Little is known about the molecular pathways regulating poor differentiation and invasion of head and neck squamous cell carcinoma (HNSCC). In the present study, we aimed to determine the role of MDA-9/Syntenin, a metastasis associated molecule in HNSCC tumorigenesis. Elevated MDA-9/Syntenin expression was evident in 67% (54/81) primary HNSCC tumors (p=0.001-0.002) and 69% (9/13) pre-neoplastic tissues (p=0.02-0.03). MDA-9/Syntenin overexpression was associated with the stage (p=0.001), grade (p=0.001) and lymph node metastasis (p=0.0001). Silencing of MDA-9/Syntenin in 3 poorly differentiated HNSCC cell lines induced squamous epithelial cell differentiation, disrupted angiogenesis and reduced tumor growth in vitro and in vivo. Background: Highly pathogenic influenza viruses cause high levels of morbidity, including excessive infiltration of leukocytes into the lungs, high viral loads and a cytokine storm. However, the details of how these pathological features unfold in severe influenza infections remain unclear. Accumulation of Gr1 + CD11b + myeloid cells has been observed in highly pathogenic influenza infections but it is not clear how and why they accumulate in the severely inflamed lung. In this study, we selected this cell population as a target to investigate the extreme inflammatory response during severe influenza infection.

Background: Aquatic biofilms often serve as environmental reservoirs for microorganisms and provide them with a nutrient-rich growth environment under harsh conditions. With regard to Cryptosporidium, biofilms can serve as environmental reservoirs for oocysts, but may also support the growth of additional Cryptosporidium stages.
Results: Here we used confocal laser scanning microscopy, scanning electron microscopy (SEM), and flow cytometry to identify and describe various Cryptosporidium developmental stages present within aquatic biofilm systems, and to directly compare these to stages produced in cell culture. The prevalence of adolescent mental illness has increased at an alarming rate over the past two decades. Due to a lack of accessible mental health care and the stigmatization of mental illness, parents of children with these disorders are often reluctant to seek help. These children often resort to negative self-coping methods among which include substance abuse, violence, and suicide. These negative self-coping methods have a harmful, lasting impact on the individual as well as the surrounding community.
Bibliotherapy, which harnesses the meditative power of books, can teach helpful coping skills to students with mental illness. When effectively utilized in …

The interrogation of T cell signaling over the past fifty years has led to the discovery of amazingly intricate cascade networks and elaborate descriptions of individual proteins' domains and functions. A complex landscape has been rendered in which proteins relay messages from the extracellular ligation of the TCR by a cognate peptide loaded MHC via changes in sub-cellular location, phosphorylation, and binding affinities and partners to enact nuclear localization of three key transcription factors required for cellular effector function and proliferation: AP-1, NF-AT, and NF-κB. Background: Machine Learning (ML) has a number of demonstrated applications in protein prediction tasks such as protein structure prediction. To speed further development of machine learning based tools and their release to the community, we have developed a package which characterizes several aspects of a protein commonly used for protein prediction tasks with machine learning.
Findings: A number of software libraries and modules exist for handling protein related data. The package we present in this work, PCP-ML, is unique in its small footprint and emphasis on machine learning. Its primary focus is on characterizing various aspects of a …

The concept of border of document allows us to deploy the significant content of a document in several regimes of meaning. A document can be studied as a semiotic mediation according its communicational intention (intention to inform, intention to signify, intention to inform oneself, intention to interpretate). After studying those values of the document, the question is about whether a documentary border persists unchanged when regimes of meaning are updated. The watershed of the Jordan River stretches over four countries – Jordan, Syria, Israel, and Lebanon – and includes the region of Palestine. With the land and its resources hotly contested by all parties involved, the dwindling supply of drinking and irrigation water from viable sources has led to scrambling exploitation, leaving the river and its connected lakes in an ever-worsening state of deterioration. The Bacterial Pathogen Yersinia Creates A Complex Structure On The Surface Of Macrophages, Wael Bahnan

During an infectious process, bacterial pathogens interact with host innate immune cells. These innate cells (primarily macrophages) play a major role in the antimicrobial defense and bacterial clearance. In this work, we identify the niche which Yersinia pseudotuberculosis creates during macrophage contact. We show that the pYV virulence plasmid is essential for the ability of the bacterium to create an acidified, partially enclosed, and durable niche along the macrophage surface. These compartments are uncoupled from the normal host cell endocytic trafficking pathway and are exposed to the extracellular medium. We present data which shows that the formation of the Yersinia …

Evolution of insect resistance to transgenic crops containing Bacillus thuringiensis (Bt) genes is a serious threat to the sustainability of this technology. However, field resistance related to the reduced efficacy of Bt maize has not been documented in any lepidopteran pest in the mainland U.S. after 18 years of intensive Bt maize planting. Here we report compelling evidence of field resistance in the fall armyworm, Spodoptera frugiperda (J.E. Smith), to Cry1F maize (TC 3507) in the southeastern region of the U.S. An F2 screen showed a surprisingly high (0.293) Cry1F resistance allele frequency in a population collected in 2011 from …

Evolution Of Acyl-Substrate Recognition By A Family Of Acyl-Homoserine Lactone Synthases, Quin H. Christensen, Ryan M. Brecht, Dastagiri Dudekula, E. Peter Greenberg, Rajesh Nagarajan

Members of the LuxI protein family catalyze synthesis of acyl-homoserine lactone (acyl-HSL) quorum sensing signals from S-adenosyl-L-methionine and an acyl thioester. Some LuxI family members prefer acyl-CoA, and others prefer acyl-acyl carrier protein (ACP) as the acyl-thioester substrate. We sought to understand the evolutionary history and mechanisms mediating this substrate preference. Our phylogenetic and motif analysis of the LuxI acyl-HSL synthase family indicates that the acyl-CoA-utilizing enzymes evolved from an acyl-ACP-utilizing ancestor. To further understand how acyl-ACPs and acyl-CoAs are recognized by acyl-HSL synthases we studied BmaI1, an octanoyl-ACP-dependent LuxI family member from Burkholderia mallei, and BjaI, an …
