Kir channels display voltage-dependent block by cytosolic cations such as Mg2+ and polyamines that causes inward rectification. In fact, cations can regulate K channel activity from both the extracellular and intracellular sides. Previous studies have provided insight into the up-regulation of Kir channel activity by extracellular K+ concentration. In contrast, extracellular Mg2+ has been found to reduce the amplitude of the single-channel current at milimolar concentrations. However, little is known about the molecular mechanism of Kir channel blockade by external Mg2+ and the relationship between the Mg2+ blockade and activity potentiation by permeant K+ ions. This study utilized magnetic resonance imaging (MRI) to monitor the real-time status of the urinary bladder in normal and diseased states following cyclophosphamide (CYP)-induced cystitis, and also examined the role of the phosphoinositide 3-kinase (PI3K) pathway in the regulation of urinary bladder hypertrophy in vivo. Our results showed that under MRI visualization the urinary bladder wall was significantly thickened at 8 h and 48 h post CYP injection. The intravesical volume of the urinary bladder was also markedly reduced. Treatment of the cystitis animals with a specific PI3K inhibitor LY294002 reduced cystitis-induced bladder wall thickening and enlarged the intravesical volumes. …

Reproduction And Population Of Porites Divaricata At Rodriguez Key: The Florida Keys, Usa, John Mcdermond

Sexual reproduction in Porites divaricata (Le Sueur, 1820) was studied over a one year period (August, 2012 to August, 2013) in a shallow water (~2m depth) carbonate bank (also described as marginal) habitat at Rodriguez Key, a small island located 1.44 km off the east coast of Key Largo. Porites divaricata is a brooding, gonochoric species with peak reproductive output (planula release) occurring in March and at least a smaller event in May. This species showed an overall female to male sex ratio of 3.5:1. However, during peak reproduction, the sex ratio was 1.2:1. Spawning (sperm release) most likely occurred …

Background. Understanding the life history of a species is essential for fully understanding its role within an ecosystem. However, many of the fish species of high ecological value have not been studied due to their less prominent roles in local recreational and commercial fisheries in comparison to other targeted species. These valuable fishes are also important trophic linkages between small neritic fishes and large, economically valuable apex predators. This paper reviews the contemporary literature of human values and valuation processes, and proposes a re-conceptualized model of a teacher’s professional development. This paper suggests a thorough analysis of the teacher’s personality and devises alternative, well thought-out strategies and approaches to alter self-knowledge, values, valuation processes, and beliefs to imbed the desired impact on their teaching practices. This paper supports the use of deliberate attempts to alter the basic motivation level of teachers and to deliberately expose them to available knowledge to modify their values, attitudes, and actions in their respective professions, organizations, and formal associations. Several groups of microorganisms are capable of producing toxins in aquatic environments. Cyanobacteria are prevalent blue green algae in freshwater systems, and many species produce cyanotoxins which include a variety of chemical irritants, hepatotoxins and neurotoxins. Production and occurrence of potent neurotoxic cyanotoxins β-N-methylamino-L-alanine (BMAA), 2,4-diaminobutyric acid dihydrochloride (DABA), and anatoxin-a are especially critical with environmental implications to public and animal health. Biomagnification, though not well understood in aquatic systems, is potentially relevant to both human and animal health effects. One of the current challenges in the systemic delivery of nanoparticles in cancer therapy applications is the lack of effective tumor localization. Iron oxide nanoparticles coated with crosslinked dextran were functionalized with the tumor homing peptide CREKA, which binds to fibrinogen complexes in the extracellular matrix of tumors. This allows for the homing of these nanoparticles to tumor tissue. The iron oxide nanoparticle core allows for particle heating upon exposure to an alternating magnetic field (AMF) while the dextran coating stabilizes the particles in suspension and decreases the cytotoxicity of the system. We reported last year (Stireman et al. 2013) in The Tachinid Times on our collaborative project, funded by the U.S. National Science Foundation, to study the phylogeny of Tachinidae on a worldwide scale. Since that report, we have made some significant progress towards our goal of assembling a robust phylogenetic tree for Tachinidae. Notably, Cerretti et al. (in press) completed a phylogenetic study using 135 morphological characters from 180 tachinid genera. Results from this study largely reflect, with some interesting exceptions, what we are finding from preliminary analyses of our molecular data. Although sampling of taxa and genes for the …

The rise of “Big Data” analytics in the private sector poses new challenges for privacy advocates. Through its reliance on existing data and predictive analysis to create detailed individual profiles, Big Data has explod-ed the scope of personally identifiable information (“PII”). It has also effectively marginalized regulatory schema by evading current privacy protections with its novel methodology. Furthermore, poor execution of Big Data methodology may create additional harms by rendering inaccurate profiles that nonetheless impact an individual’s life and livelihood. The demands on fathers to be more involved with their children have steadily been increasing since the 1970s. However, fathers have received less attention in the social science literature compared to mothers. This difference is evident regarding the parenting practices of ethnic minority fathers such as nonresident Black fathers. This dissertation investigates the effects of nonresident Black fathers’ perceptions of the coparenting relationship and coparenting support on their perceived father involvement and their perceived paternal stressors; while testing the mediating effects of maternal gatekeeping. Honey bees (Apis mellifera L.) are the most important insects for the pollination of crops and wildflowers. However, they have experienced increasing colony die-offs during the past two decades. Multiple species of parasitic mites have been described that affect honey bees. The most important species in beekeeping belong to the genus Varroa ( Varroa jacobsoniand Varroa destructor). Varroa mite parasitism of honey bees is thought to be the most significant cause of colony mortality worldwide, and mite resistance to active ingredients of acaricides has become common. Sand Point is a small cuspate foreland located along the southeastern shore of Lake Superior within Pictured Rocks National Lakeshore near Munising, Michigan. Park managers’ concerns for the integrity of historic buildings at the northern periphery of the point during the rising lake levels in the mid-1980s greatly elevated the priority of research into the geomorphic history and age of Sand Point. To pursue this priority, we recovered sediment cores from four ponds on Sand Point, assessed subsurface stratigraphy onshore and offshore using geophysical techniques, and interpreted the chronology of events using radiocarbon and luminescence dating. Extreme events such as heat waves, cold spells, floods, droughts, tropical cyclones, and tornadoes have potentially devastating impacts on natural and engineered systems and human communities worldwide. Stakeholder decisions about critical infrastructures, natural resources, emergency preparedness and humanitarian aid typically need to be made at local to regional scales over seasonal to decadal planning horizons. The potential energy surfaces (PES) for the SO3:H2CO and (SO3)2:H2CO complexes were thoroughly examined at the MP2/aug-cc-pVDZ computational level. Heterodimers and trimers are held together primarily by SO chalcogen bonds, supplemented by weaker CHO and/or OC bonds. The nature of the interactions is probed by a variety of means, including electrostatic potentials, AIM, NBO, energy decomposition, and electron density redistribution maps. The most stable dimer is strongly bound, with an interaction energy exceeding 10 kcal mol(-1). Trimers adopt the geometry of the most stable dimer, with an added SO3 molecule situated so as to interact with both of the original …

Synthetic drug-like molecules that directly modulate the activity of key clock proteins offer the potential to directly modulate the endogenous circadian rhythm and treat diseases associated with clock dysfunction. Here we demonstrate that synthetic ligands targeting a key component of the mammalian clock, the nuclear receptors REV-ERBα and β, regulate sleep architecture and emotional behaviour in mice. REV-ERB agonists induce wakefulness and reduce REM and slow-wave sleep. Interestingly, REV-ERB agonists also reduce anxiety-like behaviour. These data are consistent with increased anxiety-like behaviour of REV-ERBβ-null mice, in which REV-ERB agonists have no effect. These results indicate that pharmacological targeting of REV-ERB …

Nuclear magnetic resonance (NMR) spectroscopy has proven invaluable in the diverse field of chemometrics due to its ability to deliver information-rich spectral datasets of complex mixtures for analysis by techniques such as principal component analysis (PCA). However, NMR datasets present a unique challenge during preprocessing due to differences in phase offsets between individual spectra, thus complicating the correction of random dilution factors that may also occur. We show that simultaneously correcting phase and dilution errors in NMR datasets representative of metabolomics data yields improved cluster quality in PCA scores space, even with significant initial phase errors in the data.

D-cycloserine is an effective second line antibiotic used as a last resort to treat multi (MDR)- and extensively (XDR)- drug resistant strains of Mycobacterium tuberculosis. D-cycloserine interferes with the formation of peptidoglycan biosynthesis by competitive inhibition of Alanine racemase (Alr) and D-Alanine-D-alanine ligase (Ddl). Although, the two enzymes are known to be inhibited, the in vivo lethal target is still unknown. Our NMR metabolomics work has revealed that Ddl is the primary target of DCS, as cell growth is inhibited when the production of D-alanyl-Dalanine is halted. This report collects the documents generated as part of a two year effort to catalogue the variety of water quality and quantity data that has been collected in the Columbia Basin Trust region of British Columbia. Through the cataloguing effort, a number of important data and knowledge gaps were identified and specific recommendations developed. The available water quantity and climate data was then used in a number of analyses focused on understanding the relationship between changing climate conditions and stream discharges.

DCs are critical for initiating immunity. The current paradigm in vaccine biology is that DCs migrating from peripheral tissue and classical lymphoid-resident DCs (cDCs) cooperate in the draining LNs to initiate priming and proliferation of T cells. Here, we observe subcutaneous immunity is Fms-like tyrosine kinase 3 ligand (Flt3L) dependent. Flt3L is rapidly secreted after immunization; Flt3 deletion reduces T cell responses by 50%. Flt3L enhances global T cell and humoral immunity as well as both the numbers and antigen capture capacity of migratory DCs (migDCs) and LN-resident cDCs.
