Cell-Selective Knockout And 3d Confocal Image Analysis Reveals Separate Roles For Astrocyte- And Endothelial-Derived Ccl2 In Neuroinflammation, Debayon Paul, Shujun Ge, Yen Lemire, Evan R. Jellison, David R. Serwanski, Joel S. Pachter

UCHC Articles - Research

Background

Expression of chemokine CCL2 in the normal central nervous system (CNS) is nearly undetectable, but is significantly upregulated and drives neuroinflammation during experimental autoimmune encephalomyelitis (EAE), an animal model of multiple sclerosis which is considered a contributing factor in the human disease. As astrocytes and brain microvascular endothelial cells (BMEC) forming the blood–brain barrier (BBB) are sources of CCL2 in EAE and other neuroinflammatory conditions, it is unclear if one or both CCL2 pools are critical to disease and by what mechanism(s).

Methods

Mice with selective CCL2 gene knockout (KO) in astrocytes (Astro KO) or …

Pregnancy is a critical time to identify and address maternal mental health problems, for the health of both mother and child. Pregnant women with histories of exposure to interpersonal psychological trauma may experience a range of mental health problems including but not limited to posttraumatic stress disorder (PTSD). In a community sample of 1,581 pregnant women, 25% reported symptoms consistent with at least one of six syndromes, including PTSD, major depressive disorder (MDD), generalized anxiety disorder (GAD), or clinically significant dissociation, somatization, or affect dysregulation. Six sub-groups with distinct mental health problem profiles were identified by cluster analysis. Controlling for …

A Systems Biology Approach To Iron Metabolism, Reinhard C. Laubenbacher, Suzy V. Torti Jan 2014

A Systems Biology Approach To Iron Metabolism, Reinhard C. Laubenbacher, Suzy V. Torti

UCHC Articles - Research

Iron is critical to the survival of almost all living organisms. However, inappropriately low or high levels of iron are detrimental and contribute to a wide range of diseases. Recent advances in the study of iron metabolism have revealed multiple intricate pathways that are essential to the maintenance of iron homeostasis. Further, iron regulation involves processes at several scales, ranging from the subcellular to the organismal. This complexity makes a systems biology approach crucial, with its enabling technology of computational models based on a mathematical description of regulatory systems. Systems biology may represent a new strategy for understanding imbalances in …

Sustainable Energy In The Galapagos, Sean Dove Jan 2014

Sustainable Energy In The Galapagos, Sean Dove

Bridges: A Journal of Student Research

As the global population continues to grow, so does the dependency and need for energy. For decades, the solution to satisfy the world's demand has been the use of cheap, abundant, and inefficient sources such as coal and oil, but these sources are contributing to massive amounts of environmental degradation and the emission of greenhouse gases. The Galapagos Islands have pledged to do away with fossil fuels in an innovative "Zero Fuels Initiative" in order to help combat environmental damage to its own pristine habitats that make Galapagos one of the most sought out destinations in the world for scientific …

'I'M Coming Back Again!' The Resilience Process Of Early Career Teachers, Caroline Mansfield, Susan Beltman, Anne Price

Education Papers and Journal Articles

Early career teachers face a range of challenges in their first years of teaching and how these challenges are managed as career implications. Based on current literature, this paper presents a model of early career teacher resilience where resilience is seen as a process located at the interface of personal and contextual challenges and resources. Through a semi-structured interview the challenges faced by 13 Australian early career teachers and the resources available to manage these challenges are examined. Findings show that beginning teachers experience multiple, varied and ongoing challenges and that personal and contextual resources are both important in sustaining …

Specific Localization Of Β-Arrestin2 In Myenteric Plexus Of Mouse Gastrointestinal Tract, Hercules T. Maguma, Dipanjana D. De, Sukhada Bhave, William L. Dewey, Hamid I. Akbarali

Pharmacology and Toxicology Publications

Abstract

β-arrestin2 is a key molecule involved in signaling and internalization of activated G protein-coupled receptors including µ-opioid receptors (MOR). Previously we have shown that decreased expression of β-arrestin2 upon chronic morphine is associated with the development of opioid tolerance in the gastrointestinal tract. However, the localization of β-arrestin2 within the gastrointestinal wall is not known. In this study we found that β-arrestin2 is localized in the soma of a select group of neurons in the myenteric ganglia but not in smooth muscle. The density of β-arestin2 was significantly higher in the ileum than the colon. We identified four variants …

Activation Of Nlrp3 Inflammasomes Enhances Macrophage Lipid-Deposition And Migration: Implication Of A Novel Role Of Inflammasome In Atherogenesis, Xiang Li, Yang Zhang, Min Xia, Erich Gulbins, Krishna M. Boini, Pin-Lan Li Jan 2014

Activation Of Nlrp3 Inflammasomes Enhances Macrophage Lipid-Deposition And Migration: Implication Of A Novel Role Of Inflammasome In Atherogenesis, Xiang Li, Yang Zhang, Min Xia, Erich Gulbins, Krishna M. Boini, Pin-Lan Li

Pharmacology and Toxicology Publications

Although Nlrp3 inflammasome activation in macrophages has been shown to be critical for the development of atherosclerosis upon atherogenic stimuli, it remains unknown whether activated Nlrp3 inflammasomes by other non-atherogenic stimuli induce alterations in macrophages that may contribute in the concert with other factors to atherogenesis. Thus, the present study tested the hypothesis that activation of Nlrp3 inflammasomes by ATP, which is a classical non-lipid danger stimulus, enhances the migration of macrophage and increases lipids deposition in macrophages accelerating foam cell formation. We first demonstrated that extracellular ATP (2.5 mM) markedly increased the formation and activation of Nlrp3 inflammasomes in …

Habitat Associations Of Fish Assemblages In The Cache River, Illinois, Kristen L. Bouska, Gregory Whitledge Jan 2014

Habitat Associations Of Fish Assemblages In The Cache River, Illinois, Kristen L. Bouska, Gregory Whitledge

Publications

Fish and habitat were sampled by state agencies at 48 stations throughout the Cache River watershed, Illinois between 1992 and 2009. Two distinct fish assemblages were identified, one primarily found in the lower mainstem Cache River and a second found throughout tributaries and the upper mainstem Cache River. Using a canonical correspondence analysis, the distribution of fish species was largely explained by substrate, land use, drainage area and local habitat features. Creek chub, central stoneroller, fringed darter and fantail darter are species found to be positively associated with gravel substrate and forest. In contrast, black buffalo, gizzard shad, smallmouth buffalo, …

A Review Of Recent Perspectives On Biomechanical Risk Factors Associated With Anterior Cruciate Ligament Injury, Gerwyn Hughes Jan 2014

A Review Of Recent Perspectives On Biomechanical Risk Factors Associated With Anterior Cruciate Ligament Injury, Gerwyn Hughes

Kinesiology (Formerly Exercise and Sport Science)

There is considerable evidence to support a number of biomechanical risk factors associatiated with non-contact anterior cruciate ligament (ACL) injury. This paper aimed to review these biomechanical risk factors and highlight future directions relating to them. Current perspectives investigating trunk position and relationships between strength, muscle activity and biomechanics during landing/cutting highlight the importance of increasing hamstring muscle force during dynamic movements through altering strength, muscle activity, muscle length and contraction velocity. In particular, increased trunk flexion during landing/cutting and greater hamstring strength are likely to increase hamstring muscle force during landing and cutting which have been associated with reduced …

Modeling Tidal Marsh Distribution With Sea-Level Rise: Evaluating The Role Of Vegetation, Sediment, And Upland Habitat In Marsh Resiliency, Lisa M. Schile, John Callaway, J T. Morris, Diana Stralberg, V Thomas Parker, Maggi Kelly Jan 2014

Modeling Tidal Marsh Distribution With Sea-Level Rise: Evaluating The Role Of Vegetation, Sediment, And Upland Habitat In Marsh Resiliency, Lisa M. Schile, John Callaway, J T. Morris, Diana Stralberg, V Thomas Parker, Maggi Kelly

Environmental Science

Tidal marshes maintain elevation relative to sea level through accumulation of mineral and organic matter, yet this dynamic accumulation feedback mechanism has not been modeled widely in the context of accelerated sea-level rise. Uncertainties exist about tidal marsh resiliency to accelerated sea-level rise, reduced sediment supply, reduced plant productivity under increased inundation, and limited upland habitat for marsh migration. We examined marsh resiliency under these uncertainties using the Marsh Equilibrium Model, a mechanistic, elevation-based soil cohort model, using a rich data set of plant productivity and physical properties from sites across the estuarine salinity gradient. Four tidal marshes were chosen …

Abstract The pygmy hippopotamus Choeropsis liberiensis is an Endangered species found only in the Upper Guinea rainforests of West Africa. Using a two-phase approach, with initial semi-structured interviews followed by more extensive questionnaires, we examined local residents’ awareness of and attitudes towards the pygmy hippopotamus along the Moa River near Tiwai Island Wildlife Sanctuary in Sierra Leone. The interviews and questionnaires addressed human–hippopotamus interactions, local knowledge and awareness of pygmy hippopotamus ecology and behaviour, and public attitudes towards hippopotamus conservation. Overall, 22% of questionnaire respondents acknowledged benefits related to hippopotamus conservation; factors affecting the perception of benefits included age, livestock …

Evidence For Bluegill Spawning Plasticity Obtained By Disentangling Complex Factors Related To Recruitment, Mark A. Kaemingk, Kristopher J. Stahr, Jeffrey C. Jolley, Richard S. Holland, David W. Willis Jan 2014

Evidence For Bluegill Spawning Plasticity Obtained By Disentangling Complex Factors Related To Recruitment, Mark A. Kaemingk, Kristopher J. Stahr, Jeffrey C. Jolley, Richard S. Holland, David W. Willis

School of Natural Resources: Faculty Publications

Fishes can exhibit many forms of plasticity to maximize fitness. However, limited information exists on the ability of freshwater fish to adjust spawning behavior and characteristics (e.g., timing, duration, magnitude of spawning events) to minimize mortality of recruits and ultimately maximize fitness.Wewanted to test the life history hypothesis for bluegill (Lepomis macrochirus) (i.e., opportunistic strategy) utilizing existing literature and results from our study to further evaluate the potential for spawning plasticity in this species. Our objective was to identify bluegill recruitment bottlenecks (i.e., periods of high mortality) and factors associated with these events in a single lake during …

Temperature, Hatch Date, And Prey Availability Influence Age-0 Yellow Perch Growth And Survival, Mark A. Kaemingk, Brian D. S. Graeb, David W. Willis Jan 2014

Temperature, Hatch Date, And Prey Availability Influence Age-0 Yellow Perch Growth And Survival, Mark A. Kaemingk, Brian D. S. Graeb, David W. Willis

School of Natural Resources: Faculty Publications

Throughout their range, Yellow Perch Perca flavescens are an important ecological and economic component of many fisheries, but they often exhibit highly variable recruitment. Much research effort has been devoted to better understanding the mechanisms responsible for these erratic recruitment patterns, yet few studies have examined this process at the detail necessary to reveal complex interactions that may exist across multiple early life stages. Our current understanding of the early life recruitment patterns of Yellow Perch suggests a strong abiotic component. Using existing information, we developed three working hypotheses to examine Yellow Perch recruitment at two larval stages (5–14 and …

Does Human Predation Risk Affect Harvest Susceptibility Of White-Tailed Deer During Hunting Season?, Andrew R. Little, Stephen Demarais, Kenneth L. Gee, Stephen L. Webb, Samuel K. Riffell, Joshua A. Gaskamp, Jerrod L. Belant

School of Natural Resources: Faculty Publications

Large carnivores are considered a primary source of mortality for many ungulate populations, but harvest by hunters is the primary means of population management. However, research is needed to evaluate how human predation risk influences observability (a surrogate to harvest susceptibility) of ungulates. We determined how hunting intensity and duration influence observation rates of white-tailed deer (Odocoileus virginianus) and how deer behavior (i.e., movement rate and resource selection) affects observation rates. We sampled 37 adult (≥2 yr) male deer at 2 levels of risk (i.e., low-risk = 1 hunter/101 ha; and high-risk = 1 hunter/30 ha) during 3 …

Unscreened Water-Diversion Pipes Pose An Entrainment Risk To The Threatened Green Sturgeon, Acipenser Medirostris, Timothy D. Mussen, Dennis Chocherell, Jamilynn B. Poletto, Jon S. Reardon, Zachary Hockett, Ali Ercan, Hossein Bandeh, M. Levent Kavvas, Joseph J. Cech Jr., Nann A. Fangue

School of Natural Resources: Faculty Publications

Over 3,300 unscreened agricultural water diversion pipes line the levees and riverbanks of the Sacramento River (California) watershed, where the threatened Southern Distinct Population Segment of green sturgeon, Acipenser medirostris, spawn. The number of sturgeon drawn into (entrained) and killed by these pipes is greatly unknown. We examined avoidance behaviors and entrainment susceptibility of juvenile green sturgeon (3560.6 cm mean fork length) to entrainment in a large (.500-kl) outdoor flume with a 0.46-m-diameter water-diversion pipe. Fish entrainment was generally high (range: 26–61%), likely due to a lack of avoidance behavior prior to entering inescapable inflow conditions. We estimated that …
