Small conductance calcium-activated potassium (SK) channels belong to the family of K+ channels. They play a significant role in cell volume regulation and in lens homeostasis, and thus are associated with cataract formation. This study was designed to demonstrate the presence of the four different isoforms of SK channels (SK1, SK2, SK3, and SK4) in human lens epithelial (HLE)-B3 cell line using Western blot (WB), immunofluorescence (IF), and reverse transcriptase coupled to the polymerase chain reaction (RT-PCR). We describe and characterize a method for estimating the pressure field corresponding to velocity field measurements such as those obtained by using particle image velocimetry. The pressure gradient is estimated from a time series of velocity fields for unsteady calculations or from a single velocity field for quasi-steady calculations. The corresponding pressure field is determined based on median polling of several integration paths through the pressure gradient field in order to reduce the effect of measurement errors that accumulate along individual integration paths. Animal propulsors such as wings and fins bend during motion and these bending patterns are believed to contribute to the high efficiency of animal movements compared with those of man-made designs. However, efforts to implement flexible designs have been met with contradictory performance results. Consequently, there is no clear understanding of the role played by propulsor flexibility or, more fundamentally, how flexible propulsors should be designed for optimal performance. Structural And Functional Support In The Prediction Of Smoking Cessation In Caregivers Of Children With Asthma, Erin M. Tooley, Andrew Busch

Caregivers of children with asthma smoke at a rate similar to the general population. Research on the relative importance of structural or functional social support in smoking cessation has been mixed. Participants were smokers (N=154) who were caregivers of children with asthma. Both functional (perception of social support measured by the Interpersonal Support Evaluation List) and structural social support (living with another smoker, partner status, and the proportion of smoking friends) were measured at baseline. Participants received an asthma-education and smoking cessation intervention based on Motivational Interviewing. Biochemically-verified abstinence was assessed at 6-months post treatment. The Ozobranchidae family is the smallest and least studied hirudinean taxon. Our research includes the largest molecular dataset yet reported for marine ozobranchids (Ozobranchus margoi and Ozobranchus branchiatus) with the most number of documented turtle hosts (57) from the Atlantic and Pacific Oceans to date of any marine turtle epibiont study. Turtle species sampled in this study include loggerheads (Caretta caretta), hawksbill (Eretmochelys imbricata), olive ridley (Lepidochelys olivacea), and green turtles (Chelonia mydas). Phylogenetic analyses of mitochondrial (COI) and nuclear ribosomal (18S and 28S) genes all support the monophyly of marine Ozobranchidae leeches with speciation occurring over an extensive period …

Many remediation technologies have been developed to treat extensive subsurface contamination by chlorinated hydrocarbons (CHCs). One such technology is a permeable reactive barrier (PRB), which contains reactive media that acts as an electron donor in order to reduce CHCs. Extensive effort has been placed on finding the most suitable reactive media in PRBs, with zero-valent iron (ZVI) being the most commonly utilized media. Cytokine activation of macrophages leads to "macrophage polarizatio" The M1 subset is polarized by exposure to lipopolysaccharide (LPS) and interferon-gamma (IFN-γ) and M2 polarized cells develop after exposure to interleukin-4 (IL-4) or IL-13. In this study, the RAW 264.7 murine macrophage cell line was used to study the effect herpes simplex virus-Type I (HSV-1) and Dengue virus serotype-2 (DENV2) infections have on cultures of unpolarized (M0) or polarized subsets. It has previously been reported that AMP-activated protein kinase (AMPK) may be critical for hypoxic chemotransduction in carotid body type I cells (Wyatt et al, 2007). This study sought to determine the importance of the regulatory upstream kinase of AMPK, CamKKβ, in the acute response to hypoxia in isolated mouse type I cells.

Initial studies indicated several previously unreported artefacts associated with using the CamKKβ inhibitor STO-609 and calcium imaging techniques. Strong STO-609 autofluorescence meant that Fura-2 could not be used and X-Rhod-1 imaging revealed that STO-609 quenched fluorescence even in the absence of intracellular Ca2+. The unique optoelectronic properties of silver nanoparticles (AgNPs) have led to their explosive use in a multitude of both research and industrial settings in recent years. Localized surface plasmon resonance (LSPR) exhibited by AgNPs has been exploited extensively as a nano-scale probe in a variety of spectroscopic detection modalities, particularly in surface-enhanced Raman spectroscopy (SERS). This work studies thermal properties of magnetic nanoparticles in an external ac magnetic field. Dried iron and cobalt nanoparticles were prepared by thermal decomposition of iron pentacarbonyl (Fe(CO)5) and dicobalt octacarbonyl (Co2(CO)8), triscobalt nona(carbonyl)chloride (Co3(CO)9Cl), or tetracaobalt dodecacarbonyl (Co4(CO)12) [1]. The samples had different mean diameters: 5.6 - 21.4 nm for iron and 6.5 - 19.4 nm for cobalt. Each sample was exposed to ac magnetic field and the increase in temperature of the sample was measured. Our previous study demonstrates that cMVs of diabetic mouse are detrimental to endothelial progenitor cells (EPCs), which are known to be very important for maintaining normal endothelial function and structure. In this study, we compared the levels of circulating EPCs and EPC-derived MVs (EPC-MVs) in diabetic and healthy subjects. The newly constructed airfoil is modeled and numerically investigated. To more closely mimic the in vivo progression of HSV-1 a novel in vitro method was created. In this study HEL-30 keratinocytes were infected with a 0.1 MOI of HSV-1 and plated unto a membrane with 8μm diameter pores. The membrane insert was then placed into culture wells that contained Neuro-2a cells (N2A). The neurites from the neuronal cells made cell-to-cell contact with the infected HEL-30 cells and the virus was transmitted into the neuronal cells. Animal shelter housing is highly stressful for a dog, compromising welfare and leading to undesirable behaviors and unknown health consequences. We documented the changes in circulating numbers of white blood cells, plasma cortisol, and fecal parasite shedding of dogs housed for 10 days at a county animal shelter. White blood cell changes were most prominent on Day 10 after arrival to the shelter. Changes included increased total leukocytes, mature neutrophils, and lymphocytes, with less consistent increases in monocytes and neutrophil to lymphocyte ratio (N:L). Fecal parasite shedding was elevated and not affected by day. Cortisol levels of shelter dogs declined …

Introduction: Functional near-infrared spectroscopy (fNIRS) is a relatively young technique in the field of medical imaging. As such, it has yet to be widely implemented for clinical use, despite its promising advantages. However, unlike fMRI-its much bulkier and costly counterpart-fNIRS has yet to be proven as a standalone imaging tool within a clinical setting, particularly that of ophthalmology or physical therapy.

Previous studies show a fluoxetine/simvastatin drug combination successfully reduced infarct size and increased functional recovery when administered 20-26 hours post stroke. This project tested the hypothesis that earlier drug delivery at 6-12 hours post stroke will improve functional recovery and decrease infarct size, and that limb rehabilitation will improve functional recovery.

Pre-stroke function was determined in rats, a stroke was induced, and daily treatment began 6-12 hours post stroke. Baseline functional deficit was established, and additional testing over 90 days monitored functional recovery. Limb rehabilitation was provided for designated animals, and brain analysis measured infarct size.

The synthesis of poly(arylene ether sulfone)s, with a pendant aryl iodide, was achieved via nucleophilic aromatic substitution (NAS) polycondensation of 3-iodo-3',5'-difluoro-diphenylsulfone. The polymer series was subjected to amino acid promoted, copper catalyzed coupling reactions with various N-heterocycles. The resulting polymers became crosslinked and insoluble in any organic solvents. The iodo monomer was then subjected to an amino acid promoted, copper catalyzed coupling reaction with carbazole. The successful synthesis of 3'-(9-carbazole) 3,5-difluorodiphenylsulfone is reported. A series of PAES containing a carbazole substituent were synthesized and characterized. Assortative fertilization refers to the species-specific interactions between sperm and oocytes that affect the success of fertilization. One type of interaction is chemotaxis of sperm to oocytes. In Caenorhabditis elegans, amoeboid sperm must crawl along the uterine lining towards the spermathecae in response to oocyte-derived prostaglandin signals for fertilization. This chemotactic signaling system likely operates in other species of the Elegans-Group of Caenorhabditis as sperm of C. briggsae and C. remanei do localize to the C. elegans spermathecae. Muscle spindles (MS) and Golgi tendon organs (GTOs) are encapsulated, stretch-activated sensory receptors housed within skeletal muscles, yet their functional contributions to proprioceptive monitoring of limb movements are quite different. For example, MS are responsible for detecting stretch of muscle fibers, and GTOs provide feedback regarding muscle tension during contraction. Peripheral axons extending from proprioceptive sensory neurons (PSNs) in the dorsal root ganglia supply MS and GTOs. Field parameter measurements and water and Planktothrix rubescens samples were collected in May, August, and October, 2013 at the deepest point in the lake, to better understand these processes. At the metalimnion-hypolimnion boundary, P. rubescens produced chlorophyll and turbidity maxima. Photosynthesis produced oxygen-rich and nutrient-poor surface waters. The decay of organic matter produced anoxic, nutrient-rich water in the hypolimnion. The distribution of mercury (Hg) in the ocean is complex as a result of in situ chemical transformations and inputs from natural and anthropogenic sources. Within the ocean, inorganic Hg is methylated to monomethylmercury (MMHg), which bioaccumulates and biomagnifies in marine food webs and poses a health risk to humans who eat fish. Monomethylmercury (MMHg) can accumulate in fish to concentrations that pose a threat to the health of fish, piscivorous organisms, and humans who eat fish. A variety of environmental factors have been hypothesized to influence either the methylation of inorganic Hg or the bioaccumulation and magnification of MMHg. This study investigates the influence of selected environmental factors on MMHg concentrations in freshwater fish across a regional scale, most of the U.S. portion of Laurentian Great Lakes region. The Slc12a5 gene encodes the neuron-specific electroneutral K+Cl- cotransporter-2 (KCC2), a constitutively active Cl- extruder. In mature neurons, KCC2 functionally predominates over other members of the Slc12a family, particularly the Na+K+2Cl- cotransporter-1 (NKCC1), a ubiquitous Cl- loader. Therefore, KCC2 keeps and maintains the intracellular chloride concentration ([Cl-]i) below values predicted by thermodynamic equilibrium and in doing so, KCC2 plays a key role in neuron physiology: it makes possible the inhibitory action of neurotransmitters such as gamma-aminobutyric acid (GABA) and glycine.
