RNA therapeutics are a rising drug category with potential use for a range of conditions encompassing infectious diseases to therapies for cancer, diseases, and genetic disorders. RNA-lipid nanoparticles (RNA-LNPs) are the prominent delivery method for these therapeutics approved products include mRNA vaccines and polyneuropathy treatments. The emergency use authorizations and orphan drug status of current RNA-LNP drugs has allowed approval without finalization of the regulatory analytical procedures for quality monitoring. Background: Autonomic dysfunction represents one of the major non-motor manifestations of Parkinson’s disease (PD) and can negatively influence patients’ health-related quality of life (QoL). This study investigated the frequency and severity of autonomic symptoms in individuals with PD and examined their relationship with QoL in a single-center cohort.
Methods: Thirty patients with idiopathic PD were assessed for autonomic dysfunction, quality of life, and motor severity using validated PD–specific rating scales.
Results: Among the autonomic subdomains, urinary symptoms demonstrated the greatest degree of impairment (median score 41.67%) and showed the strongest correlation with overall PDQ-39 scores (r=0.605, p≤0.001). Higher …

Natural products remain a rich source of structurally diverse bioactive compounds for drug discovery, particularly for pain therapeutics. However, the chemical complexity of natural product extracts presents significant challenges for the efficient identification and prioritization of bioactive metabolites. This dissertation integrates untargeted LC-MS/MS-based metabolomics, molecular networking, classical natural products chemistry, and neurobiological assays to accelerate the discovery of analgesic lead compounds targeting the emerging pain-associated receptor σ₂R/TMEM97.

This study evaluates the fate and removal efficiency of Escherichia coli (E. coli) across both liquid and sludge treatment processes at a full-scale activated sludge water resource recovery facility (WRRF) in Brookings, South Dakota. Understanding pathogen reduction within wastewater treatment systems is critical for protecting public health and ensuring regulatory compliance, particularly in facilities practicing water reuse and biosolids management. Liquid and sludge samples were collected from multiple treatment stages between January and July 2025 and analyzed using the IDEXX Colilert Quanti-Tray 2000 method. This article aims to compare the osteogenic along with anti-inflammatory activity of different concentrations of pomegranate extract (PG) on human periodontal ligament-derived stem cells (hPDLSCs) in vitro.
Materials and Methods
hPDLSCs were acquired from the root surface of removed molars, cultured, and then distinguished by flow cytometry analysis and several lineage differentiation potentials. This study assessed the biological and antimicrobial properties of a calcium silicate-based material and calcium hydroxide (CH) when used as intracanal medications. MTT assay, osteogenic differentiation of human periodontal stem cells (hPDLSCs), cell mineralization-assay, and determination of ALP activity were assessed to investigate the biological properties. While the agar well diffusion, crystal violet (CRV) assay and LIVE/DEAD staining of dentin slices infected with a mature E. faecalis biofilm were used to assess the antimicrobial properties. Aim: The aim of this study is to investigate the effectiveness of the Attention U-Net architecture for teeth segmentation in panoramic dental images. By exploring the potential of Attention U-Net, the study aims to contribute to the development of more precise and automated dental imaging analysis, which could enhance diagnosis, treatment planning, and computer-aided dental systems. The Eocene-Oligocene period in the North American Cordillera represents a period of great intrigue with complex interactions among tectonics, climate, and surface processes. Although this period is generally associated with synorogenic extension associated with gravitational collapse of the Cordillera, the paleotopographic history remains debated. Existing studies offer conflicting interpretations, suggesting either sustained elevations or modest uplift during this time. Notably, recent reconstructions based on volcanic glass δD values have proposed renewed uplift during the early Oligocene, which needs to be tested with a more extensive and robust dataset. Pregnancy physiology may impact indicators of hydration, affecting exposure assessment in gestational studies with urinary chemical biomarkers.
Methods:
We aimed to characterize hydration and demonstrate the impact of different methods for standardizing urinary chemical biomarker concentrations on exposure descriptives in the Human Placenta and Phthalates Study (n = 303, 2017-2020), a prospective pregnancy cohort with eight study visits between 12 and 38 weeks of gestation. Atlantic Bluefin Tuna, Thunnus thynnus (ABFT), migrate long distances to broadcast spawn in the Gulf of Mexico (GOM) basin, the Mediterranean Sea and the Slope Sea (NW Atlantic). These areas have commonalities including a highly dynamic upper ocean eddy environment that draws nutrients from shallow thermoclines and adjacent continental shelves and can provide temporal sanctuaries from some larval predators. In the GOM basin, the Loop Current (LC) and its energetic spin—off eddies sweep weak swimming predators such as jellyfish (Pelagia noctiluca) into lines and aggregations, leaving relatively open areas as temporal sanctuaries for ABFT larvae during the vulnerable …

Southern oyster drill snails (Stramonita haemastoma complex) are nuisance organisms within the Gulf of Mexico. This is due to their damaging impact on recovering and farmed oyster (Crassostrea virginica) populations, which are one of the drills’ main food sources. Oyster reef recovery and increased aquaculture production are priorities for Mississippi and the wider northern Gulf coast. The Grand Bay National Estuarine Research Reserve (NERR) hosts multiple oyster reef restoration sites within its boundaries, yet no comprehensive survey of oyster drill abundance has been conducted in this area to date. Hard bottom coral habitats in the mesophotic zone are ecologically and economically important systems, yet questions remain about the relative abundances of benthic organisms and trophic resources available to them. Between 2006 and 2012, seagrass coverage across Sarasota Bay expanded by 1,108 ha, a 28% increase. During the subsequent years of 2012−2020, 830 ha of seagrass were lost, a 16% decline. In response to these findings, resource management efforts focused on determining the likely reason(s) why those 2 periods had such contrasting trends, to come up with strategies to reverse these losses. Based on results from the seagrass data, the 7—y period of 2006−2012 was designated a “reference period” and water quality data were compared against the subsequent 7—y designated “degraded period” of 2013−2019. Background: Short sleep and formula feeding during infancy are associated with increased risk of childhood obesity. Feeding practices and sleep arrangements vary during infancy and may also be dynamic, yet their impact on infant night sleep duration remains unclear. Understanding these relationships is crucial for formulating recommendations to support breastfeeding and address sleep concerns. Background
Suppressing estradiol (E₂) during baboon pregnancy lowers offspring skeletal muscle capillary density, vital for insulin-mediated glucose uptake, and induces insulin resistance. We examined whether E₂ deprivation also impairs microvascular flow and cardiac function.
Methods
Offspring of untreated baboons, letrozole-treated baboons, or letrozole plus E₂ (maternal s.c. injections during the second half of gestation) underwent contrast-enhanced microbubble ultrasonography to quantify microvessel flow and echocardiography to assess cardiac performance.
Results
Letrozole reduced maternal serum E₂ by 95% (p < 0.01). In letrozole offspring, microbubble flux rate (β) fell 55% (p < 0.02), replenishment was 5 s slower (p < 0.03), and microvessel flow declined 40% (p = 0.05); all were restored by added E₂. Indices of systolic (isovolumic contraction), diastolic (isovolumic relaxation), global performance (Tei index), and cardiac output were unchanged.
Conclusion
Prolonged gestational E₂ deprivation programs a reduction in microvascular flow without altering cardiac function; maternal E₂ prevents this, supporting E₂'s role in …

The hydrothermal liquefaction (HTL) process offers an energetic advantage over pyrolysis because it does not require prior drying of the biomass feedstock. However, there are significant challenges in simultaneously estimating both the yields and characteristics of products from the HTL of biomass with theoretical support. Objectives
Urinary extracellular vesicles (uEVs) are potential biomarkers for renal physiology and disease. Their isolation is challenged by Tamm-Horsfall protein (THP) contamination, particularly in high-speed ultracentrifugation (P100) fractions. Low-speed (P20) fractions contain biologically relevant uEVs, yet their proteomic composition is underexplored. This study evaluates strategies for THP removal to optimize mass spectrometry analysis and characterizes P20 and P100 uEV proteomes and its combination in healthy urine.
Methods

[Introduction] As the guest editor of the Special Issue on Ovarian Cancer Immunology Immunotherapy, I would like to include certain brief comments on the current state of the field and to introduce the Special Issue. Ovarian carcinoma and malignant melanoma are the first human tumors where immune responses, primarily restricted to autologous tumor cells, were first demonstrated. T-cell lines and clones developed in recombinant interleukin-2 (rIL-2) from tumor-infiltrating lymphocytes (TIL) from these tumors exhibited cytotoxicity and/or cytokine production, primarily restricted to autologous tumor cells. The increasing abundance of ticks poses a growing public health concern due to heightened human exposure to tick bites. The lone star tick (Amblyomma americanum: Ixodida: Ixodidae), a common human-biting species in the United States, has expanded its range in recent years. However, how its different life stages vary across time, habitats, and locations remains insufficiently understood. We analyzed tick abundance data collected in southeastern Virginia between 2009 and 2018, focusing on larval, nymphal, and adult life stages. Pain is a pervasive global health concern that affects approximately one in five adults worldwide and carries substantial psychological and economic costs. Chronic and persistent pain often co-occurs with emotional dysregulation and maladaptive coping behavior, suggesting the involvement of complex biopsychosocial mechanisms including personality pathology. Specifically, borderline personality disorder (BPD) features, have been associated with altered pain perception, increased emotional reactivity, and higher engagement in non-suicidal self-injury (NSSI). There is evidence to show that longevity and associated phenotypes of successful aging are partially determined by genetic factors (Adams et al. 2008; Barzilai et al. 2006; Barzilai et al. 2003; Murabito et al. 2013; Newman et al. 2011). Professional guidelines emphasize the importance of adapting evidence-based treatments to the cultural and contextual needs of minoritized populations. Although substantial research has examined lesbian, gay, bisexual, transgender, and queer (LGBTQ+) mental health, comparatively little is known about the psychotherapy experiences of Chinese sexual and gender minority individuals or how dialectical behavior therapy (DBT) may be delivered responsively with this population. DBT is an evidence-based treatment for emotion dysregulation, self-harm, and suicidal behavior that explicitly addresses the effects of invalidating environments. Bone regeneration is a multistage and tightly regulated process driven by coordinated interactions between the immune and skeletal systems. While inflammation is essential for initiating repair, its dysregulation contributes to delayed union, nonunion, and impaired healing commonly seen in trauma, chronic disease, and aging. Traditional interventions, including autografts and inert biomaterials, provide structural support but fail to engage or modulate the immune microenvironment that governs successful regeneration. Emerging research in osteoimmunology has revealed the centrality of immune cell populations such as macrophages, T regulatory cells, dendritic cells, and myeloid-derived suppressor cells in directing osteogenesis, angiogenesis, and the resolution of inflammation. …

The field of sustainability studies increasingly asks students to confront ecological crises alongside considerations of power, privilege, and unequal environmental harm. This work can be emotionally challenging as well as intellectually complex. Contemplative practices, such as meditation and yoga, may offer meaningful pedagogical support, helping students cultivate the attention, reflection, and emotional capacity needed to stay engaged with justice-centered sustainability learning. Although the use of contemplative pedagogies is growing in higher education, little research has examined how these practices shape justice-orientated learning outcomes in sustainability education. Across the United States, air pollution disproportionately impacts minority groups. California’s Advanced Clean Cars II (ACC II) waiver was positioned to lead to significant environmental justice impacts, such as reductions in emissions and adverse health outcomes, by establishing stricter regulations for vehicle emissions and ensuring that 100 percent of new passenger vehicle sales are zero-emission by 2035. As the federal government recently nullified ACC II, pollution, and by extension health, could worsen, especially in disadvantaged communities. Artificial intelligence (AI) holds promise for healthcare, but real-world implementation remains difficult. The Mayo clinic platform (MCP) addresses this by providing scalable, multi-institutional, de-identified data and analytical tools. Through four research projects, we demonstrate MCP's ability to support efficient cohort identification, AI model development, and real-world evidence generation. MCP enables broader accessibility and standardization compared to institutional EHRs, positioning it as a powerful platform for advancing translational research and precision medicine.
