The neurohypophysial hormone arginine vasopressin (AVP) plays important roles in fluid regulation and vascular resistance. Differences in AVP receptor expression, particularly mediated through variation in the noncoding promoter region of the primary receptor for AVP (AVPR1a), may play a role in social phenotypes, particularly social monogamy, in rodents and humans. Among primates, social monogamy is rare, but is common among New World monkeys (NWM). AVP is a nonapeptide and generally conserved among eutherian mammals, although a recent paper demonstrated that some NWM species possess a novel form of the related neuropeptide hormone, oxytocin. We therefore characterized variation in the AVP …

Hypogammaglobulinemia In Blt Humanized Mice – An Animal Model Of Primary Antibody Deficiency, Francisco Martinez-Torres, Tomonori Nochi, Angela Wahl, J. Victor Garcia, Paul W. Denton
Biology Faculty Publications
Primary antibody deficiencies present clinically as reduced or absent plasma antibodies without another identified disorder that could explain the low immunoglobulin levels. Bone marrow-liver-thymus (BLT) humanized mice also exhibit primary antibody deficiency or hypogammaglobulinemia. Comprehensive characterization of B cell development and differentiation in BLT mice revealed other key parallels with primary immunodeficiency patients. We found that B cell ontogeny was normal in the bone marrow of BLT mice but observed an absence of switched memory B cells in the periphery. PC-KLH immunizations led to the presence of switched memory B cells in immunized BLT mice although plasma cells producing PCor …

The Ties That Bind: Reevaluating The Role Of Legal Presumptions Of Paternity, Heather Kolinsky
The Ties That Bind: Reevaluating The Role Of Legal Presumptions Of Paternity, Heather Kolinsky
UF Law Faculty Publications
As Justice Brennan observed in Michael H. v. Gerald D. so many years ago, we must "identify the point at which a tradition becomes firm enough to be relevant to our definition of liberty and the moment at which it becomes too obsolete to be relevant any longer." This Article addresses one such tradition, the legal presumption of paternity, and examines it through the lens of equal protection, the changing roles of fatherhood, and the evolution of marriage.
The concept of who is a parent must change to both satisfy equal protection as well as modern scientific and societal realties. …

This article describes the phenomenon of the outpatient year experiences of the PGY(Post Graduate Year)-3 year psychiatry residents using participant interviews, focus groups, and research observation at a university health science center. A qualitative research approach was used to identify and understand psychiatric residents’ experiences of their third year. The research question was, “What are the important and valuable experiences of psychiatry residents and what meaning do they ascribe to these experiences in their acquisition of psychiatric skills?” Four themes emerged from the study: Specialty Choice Was a Momentous Decision, Observation and Reflection Should Be Modeled Prior to Practice, The …
Re-Considering Female Sexual Desire: Internalized Representations Of Parental Relationships And Sexual Self-Concept In Women With Inhibited And Heightened Sexual Desire, Eugenia Cherkasskaya
Re-Considering Female Sexual Desire: Internalized Representations Of Parental Relationships And Sexual Self-Concept In Women With Inhibited And Heightened Sexual Desire, Eugenia Cherkasskaya
Dissertations, Theses, and Capstone Projects
Background: Psychoanalytic and sociocultural thinkers and researchers suggest that the etiology of low female sexual desire, the most prevalent sexual complaint in women, is multi-determined, implicating biological and psychological factors, including women's early relational experiences and sexual self-concept that stem from gender dynamics of a patriarchal culture. Further, recent studies indicate that highly sexual women exhibit heightened sexual desire, and high levels of sexual agency and sexual esteem. The study evaluated a model that hypothesized that sexual self-concept (sexual subjectivity, self-objectification, genital self-image) explains (i.e., mediates) the relations between internalized representations of parental relationships (attachment, separation/individuation, parental identification) and sexual …

Reexamining Linguistic Relativity: What Adult Bilinguals Can Teach Us About Culture, Language, And Cognition, Natalya Petroff
Reexamining Linguistic Relativity: What Adult Bilinguals Can Teach Us About Culture, Language, And Cognition, Natalya Petroff
Dissertations, Theses, and Capstone Projects
Extending Whorf's popular notion of linguistic relativity (LR) to bilingual contexts, one would argue that a speaker's first language (L1) influences her thinking and behavior under second language (L2) conditions. According to one interpretation of LR, inter-language relativity, L1 instills in its speakers habitual ways of thinking and thus influences their perception and categorization in L2 contexts. Under intra-speaker relativity, bilinguals follow either L1 or L2 patterns of performance, depending on L2 proficiency. Finally, according to usage-based accounts of language, there is no qualitative difference between mono- and bilingual speakers, and a bilingual's performance under L2 conditions is best viewed …

The Influence Of Advanced Cognitive Ability On The Development Of Psychological Defenses And In Understanding And Managing Affect: A Study Of Latency-Aged Gifted Students, Kahlila Ife Robinson
Dissertations, Theses, and Capstone Projects
The present study examines the influence of advanced cognitive ability on the development of psychological defenses and in understanding and managing affect, using the lens of Emotional Intelligence (EI). Theories of psychological defense maturity state that defense mechanisms are influenced both by the cognitive level of the individual and by the cognitive complexity of the defense itself (Cramer, 1999; Cramer, 2009). Individuals with exceptional cognitive ability may therefore show a corresponding "match" with complex defense use. In addition to defense use, how well one is able to identify, understand, manage and use emotion to facilitate thought, abilities often labelled Emotional …
Carbon And Nitrogen Dynamics From Slow Pools Of Soil Organic Matter In A Temperate Forest: Pyrogenic Organic Matter And And Root Litter, Fernanda Dos Santos
Carbon And Nitrogen Dynamics From Slow Pools Of Soil Organic Matter In A Temperate Forest: Pyrogenic Organic Matter And And Root Litter, Fernanda Dos Santos
Dissertations, Theses, and Capstone Projects
Soil organic matter (SOM) is the dominant reservoir of organic carbon (OC) in terrestrial ecosystems, storing approximately three times the size of the C pool in the atmosphere. In temperate forests, a major fraction of the SOM consists of slowly decaying soil organic C (SOC) pools. While slowly cycling C pools constitute a large reservoir of stable C in soils, the dominant environmental factors controlling this C pool remain unresolved. This research investigates two significant, but poorly characterized slowly decaying C pools: fine root litter (< 2mm) and thermally altered plant biomass (pyrogenic organic matter, PyOM). Specifically, I used compound-specific stable isotope analysis (13C and 15N) as my main methodological approach to examine the (1) …
The Reproductive Body: Exploring Reproduction Beyond Gender, Ilyssa Ann Silfen
The Reproductive Body: Exploring Reproduction Beyond Gender, Ilyssa Ann Silfen
Dissertations, Theses, and Capstone Projects
Most of us have been taught over the course of our lives that biological sex, gender, and reproduction are inescapably linked and, over time, this has created the illusion that these are all naturally connected. However, these “natural” connections have been formed over time after generations of repetition. While it may seem impossible to separate biological sex, gender, and reproduction from one another, it is important to deconstruct this falsely organic system from both a gender and human rights perspective.
This thesis seeks to explore the complex relationship between society’s reproductive mandate and the reality of the various processes of …
Development Of Cell-Active Inhibitors And Activity-Based Probe Of Cysteine Cathepsins, Dibyendu Dana
Development Of Cell-Active Inhibitors And Activity-Based Probe Of Cysteine Cathepsins, Dibyendu Dana
Dissertations, Theses, and Capstone Projects
Cysteine cathepsins are an important class of enzymes that coordinate a variety of important cellular processes, and are implicated in various types of human diseases. Still however, many of their cellular function remain poorly understood. Chemical biology approaches employing small molecules can be utilized for this purpose. Unfortunately small molecule probes that are cell-permeable and non-peptidyl in nature are scarcely available.
In this work, first a library of sulfonyloxiranes is synthesized. From this library, 2-(2-ethylphenylsulfonyl)oxirane is identified as a selective inhibitor of cysteine cathepsins. Cell-based study reveals that 2-(2-ethylphenylsulfonyl)oxirane is a cell-permeable, covalent, and irreversible inhibitor of cathepsin B with …

A Study Of Twenty Two Caliber Long Rifle And Nine Millimeter Parabellum Bullet Ricochet From Common Substrates, Peter Diaczuk
Dissertations, Theses, and Capstone Projects
Determining the angle at which a bullet will successfully ricochet is essential information when a shooting investigation involves indirect fire. In this research, determining the critical angle and its variance was measured for six substrates, two calibers and two bullet types. This information provides the forensic scientist with fundamental data required for the scientific reconstruction and assessment of a shooting scene. Depending upon the bullet's design, the substrate, and the angle of impact, a bullet may fail to ricochet upon impact, or the bullet will ricochet. Knowledge of bullet behavior with common substrates provides valuable information for scientific investigation of …

Mental Representations, Social Exclusion, And Neurobiological Processes In Borderline Personality Disorder: A Multi-Level Study, Jeffrey K. Erbe
Dissertations, Theses, and Capstone Projects
Borderline Personality Disorder (BPD) is an ongoing public health crisis. Poor developmental quality of differentiation-relatedness of object representations and attachment insecurity have been clinically and empirically demonstrated as core patterns of intrapsychic and interpersonal dysfunction in this particular form of personality pathology. Differentiation-relatedness (D-R), which involves a complementary relationship between intrapsychic autonomy and interpersonal relatedness, has been shown to be a significant aspect of internal psychic experience that relates directly to external relationship patterns, including characteristic response to interpersonal interactions and has been a specific target for treatment of BPD. Specifically, individuals with BPD have shown lower developmental quality of …

"Fits Of Vulgar Joy": Play Anxiety In The Romantic Poets, Alison Powell
Dissertations, Theses, and Capstone Projects
"Fits of Vulgar Joy": Play Anxiety in the Romantic Poets considers the crucial but neglected role of play as a component of the imaginative faculty and as related to the development of moral sensibility in the Romantic era. It examines the Spieltrieb ("play drive") in the works of Coleridge, Rousseau, Wordsworth, Shelley, and John Clare to explore the tension between what is explicitly stated about the relationship between play, intersubjectivity, and aesthetics, and how play is actually depicted in the poetry and essays of the Romantic era. It works to identify how and with what objective the Romantics differentiated so …
Development And Applications Of Mass Spectrometric Methods For Phosphorylation Analysis, Hsin-Pin Ho
Development And Applications Of Mass Spectrometric Methods For Phosphorylation Analysis, Hsin-Pin Ho
Dissertations, Theses, and Capstone Projects
Protein phosphorylation modification regulates numerous cellular functions by a reversible and selective control of kinases and phosphatases. To understand the entire dynamic network of phosphorylation requires sensitive and reliable quantification of phosphorylation, measurements that can be achieved by mass spectrometry. In this research, we established efficient MALDI-mass spectrometric methods as strategies for single- or multi-site phosphorylation quantification without the use of isotopes, chromatography and calibration curves. The methods were assessed by analyzing peptide standards with different single-multiple phosphorylation sites, showing a wide dynamic range, good accuracy and reproducibility. This is the first label-free MALDI method without using a calibration methodology …

Venomic Characterization Of The Terebridae And Novel Terebrid Neuropeptides, Mary Elizabeth Wright
Dissertations, Theses, and Capstone Projects
Unravelling the complex mixture of neuropeptides produced by the terebrid venom duct holds the promise of discovering tomorrow's therapeutics. Cone snails have already demonstrated the potential found in the venom of these unusual marine organisms, through the commercial approval of drugs for pain and other indications. Terebrids, as the sister family to the cone snails, have been much less investigated, but have a species richness that makes them very attractive in the search for novel neuropeptides. The venomics research described in this work encompasses the first comprehensive characterization of the terebrid venom duct transcriptomes of two species, Cinguloterebra anilis and …

Staphylococcus aureus is a Gram-positive pathogen that causes a diverse range of bacterial infections. Invasive S. aureus strains secrete an extensive arsenal of hemolysins, immunomodulators, and exoenzymes to cause disease. Our studies have focused on the secreted enzyme hyaluronidase (HysA), which cleaves the hyaluronic acid polymer at the β-1,4 glycosidic bond. In the study described in this report, we have investigated the regulation and contribution of this enzyme to S. aureus pathogenesis. Using the Nebraska Transposon Mutant Library (NTML), we identified eight insertions that modulate extracellular levels of HysA activity. Insertions in the sigB operon, as well as in genes …

Within-Socket Myoelectric Prediction Of Continuous Ankle Kinematics For Control Of A Powered Transtibial Prosthesis, Samuel Farmer, M. Barbara Silver-Thorn, Philip A. Voglewede, Scott A. Beardsley
Biomedical Engineering Faculty Research and Publications
Objective. Powered robotic prostheses create a need for natural-feeling user interfaces and robust control schemes. Here, we examined the ability of a nonlinear autoregressive model to continuously map the kinematics of a transtibial prosthesis and electromyographic (EMG) activity recorded within socket to the future estimates of the prosthetic ankle angle in three transtibial amputees. Approach. Model performance was examined across subjects during level treadmill ambulation as a function of the size of the EMG sampling window and the temporal 'prediction' interval between the EMG/kinematic input and the model's estimate of future ankle angle to characterize the trade-off between model error, …
