Multisite Phosphorylation Of The Sum1 Transcriptional Repressor By S-Phase Kinases Controls Exit From Meiotic Prophase In Yeast., Daniel Corbi, Sham Sunder, Michael Weinreich, Aikaterini Skokotas, Erica S Johnson, Edward Winter

Activation of the meiotic transcription factor Ndt80 is a key regulatory transition in the life cycle of Saccharomyces cerevisiae because it triggers exit from pachytene and entry into meiosis. The NDT80 promoter is held inactive by a complex containing the DNA-binding protein Sum1 and the histone deacetylase Hst1. Meiosis-specific phosphorylation of Sum1 by the protein kinases Cdk1, Ime2, and Cdc7 is required for NDT80 expression. Here, we show that the S-phase-promoting cyclin Clb5 activates Cdk1 to phosphorylate most, and perhaps all, of the 11 minimal cyclin-dependent kinase (CDK) phospho-consensus sites (S/T-P) in Sum1. For the purposes of forest restoration, carbon (C) fixation, and economic improvement, eucalyptus (Eucalyptus urophylla) has been widely planted in South China. The understory of eucalyptus plantations is often occupied by a dense community of the fern Dicranopteris dichotoma, which intercepts tree canopy leaf litter before it reaches the ground. To understand the effects of this interception of litterfall on C cycling in eucalyptus plantations, we quantified the mass of intercepted litter and the influences of litterfall interception on litter decomposition and soil respiration. Many toxic molds synthesize and release an array of poisons, termed mycotoxins that have an enormous impact on human health, agriculture and economy [1]. These molds contaminate our buildings, indoor air and crops, cause life threatening human and animal diseases and reduce agricultural output [2]. In order to design appropriate approaches to minimize the detrimental effects of these fungi, it is essential to develop diagnostic methodologies that can rapidly and accurately determine based on fungal strains and their growth patterns, the extent of mycotoxin mediated damage caused to the environment.Here we developed a novel multi-scale predictive mathematical model that could …

Implementing a traceability plan in an herbal production plant is a complex task. In this study an attempt to integrate food safety, quality and traceability through the different stages of the organic German Chamomile (Matricaria recutita) production chain was made. First, all the current relevant international laws, regulations and standards regarding traceability, safety and quality, with emphasis on how and why they are necessary for any food production chain, were set forward. Teaching To The Test: A Pragmatic Approach To Teaching Logic, Seth C. Vannatta

The proper goal of an introductory logic course, teaching critical thinking, is best achieved by maintaining the principle of continuity between student experiences and the curriculum. To demonstrate this I explain Dewey’s naturalistic approach to logic and the process of inquiry, one which presents the elements of traditional logic in the context of student experiences. I offer an example of a logic textbook which models the maintenance of the principle of continuity I advocate. Last, I advocate a pluralistic and experimental approach to accomplish this, including methods that rely on the role of the body in learning and reasoning.

A major use of the 1000 Genomes Project (1000GP) data is genotype imputation in genome-wide association studies (GWAS). Here we develop a method to estimate haplotypes from low-coverage sequencing data that can take advantage of single-nucleotide polymorphism (SNP) microarray genotypes on the same samples. First the SNP array data are phased to build a backbone (or 'scaffold') of haplotypes across each chromosome. We then phase the sequence data 'onto' this haplotype scaffold. This approach can take advantage of relatedness between sequenced and non-sequenced samples to improve accuracy. We use this method to create a new 1000GP haplotype reference set for …

Objective: To identify the risk-predictive baseline profile patterns of demographic, genetic, immunologic, and metabolic markers and synthesize these patterns for risk prediction.
Research Design and Methods: RuleFit is used to identify the risk-predictive baseline profile patterns of demographic, immunologic, and metabolic markers, using 356 subjects who were randomized into the control arm of the prospective Diabetes Prevention Trial-Type 1 (DPT-1) study. A novel latent trait model is developed to synthesize these baseline profile patterns for disease risk prediction. The primary outcome was Type 1 Diabetes (T1D) onset.

Protein Kinase R (PKR) is a key component of the innate immune antiviral response. PKR is activated upon binding to dsRNA. However, recent studies have shown that PKR can also bind to and become activated by duplex RNAs containing complex secondary structure. The mechanism of PKR binding and activation by these RNAs is currently not known. The approach taken here to determine the mechanism of PKR binding by these RNAs is through the development of PKR constructs that are capable of covalently binding to RNAs. Mitigation activities designed to supplement nutrient and organic matter inputs to streams experiencing decline or loss of Pacific salmon typically presuppose that an important pathway by which salmon nutrients are moved to fish (anadromous and/or resident) is via nutrient incorporation by biofilms and subsequent bottom-up stimulation of biofilm production, which is nutrient-limited in many ecosystems where salmon returns have declined. The active metabolite of vitamin A, retinoic acid, is vital to several physiological functions, and is most notably associated with vision and organ development Its role in the male reproductive system is well understood, but studies in female reproduction are lacking. Recent studies show that Cyp26b1, a retinoic acid degrading enzyme, is expressed highly in the Day1-10 mouse ovary. This time point correlates with establishment of the primordial follicle pool. Interest in research on heroism has begun to increase during the past decade, but, despite the ancient roots of heroism embedded deeply in cultures around the world, empirical work on the subject is relatively sparse. Direct and explicit empirical study of heroism in organizational contexts, specifically, is especially rare. The lack of organizational heroism research is surprising. There is a preponderance of evidence that organizational wrongdoing is observed across many organizations, sometimes to great extremes that are in violation of federal law, and it can have profound social impact. Spirocycles are a class of molecules consisting of a quaternary atom al the junction of two fused rings. This structural motif is found in a number of biologically active natural products and pharmaceuticals. Current strategies to synthesize these structures include classical methods such as alkylation and rearrangement, as well as more modern methods reported in the past decade which utilize electrophilic halogen reagents. Empowerment, a core value of community psychology, is defined as a process by which people, organizations, and communities gain mastery over issues of concern to them in their lives (Rappaport, 1987). In community psychology, empowerment is understood as a construct particularly and primarily salient for minority t,>roups who hold a marginalized position in society, as psychological empowerment is a product of an individual's interaction with his or her context. This dissertation argues that Bella Swan is a representation of Barbara Creed's monstrous-feminine which serves to reinforce ideologies that insist women are abject, inherently dangerous to men, and threatening to a patriarchal status quo. Through close-textual analysis of The Twilight Saga, I demonstrate how the monstrous-feminine frames the hysterical teenage body, hypersexuality, and eternal motherhood as simultaneously unacceptable and unavoidable. These negative women's stereotypes continue to persist in dominant popular culture, and this doublebind is overcome only by the impossible perfection of vampirism. The monstrous-feminine invites constructions of teenage bodies as unstable and unreliable, women's sexuality as dangerous and …

The use of digital technologies and social media by people with serious illness to find, share, and create health information is much celebrated but rarely critiqued. Proponents laud “Health 2.0” as transforming health care practice and empowering patients. Critics, however, argue that a discourse of developing lay expertise online masks the disciplinary practices of the neoliberal state’s emphasis on individual responsibility. Understanding the relationships between human health, water, sanitation, and environmental health is a requirement to understanding the challenges that face researchers when it comes to addressing global health relating to water and sanitation. Access to improved water and sanitation is not only a precondition to health, but to all aspects of daily living. Target 7.C of the Millennium Development Goals (MDGs) addresses worldwide disparities in access to improved water and sanitation by calling for the reduction in "half of the proportion of people without sustainable access to safe drinking water and sanitation by 2015".
