Microbial communities are ubiquitous and carry out valuable functions in the environment. Decomposition of leaf material by microbial communities is important to return nutrients back to both terrestrial and aquatic organisms. Perturbations to the environment like the arrival of invasive species can have an impact on the structure and functions of the microbial community. The round goby (Neogobius melanostomus) is a Ponto-Caspian fish introduced into the Great Lakes which has since secondarily invaded tributary streams and rivers. Studies have shown they alter invertebrate communities, and these alterations have impacted organic matter decomposition. Stream studies suggest leaf litter decomposes …

Eastern hellbender (Cryptobranchus alleganiensis alleganiensis) population size has declined throughout much of its range. Previous captive-release headstarting programs have resulted in minimal success, presumably due to movement of captive-released animals away from the release site. This study aimed to increase the success of hellbender headstarting programs by assessing the effectiveness of three release methods. Releases were conducted in two stream sites within the Allegheny River drainage. Streams were similar; however stream A contained a higher boulder density. Mutation of the levy gene on chromosome 2 of Drosophila melanogaster has previously been shown to cause temperature-induced paralysis and neurodegeneration. The Suppressor of levy mutation, Su(levy), also on chromosome 2, modulates the effects of the levy mutation and partly rescues the wild-type phenotype. The goal of this research was to determine if Su(levy) mutation alleviates the effects of neurodegeneration caused by mutations associated with Alzheimer’s and Parkinson’s diseases in humans. The mutant genes used were hLRRK2, α-Synuclein, DJ1-α, Pink, Parkin, and Aβ-42. To determine this, flies carrying the above-mentioned mutant genes were …

Similar to other complex sequences of muscle activity, swallowing relies heavily upon ‘sensorimotor integration.’ It is well known that the premotor cortex and primary sensorimotor cortices provide critical sensorimotor contributions that help control the strength and timing of swallowing muscle effectors. However, the temporal dynamics of sensorimotor integration remains unclear, even when performed normally without neurological compromise. Overexpression of HOXB4 in hematopoietic stem cells (HSCs) leads to increased self-renewal without causing hematopoietic malignancies in transplanted mice. The molecular basis of HOXB4-mediated benign HSC expansion in vivo is not well understood. To gain further insight into the molecular events underlying HOXB4-mediated HSC expansion, we analyzed gene expression changes at multiple time points in Lin-Sca1+c-kit+ (LSK) cells from mice transplanted with bone marrow (BM) cells transduced with a MSCV-HOXB4-ires-YFP vector. A distinct HOXB4 transcriptional program was reproducibly induced and stabilized by 12 weeks after transplant. Tetratricopeptide (TPR) repeats are a 34-residue helix-turn-helix motif that when repeated pack into a superhelical structure. TPR domains are frequently found mediating protein-protein interactions, often through a central groove. One protein complex bearing numerous TPR repeats is the Anaphase Promoting Complex (APC). The anaphase-promoting complex (APC) is a multi-subunit complex, which orchestrates mitotic cell cycles. APC is an E3 ligase in the ubiquitin cascade, and directs the 26S proteosome degradation of cell-cycle regulators. Throughout mitotic progression, proteins that are key regulators of the cell cycle are assembled with polyubiquitin chains by APC.

Myths, misinformation, and sensationalism. These are common enemies that directly inhibit the public understanding of science. In particular, the media is often responsible for mishandling or otherwise misrepresenting scientific information, historically and presently speaking. Many sources can combat the public understanding of science through pseudoscientific means. This includes but is not limited to religion, the media, politics, or just simple hearsay. For example, Young Earth creationism is deeply rooted in Christian theology, but the beliefs hold no scientific basis. Yet, almost half of Americans still believe in Young Earth creationism. Parkin is a cytosolic E3 ubiquitin ligase that is recruited to the mitochondria during cellular stress and has been suggested to be involved in a variety of biological processes such as mitophagy. The recruitment of Parkin (PARK2) to the mitochondria is dependent upon the kinase activity and the accumulation of PINK1 on damaged mitochondria. Mutations in either PINK1 or Parkin genes disrupt this protective pathway and lead to the accumulation of damaged mitochondria. As a popular historic work with constant and worldwide performances, the sexist and racist narratives disseminated by Giacomo Puccini's opera Madama Butterfly causes harmful social and political ramifications. Many scholars point to this opera specifically when discussing the fetishization of Asian females, and mention the title character as the quintessential example of damaging stereotypes. Thus, I conduct a postcolonial and feminist reading of Madama Butterfly, through analysis of the opera's libretto, the libretto sources, and the opera's score. Multidomain enzymes often rely on large conformational motions to function. However, the conformational setpoints, rates of domain motions and relationships between these parameters and catalytic activity are not well understood. To address this, we determined and compared the conformational setpoints and the rates of conformational switching between closed unreactive and open reactive states in four mammalian diflavin NADPH oxidoreductases that catalyze important biological electron transfer reactions: cytochrome P450 reductase, methionine synthase reductase and endothelial and neuronal nitric oxide synthase. The purpose of this study was to determine whether there is a difference in Laricobius nigrinus Fender fecundity between field-collected and laboratory-reared beetles. Laricobius nigrinus used for this study were separated into four cohorts based on origin: Cohort 1 – Wild-caught from Seattle, WA – native population; Cohort 2 – Wild-caught from Banner Elk, NC – naturalized population; Cohort 3 – Laboratory-reared F1generation from Seattle, WA parents; and Cohort 4 – Laboratory-reared F1 generation from Banner Elk, NC parents.
