Arsenic is the most toxic metalloid present in the natural environment in both organic and inorganic arsenic forms. Inorganic arsenic is often more hazardous than the organic form. Arsenite and arsenate compounds are the major inorganic forms which are toxic causing severe human health dysfunction including cancer. Excretion of arsenic from the system is found elusive. Trastuzumab prolongs survival in HER2 positive breast cancer patients. However, resistance remains a challenge. We have previously shown that ADAM17 plays a key role in maintaining HER2 phosphorylation during trastuzumab treatment. Beside ADAM17, ADAM10 is the other well characterized ADAM protease responsible for HER ligand shedding. Therefore, we studied the role of ADAM10 in relation to trastuzumab treatment and resistance in HER2 positive breast cancer. ADAM10 expression was assessed in HER2 positive breast cancer cell lines and xenograft mice treated with trastuzumab. College-aged women are at an increased risk for experiencing non-consensual sexual contact (i.e. sexual assault) relative to the general population. Women who endure a sexual assault are at an increased risk for developing Post-traumatic Stress Disorder (PTSD). Intoxication by drugs and/or alcohol during an assault is linked to victims experiencing less severe PTSD symptoms; however, little is known about how the victim-offender relationship relates to PTSD symptom severity. The current study sampled 125 university women on measures that assessed degree of sexual victimization and resulting PTSD symptoms. BACKGROUND: Although genome wide studies have associated single nucleotide polymorphisms (SNP)s near PICALM with Alzheimer's disease (AD), the mechanism underlying this association is unclear. PICALM is involved in clathrin-mediated endocytosis and modulates Aß clearance in vitro. Comparing allelic expression provides the means to detect cis-acting regulatory polymorphisms. Thus, we evaluated whether PICALM showed allele expression imbalance (AEI) and whether this imbalance was associated with the AD-associated polymorphism, rs3851179.
RESULTS: We measured PICALM allelic expression in 42 human brain samples by using next-generation sequencing. Overall, PICALM demonstrated equal allelic expression with no detectable influence by rs3851179. A single sample demonstrated …

In the last several years, bacterial gene regulation via small RNAs (sRNAs) has been recognized as an important mechanism controlling expression of essential proteins that are critical to bacterial growth and metabolism. Technologies such as RNA-seq are rapidly expanding the field of sRNAs and are enabling a global view of the “sRNAome” of several bacterial species. While numerous sRNAs have been identified in a variety of both Gram-negative and Gram-positive bacteria, only a very small number have been fully characterized in the human pathogen Neisseria gonorrhoeae, the etiological agent of the STD gonorrhea. Despite growing attention to the unintended consequences of paternal incarceration for children’s wellbeing across the life course, little is known about whether and how paternal incarceration is related to food insecurity among children, an especially acute and severe form of deprivation. In this article, I use data from the Fragile Families and Child Wellbeing Study, a cohort of children born to mostly unmarried mothers, to examine the relationship between paternal incarceration and three indicators of food insecurity among young children: current food insecurity (at age five), onset of food insecurity (between ages three and five), and exit from food insecurity …

Previously, we have reported the synthesis of a homochiral l-cyclic peptide [WR]5 and its use for delivery of anti-HIV drugs and biomolecules. A physical mixture of HAuCl4 and the peptide generated peptide-capped gold nanoparticles. Here, [WR]5 and [WR]5-AuNPs were tested for their efficiency to deliver a small interfering RNA molecule (siRNA) in human cervix adenocarcinoma (HeLa) cells. Flow cytometry investigation revealed that the intracellular uptake of a fluorescence-labeled non-targeting siRNA (200 nM) was enhanced in the presence of [WR]5 and [WR]5-AuNPs by 2- and 3.8-fold when compared with that of siRNA alone …

The analysis of introgression of genomic regions between divergent populations provides an excellent opportunity to determine the genetic basis of reproductive isolation during the early stages of speciation. However, hybridization and subsequent gene flow must be relatively common in order to localize individual loci that resist introgression. In this study, we used next-generation sequencing to study genome-wide patterns of genetic differentiation between two hybridizing subspecies of rabbits (Oryctolagus cuniculus algirus and O. c. cuniculus) that are known to undergo high rates of gene exchange. The notion that smoking is in direct relation to the character trait of impulsivity is widely agreed upon. This study examines the individual character traits of the UPPS-P impulsivity scale that comprise the construct of impulsivity as they relate to cigarette smoking. This study seeks to illustrate whether or not there is determination of smoking behavior as a result of high levels of one or more of these impulsive traits. Analysis will be evaluated through means of several different analyses of variance to determine if any relationships are present.

High Sensitive Detection Of Cd(Ii) And Pb(Ii) Based On Antimony-Film Covered Pencil Core Electrodes, Shan Chen, Zhao-Yan Zheng, Yi-Min Fang, Li-Qing Zheng, Jian-Jun Sun

In this paper, a pencil core was used as the substrate for an antimony-film electrode for simultaneous detection of Cd(II) and Pb(II) by square-wave anodic stripping voltammetry (SWASV). With the deposition time of 180 s, the low limit of detection (LOD) for the electrode was determined to be 0.075·g?L-1 for Cd(II), while 0.13·g·L-1 for Pb(II) with good reproducibility in low pH solutions (pH = 2). The results show a much lower LOD than that of bismuth-film pencil core electrode. Finally, this antimony-film electrode was successfully applied to determine Cd(II) and Pb(II) in tap water.

A label-free electrochemical sensing electrode for highly sensitive detection of adenosine was constructed based on the signal amplification of carbon nanotubes (CNTs). The change in the interfacial feature of the modified electrode was characterized by electrochemical impedance spectroscopy. Using [Ru(NH3)6]3+ as the signaling moiety, adenosine with concentrations as low as 0.027 nmol·L-1 can be selectively detected. Additionally, the fabrication of this present aptasensor was simple, time-saving and cost-effective. Compared with other reported aptasensors, the proposed aptasensor had advantages of excellent sensitivity, selectivity and simplicity, which plays a potential role in development of aptasensor.

We represent the protein structure of scTIM with a graph-theoretic model. We construct a hierarchical graph with three layers - a top level, a midlevel and a bottom level. The top level graph is a representation of the protein in which its vertices each represent a substructure of the protein. In turn, each substructure of the protein is represented by a graph whose vertices are amino acids. Finally, each amino acid is represented as a graph where the vertices are atoms. In this dissertation, I first established a novel non viral piggyBac transposon mediated transgenesis approach delivered by in utero electroporation for radial glia lineage tracing. Furthermore, I developed a novel central nervous system tumor model in rat using piggyBac IUE. By directing expression of oncogenic HRasV12/AKT in radial glia, astrocytes and oligodendrocytes, glioblastoma multiforme and anaplastic oligoastrocytoma were induced. Also addition of neurogenic bHLH transcription factors Neurogenin 2 and Neural differentiation 1 to GBM inducing HRasV12/AKT, malignant pediatric atyical teratoid rhabdoid tumor like tumor was induced. Increasingly, high-dimensional genomics data are becoming available for many organisms.Here, we develop OrthoClust for simultaneously clustering data across multiple species. OrthoClust is a computational framework that integrates the co-association networks of individual species by utilizing the orthology relationships of genes between species. It outputs optimized modules that are fundamentally cross-species, which can either be conserved or species-specific. We demonstrate the application of OrthoClust using the RNA-Seq expression profiles of Caenorhabditis elegans and Drosophila melanogaster from the modENCODE consortium. A potential application of cross-species modules is to infer putative analogous functions of uncharacterized elements like non-coding RNAs based on guilt-by-association.

Room temperature ionic liquid (RTIL) is used as a soft-template to organize a meso-macroporous material constructed by self-assembled giant gold nanoparticles which are capped by L-cysteine. First, L-cysteine capped gold nanoparticles can self-assembly to form nanowires and sub-micrometer spherical giant particles due to the static interaction and/or the condensation reaction between the carboxyl and amino groups at the outer terminal of the ligand. Second, the spherical assembled particles can form a quasi-solid gel when grinding with a hydrophobic RTIL, 1-octyl-3-metyllimidazolium hexafluorophosphate. Nucleic acid bioconjugates have gained widespread use in medicinal chemistry research programs aimed at fighting human malignancies such as cancer, diabetes, genetic and infectious diseases. Their popularity stems from their ability to accelerate the drug development process by conjugating chemical functionality that may improve the pharmacology of a bioactive nucleoside. Moreover, this strategy has been proven to be effective with small molecule nucleoside analogs and those derived from lengthy oligonucleotide sequences. Considering these fruitful applications, my research and this thesis aims to explore the synthesis, characterization and therapeutic potential of novel classes of nucleic acid bioconjugates.
