Characterization Of The Disulfide In The Ectodomain Of Anthrax Toxin Receptor 2, Pedro Raymundo Jacquez

Anthrax toxin receptors act as molecular clamps or switches that control anthrax toxin entry, pH-dependent pore formation, and translocation of enzymatic moieties across the endosomal membranes. The ectodomain of anthrax toxin receptor 2 (ANTXR2) is composed of a von Willebrand factor A (VWA) domain that binds to anthrax toxin protective antigen (PA) and a newly defined immunoglobulin-like (Ig) domain, in which the disulfide bonds are required for PA pore formation and for the folding of ANTXR2. While the VWA domain has been well characterized, the structure and function of the whole ectodomain (VWA-Ig) are poorly defined, this is mainly due …

Proteins are ubiquitous in all living organisms, executing the majority of cellular functions in distinct ways. Understanding a protein's role necessitates investigating its structure and function, which are closely related. My research couples these two aspects by delving into the biochemical and structural characterization of proteins in four distinct systems, all playing central roles in numerous significant disease progressions. These four original research endeavors were all targeted for structural studies with a unifying relationship to establish our new structural biochemistry lab. The potential for a highly pathogenic influenza pandemic remains a concern for global health. The ability of the influenza virus to undergo antigenic shift and genetic drift give circulating strains of influenza the high probability of developing resistance to current antiviral therapies. Emerging strains of higher virulence, to which the infected person has not had any previous exposure to viral antigens, pose a greater threat of serious illness and death. Since the last decade, new techniques have made optical microscopy break the diffraction barrier of resolution where all of them are based on molecular fluorescence. Among them, stimulated emission depletion microscopy (STED) has reached less than 25 nm resolution by engineering the point spread function. However, the existing obstacles associated with fluorescence tagging makes it desirable to achieve label-free imaging. Recently the pump-probe method has made it possible to obtain image contrast from molecular absorption and vibration signatures that do not depend on fluorescence. This project is a focus on characterizing the process for actualizing three dimensional structures out of a carbon nanostructure composite via a direct print additive manufacturing process. Manufacturing parts additively enables for realization of geometrically complex shapes that often times cannot be manufactured any other way. The specificity of a material's properties have to be such, that the processing method can precisely place and bond material to itself in a highly repeatable manner. The anti-cancer properties of curcumin, the natural yellow pigment in the roots of turmeric (Curcuma longa), have been extensively described over the last century. It has been shown to interfere with multiple cell signaling pathways, including execution of apoptosis, anti-proliferation, anti-angiogenic, and anti-inflammatory properties. However, curcumin's cytotoxic potential is limited when administered in vivo. With the idea of preserving curcumin's characteristics, but improving its bioavailability, analogues have been developed and evaluated in the last decade. Most of the analogues have shown very good anti-cancer activity in various cell lines and one analogue is now in a clinical trial. Nonetheless, this …

The objective of this study was to conduct an avian survey of five water sources (four ephemeral former cattle tanks and one perennial spring), determine how many species inhabit or use Indio Mountains Research Station, and show a hierarchal similarity between the survey sites. This survey was conducted in a Chihuahuan Desert scrub landscape on Indio Mountains Research Station (IMRS) in Hudspeth County, Texas. There have been no previous formal studies of the avifauna at IMRS. All species accounts or sightings have been from casual sightings.

Neurodegeneration, a progressive loss of nerve cells (neurons), occurs in many neurological disorders, including Alzheimer's and Parkinson's diseases, as well as in the aging brain. Disruption of microtubules in neurons and the aggregation of proteins associated with them is the hallmark of neurodegeneration. Nevertheless, the cause of this disorder is largely unknown, and no effective drugs are available to treat the disease processes. Therefore, there is a need to understand the molecular mechanisms that drive the assembly and disassembly of microtubules during neurite outgrowth and differentiation. The detection and determination of viable phytoplankton are fundamental assays in oceanographic and limnological fields. Assessing phytoplankton physiology allows for insight into how cells might respond to different environmental or physical stressors, making it an important subject for ecological research and applied sciences, where the physiological status of photautotrophs must be known. In particular, maritime operations have been identified as major vectors in the spread of aquatic invasive species through ballast water transport. Vesicular horizons are common at the surface of arid and semiarid soils and play a critical role in regulating infiltration. Most methods for examining pore morphology in the vesicular horizon involve physical sectioning of the sample and individual measurement of pores, which is time-consuming and provides an incomplete view of the pores. The objectives of this study were to (i) develop methods for the classification and characterization of pores in the vesicular horizon using high resolution X-ray computed tomography (HRXCT) and (ii) use these methods to examine the distribution of pores within vesic-ular peds. Since federal regulations authorized single-sex education in 2006, there has been an explosion of single-sex schools and classes. Although the Supreme Court has not ruled, three federal court decisions have addressed the constitutionality of single-sex classes, and the issue will percolate toward Supreme Court review soon. The arguments are that parents should have choices and “diversity” of educational options, that “brain research” shows that boys and girls are so biologically different to need sex-specific educational environments, that educational outcomes are better, and single-sex learning environments allows boys and girls to break through gender stereotypes. Glyco-Modification Of Protein With O-Cyanate Chain-End Functionalized Glycopolymer Via Isourea Bond Formation, Valentinas Gruzdys, Hailong Zhang, Xue-Long Sun

Glycoengineering aimed at addition of carbohydrates to proteins is an attractive approach to alter pharmacokinetic properties of proteins such as enhancing stability and prolonging the duration of action. We report a novel protein glyco-modification of BSA and recombinant thrombomodulin with O-cyanate chain-end functionalized glycopolymer via isourea bond formation. The protein glycoconjugates were confirmed by SDS-PAGE, western blot, and MALDI-TOF Mass Spectrometry. Protein C activation activity of the glyco-modified recombinant thrombomodulin was confirmed, proving no interference to activity from the glycopolymer modification. The isourea bond formation under mild conditions was demonstrated as an alternative method for protein modification with polymers.

Background: Several previous investigations have determined potential risk factors for stress fractures in athletes and military personnel.
Purpose: To determine factors associated with the development of stress fractures in female athletes.
Study Design:
