Establishment of protoplasts system provides a useful platform for cloning and genetic manipulation of ginger plants. In this study, an efficient protocol for developing protoplast isolation and culture for Boesenbergia rotunda has been established. B. rotunda embryogenic cell suspension cultures showed good growth rate (μ = 0.1125) when cultured in plant growth regulator (PGR)-free liquid Murashige and Skoog (MS) basal medium supplemented with 3 % sucrose, where no promotive effect were observed in the presence of any concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) and sonication treatment. Six mushroom samples (Ganoderma lucidum, Cordyceps militaris, Lignosus rhinocerotis, Pleurotus giganteus, Pleurotus floridanus and Auricularia polytricha) were screened for their anticoagulant activity. Mushroom samples provided were freeze-dried and blended into powder form. Different concentrations of aqueous mushroom extracts were added to one ml of fresh bovine blood and well mixed. From the observations, among the six mushroom samples, only two samples namely A. polytricha and L. rhinocerotis showed anticoagulant activities in the preliminary screening. The present study deals with tissue culture of Punica granatum L. Nodal stem explant was found to be the best explant which gave the highest shoot number (3.8±0.2) on MS medium supplemented with 1.5 g/L BAP, while shoot tip explant (1.9±0.1cm) on MS medium with 2.0 mg/L BAP gave the best shoot height. MS supplemented with 1.5 mg/L BAP was observed to give the highest shoot production (4.1±0.2) with combination of 40 g/L of sucrose in shoot tip explant. Liposomes have been used widely as carriers for active ingredients in cosmetic because of their ability to encapsulate both hydrophilic and hydrophobic compounds. In this work, fatty acid liposomes were prepared and introduced into olive oil-in-water emulsions stabilized by C14–C18 sucrose ester mixtures at pH 8.5. Light microscopy images of the emulsions showed evidence of the coexistence of oleic acid liposomes and the emulsion droplets. As alkyl chain length of sucrose ester increased, the average droplet size decreased, while the zeta potential became more negative. In the present study, concentration of minerals Na, Mg, K, Ca, Mn, Fe, Cu and Zn in selected commercially available brown rice and white rice from Malaysia were determined using microwave assisted digestion with inductively coupled plasma mass spectrometry. The data obtained were evaluated using chemometric tools. Hierarchical cluster analysis and principal component analysis visually reveal the natural clustering of rice sample into two distinct clusters where brown rice are associated with higher mineral concentration. The statistics show that all the mineral concentrations found in brown rice are significantly higher than those found in white rice (P<0.001). Actinomycetes, mainly Streptomyces spp., have been extensively studied as potential biocontrol agents against plant pathogenic fungi. This study was aimed at isolating and screening Streptomyces strains from rhizosphere soils of Hylocereus polyrhizus collected in Kuala Pilah for potential in vitro antifungal activity. A total of 162 putative strains of actinomycetes was isolated from moist-heat treated soil plated on starchcasein-nitrate agar, humic-acid-vitamin agar and raffinose-histidine agar. Based on the ability to produce abundant aerial mycelium, 110 strains were categorised as Streptomycete-like. The present study investigated the effects of green, white and black tea (Camellia sinensis; 2% w/v) on the fermentation of milk and antioxidant potential of yogurt. Each tea (water extract) was added into milk-starter culture mixture and incubation was carried out at 42°C until pH was reduced to 4.5. The yogurts were then refrigerated (4°C) for up to 21 days and samples were analysed for antioxidant potential (diphenyl picrylhydrazyl (DPPH) radical scavenging, ferric reducing antioxidant power (FRAP) and ferrous ion chelating (FIC) assays), pH, titratable acid and viable yogurt bacteria counts. Tea yogurts had higher antioxidant potentials (p < 0.05) than plain yogurt with green tea yogurt (GTY) having the highest FRAP (2.49 - 2.98 mmol Fe2+ E/L) and black tea yogurt (BTY) having the highest FIC (87.50 - 89.87 %) activity throughout the storage period. Both GTY (90.07 - 96.74%) and white tea yogurt (WTY; 89.83 - 96.39%) showed the highest DPPH radical scavenging activity throughout the storage period. In this study, a semi automated cell suspension culture protocol for Musa acuminata cv. ‘Berangan’ using bioreactor has been established. Flower clusters at the positions of 8 to 10 were the most responsive (70 %) towards embryogenesis. After eight months of culture, embryogenic calli generated from male inflorescences were transferred to liquid suspension medium. Embryogenic cell suspension culture was successfully established after two months of culture in liquid M2 medium. Mosaic Virus (CMV) is a significant plant pathogen affecting various crops and plants in Malaysia. A single chain variable fragment (scFv) anti-CMV antibody was successfully developed via a scFv library constructed with mRNA from the spleen cells of a CMV coat protein-immunized mouse and transformed by Agrobacterium tumefaciens into tobacco plants (Nicotiana tabacum L. cv. White burley). In this study, the performance of primary transformants and 3 successive generations of Nicotiana tabacum expressing anti-CMV scFv were evaluated. The rhizomes and leaves of Alpinia conchigera were extracted by supercritical fluid extraction (SFE) technique using liquid carbon dioxide at constant temperature (40°C) with two different pressures (A1: leaves extract at 1500 psi; A2: leaves extract at 5000 psi; B1: rhizomes extract at 1500 psi; B2: rhizomes extract at 5000 psi). The volatile constituents of the extracts were detected by GCMS. The major compound was identified as acetoxychavicol acetate in all the extracts. We studied temporal variation of picocyanobacteria and their production (μ) and loss (g) rates for over two years of period at Port Klang (PK) estuarine waters [03º00.1'N, 101º23.4'E] and Port Dickson (PD) coastal waters [02º29.5'N, 101º50.3'E] along the Straits of Malacca. PK waters showed higher level of eutrophication, lower euphotic depth and higher TSS level as compared to PD. Heterotrophic bacterial abundance at PK (2.78 ± 1.58 × 106 cell ml–1) was generally higher than PD (1.39 ± 0.49 × 106 cell ml–1) (Student’s t-test: t = – 5.30, df = 47, p < 0.001). In contrast, picocyanobacterial abundance at PD (1.33 ± 0.47 × 105 cell ml–1) was always higher than at PK (0.28 ± 0.17 × 105 cell ml–1) (Student’s t-test: t = 10.44, df = 30, p < 0.001). μ and g of picocyanobacteria were tightly coupled with each other (R2 = 0.47, df = 459, p < 0.001) and similar at both sites (p > 0.05). μ ranged from – 0.03 to …

Lipid nanoparticles are colloidal carrier systems that have extensively been investigated for controlled drug delivery, cosmetic and pharmaceutical applications. In this work, a series of stearic-oleic acid nanoparticles (SON) with different oleic acid compositions were successfully prepared by melt-emulsification combined with ultrasonication method. The SONs prepared have average particle size range from 200 nm to 250 nm depends on the oleic acid concentration in the mixture. The structure of these complexes has been confirmed by X-ray crystallography to give a trinuclear oxo-centred carboxylate. Fibrinolytic activity was identified in the fruiting bodies of the medicinal mushrooms L. rhinocerotis and A. polytricha. However, the present study emphasised on the evaluation of the fibrinolytic activity from L. rhinocerotis and its partially purified enzyme only. Extracellular lipase of the yeast Candida rugosa was produced via high cell density fed-batch fermentations using palm oil as the sole source of carbon and energy. Feeding strategies consisted of a pH-stat operation, foaming-dependent control and specific growth rate control in different experiments. Compared to foaming-dependent feeding and the pHstat operation, the specific growth rate control of feeding proved to be the most successful. At the specific growth rate control set point of 0.05 h1, the final lipase activity in the culture broth was the highest at 700 U L1.
