Critical loads (CLs) and dynamic critical loads (DCLs) are important tools for the management of ecosystems that are impacted by high sulfate, nitrate and ammonium deposition. In this study, a biogeochemical model (PnET-BGC) was applied to 20 watersheds in the Adirondack region and 12 watersheds in Great Smoky Mountain National Park (GRSM) to calculate CLs and DCLs. I evaluated ecosystem changes in response to historical and potential future changes in acidic deposition. I analyzed factors affecting CLs and DCLs for acidification in the acid impacted Constable Pond Watershed in the Adirondack region, specifically evaluating trade-offs of sulfate and nitrate deposition, …

Thermohaline circulation is responsible for the global transport of heat in surface waters of the ocean, and variability in this circulation has been linked to both short- and long-term climate change on time scales ranging from centuries to tens of thousands of years. Accurate reconstructions of past variability are thus critical to understanding modern and future thermohaline circulation changes and the response of the global climate system. The main circulation feature in the Atlantic is Atlantic Meridional Overturning Circulation (AMOC), and has been reconstructed using a variety of static proxies, but more recently through the use of sedimentary 231Paxs …

Exposing Zostera marina to fertilized porewater has been shown to increase vegetative shoot density and leaf growth rates, as well as affect morphological shoot characteristics including increasing leaf length, leaf area and the number of leaves per shoot. However, aside from changes in shoot density, records of morphological changes in reproductive shoots subjected to increased nutrient availability are lacking. To address this, five replicate ambient and nutrient enriched patches, each between 0.5 and 2.0 m2, were selected within a developing Z. marina meadow in Shinnecock Bay, NY. Porous media, such as rocks and soil, make up the largest proportion of mass in nature. Pore-network model is a widely used model to describe the tiny and complicated inner structure of porous media. Assisted by the development of X-ray computed microtomography in recent years, researchers are able to extract the geometric characters of inner pores and throats from porous media, then further explore two-phase flow in porous media more exactly. The excessive delivery of nitrogen from land into coastal waters can lead to a host of environmental problems including algal blooms, hypoxic zones, habitat loss, and acidification. While many of these environmental problems have manifested themselves within Long Island's coastal bays, the quantity and sources of nitrogen are largely unknown in much of this region, making the development of effective management plans to ameliorate these problems exceedingly difficult. Approximately every other year during the austral summer a larger phytoplankton bloom propagates off of the southeast tip of Madagascar. The cause of the interannual variability of this distinct oceanic feature remains unclear, so here we improve the analysis of the interannual variability of the bloom with a longer ocean color dataset, re-examine the existing mixed layer depth and iron fertilization hypotheses formerly used to explain bloom formation and analyze the variability in the larger ocean-atmosphere system in the region in relation to bloom formation. Trait diversity often underlies complexity observed in biological systems. Whether it is behavior, life history, form or function, variation imparts differential performance on the organism, which may influence processes at higher levels of organization. Using two approaches, my dissertation research concerns the appraisal of diversity in fishes and its consequences for their ecology and evolution. The first two chapters focus on variety of spatial behaviors in northwest Atlantic marine communities. Temporal changes in community habitat utilization, as quantified by the interspecific abundance-occupancy (A-O) relationship, were the focus of chapter one. In the present work, poly lactic acid (PLA) is used as a model system to investigate the mechanical behavior of 3-D printed foams with triangular cells. Solid PLA tension and compression specimens and foams made of PLA were fabricated using fused deposition 3-D printing technique. The solid PLA tension specimens were characterized for their densities and found to be about 10% lower in density as compared to their bulk counter parts. PLA is a material easy to manufacture. The biodegradability makes it a perfect material for tissue engineering. Several conditions for biodegradability experiments for spin-coating Polylactic acid thin films were tried. Polylactic acid thin films were immersed in different solution for different times. Thickness, morphology and mechanical properties were analyzed after the Polylactic acid thin films immersing test. Dermal fibroblasts were plated on the Polylactic acid thin films, culturing with conditioning medium. Biomineralization is a process in which living organism grow composite materials consisting of inorganic and organic materials. This produces a composite material consisting of both inorganic and organic components, with superior mechanical properties. In the human body bone and dentin are both examples of biominerals. In this research Raman spectroscopy was used to characterize dentin from mice and human teeth, to determine composition. In the mouse tooth samples areas of irregular dentin were found, along the inside of the tooth, to be in the process of mineralization. Engineered Polymer hydrogels and hydrogels from Bio macromolecules have visco-elastic properties that can be measured using Oscillatory Shear Rheology. Manipulation and measurement of physical properties in gels including F-127 Pluronic Block Co-Polymer and Poly(N-isopropylacrylamide)-Clay are shown through OSR by addition of salts, clays and glucose at physiological levels. Rheological analysis of f-127 illustrates changes in G' reduction with phase transition temperature. Measurements also indicate physical changes due to the aforementioned additives vary as a function of the gel physical and chemical structure. In particular, non-enzymatic glycation is shown to change the modulus of elasticity in both of the gels tested. …

Polyaniline (PANI) belongs to the conducting polymers and has been studied for so many years. PANI has three different states depending on the redox process. In this thesis, we used the completely reduced form leucoemeraldine base (LEB) to synthesize the Chemomechanical actuator. Cellulose Acetate (CA) was used to be the matrix because CA can provide a strong mechanical property for the film synthesizing. PANI and CA molecules interconnected and form the Chemomechanical actuator, which has the selectivity for acetone vapor. This dissertation examines the issue of " place" in the lives and works of three contemporary women artists of the African Diaspora: Maria Magdalena Campos-Pons, IngridMwangiRobertHutter, and Wangechi Mutu. Place as a location is a core challenge of globalism: though fundamental to our existence, it has ceased to be permanent or definite. However, place is more than location--it is cultural, gendered, historical, interactive, personal and social, among other things--and can only be fully understood in considering several of these different facets. The human de novo purine biosynthetic pathway consists of six proteins, which come together to form a protein complex called the purinosome. The tri-functional enzyme, TGART, which contains three domains, is one of the purinosome proteins. Protein-protein interaction studies have shown that TGART, along with two other de novo purine pathway proteins FGAMS and PPAT, is at the core of purinosome complex. Little is known, however about the interacting surface, stoichiometry or interaction mechanism. The production of lipids and proteins for growth, as well as nucleotides for the replication of the genome, are key requirements that loom as importantly as the need for the production of energy in the transformed metabolic state of cancer cells. The remodeling of the TCA cycle (extensive anaplerosis and cataplerosis) reflects this need for biosynthetic macromolecules. Recombination is an essential meiotic process to ensure proper chromosome distribution. Recombination is promoted by the synaptonemal complex, a structure which bridges two pairs of homologous sister chromatids, bringing them into close proximity. The synaptonemal complex is formed by condensation of the sister chromatids on protein cores called axial elements. Homologous axial elements are then held together in budding yeast by a protein called Zip1. Zip1 is part of the ZMM group of proteins that work together during meiosis to promote the formation of crossovers which are distributed throughout the genome by interference. Cells require purines for critical biological processes that can be created by the de novo biosynthetic pathway or maintained by the salvage pathways. The de novo pathway in humans requires 6 enzymes, some of which are multifunctional. Adenylosuccinate lyase (ASL) catalyzes step 8 of the de novo pathway converting susuccinylaminoimidazolecarboxamide ribonucleotide (SAICAR) to aminoimidazolecarboxamide ribotide (AICAR). It also catalyzes the second step of the purine nucleotide cycle by converting SAMP to AMP. A short isoform of ASL (sASL) is ubiquitously expressed in all cell types at a lower concentration. Breast cancer is the second deadliest cancer in the United States and is classified into four main subtypes including HER2-positive breast cancer. There have been a number of HER2-targeted therapeutic treatments that were discovered, but they are expensive and patients frequently exhibit a resistance for these treatments. Oncogene expression in normal cells can cause an irreversible growth arrest called oncogene-induced senescence (OIS), which serves as a barrier to cancer initiation. Elevated reactive oxygen species (ROS) have been detected in most senescent cells and are thought to play a causal role in OIS. ROS are believed to have a second messenger function in transducing signals, however their molecular role in senescence remains elusive. High-capacity rechargeable batteries play a central role in a variety of emerging technologies, including mobile electronics, plug-in and hybrid electric vehicles, and grid-scale storage. All of these technologies will greatly benefit from the design and discovery of next-generation cathode materials that can deliver substantially higher energy densities at reduced cost. All-carbon materials have been widely investigated because of their potential thermal, electrical and physical properties. Carbyne, which would have all sp hybridized carbons, has received considerable attention. Polydiiododiacetylene (PIDA), which has a carbon backbone and iodine atom substituents, serves as a potential precursor to carbyne. We propose that carbyne could result from complete deiodination of PIDA in the presence of Lewis bases such as triethyl amine, pyridine and pyrrolidine. Sulfur rich metal coordinated active sites is a prominent feature of many metalloenzymes (hydrogenase, nitrogenase, carbon monoxide dehydrogenase) involved in catalytically producing and/or absorbing helpful and/or toxic gases. As the mechanism of gas production and/or absorption heavily depends on the structural intricacies, biomimetic modelling of the active sites is of paramount importance to understand the structure-function relationship.
