Liver receptor homolog 1 (LRH1), directs the development and differentiation of embryonic pancreas, and is overexpressed in pancreatic cancer (PC). We hypothesized that LRH1 promotes PC growth. Cell proliferation and tumorigenicity in nude mice were compared between empty vector-transfected (control) and stable LRH1-overexpressed PC cell lines. The subsequent tumor burden, vasculature development, and histologic features were evaluated. LRH1 overexpression enhanced the expression of downstream target genes (cyclin D1/E1) and stimulated cell proliferation in PC cell lines. LRH1 upregulated cyclin E1 truncated T1/T2 isoforms expression which may occur through ERα–calpain1 signaling. Compared with the control, LRH1 overexpressing stable cells generated tumors …

The preventive effects of the American cranberry (Vaccinium macrocarpon) against urinary tract infections are supported by extensive studies which have primarily focused on its phenolic constituents. Herein, a phenolic-free carbohydrate fraction (designated cranf1b-F2) was purified from cranberry fruit using ion exchange and size exclusion chromatography. MALDI-TOF-MS analysis revealed that the cranf1b-F2 constituents are predominantly oligosaccharides possessing various degrees of polymerisation and further structural analysis (by GC–MS and NMR) revealed mainly xyloglucan and arabinan residues. In antimicrobial assays, cranf1b-F2 (at 1.25 mg/mL concentration) reduced biofilm production by the uropathogenic Escherichia coli CFT073 strain by over 50% but did not …

Flavin-containing monooxygenase-3 (FMO3) catalyzes metabolic reactions similar to cytochrome P450 monooxygenase, however, most metabolites of FMO3 are considered non-toxic. Recent findings in our laboratory demonstrated Fmo3 gene induction following toxic acetaminophen (APAP) treatment in mice. The goal of this study was to evaluate Fmo3 gene expression in other diverse mouse models of hepatic oxidative stress and injury. Fmo3 gene regulation by Nrf2 was also investigated using Nrf2 knockout (Nrf2 KO) mice. In our studies, male C57BL/6J mice were treated with toxic doses of hepatotoxicants or underwent bile duct ligation (BDL, 10 days). Within its 150 year history, public health has grown from a focus on local communities, to include country-wide, then international and now global perspectives. Using the United Nation’s Millennium Goals as its primary framework, this article provides an overview of global public health within the broadest possible context of the world and all of its peoples. Also provided are discussions of the global burden of disease as measured in disability-adjusted life years (DALYs), global health statistics, current health priorities and recommendations for action by social workers and other health professionals.

Introduction

Vein graft adaptation to the arterial circulation entails many changes at the cellular/molecular levels, including both cell proliferation and extracellular matrix (ECM) deposition/remodeling, resulting in thickening of the vein wall, and in excessive cases, pathological neointimal hyperplasia. Eph-B4 is a receptor tyrosine kinase and embryonic determinant of veins that, when stimulated, has been shown by our lab to decrease vein graft wall thickening. We hypothesize that among other mechanisms, Eph-B4 exerts this effect through modulating extracellular matrix production and/or degradation, and that Eph-B4 interacts directly with eNOS to mediate some of its downstream effects.

Objective. To determine if poractant alfa (Curosurf®) could serve as a viable vehicle for silencing RNA (siRNA) delivery. Specifically, we hypothesize that use of Curosurf® as an enhanced delivery vehicle for C/EBP homologous protein (CHOP) and Angiopoietin 2 (Ang2) siRNA would augment gene silencing, preventing cell death and bronchopulmonary dysplasia (BPD) in the mouse lung.

Design. Transfection efficiency of Curosurf® was compared to naked siRNA and Lipofectamine 2000®. MLE-12 cells were transfected with CHOP and Ang2 siRNA using Curosurf®, then exposed to hyperoxic conditions. Cultures were evaluated for respective protein expression and markers of cell death. The objective of this project is to report oncologic and functional outcomes for a cohort of patients with supraglottic squamous cell carcinoma (SSCC) treated in a multidisciplinary setting including the use of transoral laser microsurgery (TLM).

A retrospective observational study at Yale New Haven Hospital, academic teaching hospital was performed. A total of 56 patients without evidence of distant metastasis at presentation treated for SSCC between January 1, 2003 and December 31, 2007 were identified. The main outcome measures include overall survival (OS), locoregional and distant recurrence, and the incidence of tracheostomy and gastrostomy tube requirement.

Dupuytren's disease is a common heritable connective tissue disorder of poorly understood etiology. It is thought that oxidative stress pathways may play a critical role in the development of Dupuytren's disease, given the various disease associations that have been observed. We sought to sequence the mitochondrial and nuclear genomes of patients affected with Dupuytren's disease using next-generation sequencing technology to potentially identify genes of potential pathogenetic interest. Additionally, we sought to compare the genomes between diseased tissue and blood to look for potential somatic mutations in the diseased tissue. Background: Isocyanates are one of the most commonly reported causes of occupational asthma; however, the risks of developing isocyanate asthma in modern production facilities remain poorly defined. The purpose of the present investigation was to evaluate TDI exposure and respiratory health among an inception cohort of workers during their first year of employment at a new polyurethane foam production factory.

Methods: Forty-nine newly hired workers were evaluated pre-employment, 6-months, and 12-months post-employment through questionnaire, spirometry and TDI-specific serology. 30 years ago, the Church of Ireland allowed women’s ordination as deacons, then later priests and bishops. Leading to this doctrinal and social development, the Church of Ireland wrestled with big questions on the nature of Christian ministry and women’s roles in the church. Have the Bible and church tradition always upheld male headship? Are women being called by God to serve as deacons, priests and bishops? Can groups with major theological differences reconcile and remain one church?
