Novel Approaches Reveal That Toxoplasma Gondii Bradyzoites Within Tissue Cysts Are Dynamic And Replicating Entities In Vivo, Elizabeth Watts, Yihua Zhao, Animesh Dhara, Becca Eller, Abhijit R. Patwardhan, Anthony P. Sinai Sep 2015

Novel Approaches Reveal That Toxoplasma Gondii Bradyzoites Within Tissue Cysts Are Dynamic And Replicating Entities In Vivo, Elizabeth Watts, Yihua Zhao, Animesh Dhara, Becca Eller, Abhijit R. Patwardhan, Anthony P. Sinai

Microbiology, Immunology, and Molecular Genetics Faculty Publications

Despite their critical role in chronic toxoplasmosis, the biology of Toxoplasma gondii bradyzoites is poorly understood. In an attempt to address this gap, we optimized approaches to purify tissue cysts and analyzed the replicative potential of bradyzoites within these cysts. In order to quantify individual bradyzoites within tissue cysts, we have developed imaging software, BradyCount 1.0, that allows the rapid establishment of bradyzoite burdens within imaged optical sections of purified tissue cysts. While in general larger tissue cysts contain more bradyzoites, their relative "occupancy" was typically lower than that of smaller cysts, resulting in a lower packing density. The packing …

USF Patents

Provided herein is a method of reducing a microbial infection of a heparinized surface wherein heparin precipitation is reduced as compared to a control. In some embodiments a composition comprising ethanol and isopropanol is applied to the surface. In other embodiments, two or more ethanol compositions are applied and removed sequentially from the surface prior to contacting the surface with a biological fluid, tissue or vessel, wherein the composition in each successive administration comprises an increased amount of ethanol. Also provided is a method of reducing a microbial infection of a surface comprising applying a mucilage extract from an Opuntia …

Methods And Products For Biasing Cellular Development, Kiminobu Sugaya, Angel Alvarez, Amelia Marutle Sep 2015

Methods And Products For Biasing Cellular Development, Kiminobu Sugaya, Angel Alvarez, Amelia Marutle

UCF Patents

Methods are described that bias cells, such as potent and multipotent stem cells, by transfection with a nucleic acid sequence, to differentiate to a desired end-stage cell or a cell having characteristics of a desired end-stage cell. In particular embodiments, human neural stem cells are transfected with vectors comprising genes in the homeobox family of transcription factor developmental control genes, and this results in a greater percentage of resultant transformed cells, or their progeny, differentiating into a desired end-stage cell or a cell having characteristics of a desired end-stage cell.

Biocompatible Nano Rare Earth Oxide Upconverters For Imaging And Therapeutics, Michael Bass, Sudipta Seal, William Self Sep 2015

Biocompatible Nano Rare Earth Oxide Upconverters For Imaging And Therapeutics, Michael Bass, Sudipta Seal, William Self

UCF Patents

Methods and systems for doping cerium oxide and other therapeutically valuable nanoparticles with rare earth ions either singly or in combinations so that infrared light is absorbed and visible light is emitted through the process of up conversion. A novel type of therapeutic nanoparticle with rare earth doping to provide for up conversion emission is invented that makes possible identification, tracking and evaluating the functionality of the nanoparticles and further elucidation of ROS scavenging mechanism. The present invention may be applied to a wide variety of therapeutic agents under investigation.

Novel Pcr Assays Complement Laser Biosensor-Based Method And Facilitate Listeria Species Detection From Food, Kwang-Pyo Kim, Atul K. Singh, Xingjian Bai, Lena Leprun, Arun K. Bhunia Sep 2015

Novel Pcr Assays Complement Laser Biosensor-Based Method And Facilitate Listeria Species Detection From Food, Kwang-Pyo Kim, Atul K. Singh, Xingjian Bai, Lena Leprun, Arun K. Bhunia

Department of Food Science Faculty Publications

The goal of this study was to develop the Listeria species-specific PCR assays based on a house-keeping gene (lmo1634) encoding alcohol acetaldehyde dehydrogenase (Aad), previously designated as Listeriaadhesion protein (LAP), and compare results with a label-free light scattering sensor, BARDOT (bacterial rapid detection using optical scattering technology). PCR primer sets targeting the lap genes from the species ofListeria sensu stricto were designed and tested with 47 Listeria and 8 non-Listeria strains. The resulting PCR primer sets detected either all species of Listeria sensu stricto or individual L. innocua, L. ivanovii and L. seeligeri, …

Breast cancer is a leading cause of death for women, with mortality resulting from metastasis. Metastases are often detected once tumour cells affect the function of solid organs, with a high disease burden limiting effective treatment. Here we report a method for the early detection of metastasis using an implanted scaffold to recruit and capture metastatic cells in vivo, which achieves high cell densities and reduces the tumour burden within solid organs 10-fold. Recruitment is associated with infiltration of immune cells, which include Gr1hiCD11b+cells. We identify metastatic cells in the scaffold through a label-free detection system using inverse spectroscopic …

Prehistoric Drawings In Mammoth Cave, Logan Kistler Sep 2015

Prehistoric Drawings In Mammoth Cave, Logan Kistler

Kaleidoscope

During a recent Earthwatch Institute survey of archaeological remains in Mammoth Cave, a project was begun to find and record prehistoric images on the cave walls. I chose to analyze petroglyphs and pictographs on three panels in Main Cave. This article offers a hypothesis for the circumstances surrounding the rock art’s production: the geometric and anthropomorphic figures in Mammoth Cave are representative of a series of visual percepts experienced cross-culturally and caused by various conditions — including sensory deprivation, fatigue, and psychoactive drug use — acting on the ocular anatomy and nervous system. That is, the glyphs might be visual …

Nietzsche Contra Wright: On Becoming What You Are, Jordan Rodgers Sep 2015

Nietzsche Contra Wright: On Becoming What You Are, Jordan Rodgers

Kaleidoscope

Robert Wright’s recent book on evolutionary psychology, The Moral Animal, is concerned largely with the ethical implications of recent evolutionary science, and espouses a form of utilitarianism as the ethical theory that should naturally follow evolutionary insights into human psychology. This paper challenges that notion, with constant reference to the work of the philosopher Friedrich Nietzsche, on the basis that such an ethical theory places far too little emphasis on the individual as such, and is tantamount to a form of nihilism. This paper also argues that, while seeking for the happiness of other people is a good thing, our …
