Retrotransposons (RTs) comprise approximately half of the human genome and contribute to chromatin structure, regulatory motifs, and protein-coding sequences. Since RT insertions can disrupt functional genetic elements as well as introduce new sequence motifs to a region, they have the potential to effect the function of genes that harbour insertions as well as those nearby. A detailed characterization of RT density in genes could help inform predictions of the functional consequence of de novo as well as polymorphic insertions. High temperature stress results in yield loss and alterations to seed composition during seed filling in oilseed rape (Brassica napus). However, the mechanism underlying this heat response is poorly understood. In this study, global transcription profiles of 20 d-old siliques of B. napus were analyzed after heat stress using a Brassica 95k EST microarray. The up-regulated genes included many HSF/HSP transcripts and other heat-related marker genes, such as ROF2, DREB2a, MBF1c and Hsa32, reflecting the conservation of key heat resistance factors among plants. In the broadest context, biological markers, or biomarkers, are molecules that characterize a biological system or process. In the setting of cardiovascular disease, a number of biomarkers have become an integral part of diagnostic and risk stratification strategies. In this review, we will discuss classic and emerging biomarkers of cardiovascular disease and the role of these biomarkers in the diagnosis and prognosis of elderly patients presenting with acute myocardial infarction.

This study examined the impact of several noise conditions on speech articulator movements during a sentence repetition task. Sixty participants in three age groups ranging from 20 to 70 repeated a sentence under five noise conditions. Lower lip movements during production of a target sentence were used to compute the spatiotemporal index (STI). It was hypothesized that STI would be lower (indicating greater stability) in the silent baseline condition. There were changes in speech production under several of the noise conditions. Understanding vocal fold mechanics is an integral part of voice research and synthetic vocal fold models are an essential tool in characterizing vocal fold mechanics. These models contain multiple layers with varying stiffness, much like human vocal folds. The purpose of this thesis is to improve the current models and modeling techniques, as well as investigate the impact of asymmetry on model vibration. A new design for an MRI-based model is detailed. This model has a more realistic geometry than the simplified models and mimics some of the vibratory characteristics observed in human vocal folds. Over the course of more than a century, the St. Petersburg Pier has become the visual symbol of the city. During that period, the pier has gone through three distinct iterations. First came a functional pier for cargo and passengers. In the 1920s, a grand Mediterranean Revival-style Million Dollar Pier was built. In 1967, it was torn down. In the 1970s, a strikingly modern Inverted Pyramid was built on the same spot. Today, the city seeks a new pier that would be “iconic.” However, the meaning of the term has not been made clear by city leaders. By studying the …

Seed morph, abiotic conditions and time of germination can affect plant fitness, but few studies have tested their combined effects on plasticity of plant life history traits. Thus, we tested the hypothesis that seed morph, germination season and watering regime influence phenotypic expression of post-germination life history traits in the diaspore-heteromorphic cold desert winter annual/spring ephemeral Diptychocarpus strictus. The two seed morphs were sown in watered and non-watered plots in late summer, and plants derived from them were watered or not-watered throughout the study. Phase variation in the Gram-negative human pathogen Vibrio vulnificus involves three colonial morphotypes- smooth opaque colonies due to production of capsular polysaccharide (CPS), smooth translucent colonies as the result of little or no CPS expression, and rugose colonies due to production of a separate extracellular polysaccharide (EPS), which greatly enhances biofilm formation. Previously, it was shown that the brp locus, which consists of nine genes arranged as an operon, is up-regulated in rugose strains in a c-di-GMP-dependent manner, and that plasmid insertions into the locus resulted in loss of rugosity and efficient biofilm production. Mitochondrial Ca(2+) controls numerous cell functions, such as energy metabolism, reactive oxygen species generation, spatiotemporal dynamics of Ca(2+) signaling, cell growth and death in various cell types including neurons. Mitochondrial Ca(2+) accumulation is mainly mediated by the mitochondrial Ca(2+) uniporter (MCU), but recent reports also indicate that mitochondrial Ca(2+)-influx mechanisms are regulated not only by MCU, but also by multiple channels/transporters. The modern European climatic regime is peculiar, due to its unitary winter but diverse summer climates and a pronounced Mediterranean climate in the south. However, little is known on its evolution in the deep time. Here we reconstruct the European summer climate conditions in the Tortonian (11.62-7.246 Ma) using plant fossil assemblages from 75 well-dated sites across Europe. Our results clearly show that the Tortonian Europe mainly had humid to subhumid summers and no arid climate has been conclusively detected, indicating that the summer-dry Mediterranean-type climate has not yet been established along most of the Mediterranean coast at least by …

Purpose: High sensitivity C-reactive protein (CRP) is a risk factor for cardiovascular disease based on finding in primarily non-Hispanic White populations. Obesity, another risk factor for cardiovascular disease, is higher in Blacks as compared to non-Hispanic Whites. The objective of this study was to assess the relationship between CRP, a marker of systemic inflammation, and obesity indicators by ethnicity, diabetes status and gender for two Black ethnicities.

Methods: Anthropometrics and venous blood were collected for African and Haitian Americans with and without type 2 diabetes in a cross-sectional study. A total of 434 participants; 190 African Americans, 244 Haitian Americans, …

Phreatic overgrowths on speleothems (POS) are one of many sea-level proxies available to Quaternary geologists in Mallorca; these carbonate encrustations form at the air-water interface in cave passages flooded with brackish water. POS are ideal for reconstruction of western Mediterranean sea level because they are widespread in Mallorca's caves, can be precisely dated by U-series methods, constrain sea-level stands to sub-meter elevation, and are well preserved and accessible in the subterranean environment.

The Walter Reed Institute of Army Research developed the antimalarial drug mefloquine then collaborated with Hoffman-La Roche to produce the drug under its brand name "Lariam," after Food and Drug Administration (FDA) approved licensure in 1989. For over twenty years, the Army used this pill as its "drug of choice" for soldiers deployed to endemic regions until 2009, and in 2013 the Food and Drug Administration warned that the drug's neurotoxic effects could be lasting, if not permanent. Experts and organizations involved in freshwater resources management have emphasized the importance of long-term urban resource planning and management that considers the tight coupling which exists between human - nature - technology systems. The resistance of contemporary urban growth efforts to consider resource carrying capacities and ecosystem requirements has led to costly unintended consequences including the deterioration of natural capital and their associated ecosystem services, and the degradation of water resource flows. We propose methodology for exact statistical tests of hypotheses for models of network dynamics. The methodology formulates Markovian exponential families, then uses sequential importance sampling to compute expectations within basins of attraction and within level sets of a sufficient statistic for an over-dispersion model. Comparisons of hypotheses can be done conditional on basins of attraction. Examples are presented.

Flicker electroretinograms are steady-state electroretinograms (ERGs) generated by high rate flash stimuli that produce overlapping periodic responses. When a flash stimulus is delivered at low rates, a transient response named flash ERG (FERG) representing the activation of neural structures within the outer retina is obtained. Although FERGs and flicker ERGs are used in the diagnosis of many retinal diseases, their waveform relationships have not been investigated in detail. A number of novel spiro-pyrrolidines/pyrrolizines derivatives were synthesized through [3+2]-cycloaddition of azomethine ylides with 3,5-bis[(E)-arylmethylidene]tetrahydro-4(1H)-pyridinones 2a–n. Azomethine ylides were generated in situ from the reaction of 1H-indole-2,3-dione (isatin, 3) with N-methylglycine (sarcosine), phenylglycine, or proline. All compounds (50 μM) were evaluated for their antiproliferative activity against human breast carcinoma (MDA-MB-231), leukemia lymphoblastic (CCRF-CEM), and ovarian carcinoma (SK-OV-3) cells. N-α-Phenyl substituted spiro-pyrrolidine derivatives (5a–n) showed higher antiproliferative activity in MDA-MB-231 than other cancer cell lines. Ovarian cancer, the deadliest of gynecologic cancers, is usually not diagnosed until advanced stages. Although carboplatin has been popular for treating ovarian cancer for decades, patients eventually develop resistance to this platinum-containing drug. Expression of neurogenic locus notch homolog 3 (Notch3) is associated with chemoresistance and poor overall survival in ovarian cancer patients. Overexpression of NICD3 (the constitutively active form of Notch3) in OVCA429 ovarian cancer cells (OVCA429/NICD3) renders them resistance to carboplatin treatment compared to OVCA429/pCEG cells expressing an empty vector. This research uses Bayesian analysis of fluorescence spectroscopy results to determine if wastewater from the Heber Valley Special Service District (HVSSD) lagoons in Midway, UT has seeped into the adjacent Provo River. This flow cannot be directly measured, but it is possible to use fluorescence spectroscopy to determine if there is seepage into the river.Fluorescence spectroscopy results of water samples obtained from HVSSD lagoons and from upstream and downstream in the Provo River were used to conduct this statistical analysis. The electrical properties of proteins in solution are important for their structure and function. Computational biophysics studies of proteins need accurate parameters to ensure that numerical simulations match physical reality. Past work in this eld has compared the electrical properties of proteins obtained from dielectric spectroscopy to numerical simulations of proteins in water with adjustment of pKa values to try to capture the inevitable changes in electrical conformation that will occur in a complex structure such as a folded protein. Context and Objective: Adipose tissue in insulin resistant subjects contains inflammatory cells and extracellular matrix components. This study examined adipose pathology of insulin resistant subjects who were treated with pioglitazone or fish oil.

Design, Setting and Participants: Adipose biopsies were examined from nine insulin resistant subjects before/after treatment with pioglitazone 45 mg/day for 12 weeks and also from 19 subjects who were treated with fish oil (1,860 mg EPA, 1,500 mg DHA daily). These studies were performed in a clinical research center setting.

Results: Pioglitazone treatment increased the cross-sectional area of adipocytes by 18% (p = 0.01), and also increased …
