Novel Mode Of Defective Neural Tube Closure In The Non-Obese Diabetic (Nod) Mouse Strain, J. Michael Salbaum, Claudia Kruger, Jacalyn Macgowan, Nils J. Herion, David Burk, Claudia Kappen Nov 2015

Novel Mode Of Defective Neural Tube Closure In The Non-Obese Diabetic (Nod) Mouse Strain, J. Michael Salbaum, Claudia Kruger, Jacalyn Macgowan, Nils J. Herion, David Burk, Claudia Kappen

Faculty Publications

Failure to close the neural tube results in birth defects, with severity ranging from spina bifida to lethal anencephaly. Few genetic risk factors for neural tube defects are known in humans, highlighting the critical role of environmental risk factors, such as maternal diabetes. Yet, it is not well understood how altered maternal metabolism interferes with embryonic development, and with neurulation in particular. We present evidence from two independent mouse models of diabetic pregnancy that identifies impaired migration of nascent mesodermal cells in the primitive streak as the morphogenetic basis underlying the pathogenesis of neural tube defects. We conclude that perturbed …

A Combination Approach To Treating Fungal Infections, Sanjib K. Shrestha, Marina Y. Fosso, Sylvie Garneau-Tsodikova

Pharmaceutical Sciences Faculty Publications

Azoles are antifungal drugs used to treat fungal infections such as candidiasis in humans. Their extensive use has led to the emergence of drug resistance, complicating antifungal therapy for yeast infections in critically ill patients. Combination therapy has become popular in clinical practice as a potential strategy to fight resistant fungal isolates. Recently, amphiphilic tobramycin analogues, C12 and C14, were shown to display antifungal activities. Herein, the antifungal synergy of C12 and C14 with four azoles, fluconazole (FLC), itraconazole (ITC), posaconazole (POS), and voriconazole (VOR), was examined against seven Candida albicans strains. All tested strains …

Direct And Indirect Effects Of The Caribbean Cymothoid Parasite Anilocra Haemuli On Host Energetics, Behavior, And Trophic Interactions, Rachel Lynn Welicky Nov 2015

Direct And Indirect Effects Of The Caribbean Cymothoid Parasite Anilocra Haemuli On Host Energetics, Behavior, And Trophic Interactions, Rachel Lynn Welicky

Student Theses and Dissertations

The majority of coral reef research focuses on organisms that are familiar to humans, yet 80% of coral reef animals are parasites. Research on the direct effects of parasitism has suggested that hosts may undergo both behavioral and physiological changes. These changes may manifest at the ecosystem level, yet evidence of such changes are scarce. The objectives of this study were to investigate the direct and indirect effects of the Caribbean cymothoid parasite Anilocra haemuli on Haemulon flavolineatum (French grunt) energetics, behavior and trophic interactions. I conducted field surveys to measure A. haemuli prevalence of infection and determine its association …

Soybean Irrigation Initiation Timing Using Evapotranspiration And Soil Moisture Sensor Cues, Justin Chlapecka Nov 2015

Soybean Irrigation Initiation Timing Using Evapotranspiration And Soil Moisture Sensor Cues, Justin Chlapecka

Student Theses and Dissertations

Expanded use of irrigation management tools is needed to improve water use efficiency in eastern Arkansas soybean (Glycine max) production. Irrigation initiation timing was studied on a furrow irrigated, sandy loam commercial field in Mississippi County. A major objective was to develop, validate, and expand use of irrigation timing cues, incorporating local weather station data, atmometers, and soil moisture measurements. Four timing treatments were evaluated: early, recommended, late, and rainfed, and the crop and pest response to each. With above average rainfall in 2014 and 2015, results showed little variation of soybean yield. Yield monitor data indicated penalties only within …

Engineering Novel Glycopeptides As A Molecular Carrier For Thermostable E1 Endoglucanase Expressed In Planta, Hong Fang

Student Theses and Dissertations

Reengineering plant cell walls with cell wall-modifying enzymes (CWMEs) represents a promising solution to overcome the recalcitrance for lignocellulosic biomass efficient conversion to liquid biofuels. This project investigated innovative application of hydroxyproline (Hyp)-O-glycosylation to engineer novel glycopeptide tags for heterologous CWMEs to maximize their hydrolytic functions in cell walls without disrupting composition and architecture. I addressed the hypothesis that a strategically designed glycopeptide tag fused to thermostable E1 endoglucanase and expressed in tobacco will facilitate the enzyme deposition into cell wall matrix to make the resulting biomass more subject to biochemical saccharification, while not adversely affecting the normal growth of …

Despite Buffers, Experimental Forest Clearcuts Impact Amphibian Body Size And Biomass, Jessica S. Veysey Powell, Kimberly J. Babbitt Nov 2015

Despite Buffers, Experimental Forest Clearcuts Impact Amphibian Body Size And Biomass, Jessica S. Veysey Powell, Kimberly J. Babbitt

New Hampshire Agricultural Experiment Station Publications

Forest buffers are a primary tool used to protect wetland-dependent wildlife. Though implemented widely, buffer efficacy is untested for most amphibian species. Consequently, it remains unclear whether buffers are sufficient for maintaining amphibian populations and if so, how wide buffers should be. We present evidence from a six-year, landscape-scale experiment testing the impacts of clearcutting, buffer width, and hydroperiod on body size and condition and biomass of breeding adults for two amphibian species at 11 vernal pools in the northeastern United States. We randomly assigned treatments (i.e., reference, 100m buffer, 30m buffer) across pools, clearcut to create buffers, and captured …

Biology Faculty Publications and Presentations

Mutations in WNK1 and WNK4, and in components of the Cullin-Ring Ligase system, kelch-like 3 (KLHL3) and Cullin 3 (CUL3), can cause the rare hereditary disease, Familial Hyperkalemic Hypertension (FHHt). The disease is characterized by overactivity of the renal sodium chloride cotransporter (NCC), which is phosphorylated and activated by the WNK-stimulated Ste20-type kinases, SPAK and OSR1. WNK kinases themselves can be targeted for ubiquitination and degradataion by the CUL3-KLHL3 E3 ubiquitin ligase complex. It is unclear, however, why there are significant differences in phenotypic severity among FHHt patients with mutations in different genes. It was reported that kelch-like 2 (KLHL2), …

Resource Recovery Through Halophyte Production In Marine Aquaponics: An Evaluation Of The Nutrient Cycling And The Environmental Sustainability Of Aquaponics, Suzanne Boxman

USF Tampa Graduate Theses and Dissertations

Aquaculture, the farming of aquatic animals and plants, is an important component of global food production, which supplies a nutritious protein source for millions of people. Interest in improving the sustainability of aquaculture has led to the development of aquaponics in which fish production is combined with plant production to create zero-discharge systems. A need for more fundamental science and engineering research on marine aquaculture and growing interest in production of halophytes motivated this novel research on marine aquaponics. One objective was to evaluate the growth and nutrient removal capacity of halophytes in marine aquaponics. Bench-scale studies were conducted to …

Synthesis Of Agents For The Treatment And Analysis Of Tropical Diseases, Jeanine Yacoub Nov 2015

Synthesis Of Agents For The Treatment And Analysis Of Tropical Diseases, Jeanine Yacoub

USF Tampa Graduate Theses and Dissertations

Toxoplasmosis is an opportunistic disease caused by the protozoan parasite Toxoplasma gondii. The parasite is usually staved off by a healthy immune system and remains dormant in the body. In immunocompromised patients, the parasite can become active and spread throughout the body causing symptoms such as encephalitis, cognitive disorders, seizures, and death. Combination drug therapy is the usual treatment for toxoplasmosis; however, patients suffer from problems of intolerance, allergic reactions, and cytotoxicity. In an effort to identify new drug targets for toxoplasmosis, a series of compounds have been synthesized that can be used as tools to probe the unique …

Eosinophilic Pneumonia In A Patient With Bronchial Myiasis : Case Report And Literature Review, Arindom Aich, Suad Al-Ismaili, Fatma A. Al-Wardi, Talal Hm, Quasem Al-Salmi, Hilal Al-Hashami

Sultan Qaboos University Medical Journal

Pulmonary myiasis is an unusual form of myiasis in humans and has been recently identified as a cause of eosinophilic pneumonia. We report the case of a 13-year-old Omani boy who presented to the Royal Hospital, Muscat, Oman, in October 2014 with respiratory distress. Bronchial aspirates revealed features of eosinophilic pneumonia. Possible larvae identified in the cytology report, a high immunoglobulin E level and the patient history all indicated bronchial myiasis. The patient was treated with steroids and ventilation and has since been diseasefree with no long-term side-effects. To the best of the authors’ knowledge, this is the first case …

The Impact Of Pedagogical Agents' Conversational Formality On Learning And Learner Impressions, Haiying Li Nov 2015

The Impact Of Pedagogical Agents' Conversational Formality On Learning And Learner Impressions, Haiying Li

Electronic Theses and Dissertations Archive

The dissertation examined the impact of agents' conversational formality on the learning of summarization skills and learner impressions in an intelligent tutoring system (ITS) with trialogues (a teacher agent, a student agent, and a human learner). Formal language is in the informational genre (as opposed to narrative) and increases with word abstractness, syntactic complexity, referential cohesion, and deep cohesion. At the other end of the continuum, informal discourse tends to have concrete words, simple syntax, low cohesion (because knowledge-based inferences can fill the gaps), and high narrativity. There have been no experimental investigations that have maniputed teacher language because it …

Molecular Role Of Dab2ip Gtpase Activating Protein In Brain Development, Farimah Salami Nov 2015

Molecular Role Of Dab2ip Gtpase Activating Protein In Brain Development, Farimah Salami

Electronic Theses and Dissertations Archive

Dab2ip is the member of Ras GTPase activating protein family that interacts with Disabled-1/2 famlily proteins, which play a critical role in brain development. Dab2ip is mainly known as a tumor suppressor, however recent studies have shown that it has important functions in the mammalian brain. In this study, using bioinformatics and experimental techniques, I characterized the mouse Dab2ip gene structure. Unlike previous reports, I found that the mouse Dab2ip gene has 20 exons and 3 CpG islands. I identified three transcript variants of Dab2ip which starts from exon one. Alternative splicing at the 5' end of these transcripts result …
