Characterization Of Suf Pathway For Fe-S Cluster Assembly In Escherichia Coli, Yuyuan Dai
Characterization Of Suf Pathway For Fe-S Cluster Assembly In Escherichia Coli, Yuyuan Dai
Theses and Dissertations
Fe-S clusters are critical metallocofactors required for cell function. Because of the toxicity of ferrous iron and sulfide to the cell, in vivo Fe-S cluster assembly is carried out by multiprotein biosynthetic pathways. Escherichia coli contains a stress-responsive Fe-S cluster assembly system, the SufABCDSE pathway, working under iron starvation and oxidative stress conditions. The cysteine desulfurase SufS and its accessory protein SufE work together to mobilize persulfide from L-cysteine. We collaborated with Dr. Laura S. Busenlehner to use hydrogen/deuterium exchange mass spectrometry (HDX-MS) to characterize SufS-SufE interactions and protein dynamics. HDX-MS analysis shows that SufE binds near the SufS active …
High Order Statistics Of Natural And Manmade Sounds, Rahul Narayan
High Order Statistics Of Natural And Manmade Sounds, Rahul Narayan
Master's Theses
Environmental sounds, both man-made and natural, vary on multiple time and frequency scales generating a large range of temporal, spectral and amplitude modulations that are evident in the high-order statistics of the sound spectrogram. Healthy hearing humans perceive high-order statistical regularities and use this information to categorize and discriminate sounds. This paper tests the hypothesis that biologically motivated sound statistics can enable/enhance discrimination and identification of sound categories from a computational standpoint. A large catalogue of natural and man-made sounds and their associated high-order contrast and intensity statistics were developed, and the information carrying content of each statistic for sound …
Suzuki-Miyaura Reactions Are Used To Synthesize Z-Alkenes From Cyclic Boronic Half-Acids, Daniel Lee French
Suzuki-Miyaura Reactions Are Used To Synthesize Z-Alkenes From Cyclic Boronic Half-Acids, Daniel Lee French
Undergraduate Honors Thesis Collection
Cis-Alkenes provide the basic structure and reactivity for a variety of compounds, from the common chemicals found in any organic laboratory to advanced drugs used to treat cancer or suppress the immune system (Littke & Fu, 2002). Synthetically, alkenes are also of use in that they serve as a site for further chemistry. Historically, Z-alkenes have been more difficult to synthesize than their E-counterparts (McNulty et al., 2010; Siau, Zhang, & Zhao, 2012). Furthermore, in many reactions, a mixture of E- and Z- conformations of the alkene is produced. Such differences may seem trivial upon first inspection, but because there …

The concurrence of tomato yellow leaf curl virus (TYLCV) with the spread of its vector Bemisia tabaci Q rather than B in China suggests a more mutualistic relationship between TYLCV and Q. Here, we investigated the hypothesis that viruliferous B and Q have different effects on plant defenses. We found the fecundity of nonviruliferous B, nonviruliferous Q, viruliferous Q and viruliferous B was 11.080, 12.060, 10.760, and 11.220 respectively on plants previously attacked by the other biotype, however, on their respective noninfested control leaves fecundity was 12.000, 10.880, 9.760, and 8.020 respectively. Only viruliferous B had higher fecundity on viruliferous …

A Thermal Window For Yawning In Humans: Yawning As A Brain Cooling Mechanism, Jorg J. M. Massen, Kim Dusch, Omar T. Eldakar, Andrew C. Gallup
Biology Faculty Articles
The thermoregulatory theory of yawning posits that yawns function to cool the brain in part due to counter-current heat exchange with the deep inhalation of ambient air. Consequently, yawning should be constrained to an optimal thermal zone or range of temperature, i.e., a thermal window, in which we should expect a lower frequency at extreme temperatures. Previous research shows that yawn frequency diminishes as ambient temperatures rise and approach body temperature, but a lower bound to the thermal window has not been demonstrated. To test this, a total of 120 pedestrians were sampled for susceptibly to self-reported yawn contagion during …
Mechanism Of Insulin Aggregation: Applied To Alzheimer's Disease, Milos Atz
Mechanism Of Insulin Aggregation: Applied To Alzheimer's Disease, Milos Atz
Honors Scholar Theses
Alzheimer’s disease, a debilitating neurodegenerative illness, is caused by the irreversible aggregation of beta-amyloid proteins in the brain. In Alzheimer’s brains, the protein can become disfigured, causing it to aggregate into long, insoluble fibers that deposit on brain tissue. Studying the aggregation mechanisms of amyloid proteins can lead to a deeper understanding of the progression of Alzheimer’s disease and possibly point towards a potential cure or treatment for the disease. Heat induced aggregation of insulin provides a model system to study the aggregation of amyloidogenic proteins. This study investigates the early stages of heat induced insulin aggregation using dynamic light …

Tidal Energy Conversion In A Global Hot Spot: On The 3-D Dynamics Of Baroclinic Tides At The Celtic Sea Shelf Break, Vasiliy Vlasenko, Nataliya Stashchuk, Mark E. Inall, Joanne E. Hopkins
School of Biological and Marine Sciences
Globally, the Celtic Sea shelf break is ranked highest as an energetic "hot spot" of tidal energy conversion, therefore making it the most significant contributor to global internal tidal energy flux. In this paper, the three-dimensional dynamics of baroclinic tides in the shelf-slope area of the Celtic Sea was investigated numerically and using observational data collected on the 376th cruise of the RV "RRS Discovery" in June 2012. The time series recorded at a shelf break mooring showed that semidiurnal internal waves were accompanied by packets of internal solitary waves with maximum amplitudes up to 105 m, the largest internal …

Is Retention Enough? Learning And Satisfaction Of First-Generation College Seniors, David Mahan, Kristin B. Wilson, Joseph M. Petrosko Jr., Michael R. Luthy
Kentucky Journal of Higher Education Policy and Practice
Progress has been made in retaining and graduating traditional-age first-generation college students at four-year institutions. First-generation students, however, often experience college differently because of external factors, which can negatively influence their learning experience and overall satisfaction. This study explored learning and satisfaction measures of seniors at a small private university in the Midwest. Using Astin’s I-E-O model (1970), the following variables were considered: precollege student characteristics (input); academic engagement, co-curricular activities, campus relationships (environment); and satisfaction, learning (outcome). The sample consisted of seniors participating in NSSE and/or ETS MAPP (n=342). Findings confirmed that first-generation students typically worked more for pay, …

Bacterial Migration Through Low-Permeability Fault Zones In Compartmentalised Aquifer Systems: A Case Study In Southern Italy, Antonio Bucci, Emma Petrella, Gino Naclerio, Sabrina Gambatese, Fulvio Celico
International Journal of Speleology
The aim of this study was to experimentally verify the significance of microbial transport through low-permeability fault zones in a compartmentalised carbonate aquifer system in Southern Italy.
The temporal variability of microbial communities in two springs fed by the same aquifer system, but discharging up- and down-gradient of two low-permeability fault zones, was analysed using a 16S rDNA polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE)-based approach. At both springs, a remarkable temporal variation in PCR-DGGE profiles was detected throughout the observation period. When comparing the PCR-DGGE profiles of the two springs, a synchronous evolution over time was observed. Moreover, the …
Beyond The Canonical Function Of Receptor Tyrosine Kinase: New Perspectives For Erbb2 (Her2) Signaling Control Across The Cell Membrane, Qian Zhang
Open Access Dissertations
The complex of ligand-binding deficient ERBB2 with kinase-dead ERBB3 represents the most potent signaling partner among the ERBB receptor family. The canonical model of receptor activation only provides an explanation for ERBB3 phosphorylation, leaving the ERBB2 part unresolved.Traditional therapeutic agents including monoclonal antibodies or kinase inhibitors against ERBB2 are effective in treatment, but their mechanism of action remains unknown. Meanwhile, patients often develop high levels of drug resistance over time. Since the process of ERBB2 signaling activation and transduction is elusive, we are interested in looking into the underlying molecular mechanisms, and thereby developing new medications in an effort to …

Effects Of Eph/Ephrin Mutations On Pre Pulse Inhibition In Mice, Andrea Marie Liuzzo
Effects Of Eph/Ephrin Mutations On Pre Pulse Inhibition In Mice, Andrea Marie Liuzzo
Dissertations, 2014-2019
The acoustic startle response (ASR) is a reliable reflexive behavioral response in mammals elicited by an unexpected intense acoustic startle eliciting stimulus (SES). It is mediated by a sub-cortical pathway that includes the inferior colliculus (IC). The ASR amplitude can be measured with an accelerometer beneath the subject attached to the cage, and can be decreased in amplitude by presenting a less intense, non-startling stimulus 20-300 ms before the SES. This reflexive decrement in ASR is called pre pulse inhibition (PPI) and indicates that the relatively soft pre pulse was heard. Murine species have been used to study this response …
The Neural Representation And Behavioral Detection Of Frequency Modulation, Daniel Elliott Shearer
The Neural Representation And Behavioral Detection Of Frequency Modulation, Daniel Elliott Shearer
Dissertations, 2014-2019
Understanding a speech signal is reliant on the ability of the auditory system to accurately encode rapidly changing spectral and temporal cues over time. Evidence from behavioral studies in humans suggests that relatively poor temporal fine structure (TFS) encoding ability is correlated with poorer performance on speech understanding tasks in quiet and in noise. Electroencephalography, including measurement of the frequency-following response, has been used to assess the human central auditory nervous system’s ability to encode temporal patterns in steady-state and dynamic tonal stimuli and short syllables. To date, the FFR has been used to investigate the accuracy of phase-locked auditory …
Exploration Of Genomic Determinants For Host Range In Phages Of Pathogenic And Non-Pathogenic Mycobacteria, Anisa Christine Tracy
Exploration Of Genomic Determinants For Host Range In Phages Of Pathogenic And Non-Pathogenic Mycobacteria, Anisa Christine Tracy
Masters Theses, 2010-2019
The genus Mycobacterium is diverse, ranging from slow-growing human pathogens such as M. tuberculosis to fast-growing saprophytes such as M. smegmatis. M. ulcerans is the causative agent of the neglected disease known as Buruli ulcer, an emerging pathogen first described in Buruli County, Uganda. Antibiotic resistance among Mycobacterial pathogens is growing at an alarming rate and is creating an urgent need for novel diagnostic and therapeutic approaches. Elucidating the details of virus/host interactions among the mycobacteria and their phages thus has the potential to affect how we think about the role of these bacteria in both clinical and environmental settings. …
The Influence Of A Cyp1a2 Polymorphism On The Hemostatic Response To Caffeine, Joseph James Aiello
The Influence Of A Cyp1a2 Polymorphism On The Hemostatic Response To Caffeine, Joseph James Aiello
Masters Theses, 2010-2019
Research has continued to provide conflicting results of caffeine’s effect on acute coronary events; although it is now apparent that a single nucleotide polymorphism at intron 1 of the cytochrome P450 (CYP1A2) gene may explain these equivocal findings. Despite the fact that markers of coagulation potential and fibrinolytic potential are associated with risk for myocardial infarction, the extent to which caffeine affects these markers is unknown. The purposes of this study were to determine the acute effects of caffeine on coagulation and fibrinolytic potential at rest and following maximal exercise and to determine if hemostatic responses engendered by caffeine are …

Motor Axon Synapses On Renshaw Cells Contain Higher Levels Of Aspartate Than Glutamate, Dannette Shanon Richards, Ronald W. Griffith, Shannon H. Romer, Francisco J. Alvarez
Neuroscience, Cell Biology & Physiology Faculty Publications
Motoneuron synapses on spinal cord interneurons known as Renshaw cells activate nicotinic, AMPA and NMDA receptors consistent with co-release of acetylcholine and excitatory amino acids (EAA). However, whether these synapses express vesicular glutamate transporters (VGLUTs) capable of accumulating glutamate into synaptic vesicles is controversial. An alternative possibility is that these synapses release other EAAs, like aspartate, not dependent on VGLUTs. To clarify the exact EAA concentrated at motor axon synapses we performed a quantitative postembedding colloidal gold immunoelectron analysis for aspartate and glutamate on motor axon synapses (identified by immunoreactivity to the vesicular acetylcholine transporter; VAChT) contacting calbindin-immunoreactive (-IR) Renshaw …

Individuals that were born in a foreign country, have a bachelor’s degree or higher from a foreign post-secondary education institution, and are not working at their full level of expertise, are referred to more concisely as “internationally trained professionals” (ITPs). Social workers have called becoming informed about immigrants’ integration needs a new frontier of social service and professional development. All fields of human services will benefit from any data resulting from descriptive exploration of the ITP integration issue. The bounded system that serves for this case study is the current limited response to the ITP issue seen in Virginia, USA. …

The Science Of Charity: The Social Network That Restructured Law And Order In Baltimore, 1881-1901, Hope Elizabeth Byers
Masters Theses, 2010-2019
Social networks undeniably build more cohesive ideologies between its members. Historians can better understand the path that urban reform took at the end of the nineteenth by focusing on the social networks that participated in many different reform efforts. In Baltimore, a group of elite businessmen began a variety of association and societies to aid the poor in their midst. The Charity Organization Society best combined this group of men. The Charity Organization Society of Baltimore sought to uplift the poor through advice rather than monetary aid. The group’s campaign to remove alms distribution from the police department in the …
