Computational Investigation On The Structural Properties Of Neurofilaments And Their Sidearms, Lakshmi Jayanthi Jan 2014

Computational Investigation On The Structural Properties Of Neurofilaments And Their Sidearms, Lakshmi Jayanthi

Wayne State University Dissertations

Neurofilaments (NFs) are class IV intermediate filaments, abundantly found in the large myelinated axons. They are the key determinants of axonal diameter and consequently the nerve conduction properties. On the other hand, abnormal NF accumulation has been the hallmark of debilitating neurodegenerative disorders. NF compaction is also one of the pathological manifestations of traumatic axonal Injury. However, the exact relation between the disorganized NFs and the etiology of the neurodegenerative disorders is yet to be fully understood. NFs are assembled from three subunits: Low (NFL), Medium (NFM) and Heavy (NFH). These subunits are characterized by a common alpha helical rod …

Microvortices In Droplets: Generation & Applications, Gopakumar Kamalakshakurup Jan 2014

Microvortices In Droplets: Generation & Applications, Gopakumar Kamalakshakurup

Wayne State University Dissertations

The emerging field of droplet microfluidics deals with the manipulation of nL-fL droplets encapsulated within an immiscible carrier phase. The droplets are used as reaction containers for biochemical assays, enabling drastic reduction in assay volumes needed for modern life sciences research. To achieve this, basic laboratory processes such as mixing, detection, and metering must be emulated in the droplet format. Three important unit operations relevant to high throughput screening include 1) the concentration of particles and species within droplets, which is necessary for heterogeneous assays; 2) sensing the biochemical contents of a droplet; and 3) the sorting of droplets based …

An Analytics Approach To Reducing Hospital Readmission, Issac Shams Jan 2014

An Analytics Approach To Reducing Hospital Readmission, Issac Shams

Wayne State University Dissertations

One of the significant sources of waste in the Unites States health care systems is preventable hospital readmission. About 2.3 million Medicare fee-for-service beneficiaries are re-hospitalized within 30 days after discharge which incurs an annual cost of $17 billion. However, it is reported by the Medicare Payment Advisory Commission that about 75% of such readmissions can and should be avoided because they are the results of factors such as poor planning for follow up care transitions, inadequate communication of discharge instructions, and failure to reconcile and coordinate medications. Hence, reducing unnecessary rehospitalization through care transition and systems engineering principles has …

Computational Studies Of Enzymatic Reactions And Inorganic Compounds, Dong Fang Jan 2014

Computational Studies Of Enzymatic Reactions And Inorganic Compounds, Dong Fang

Wayne State University Dissertations

Quantum mechanics (QM) calculations have been demonstrated to give very accurate results in studying a variety of chemical reactions. However, the current computing resources limits its use in large-size systems such as enzymes. Compared to QM, Molecular Mechanics (MM) have an advantage in computing time but disadvantage in accuracy, and MM cannot be used in reactions involving bonding forming and breaking. To combine the advantages of these two types of methods, QM/MM has been introduced to study reactions and processes in large-size systems, and it has been proven to give accurate results with moderate computational expense. When studying enzymatic reactions, …

Associations Between Maternal Maltreatment-Specific Shame, Maternal-Infant Interactions, And Infant Emotion Regulation, Rena A. Menke Jan 2014

Associations Between Maternal Maltreatment-Specific Shame, Maternal-Infant Interactions, And Infant Emotion Regulation, Rena A. Menke

Wayne State University Dissertations

The current study focuses on maltreatment-specific shame as a potential mechanism by which mothers' histories of childhood maltreatment might influence parenting and infant emotion regulation. Shame is a common reaction to childhood maltreatment, and the persistence of maltreatment-specific shame is associated with psychopathology and other psychosocial problems long after the abuse ends (Andrews, Brewin, Rose, & Kirk, 2000; Feiring, Taska, & Lewis, 2002a; Feiring & Taska, 2005). Despite being associated with psychopathology (e.g., depression, PTSD), shame is a conceptually distinct abuse-specific reaction that can interfere with self and interpersonal development (Feiring, Cleland & Simon, 2010; Feiring, Simon, Cleland, 2009; Feiring, …

The Role Of Pdgf C And Its Splice Variant In Breast Cancer, Alyssa Bottrell Jan 2014

The Role Of Pdgf C And Its Splice Variant In Breast Cancer, Alyssa Bottrell

Wayne State University Dissertations

The PDGF family consists of four members; while PDGF A and B are secreted as active dimers, PDGF C and D are secreted as latent dimers that undergo serine protease-mediated extracellular proteolytic activation. Gene expression analysis of breast cancer cell lines showed that PDGF C expression is associated with Basal B subtype breast cancer cells which have cancer stem cell-like characteristics. Furthermore, PDGF C expression is associated with triple-negative (estrogen receptor-, progesterone receptor- and HER2/neu-negative) breast cancer cells, a challenging type of breast cancer to treat. During the course of our study, we discovered a splice variant of PDGF C …

Health-Related Behaviors In Low-Income, Minority Youth: The Role Of Motivation, Basic Needs, Mental Health, And Environmental Barriers, Brittany A. Kohlberger

Wayne State University Dissertations

Engaging in health-related behaviors, such as exercise, healthy eating, and sleep hygiene, are associated with fewer externalizing and internalizing behavior problems and higher self-esteem, suggesting that there may be large benefits to engaging in health-related behaviors during the transitional developmental phase of adolescence (Ong, Wickramaratne, Tang, & Weissmann, 2006; Stathopoulou, Power, Berry, Smits, & Otto, 2006). The purpose of the current study was to identify key psychological processes and external factors that may boost or impede youths' motivation and engagement in health-related behaviors. Specifically, we expected that higher levels of psychological symptoms and environmental barriers will be significantly, negatively associated …

A Cross-Sectional Study Of The Relations Between Parental Factors And Academic Achievement In High School Adolescents, Patricia Marie Lanzon Jan 2014

A Cross-Sectional Study Of The Relations Between Parental Factors And Academic Achievement In High School Adolescents, Patricia Marie Lanzon

Wayne State University Dissertations

Bronfenbrenner's (1979) ecological model was used as the theoretical model for this study of parental achievement factors that influence adolescent academic achievement. Many predictors are related to a child's potential for academic achievement. Of the most proximal are parental and family factors, which were the focus in this study. The proximal parental and family factors for this study were as follows: family structure, maternal and paternal parenting styles and parental involvement, parents' school and community involvement, parental attitude toward achievement, and free and reduced lunch status. Achievement as determined by the student's actual grade point average was the dependent variable. …

Structural And Functional Characterization Of Proteins Of Unknown Function (Hp0495, Hp0100 And Hp1259) In Helicobacter Pylori, Shirin Fatma Jan 2014

Structural And Functional Characterization Of Proteins Of Unknown Function (Hp0495, Hp0100 And Hp1259) In Helicobacter Pylori, Shirin Fatma

Wayne State University Dissertations

H. pylori is missing the glutaminyl- and asparaginyl-tRNA synthetases (GlnRS and AsnRS, respectively). Consequently, H. pylori uses an indirect aminoacylation pathway to generate Gln-tRNAGln and Asn-tRNAAsn. Within this process, Asn-tRNAAsn is produced by misacylation of tRNAAsn with aspartate by a non-discriminating aspartyl-tRNA synthetase (ND-AspRS). Next, the heterotrimeric, glutamine-dependent amidotransferase (called AdT or GatCAB) converts the misacylated Asp-tRNAAsn into Asn-tRNAAsn. A parallel pathway exists for the synthesis of Gln-tRNAGln, wherein misacylation of tRNAGln with glutamate is catalyzed by a tRNAGln-specific glutamyl-tRNA synthetase (GluRS2) to generate Glu-tRNAGln; this misacylated intermediate is converted to Gln-tRNAGln by the same AdT. This dependence on misacylated …

The purpose of this study was to investigate school administrators' responses to bullying situations and explore variables that predict their responses. Elementary, middle and high school administrators (n = 126) from school districts throughout Michigan participated in the study. Data were collected during the 2013-2014 school year.

Differences between beliefs about the causes of victimization were found in that school administrators were less likely to endorse normative beliefs about the causes of victimization than assertive or avoidant beliefs. School administrators were more likely to endorse assertive beliefs for girls than for boys. Empathy towards victims, perceived seriousness of the bullying …

Dendrimer Based Nanotherapeutics For Ocular Drug Delivery, Siva Pramodh Kambhampati Jan 2014

Dendrimer Based Nanotherapeutics For Ocular Drug Delivery, Siva Pramodh Kambhampati

Wayne State University Dissertations

PAMAM dendrimers are a class of well-defined, hyperbranched polymeric nanocarriers that are being investigated for ocular drug and gene delivery. Their favorable properties such as small size, multivalency and water solubility can provide significant opportunities for many biologically unstable drugs and allows potentially favorable ocular biodistribution. This work exploits hydroxyl terminated dendrimers (G4-OH) as drug/gene delivery vehicles that can target retinal microglia and pigment epithelium via systemic delivery with improved efficacy at much lower concentrations without any side effects.

Two different drugs Triamcinolone acetonide (TA) and N-Acetyl Cysteine (NAC) conjugated to G4-OH dendrimers showed tailorable sustained release in physiological relevant …

Center for Advanced Land Management Information Technologies: Publications

Great care is needed to obtain spectral data appropriate for phenotyping in a scientifically rigorous manner. This paper discusses the procedures and considerations necessary and also suggests important pre-processing and analytical steps leading to real-time, non-destructive assessment of crop biophysical characteristics. The system has three major components: (1) data-collection platforms (with a focus on backpack and tractor-mounted units) including specific instruments and their configurations; (2) data-collection and display software; and (3) standard products depicting crop-biophysical characteristics derived using a suite of models to transform the spectral data into accurate, reliable biophysical characteristics of crops, such as fraction of green vegetation, …

Pre- And Post-Harvest Interventions To Reduce Pathogen Contamination In The U.S. Beef Industry, T. L. Wheeler, Norasak Kalchayanand, Joseph M. Bosilevac

Roman L. Hruska U.S. Meat Animal Research Center: Reports

Significant effort has been targeted at reducing the risk of pathogens in U.S. beef products since the mid-1990s. These efforts were focused on Escherichia coli O157:H7 after it was declared an adulterant in ground beef or its components. Post-harvest interventions applied to hides and carcasses by beef processors resulted in significant progress. Effective pre-harvest approaches proved hard to identify and implement. Six additional pathogenic E. coli serogroups were made adulterants in some beef products in 2012 and discussion regarding Salmonella is ongoing. Success to date has resulted from the combination of regulatory, research, and industry efforts to reduce the presence …

The Problem Of Having Both A Body And A Name In The Work Of Jean-Luc Marion: Names, Fathers And The Hopeful Possibilities Of A Queer Phenomenology, Colby Dickinson Jan 2014

The Problem Of Having Both A Body And A Name In The Work Of Jean-Luc Marion: Names, Fathers And The Hopeful Possibilities Of A Queer Phenomenology, Colby Dickinson

Theology: Faculty Publications and Other Works

In this essay, and following upon both Jacques Lacan’s and Jacques Derrida’s personal struggles with fatherhood and the naming of their children, I take up what I consider to be Jean-Luc Marion’s failure to deal with the embodiment of fatherhood through an examination of patriarchal signification, or, specifically, the naming of one’s children after the father—at least insofar as Marion’s brief analysis of this symbolic act points toward his failure to think through the various potential and lived embodiments of the father. I aim to illuminate how his efforts to continue this naming of the child with the father’s name …

Government Regulation: From Independency To Dependency, Part 2, Steven Alan Samson

Faculty Publications and Presentations

What Robert Bellah calls ‘expressive individualism’ has led to unprecedented social legislation in America and expanded government employment since the 1960s, helping to produce a generous supply of public services, policy entrepreneurs, and clientele groups. The legal scholar Lawrence M Friedman notes that ‘the right to be ‘oneself,’ to choose oneself, is placed in a special and privileged position.’ As a consequence, ‘achievement is defined in subjective, personal terms, rather than in objective, social terms.’ When the claims of expressive individualism are considered in tandem with the increasing reach of the modern social service state, a case may be made …

Characterization Of A Human Powered Nebulizer Compressor For Resource Poor Settings, Christopher J. Hallberg, M Therese Lysaught, Christopher E. Zmudka, William K. Kopesky, Lars E. Olson Jan 2014

Characterization Of A Human Powered Nebulizer Compressor For Resource Poor Settings, Christopher J. Hallberg, M Therese Lysaught, Christopher E. Zmudka, William K. Kopesky, Lars E. Olson

Institute of Pastoral Studies: Faculty Publications and Other Works

Respiratory disease accounts for three of the ten leading causes of death worldwide. Many of these diseases can be treated and diagnosed using a nebulizer. Nebulizers can also be used to safely and efficiently deliver vaccines. Unfortunately, commercially available nebulizers are not designed for use in regions of the world where lung disease is most prevalent: they are electricity-dependent, cost-prohibitive, and not built to be reliable in harsh operating conditions or under frequent use.

To overcome these limitations, the Human Powered Nebulizer compressor (HPN) was developed. The HPN does not require electricity; instead airflow is generated manually through a hand-crank …

The recent identification of a novel binding site for angiotensin (Ang) II as the peptidase neurolysin (E.C. 3.4.24.16) has implications for the renin-angiotensin system (RAS). This report describes the distribution of specific binding of 125I-Sarcosine1, Isoleucine8 Ang II (125I-SI Ang II) in neurolysin knockout mouse brains compared to wild-type mouse brains using quantitative receptor autoradiography. In the presence of p-chloromercuribenzoic acid (PCMB), which unmasks the novel binding site, widespread distribution of specific (3 µM Ang II displaceable) 125I-SI Ang II binding in 32 mouse brain regions was observed. Highest levels of binding >700 fmol/g initial wet weight were seen in …

Neighborhood-Level Influences On Delays In Diagnostic Follow-Up From Mammography Screening In African-American Women: A Systematic Review, Barbara A. Fowler Jan 2014

Neighborhood-Level Influences On Delays In Diagnostic Follow-Up From Mammography Screening In African-American Women: A Systematic Review, Barbara A. Fowler

Nursing Faculty Publications

Although there has been a steady decrease in breast cancer deaths among all women (pooled data) age 40 to 65 years during the past two decades in the United States, breast cancer deaths remain substantially higher among African-American women than White women (32.4% vs. 23.9%), resulting in an overall lower five-year survival rate (78.4% vs. 91.2%). These differences are primarily due to delays in diagnostic follow-up from mammography screening (MS) (ACS, 2012]. Following the secondary research data methods by analyzing the popular medical databases and articles published in the peer-reviewed medical journals in US during 2004-2014 the author would like …

Effect Of The Clostridium Difficile Tcde Protein On Membranes Of Escherichia Coli, Sophie Hollenberg Jan 2014

Effect Of The Clostridium Difficile Tcde Protein On Membranes Of Escherichia Coli, Sophie Hollenberg

Inquiry: The University of Arkansas Undergraduate Research Journal

Clostridium difficile infects millions of hospitalized patients worldwide, causing pseudomembranous colitis, a disease with symptoms attributable to two toxins produced by the bacterium. The PaLoc gene locus in the C. difficile genome is responsible for toxin production and activation, but it is not known how the toxins are released from the pathogen. Within PaLoc is a gene for the TcdE protein, which shares striking similarity to a class of membrane pore-forming proteins called holins. To assess whether TcdE catalyzes toxin release, the engineered tcdE gene was expressed in Escherichia coli. An arabinose promoter upstream of the tcdE gene was manipulated to …

Distribution Of Intravascular And Extravascular Extracellular Volume Fractions By Total Area Under Curve For Neovascularization Assessment By Dynamic Contrast-Enhanced Magnetic Resonance Imaging, Yi Sun

Publications and Research

In this paper, we propose and investigate distribution of intravascular and extravascular extracellular volume fractions (DIEEF) as a noninvasive biomarker for neovascularization assessment by dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI). A generalized two-compartment exchange model (G2CXM) that uniformly includes the Patlak model, Tofts model, extended Tofts model, and recent two-compartment exchange model as special instances is first presented. Based on the total area under curve of the G2CXM a method of DIEEF estimation without knowing the artery input function is proposed. The mean square error of DIEEF estimate in the presence of noise and with incomplete DCE-MRI data is analyzed. …

The Three Dimensional Shape And Roughness Of Mineral Dust, Xinxin Woodward

Dissertations, Master's Theses and Master's Reports - Open

Mineral dust shape and roughness are important for a multitude of processes; it is known for aspherical shape but the true measurements in three dimensions are rare. Atomic Force Microscope was used for determine both 3D shape and roughness for two dust which are commonly used in laboratory experiments – Arizona Test Dust (ATD) and Kaolinite. We determined both of them are rather flat and round; an oblate spheroid would be a good model. Loess Filter was used to smooth the particles' surface and correlation analysis was used to examine the surfaces' properties of the dust; we found no features …
