Metabolomics is the science of comprehensive evaluation of changes in the metabolome with a goal to elucidate underlying biological mechanisms of a living system. There is an opinion in the field of metabolomics, the study of the set of metabolites present within an organism, cell, or tissue, that the future development of the field is contingent upon two factors. One of the factors is the advancement of analytical instrumentation, and the other is developing data mining methodologies for extracting meaningful and interpretable experimental results. Background: Human-associated microbial communities include fungi, but we understand little about which fungal species are present, their relative and absolute abundances, and how antimicrobial therapy impacts fungal communities. The disease cystic fibrosis (CF) often involves chronic airway colonization by bacteria and fungi, and these infections cause irreversible lung damage. Fungi are detected more frequently in CF sputum samples upon initiation of antimicrobial therapy, and several studies have implicated the detection of fungi in sputum with worse outcomes. Thus, a more complete understanding of fungi in CF is required. Results: We characterized the fungi and bacteria in expectorated sputa from six …

The aggregation of amyloid-β (Aβ) peptide and its deposition in parts of the brain form the central processes in the etiology of Alzheimer disease (AD). The low-molecular weight oligomers of Aβ aggregates (2 to 30 mers) are known to be the primary neurotoxic agents whose mechanisms of cellular toxicity and synaptic dysfunction have received substantial attention in the recent years. However, how these toxic agents proliferate and induce widespread amyloid deposition throughout the brain, and what mechanism is involved in the amplification and propagation of toxic oligomer species, are far from clear. Autoantibodies To The Ny-Eso-1 Tumor Antigen In Metastatic Melanoma: Sialylation Of The Fc Region Of Immunoglobulin G Induces Differential Expression Signatures Of Inflammatory Molecules During Dendritic Cell Differentiation And Maturation, Martin Oaks, Nathaniel Rein, John O. Richards, James Shaffer
Journal of Patient-Centered Research and Reviews
Purpose: We tested the hypothesis that different glycoforms of antibodies from patients with metastatic melanoma have different functional effects on human dendritic cell differentiation and maturation.
Methods: Antibodies to the cancer antigen NY-ESO-1 were affinity-purified from patients with melanoma and further fractionated into different glycoforms by lectin chromatography. Sialic acid-rich and sialic acid-poor fractions of these immunoglobulin G antibodies (IgG) were added to dendritic cell cultures during both differentiation and maturation, and the resulting cellular messenger RNA (mRNA) and culture supernatants were tested by microarray and enzyme-linked immunoassay for molecules related to inflammatory pathways.

As STEM areas increasingly rely on pre-existing data, either to validate or extend the scientific body of knowledge, students who have baseline knowledge of how to find, evaluate, and access data will have an advantage. Accordingly, undergraduate STEM curricula is increasingly focused on research-based group projects that develop professional skills, building the professional portfolio needed for early career scientists, technologists, and engineers. This project works to develop new tools to implement basic data skills within the undergraduate disciplinary curricula. Background:
The homeodomain leucine zipper (HD-Zip) transcription factor family is one of the largest plant specific superfamilies, and includes genes with roles in modulation of plant growth and response to environmental stresses. Many HD-Zip genes are characterized in Arabidopsis (Arabidopsis thaliana), and members of the family are being investigated for abiotic stress responses in rice (Oryza sativa), maize (Zea mays), poplar (Populus trichocarpa) and cucumber (Cucmis sativus). Findings in these species suggest HD-Zip genes as high priority candidates for crop improvement.

The Qualitative Report
A problem with the popular desire to legitimate one’s research through the inclusion of reflexivity is its increasingly uncritical adoption and practice, with most researchers failing to define their understandings, specific positions, and approaches. Considering the relative recentness with which reflexivity has been explicitly described in the context of grounded theory, guidance for incorporating it within this research approach is currently in the early stages. In this article, we illustrate a three-stage approach used in a grounded theory study of how parents of children with autism navigate intervention. Certain N95 FFR models that staple the head straps directly onto filtering material are commercially available. This method of assembly can tear or reduce fiber density in the immediate area surrounding the staple punctures. Five N95 FFR models were evaluated to determine if staple punctures on the filter medium reduce the protection offered by the respirators.
Total penetrations were measured with the staple punctures intact and when stretching the head straps a distance equivalent to a 95% male head circumference. Filter penetration were measured by sealing the staple punctures. Aerosols of 200, 500, and 1000 nm were used to challenge …

Numerous nanocarriers are currently being investigated as drug delivery vehicles for transporting chemotherapeutics to cancer cells. Our research group has recently synthesized a new generation of echogenic liposomes based on the concept of encapsulating one or more nanoemulsion droplets inside a liposome (called an “eLiposome”). The concept is to use a nanoemulsion droplet with a low boiling point near body temperature, thus requiring only a small acoustic nudge to vaporize the droplet from liquid to gas and break open the eLiposome, thus releasing its contents. The cost of reproduction hypothesis suggests that current reproduction has inherent tradeoffs with future reproduction. These tradeoffs can be both in the form of energy allocated to current offspring as opposed to somatic maintenance and future reproduction (allocation costs), or as an increase in mortality as a result of morphological or physiological changes related to reproduction (viability costs). Individuals may be able to decrease viability costs by altering behavior. Female western mosquitofish, Gambusia affinis experience a reduction in swimming ability as a consequence of pregnancy. We test for a viability cost of reproduction, and for behavioral compensation in pregnant female …

Going From Microbial Ecology To Genome Data And Back: Studies On A Haloalkaliphilic Bacterium Isolated From Soap Lake, Washington State, Melanie R. Mormile

Soap Lake is a meromictic, alkaline (∼pH 9.8) and saline (∼14-140 g liter-1) lake located in the semiarid area of eastern Washington State. Of note is the length of time it has been meromictic (at least 2000 years) and the extremely high sulfide level (∼140 mM) in its monimolimnion. As expected, the microbial ecology of this lake is greatly influenced by these conditions. A bacterium, Halanaerobium hydrogeniformans, was isolated from the mixolimnion region of this lake. Halanaerobium hydrogeniformans is a haloalkaliphilic bacterium capable of forming hydrogen from 5- and 6-carbon sugars derived from hemicellulose and cellulose. Due …

Portfolio selection (optimization) problem is a very important and widely researched problem in the areas of finance and economy. Literature review shows that many methods and heuristics were applied to this hard optimization problem, however, there are only few implementations of swarm intelligence metaheuristics. This paper presents artificial bee colony (ABC) algorithm applied to the cardinality constrained mean-variance (CCMV) portfolio optimization model. By analyzing ABC metaheuristic, some deficiencies such as slow convergence to the optimal region, were noticed.
