Coronary Artery Disease (CAD), the most prevalent form of heart disease, is the result of clogged or damaged coronary arteries and claims around 380,000 Americans annually. A common treatment for CAD involves placing a stent into the artery in order to open the lumen and support the native tissue—a procedure that drastically reduces patient recovery times in comparison to heart bypass surgery. However, stents do not always interact well with the body and require additions such as surface coatings or drug elution in order for additional biocompatibility. The uterine endometrium undergoes cyclical phases of cell proliferation, cell differentiation, and menstruation under the influence of the ovarian hormones, estrogen and progesterone. Since the data necessary to create a classical kinetic model of these signaling pathways is lacking, we used a Boolean network approach that includes the influences of various growth factors and the differential expression of their receptors under the influence of estrogen and progesterone. Results show a gain in endometrial tissue and loss of tissue during menstruation that mirrors what can be expected over the course of a normal menstrual cycle in women, where the endometrium typically …

Genome-Wide Association Studies (GWAS), whole genome sequencing, and high-throughput omics techniques have generated vast amounts of genotypic and molecular phenotypic data. However, these data have not yet been fully explored to improve the effectiveness and efficiency of drug discovery, which continues along a one-drug-one-target-one-disease paradigm. As a partial consequence, both the cost to launch a new drug and the attrition rate are increasing. Systems pharmacology and pharmacogenomics are emerging to exploit the available data and potentially reverse this trend, but, as we argue here, more is needed. Human epidermal growth factor receptor 2 (HER2) is overexpressed in approximately 30% of breast cancer patients with poor prognosis. In addition, microRNAs are small non-coding RNA that have been linked to many cancers. Here we investigate which miRNAs are differentially regulated by HER2 overexpression. Using quantitative reverse-transcription prolymerase chain reaction (QRT-PCR) and matching it with the clinical data acquired from Dvinge, we find five candidate miRNAs. When comparing the miRNAs’ effect on patient survival, only three miRNAs stand as good predictors of patient survival outcome. These miRNAs are miR-146a-5p, miR-181d, and miR-195-5p. The purpose of this proposed study was to determine the potential changes in testosterone (free and total) and estradiol over a four-week intervention period using D-aspartic acid (DAA) in men. Twenty-four physically active men, aged 18-39 years, were randomly assigned to two groups (conditions) using a placebo-controlled, double-blind design. Baseline values for all hormones were compared to values measured at the end of two and four weeks of DAA administration, provided at a dosage of 3.12g, compared to the placebo. Nitrate ingestion improves aerobic exercise performance, but the efficacy in resistance training has not been established. The purpose of this study was to investigate whether consuming whole beetroot, as a means to ingest nitrate, had an ergogenic benefit in muscular power and endurance in resistance training in powerlifters and physique competitors. Thirteen men and women (mean ± SE age, 21.2 ± 0.8 y; mass, 83.1 ± 5.9 kg) that had competed in a powerlifting or a physique competition in the previous year were studied in a randomized double blind controlled crossover trial with a seven day washout period. For many years, it has been proposed that oxidative stress within the mitochondria causes mutations to the mitochondrial genome, resulting in changes in copy number per single cell. Ultimately, this compensatory increase in mtDNA, coupled with an increase in mutations, has been suggested to play a major role in the pathobiology of aging. However, evidence for this phenomenon is somewhat controversial. An integrated dual-mode instrument that combines grating-coupled surface plasmon resonance (GCSPR) and grating-coupled surface plasmon coupled emissions (GCSPCE) should allow for the efficient detection of toxic analytes in a high-content system using gold-coated superparamagnetic nanobeads. Background: Rhabdomyolysis is a potentially life-threatening syndrome characterized by the breakdown of skeletal muscle fibers resulting in the release of proteins into general circulation. Common practice for diagnosing exertional rhabdomyolysis (ER) is using biochemical markers for muscle damage; creatine kinase (CK) and myoglobin (Mb) are proteins detected in the bloodstream after muscle injury or trauma. Serum creatinine is the best biochemical marker for calculating renal function. Measuring GFR is not common when assessing ER. Purpose: The aim of this study is to help predict ER. Thanks to advances in technology, the survival rate of infants and children with critical illnesses has improved. One consequence has been an increase in the acuity levels of hospitalized pediatric patients, which may heighten the risk of in-hospital cardiopulmonary arrest. Patient safety can be compromised by nurses who do not detect subtle clinical changes in a patient's condition, do not perform accurate interventions, or do not apply critical thinking (failure to rescue). The Institute of Medicine (10M) in 1999 estimated that approximately 100,000 people died each year as the result of poor care.

Central arterial stiffness, indicated by pulse wave velocity (PWV), is increased during the hour post ultramarathon (ULT), but it's currently unknown if PWV remains elevated 24 hours after ULT or marathon (MAR). PURPOSE: To determine the effects of ULT and MAR on C-reactive protein (CRP), aortic blood pressure (BP), aortic augmentation index (AIx), PWV, and LF:HF ratio. We hypothesized that CRP, AIx, PWV and LF:HF ratio would be elevated 24 hours post-race. Our secondary hypothesis was that changes in PWV would be inversely correlated to the percentage of moderate activity (3-6 METs) during competition. METHODS: Applanation tonometry was used …

Elafin, an endogenous serine protease inhibitor, is a critical component of the epithelial barrier against neutrophil elastase (NE) activity. The central hypothesis examined in this dissertation was that elafin has tumor suppressive properties in breast cancer. In support of this hypothesis, immunohistochemical (IHC) analysis revealed that elafin was downregulated in the majority of invasive breast tumors and a subset of pre-invasive ductal carcinoma in situ (DCIS) compared to elafin expression in the normal mammary epithelium. The current model for predicting human total energy expenditure (TEE), the Factorial Method, significantly underestimates actual TEE, particularly among highly active populations. In this study, the Allocation Model is presented for predicting TEE. Unlike the Factorial Method, the Allocation Model includes metabolic cost terms for both thermoregulation and the thermic effect of food, as well as using more accurate basal metabolic rate and activity cost estimations. Guided bone regeneration (GBR) membranes function as barriers in preventing soft tissue migration into dental osteogenic spaces. Chitosan is a biocompatible and degradable natural polysaccharide, which has shown potential in tissue regeneration and GBR applications. Trifluoroacetic acid (TFA) is a common solvent used to make the chitosan polymer solution for electrospinning process. Impaired responsivity to hypercapnia or hypoxia is commonly considered a mechanism of failure in Sudden Infant Death Syndrome (SIDS). We used a model of developmental cerebellar Purkinje cell loss to determine if such loss influenced compensatory ventilatory responses to hypercapnic and hypoxic challenges. Twenty-four Lurcher mutant mice and wildtype controls were sequentially exposed to 2% increases in CO2 (0%-8%) or 2% reductions in O2 (21%-13%) for four-minutes, with return to room air between each exposure. Whole-body plethysmography was used to continuously monitor tidal volume and breath frequency. Tidal volume increases to CO2 elevation were reduced in Mutants, and slower to …

Neurotrophic factors are proteins that play an important role in the development and maintenance of neurons. Recent studies have shown that neurotrophic factors may hold promise for treating damage to the nervous system caused by trauma or diseases, such as amyotrophic lateral sclerosis or Parkinson’s disease. Glial cell line-derived neurotrophic factor (GDNF) is expressed in skeletal muscles and affects peripheral motor neurons. The current study aimed to examine the influence of body fat percentage on thermal responses to water immersion at the two temperature extremes used in athletic recovery, by comparing the thermal responses of low- and high-fat individuals to immersion in hot water (HWI) or cold water (CWI). Thirty-nine subjects completed a body composition assessment (to determine low-fat and high-fat groups), and 15 minutes of either CWI (n = 20) or HWI (n = 19), followed by 30 minutes of passive rest. Antioxidants, when consumed, have the ability to neutralize free radicals, suggesting their presence may protect human physiology from destructive oxidative processes1. Several assays have been developed that quantify the antioxidant capacity of compounds, and one assay that is commonly utilized by researchers and industry is the Oxygen Radical Absorbance Capacity (ORAC) assay2. Despite its popularity, significant differences in reported ORAC values occur between laboratories, leading to difficulties in comparing research. Dr. Parker and his lab conducted a series of tests to attempt to standardize and optimize the ORAC assay.

A fundamental question in biology is how proteins, which are synthesized by the ribosome as a linear sequence of amino acids, fold into their native functional state. Many proteins require the assistance of molecular chaperones to maneuver through the folding process to protect them from aggregation and to help them reach their native state in the very concentrated protein environment of the cell.
