The protein tau is a major contributor in some of the most prevalent neurodegenerative diseases, including the most common form of dementia, Alzheimer's Disease (AD). As a member of the microtubule-associated protein family, tau in enriched in the axons of mature and growing neurons, though under certain conditions, can become hyperphosphorylated and accumulate into toxic oligomeric species and aggregates. In the studies outlined here, we sought to directly target the protein tau using Antisense Oligonucleotides (ASOs) to reduce total expression of tau in vivo and assess if such a reduction could be therapeutically beneficial. HSPA9 was previously identified as a candidate gene in a commonly deleted region (CDR) associated with myelodysplastic syndrome (MDS), a clonal hematopoietic stem cell disorder. Cytogenetic abnormalities occur in ~50% of MDS patients and an interstitial deletion or loss of chromosome 5 containing HSPA9 is the most common, occurring in up to 25% of patients. In order to understand the role of HSPA9 in hematopoiesis and disease development, we created an Hspa9 knockout mouse model. We characterized hematopoiesis of heterozygous mice (Hspa9+/-), which have a 50% reduction in Hspa9 expression, modeling the heterozygous loss of HSPA9 and 50% reduction in …

The cyclin dependent kinase inhibitor, p21, plays a key role in cell cycle. Additionally, the p21 gene, Cdkn1a (hereafter p21) is often used as a marker of cellular stress. To investigate p21 promoter activity under basal conditions and in response to various forms of stress, we generated knock-in imaging reporter mice that express firefly luciferase (FLuc) under the control of the endogenous p21 promoter. I demonstrated that FLuc expression and bioluminescence detection mirrored endogenous p21 protein levels and promoter activity in vivo. Contrary to previously known roles for p53-mediate expression of p21, imaging of reporter cells demonstrated that p53 prevented …

PART I: Investigation and Optimization of Extraordinary Electroconductance (EEC) Sensors

This thesis presents an investigation and geometric optimization of Extraordinary Electroconductance (EEC) sensors, a member of an established class of sensors that exhibit `extraordinary' phenomenon driven by interfaces that maximize current redistribution under an applied perturbation. EEC sensors are responsive to both applied electrical fields and optical illumination and show promising application in the detection of complex biological signals that can aid in conclusive diagnoses.

The EEC device response to nonuniform illumination establishes greater versatility for EEC devices by allowing for position-dependent light sensing while under local illumination. Additionally, an …

In response to genotoxic stress, cells activate DNA damage checkpoint pathways that maintain genetic fidelity, promote cell survival, and prevent malignant transformation. Checkpoint kinase 2 (CHK2) is an evolutionarily conserved serine/threonine protein kinase that is activated in response to DNA double strand breaks. Upon activation by the DNA damage transducer kinase, ATM, CHK2 phosphorylates a wide variety of down stream effectors to promote cell cycle arrest, DNA repair and apoptosis. This dissertation includes three papers using quantitative models to sensibly describe what kinds of preferences political actors will or actually do hold when existing theory offers no insight. The first two papers use evolutionary game theory to predict ways in which politicians, artificially selected on the basis of good performance to remain in office, will in the long run diverge from instrumental rationality as ordinarily assumed in game theory. Multiple modalities are clinically used to quantify cardiovascular function. Most clinical indexes derived from these modalities are empirically derived or correlation- based rather than causality based. Hence these indexes don't provide insight into cardiac physiology and the mechanism of dysfunction. Our group has previously developed and validated a mathematical model using a kinematic paradigm of suction- initiated ventricular filling to understand the mechanics of early transmitral flow and the associated physiology/ pathophysiology. The model characterizes the kinematics of early transmitral flow analogous to a damped simple harmonic oscillator with lumped parameters- ventricular stiffness, ventricular viscoelasticity/ relaxation and ventricular load.

Anthropogenic activities associated with the production of nuclear materials have resulted in uranium contaminated soil and groundwater. The carcinogenic and toxic effects of uranium contamination pose a significant risk to the environment and human health. Phosphate addition to uranium-contaminated subsurface environments has been proposed as a strategy for in situ remediation. Addition of phosphate amendments can result in uranium sequestration in its oxidized +VI state without sustaining reducing conditions as is needed for in situ immobilization via chemical or biological reduction of U(VI) to less soluble U(IV) species. ISG15 is a diubiquitin-like posttranslational modifier and one of the most rapidly induced genes upon type I interferon stimulation. Hundreds of host proteins have been identified as targets of ISG15 conjugation after interferon stimulation, and a number of viral proteins have been shown to be modified by ISG15 during infection. Given the known importance of posttranslational protein modification in the regulation of cellular biology, understanding how ISG15 affects the interferon response and virus replication has been of considerable interest. Critical in determining transplantation outcome, whether tolerance is achieved or not, are CD4+ T cells that can recognize peptides presented on allogeneic (non-self) Major Histocompatibility Complex (MHC) molecules in addition to their conventional ligands, peptides presented on self MHC. It is an enigma as to how these alloreactive T cells can bind to allogeneic MHC given that T cells undergo stringent positive selection during development to bind to peptides presented on self MHC. Niemann-Pick Type C (NPC) disease is a fatal pediatric cholesterol storage disease that is caused by mutations in either the npc1 or npc2 genes. Loss of function of either gene results in toxic storage of free cholesterol in the lysosomes due to a cholesterol trafficking defect. In humans the free cholesterol accumulation leads to Purkinje cell loss in the cerebellum and is accompanied by neurodegenerative symptoms that include ataxia, vertical supranuclear gaze palsy, and decreased motor function. Most NPC1 patients succumb to the disease within the first two decades of life.

The main function of parathyroid hormone (PTH) is to regulate serum calcium levels, in part by stimulating osteoclast mediated bone resorption. However, PTH can stimulate bone formation if given in intermittent doses and it is currently used as a bone anabolic agent to treat osteoporosis. The mechanisms by which PTH stimulates new bone formation are not yet completely clarified; PTH targets not only osteoblasts, where PTH receptors are abundantly expressed, but also immune cells. In this thesis, I examined the role of the calcium dependent cell-cell adhesion molecule, N-cadherin, in PTH signaling and bone anabolic responses. The findings presented in …
