Family-Specific, Novel, Deleterious Germline Variants Provide A Rich Resource To Identify Genetic Predispositions For Brcax Familial Breast Cancer., Hongxiu Wen, Yeong C. Kim, Carrie Snyder, Fengxia Xiao, Elizabeth A. Fleissner, Dina Becirovic, Jiangtao Luo, Bradley Downs, Simon Sherman, Kenneth Cowan, Henry T. Lynch, San Ming Wang

Journal Articles: Eppley Institute

BACKGROUND: Genetic predisposition is the primary risk factor for familial breast cancer. For the majority of familial breast cancer, however, the genetic predispositions remain unknown. All newly identified predispositions occur rarely in disease population, and the unknown genetic predispositions are estimated to reach up to total thousands. Family unit is the basic structure of genetics. Because it is an autosomal dominant disease, individuals with a history of familial breast cancer must carry the same genetic predisposition across generations. Therefore, focusing on the cases in lineages of familial breast cancer, rather than pooled cases in disease population, is expected to provide …

Evolution And Equilibration Of Artificial Morphologic Perturbations In The Form Of Nearshore Berm Nourishments Along The Florida Gulf Coast, Katherine Emily Brutsché Jun 2014

Evolution And Equilibration Of Artificial Morphologic Perturbations In The Form Of Nearshore Berm Nourishments Along The Florida Gulf Coast, Katherine Emily Brutsché

USF Tampa Graduate Theses and Dissertations

Inlets and channels are dredged often to maintain navigation safety. It is beneficial to reintroduce the dredged material back into the littoral system, in the form of beach or nearshore nourishments. Nourishment in the nearshore is becoming an increasingly utilized method, particularly for dredged material that contains more fine sediment than the native beach. This research examines the morphologic evolution of two different nearshore nourishments. A nearshore berm was constructed at Fort Myers Beach, Florida using mixed-sized sediment dredged from a nearby channel. The nearshore berm was placed in water depths between 1.2 and 2.4 m with the berm crest …

Manipulating The Tumor Microenvironment For Therapeutic Benefit, Kate M. Bailey Jun 2014

Manipulating The Tumor Microenvironment For Therapeutic Benefit, Kate M. Bailey

USF Tampa Graduate Theses and Dissertations

The physical tumor microenvironment contributes significantly to carcinogenesis, cancer progression and metastatic dissemination. Two main components of the tumor microenvironment, hypoxia and acidosis, are present in nearly every solid tumor and act as powerful selection forces against the tumor. Hypoxia and acidosis promote tumor heterogeneity and contribute to chemotherapy and radiotherapy resistance. This dissertation interrogates methods to target the tumor microenvironment including two novel studies describing mechanisms of buffer therapy resistance and targeting tumor hypoxia with vasodilators to enhance the efficacy of a hypoxia activated prodrug, TH-302.

In the first study, mechanisms of buffer therapy resistance were identified and detailed. …

Just Love: A Collaborative Evaluation Of A Faith-Based School-Family-Community Partnership Through The Voices Of The Children, Lynette M. Henry

USF Tampa Graduate Theses and Dissertations

Abstract

Faith-based school-family-community partnerships have been a federal mandate over the past decade, yet little has been written about the outcomes of these faith-based partnerships. A need exists to understand if the potential in these faith-based partnerships is indeed realized in positive outcomes for students and schools. The purpose of this study was to evaluate a faith-based school-family-community partnership, Just Love. Just Love is a faith-based school-family-community partnership between a large suburban church and a Title 1, urban elementary school, Charisma Elementary School (Charisma ES; pseudonym). It was implemented in what is considered a "failing school". Just Love's purpose was …

Cancer And Infection, Kathleen Hope Plummer Jun 2014

Cancer And Infection, Kathleen Hope Plummer

USF Tampa Graduate Theses and Dissertations

E. coli is the most frequently isolated Gram negative pathogen from bacteremia in cancer patients and is repeatedly recovered from many other extraintestinal illnesses. These infections are commonly endogenous in nature and interfere with the treatment of cancer resulting in increased healthcare costs, morbidity, and mortality rates. Cancer and the treatments related to cancer cause alterations in the microbiome of the gut and other organs. Despite this point, there is a serious lack of knowledge about the genetic types of E. coli infecting cancer patients. This gap results in vague prevention strategies and limited treatment options for cancer patients. Multi …

The Development And Initial Validation Of An Instrument Measuring The Cognitive Domain Of Intercultural Maturity, Melanie Lynne Wicinski Jun 2014

The Development And Initial Validation Of An Instrument Measuring The Cognitive Domain Of Intercultural Maturity, Melanie Lynne Wicinski

USF Tampa Graduate Theses and Dissertations

Abstract

Through a two-phase process an instrument was created to measure the cognitive domain as proposed by King and Baxter Magolda in their Developmental Model of Intercultural Maturity (2005). The first phase involved expert panels who identified the competencies which exist in the cognitive domain, identified situations which might exist between individuals from different cultures, validated scenarios created from the identified situations, and created responses which corresponded to the three developmental levels (Initial, Intermediate, and Mature) defined in the Developmental Theory of Intercultural Maturity.

Within the second phase, the created instrument was administered to 371 individuals representing eight geocultural world …

Crohn’S Disease Linked Polymorphisms Associated With Autophagy Contribute To Th17 Cell Induction Through Increasing The Expression Of Il-1Β And Tnf-Α, Daniel Clayton Morse Jun 2014

Crohn’S Disease Linked Polymorphisms Associated With Autophagy Contribute To Th17 Cell Induction Through Increasing The Expression Of Il-1Β And Tnf-Α, Daniel Clayton Morse

Pursuit - The Journal of Undergraduate Research at The University of Tennessee

Genome wide association studies (GWAS) have linked polymorphisms in autophagy genes to Crohn’s Disease (CD). Interestingly, recent studies have shown that defective autophagy leads to increased levels of the cytokines IL-1b and TNF-a, which play a critical role in Th17 cell differentiation. This is significant because CD is marked by a Th17 cell mediated inflammatory response. This article examines the manners in which defective expression of autophagy linked proteins, can indirectly amplify and sustain Th17 cell induction through increasing the production of Th17 positively differentiating cytokines (IL-1b and TNF-a) and decreasing the production of a Th17 down regulating cytokine, IL-10.

Mathematically Modeling Fetal Electrocardiograms, Samuel Estes, Kiersten Utsey, Erick Kalobwe Jun 2014

Mathematically Modeling Fetal Electrocardiograms, Samuel Estes, Kiersten Utsey, Erick Kalobwe

Pursuit - The Journal of Undergraduate Research at The University of Tennessee

Abstract

Some of the most common and fatal birth defects are those related to the heart. In adults, possible heart conditions are often identified through the use of an electrocardiogram (ECG). However, due to the presence of other signals and noise in the recording, fetal electrocardiography has not yet proven effective in diagnosing these defects. This paper develops a mathematical model of three-dimensional heart vector trajectories, which was used to generate synthetic maternal and fetal ECG signals. The dipole model is a useful simplification in which the electrical activity of the heart is viewed as a single time-varying vector originating …

Fty720 (Fingolimod) Provides Insight Into The Molecular Mechanisms Of Multiple Sclerosis, Madelyn Elizabeth Crawford

Pursuit - The Journal of Undergraduate Research at The University of Tennessee

Multiple sclerosis (MS) is a neurodegenerative disorder caused by a prolonged immune- mediated inflammatory response that targets myelin. Nearly all of the drugs approved for the treatment of MS are general immunosuppressants or only function in symptom management. The oral medication fingolimod, however, is reported to have direct therapeutic effects on cells of the central nervous system in addition to immunomodulatory functions. Fingolimod is known to interact with sphingosine-1-phosphate (S1P) receptors, and the most widely- accepted theory for its mechanism of action is functional antagonism of the receptor. This review examines significant neuromodulatory effects achieved by functional antagonism of the …

Cognitive Mapping And The Adventures Of The Postmodern, Kellner Douglas Jun 2014

Cognitive Mapping And The Adventures Of The Postmodern, Kellner Douglas

Theoretical Studies in Literature and Art

During the 1980s and 1990s, debates about the differences between modernity and postmodernity as historical epochs; modernism and postmodernism in the arts; and wars between modern and postmodern theory defined key theoretical problematics in intellectualized spaces of the West, like the University. Fredric Jameson was one of the initiators of debates over the postmodern, and introduced the concept of cognitive mapping to describe theoretical accounts of the contemporary moment. I discuss conceptions of theory and mapping in the modern and postmodern debates and sort out appropriate uses of the discourse of the postmodern, and the usefulness of the postmodern turn …

Alcohol Drinking Patterns And Differences In Alcohol-Related Harm: A Population-Based Study Of The United States, D. Antai, G. B. Lopez, J. Antai, D. S. Anthony

Publications and Research

Alcohol use and associated alcohol-related harm (ARH) are a prevalent and important public health problem, with alcohol representing about 4% of the global burden of disease. A discussion of ARH secondary to alcohol consumption necessitates a consideration of the amount of alcohol consumed and the drinking pattern. This study examined the association between alcohol drinking patterns and self-reported ARH. Pearson chi-square test (χ2) and logistic regression analyses were used on data from the National Comorbidity Survey Replication (NCS-R). The NCS-R is a cross-sectional nationally representative sample. Data was obtained by face-to-face interviews from 9282 adults aged ≥18 years in the …

Makara Journal of Science

Papua New Guinea (PNG) mutation is a point mutation that occurs in noncoding region of alpha globin clusters. Polymorphism promotes an additional recognition site for transcription factor (GATA-1) which presumably downregulates alpha globin synthesis. The aim of this research is to detect PNG mutation in other populations in Indonesia, thus the results will be used for completing standard diagnoses in detecting alpha thalassemia mutation based on ethnic background. The method used in detecting PNG mutation was PCR-RFLP. Detection of 399 samples (MCH

Makara Journal of Science

Soil-transmitted helminth (STH) infections (Necator americanus (hookworm), Ancylostoma duodenale (hookworm), and Ascaris lumbricoides) can lead to anemia, malnutrition, and iron deficiency. Traditionally, STH infections have been diagnosed using microscopy to detect eggs in human fecal samples. However, there are several limitations of this method. The aim of this research was to detect the percentage of submicroscopic STH infections from human fecal samples (children, 5–18 years old) in Nangapanda, Ende, using the real-time polymerase chain reaction (PCR) method. The fecal samples were collected in two time periods, which were before and after treatment, using 400 mg of Albendazole. There were 242 …

Rna Detection Technology For Applications In Marine Science: Microbes To Fish, Robert Michael Ulrich Jun 2014

Rna Detection Technology For Applications In Marine Science: Microbes To Fish, Robert Michael Ulrich

USF Tampa Graduate Theses and Dissertations

The accurate identification of taxa from mixed assemblages using genetic analysis remains an important field of molecular biology research. The common principle behind the development of numerous documented genetic detection technologies is to exploit specific nucleotide sequences inherent to each taxon. This body of work focuses on practical applications of real-time nucleic acid sequence-based amplification (RT-NASBA) in marine science, and is presented in four case studies. Each study represents novel work in the genetic identification of respective taxa of interest using RT-NASBA. Two case studies documented the development of an assay targeting mitochondrial 16S rRNA to discern legally salable grouper …
