The Roles Of Mir-155 And Mir-15/16 In Natural Killer Cell Function And Maturation, Ryan Patrick Sullivan

Natural Killer (NK) cells are a crucial component of the immune system, with a role in both early host defense against pathogens and tumor immunosurveillance. NK cells develop in the bone marrow from a common lymphoid precursor, along with T and B cells, but undergo a separate education process. Additional maturation in the periphery makes NK cells fully competent to protect the host. NK cells protect the host via two primary effector pathways - direct cytotoxicity, and indirect cytokine/chemokine production (e.g. IFN-g), which shapes the subsequent adaptive immune response. The brain contains an enormous number of neurons with diverse gene expression, morphology, and connectivity. These neurons exhibit distinct activity in the course of behaviors. The study of neural coding of a specific behavior necessitates recording activity from multifarious neurons in the circuit.One appealing approach is to simultaneously image the activity of a very large neuronal population at cellular resolution. However, recording calcium signals from tens of thousands of neurons at one time is not trivial. The gold standard technique, two-photon laser-scanning microscopy, typically permits recording from hundreds of neurons. Immune surveillance requires efficient trafficking of leukocytes throughout the body. To achieve this, leukocytes have evolved to be highly migratory and responsive to environmental cues, which provide guidance for proper tissue distribution. The translation of external environmental cues to intracellular physical changes in leukocytes requires a cascade of receptors, signal transducers, and mechanical effectors. My doctoral research focused on using T-cells as a model to study the unique cellular process of how signal transducers interact with and regulate mechanical effectors in fast migrating immune cells. The last decade has seen the advent of brain computer interfaces able to extract precise motor intentions from cortical activity of human subjects. It is possible to convert captured motor intentions into movement through coordinated, artificially induced, neuromuscular stimulation using peripheral nerve interfaces. Our lab has developed and tested a new type of peripheral nerve electrode called the Macro-Sieve electrode which exhibits excellent chronic stability and recruitment selectivity. Work presented in this thesis uses computational modeling to study the interaction between Macro-Sieve electrodes and regenerated peripheral nerves. These are two data sets that were compiled during a UAF student research project, Landscape Ecology class 469/669 (eLearning). They represent a value-added data set and can easily be mapped in a Geographic Information System (GIS) etc. For rabies in Alaska, 237 confirmed cases were found of which 158 had complete information (year, coordinates and vector). The rabies cases in this database are from 1914 til 2013; vectors include Dog, Wolf, Red Fox, Coyote, Arctic Fox, Cat, Caribou, Little Brown Bat, Keen's Myotic Bat and Wolverine. The Alaskan trichinosis data cover 1976-2012 and with various details. Species covered are: Walrus …

Objective To screen different solvent extracts of Elaeagnus kologa (E. kologa) leaf to determine the phytochemicals, potent antioxidant and antibacterial activity to find out the possible source of applied pharmaceutical formulations. Methods Solvent extracts of leaf material were prepared using the Soxhlet apparatus. A study was performed on antioxidant activity of methanolic extract of leaf by 1-1-diphenyl-2-picrylhydrazyl method. The phenolic and flavonoid content of all the fractions were determined using high performance liquid chromatography. Leaves were also subjected to protein and carbohydrate test. Several homo- and heterobimetallic NiII–MII complexes (MII = Fe, Co, Ni, Zn) supported by an unsymmetric polydentate ligand (L13−) are reported (L13− is the trianion of 2-[bis(2-hydroxy-3,5-tert-butylphenyl)aminomethyl]-4-methyl-6-[(2-pyridylmethyl)iminomethyl]phenol). The L13− chelate provides two distinct coordination environments: a planar tridentate {N2O} site (A) and a tetradentate {NO3} site (B). Reaction of L13− with equimolar amounts of NiII and MII salts provides bimetallic complexes in which the NiII ion exclusively occupies the tetragonal A-site and the MII ion is found …

During the cell cycle, a cell must replicate its DNA, segregate the genome and divide into two identical daughter cells with full chromosomes. This process needs to be specifically coordinated to ensure that cell division does not occur before DNA replication has finished and that the cell septates precisely at midcell to segregate one chromosome to each daughter cell. If cells divide before the chromosome is fully replicated, the chromosome could be guillotined or cell division could occur resulting in cells without chromosomes. If division does not occur properly, the cells elongate but do not divide and form multinucleate filaments. …

Differentiation and cell-specific functions are coupled with metabolic alterations to meet the needs of the cell. In this thesis, I have investigated the alterations of cellular metabolism in osteoblast-lineage cells in response to two different bone anabolic signals, WNT and PTH. I have further elucidated the mechanism underlying the metabolic changes, and have explored the functional importance of such changes for bone anabolism.

Osteoblasts, the principal bone-forming cells, are differentiated from mesenchymal progenitor cells through sequential stages. Protein aggregation is the hallmark of protein conformational disorders such as Alzheimer's disease and prion diseases. Prions are infectious proteins that propagate a self- templating amyloid structure, and have become a model for studying these diseases. Interestingly, a single protein can form a variety of distinct amyloid structures, a phenomenon referred to as amyloid polymorphism. In prion diseases, these different structures, called prion strains, dictate variation in pathology. However, the underlying basis for how structural variation modulates pathology remains unclear.

I. Podocytes are specialized epithelial cells in the kidney glomerulus that play important structural and functional roles in maintaining the filtration barrier. In nephrotic syndrome, a major breakdown of the kidney filtration barrier associated with proteinuria, hyperlipidemia, and edema, podocytes undergo changes in morphology and appear to internalize serum proteins. We postulated that fluid-phase uptake by podocytes might play a role in maintaining the integrity of the filtration barrier. Using fluid-phase tracers, we show that podocytes in vivo actively internalize fluid from the serum and that the rate of internalization is enhanced when the filtration barrier is disrupted. Phase Transitions On Random Lattices: How Random Is Topological Disorder?, Hatem Barghathi, Thomas Vojta

We study the effects of topological (connectivity) disorder on phase transitions. We identify a broad class of random lattices whose disorder fluctuations decay much faster with increasing length scale than those of generic random systems, yielding a wandering exponent of ω = (d−1)/(2d) in d dimensions. The stability of clean critical points is thus governed by the criterion (d+1)ν > 2 rather than the usual Harris criterion dν > 2, making topological disorder less relevant than generic randomness. The Imry-Ma criterion is also modified, allowing first-order transitions to survive in all dimensions d > 1. These results explain a host of puzzling violations …

Because we found UV-exposed oral tissue cells have reduced DNA repair and apoptotic cell death compared with skin tissue cells, we asked if a correlation existed between personal UV dose and the incidences of oral and pharyngeal cancer in the United States. We analyzed the International Agency for Research on Cancer's incidence data for oral and pharyngeal cancers by race (white and black) and sex using each state's average annual personal UV dose. We refer to our data as ‘white’ rather than ‘Caucasian,’ which is a specific subgroup of whites, and ‘black’ rather than African-American because blacks from other countries …

Neonatal sepsis poses a major challenge to achieving the MDG-4 due to lack of facilities to implement proposed management guidelines. Identifying risk factors of neonatal sepsis will help put strategies in place to prevent sepsis. This prospective case control study investigated risk factors of neonatal sepsis in the Asutifi District a typical rural setting of the Brong Ahafo Region of Ghana. A semi-structured check list was used to collect clinical and demographic data from 196 neonates (96 with sepsis as case and 100 without sepsis as control) and respective mothers. Ghana is a known endemic area for hepatitis B virus (HBV) infections, yet the consequences of HBV infection on pregnancy outcomes are unknown. This prospective cohort study was thus conducted among 512 pregnant women attending antenatal clinic in the Cape Coast Teaching Hospital, Ghana, between January, 2011 and December, 2013 to determine the effects of hepatitis B during pregnancy on birth outcomes in Ghana. The HBsAg status of all pregnant women was determined by the latex agglutination test while a researcher administered semi-structured checklist was used to collect demographic/obstetric/medical data of respondents. Undernutrition is an underlying determinant of 45% of all childhood deaths annually, resulting in 3.1 million deaths to children less than five years globally (Black et al., 2013). The adverse effects of undernutrition, especially chronic undernutrition, could cause impaired physical growth, mental development retardation, low productivity and poverty during adulthood, and undernutrition of next generation. Worldwide, over 200 million children are undernourished (Black et al., 2013). Thus, there is an imperative to identify effective preventive actions or interventions for child undernutrition.
