC contamination of beef is a significant food safety concern in the U.S. On-farm STEC control measures may reduce the incidence of contamination in processing facilities. The objective of this study was to determine prevalence of O157 and non-O157 STEC on beef cattle farms in eastern Arkansas, and to possibly identify farm management and/or environmental risk factors associated with STEC prevalence in cattle. Rectal swab samples (10 cows per …

(-)-Oleocanthal Rapidly And Selectively Induces Cancer Cell Death Via Lysosomal Membrane Permeabilization, Onica Legendre, Paul A. S. Breslin, David A. Foster

Publications and Research

(-)-Oleocanthal (OC), a phenolic compound present in extra-virgin olive oil (EVOO), has been implicated in the health benefits associated with diets rich in EVOO. We investigated the effect of OC on human cancer cell lines in culture and found that OC induced cell death in all cancer cells examined as rapidly as 30 minutes after treatment in the absence of serum. OC treatment of non-transformed cells suppressed their proliferation but did not cause cell death. OC induced both primary necrotic and apoptotic cell death via induction of lysosomal membrane permeabilization (LMP). We provide evidence that OC promotes LMP by inhibiting …

Real-Time Laser Beam Alignment To A Remote Target Using Blob Detection And Filtration For Unmanned Mobile Spectroscopy Platform, Jarjees Ahmed Khidir

Theses and Dissertations

In a project supported by NASA, a new unmanned air quality monitoring system has been proposed to look for signs of life on Mars. The system development and tests are specially directed to build an unmanned open-path spectrometer for near-ground biogenic gas concentration detection on Mars. Moved by a mobile robot platform, the spectrometer shoots an infra-red laser beam to a remote retro-reflector and examines the reflected signal looking for the absorption peak of the investigated gas. The problem is that the laser beam needs to be constantly aligned to the retro-reflect (the target) while the mobile robot is moving. …

Novel Selected Hetero-Atom Doped Carbon From Renewable Resource Materials For Fuel Cell And Supercapacitor Applications, Sunil Kumar Ramasahayam Sep 2015

Novel Selected Hetero-Atom Doped Carbon From Renewable Resource Materials For Fuel Cell And Supercapacitor Applications, Sunil Kumar Ramasahayam

Theses and Dissertations

Doped carbon materials have gained a lot of attention recently due to their wide array of applications, for example, in dye degradation, energy conversion and storage, electrocatalysis, sensors and electronics. These doped carbon materials are claimed to have a tremendous potential to substitute expensive metal based electrocatalysts in fuel cell and supercapacitor applications. A recently established microwave assisted technique, which does not make use of any inert or reducing gas during the synthesis, was utilized to synthesize doped carbon materials. The method is simple, rapid, economical and high yielding. Renewable resource materials like tannin, spent coffee grounds and thiamine were …

The Effect Of Surface Modification Of Aligned Poly-L-Lactic Acid Electrospun Fibers On Fiber Degradation And Neurite Extension, Nicholas J. Schaub, Clémentine Le Beux, Jianjun Miao, Robert J. Linhardt, Johan G. Alauzun, Danielle Laurencin, Ryan J. Gilbert Sep 2015

The Effect Of Surface Modification Of Aligned Poly-L-Lactic Acid Electrospun Fibers On Fiber Degradation And Neurite Extension, Nicholas J. Schaub, Clémentine Le Beux, Jianjun Miao, Robert J. Linhardt, Johan G. Alauzun, Danielle Laurencin, Ryan J. Gilbert

Chemical and Biochemical Engineering Faculty Research & Creative Works

The surface of aligned, electro spun poly-L-lactic acid (PLLA) fibers was chemically modified to determine if surface chemistry and hydrophilicity could improve neurite extension from chick dorsal root ganglia. Specifically, diethylenetriamine (DTA, for amine functionalization), 2-(2-aminoethoxy) ethanol (AEO, for alcohol functionalization), or GRGDS (cell adhesion peptide) were covalently attached to the surface of electro spun fibers. Water contact angle measurements revealed that surface modification of electro spun fibers significantly improved fiber hydrophilicity compared to unmodified fibers (p < 0.05). Scanning electron microscopy (SEM) of fibers revealed that surface modification changed fiber topography modestly, with DTA modified fibers displaying the roughest surface structure. Degradation of chemically modified fibers revealed no change in fiber diameter in any group over a period of seven days. Unexpectedly, neurites from chick DRG were longest on fibers without surface modification (1651 ± 488 μm) and fibers containing GRGDS (1560 ± 107 μm). Fibers modified with oxygen plasma (1240 ± 143 μm) or DTA (1118 ± 82 μm) produced shorter neurites than the GRGDS or unmodified fibers but were not statistically shorter than unmodified and GRGDS modified fibers. Fibers modified with AEO (844 ± 151 μm) were significantly shorter than unmodified and GRGDS modified fibers (p< 0.05). Based on these results, we conclude that fiber hydrophilic enhancement alone on electro spun PLLA fibers does not enhance neurite outgrowth. Further work must be conducted to better understand why neurite extension was not improved on more hydrophilic fibers, but the results presented here do not recommend hydrophilic surface modification for the purpose of improving neurite extension unless a bioactive ligand is used.

Autophagy is important for maintaining cellular homeostasis, and thus its deficiency is implicated in a broad spectrum of human diseases. Its role in platelet function has only recently been examined. Our biochemical and imaging studies demonstrate that the core autophagy machinery exists in platelets, and that autophagy is constitutively active in resting platelets. Moreover, autophagy is induced upon platelet activation, as indicated by agonist-induced loss of the autophagy marker LC3II. Additional experiments, using inhibitors of platelet activation, proteases, and lysosomal acidification, as well as platelets from knockout mouse strains, show that agonist-induced LC3II loss is a consequence of platelet signaling …

Neurobiology Of Seasonal Life-History Transitions, Ashley Rae Lucas Sep 2015

Neurobiology Of Seasonal Life-History Transitions, Ashley Rae Lucas

Dissertations and Theses

Many animals exhibit seasonal changes in life-history stages, and these seasonal transitions are often accompanied by dramatic switches in behavior. While the neuroendocrine mechanisms that regulate such behavioral transitions are poorly understood, arginine vasotocin (AVT) and neuropeptide Y (NPY) are excellent candidates because they regulate reproductive and feeding behavior, respectively. In this study, I asked if seasonal changes in AVT and/or NPY are concomitant with spring migration away from the breeding grounds, as male and female red-sided garter snakes (Thamnophis sirtalis parietalis) are transitioning from reproductive to non-reproductive behavior during this time. To address this question, I collected …

What Difference Does It Make? An Essay Review Of Beyond Versus: The Struggle To Understand The Interaction Of Nature And Nurture; James Tabery; Mit Press: Cambridge, Ma, 2014, Peter J. Taylor

Working Papers on Science in a Changing World

Beyond versus makes its contribution to the thriving industry of books that clarify or recast nature-nurture issues through seven conceptual moves. The first is to posit a divide between sociological and philosophical inquiry. As Tabery depicts them, commentators on the science invoked in nature-nurture debates often focus on the racist or other political views of disputants or on their flawed understanding of scientific concepts. Tabery, in contrast, as a philosopher of science, explains past and present disagreements as stemming from “a disagreement concerning how explanation works in science.” (The other moves include explanatory and terminological divides, connecting associations to mechanisms, …
