CROSSING BORDERS - WOMEN AGENT'S OF BRITAIN'S SPECIAL OPERATIONS EXECUTIVE (SOE) IN WORLD WAR TWO This thesis will explore the experiences of Odette Sansom Churchill Hallowes, a Franco-British woman recruited as a secret agent during World War II by the Special Operations Executive (SOE), a branch of the British Foreign Office, to infiltrate German-occupied France for the purpose of assisting the French Resistance in disrupting vital German operations in the lead-up to the Allied invasion of the continent. Design, synthesis and biological evaluation of novel 2,5,6-trisubstituted benzimidazoles targeting FtsZ as antitubercular agents Filamenting temperature-sensitive protein Z (FtsZ), an essential cell division protein, is a promising target for the drug discovery of new-generation antibacterial agents against various bacterial pathogens. As a part of SAR studies on benzimidazoles, we have synthesized a library of 376 novel 2,5,6-trisubstituted benzimidazoles, bearing ether or thioether linkage at the 6-position. In a preliminary HTP screening against Mtb H37Rv, 108 compounds were identified as hits at a cut off values of 5 ìg/mL. Among those hits, 10 compounds exhibited MIC values in the …

Telomeres are repetitive nucleoprotein complexes at the ends of chromosomes that provide a protective cap for genomic DNA. Telomere shortening leads to cellular senescence or apoptosis. Accordingly, telomere length is considered both a mechanism and marker for aging. Increased exposure to chronic stress is associated with shorter telomere length. In addition, differences in stress reactivity have been associated with telomere length. Here I will describe several studies identifying neural circuits associated with such telomere shortening, an evaluation of saliva as an appropriate biomaterial to measure telomere length, and an exploration of the role of microRNAs as mediators of stress-related telomere …

Squamous cell carcinoma antigens (SCCAs) are members of the serpin family of endogenous serine/cysteine protease inhibitors. First identified in the serum of a patient with squamous cell carcinoma of the uterine cervix, both SCCA1 and SCCA2 have now been found to be elevated in cancers of the lung, head and neck, and liver. Despite SCCA's long known involvement with human cancer, little is known about SCCA's functional role within tumor development and progression. I began my study examining SCCA expression in numerous epithelial cancers. In assessing 1,300 breast cancer samples, SCCA expression was found to correlate with high-grade, advanced-stage cancers. …

Connexin (Cx) proteins form intercellular gap junction channels to facilitate the exchange of ions and second messengers between adjacent cells. Mutations in connexins cause several human diseases by interrupting cell-to-cell communication and coupling. Connexins also form functional hemichannels in nonjunctional membranes that have an unclear role in normal physiology but have altered activity in pathological states. Mutations in GJB2, the gene encoding connexin26 (Cx26), are linked to sensorineural hearing loss as well as syndromic deafness associated with skin disorders. In contrast to Cx26 mutations causing nonsyndromic deafness, those that also cause skin disease are exclusively gain-of-function single amino acid changes …

Gas hydrate technology is one of research focus in recent years, and can be applied to solve problems in energy, environment, and other areas. Previous views had been that most gas hydrates decompose into gas and water (or ice) under 1 GPa or so, thus, research on hydrate structures in past few decades were conducted under low pressure conditions. Submarine groundwater discharge (SGD) from oceanic islands has been estimated to contribute over a third of the global SGD due to orographic precipitation, short aquifer pathways and poorly developed surface drainage. This seepage of groundwater across the sea floor connects land and coastal ocean resources, and is hereby proposed as a parameter to evaluate the interconnections between coastal environmental quality and coastal populations and development. Relatively few islands have been studied, but SGD is typically found to be an important, and often the only, source of nutrients to coastal waters. Honeybee pollination accounts annually for over $14 billion in United States agriculture alone. Within the past decade there has been a mysterious mass die-off of honeybees, an estimated 10 million beehives and sometimes as much as 90% of an apiary. There is still no consensus on what causes this phenomenon, called Colony Collapse Disorder, or CCD. Several mathematical models have studied CCD by only focusing on infection dynamics. We created a model to account for both healthy hive dynamics and hive extinction due to CCD, modeling CCD via a transmissible infection brought to the hive by foragers. The system of …

Background: Mitochondrial DNA (mtDNA) deletions cause disease and accumulate during aging, yet our understanding of the molecular mechanisms underlying their formation remains rudimentary. Guanine-quadruplex (GQ) DNA structures are associated with nuclear DNA instability in cancer; recent evidence indicates they can also form in mitochondrial nucleic acids, suggesting that these non-B DNA structures could be associated with mtDNA deletions. Currently, the multiple types of GQ sequences and their association with human mtDNA stability are unknown. Results: Here, we show an association between human mtDNA deletion breakpoint locations (sites where DNA ends rejoin after deletion of a section) and sequences with G-quadruplex …

First Proof Of Concept Of Sustainable Metabolite Production From High Solids Fermentation Of Lignocellulosic Biomass Using A Bacterial Co-Culture And Cycling Flush System, Wanying Yao, Sue E. Nokes

To improve the lignocellulose conversion for ABE in high solids fermentation, this study explored the feasibility of cycling the process through the cellulolytic or/and solventogenic phases via intermittent flushing of the fermentation media. Five different flushing strategies (varying medium ingredients, inoculum supplement and cycling through phases) were investigated. Flushing regularly throughout the cellulolytic phase is necessary because re-incubation at 65 °C significantly improved glucose availability by at least 6-fold. The solvents accumulation was increased by 4-fold using corn stover (3-fold using miscanthus) over that produced by flushing only through the solventogenic phase. Tissue engineered constructs have proven to possess comparable mechanical and physical characteristics to autologous tissues making them suitable replacements fordiseased anatomy when autologous tissue transplants are unavailable. However, difficulty in reseeding the constructs with cells prevents the tissue-engineered scaffoldsfrom becoming an adequate, equivalent replacement for diseased tissues when compared to autologous transplants. Cells adhere to the outer surfaces of scaffolds but are incapable of penetrating the constructs to provide complete cellularization. If cells were able to populate the inner layers of the tissue, then they could likely diffuse into the intimal and external layers of the construct to create a …

Human neurosphere stem cells offer promising potential for the treatment of neurodegenerative diseases. Their well characterized multi-potency of differentiating into neurons, astrocytes, and oligodendrocytes when exposed to the optimum exogenous growth factors make them an exciting area of study (38). Finding novel endogenous methods of modulating stem cell metabolism will allow for the safer treatment of various brain disorders (34). In this experiment, melatonin, N-acetylserotonin, L-tryptophan, and L-DOPA are added in three different concentrations to neurospheres suspended in HNSC/GBM media with less than optimal concentrations of exogenous epidermal growth factor (EGF) and fibroblast growth factor (FGF). Community-acquired, methicillin-resistant Staphylococcus aureus strains often cause localized infections in immune-compromised hosts, but some strains show enhanced virulence leading to severe infections even among healthy individuals with no predisposing risk factors. The genetic basis for this enhanced virulence has yet to be determined. S. aureus possesses a wide variety of virulence factors, the expression of which is carefully coordinated by a variety of regulators. Several virulence regulators have been well characterized, but others have yet to be thoroughly investigated. The study aimed at exploring and understanding the role of the CPD provided the SESEMAT (on teachers’ practice). SESEMAT is the Secondary Science and Mathematics Teachers’ Program in Uganda. The motion and dissolution of bubbles in confined microgeometries is of pivotal importance for many natural and industrial flow processes such as microchemical systems and the development of models for unveiling the fundamentals of oil recovery in porous-like media. The Andean Ibis (Theristicus branickii) of the highland grasslands of Ecuador, Peru, and Bolivia is listed globally as Near Threatened and Critically Endangered in Ecuador. The Ecuadorian population is estimated at 100 individuals and is restricted to the vicinities of Volcán Antisana and Volcán Cotopaxi. Knowledge of the nesting biology of the Andean Ibis in Ecuador consists of a single filming in 1989. Much of its general ecology and movements is also unknown.
