Multiple myeloma is a hematological malignancy of plasma cells that disseminates throughout the body. Crosstalk between myeloma and host cells is critical to the establishment of a microenvironment conducive to tumor growth and progression. An important mediator of this crosstalk is syndecan-1, a heparan sulfate proteoglycan. Syndecan-1 is proteolytically shed from the surface of myeloma cells and is abundant in the bone marrow microenvironment and serum of myeloma patients. Shed syndecan-1 facilitates tumor-host crosstalk within the microenvironment to drive tumor growth, angiogenesis, and metastasis. Next-generation sequencing (NGS) technologies reveal unprecedented insights about genome, transcriptome, and epigenome. However, existing quantification and statistical methods are not well prepared for the coming deluge of NGS data. In this dissertation, we propose to develop powerful new statistical methods in three aspects. First, we propose a Hidden Markov Model (HMM) in Bayesian framework to quantify methylation levels at base-pair resolution by NGS. Second, in the context of exome-based studies, we develop a general simulation framework that distributes total genetic effects hierarchically into pathways, genes, and individual variants, allowing the extensive evaluation of existing pathway-based methods. In eukaryotic cells, genomic DNA is packaged into a chromatin structure by association with histone and non-histone proteins. Posttranslational modifications of histones play important roles in the regulation of chromatin structure and function. Ubiquitination of histone H2A (ubH2A) represents a predominate modification, occurring on ~10% of total cellular H2A. While H2A ubiquitination is primarily conferred by Polycomb Repressive Complex 1 (PRC1), H2A deubiquitination has been attributed to multiple H2A deubiquitinases. Our laboratory previously reported the purification and functional characterization of a H2A-specific deubiquitinase, USP16 (initially named as Ubp-M) in human cells. Insulin resistance in diabetics comprises of a reversible component treatable with intensive glycemic control and a non-reversible component present in pre-diabetes that may lead to the development of overt diabetes. The reversible component is induced and exacerbated by hyperglycemia in both uncontrolled type 1 and type 2 diabetes, a process known as glucose-induced insulin resistance (GIIR) or glucose toxicity. The tribbles homolog 3 (TRIB3) has been identified as a pseudokinase that inhibits Akt activity by physically blocking its phosphorylation site. Multiple studies have demonstrated that single-nucleotide polymorphisms (SNPs) in the ITGAM locus (including the non-synonymous SNPs rs1143679, rs1143678 and rs1143683) are associated with SLE. ITGAM encodes the protein CD11b, a subunit of the ß2 integrin Mac-1. The purpose of this work was to determine the effects of ITGAM genetic variation on the biological functions of neutrophil Mac-1. Neutrophils from ITGAM genotyped and sequenced healthy donors were isolated for functional studies. The phagocytic capacity of neutrophil ITGAM variants was probed with complement coated erythrocytes, serum treated zymosan, heat treated zymosan and IgG coated erythrocytes. Generalized estimating equation (GEE) and generalized linear mixed model (GLMM) are two statistical modeling methods commonly used in the analyses of correlated outcomes from cluster-randomized trials (CRTs). The fact that most CRTs involve only a small number of clusters makes the small sample adjustment critical to preserve the type I error rates in testing the null hypothesis of intervention effects. To improve the small sample performance of GEE, some bias-corrected sandwich estimators have been proposed. Monolithic zirconia restorations are popular but removing these crowns is difficult. The purpose of the study is to measure and compare the cutting effectiveness of 10 types of diamond burs through monolithic sintered zirconia. Ten types of diamond burs (n=8) were compared for the cutting effectiveness in Zirconia (CERCON; DeguDent) using a high speed computer controlled cutting device with water spray (5.45L/min). The turbine used to cut the blocks rotated at 150,000 rpm and with the burs at a depth of 2mm into the zirconia during cutting. A 100 gm load was applied during cutting for each bur which made …

The purpose of these dissertation studies was to 1) determine whether influenza infection alters CFTR activity in polarized epithelium; 2) measure CFTR expression and activity in cells co-expressing influenza M2 protein; and 3) examine the role of M2 on the alteration of CFTR during viral infection. We determined that infection of polarized primary epithelial cells with influenza decreases CFTR expression and activity. In addition, individual cells infected with influenza had decreased CFTR conductance as measured by whole-cell patch clamp. Methylmercury (MeHg) is a known neurotoxicant and bioaccumulates in fish, with exposure to humans in utero being of highest concern. Iron deficiency (ID) accounts for approximately 50% of anemia cases. Anemia is estimated to affect 1.62 billion people worldwide, and is particularly problematic during pregnancy and early life. Previous studies have shown that ID exacerbates toxicity associated with exposure to metals, including lead, manganese, and cadmium. For over 30 years, respirator sizing has been based on an anthropometric survey performed by LANL using the young and predominantly Caucasian US Air Force personnel. NIOSH, having observed the increase in diversity in the workplace, began a new survey in 2003 weighted to the US census. Finding that the LANL panel was no longer representative of the modern workforce, a new fit test panel recommending five sizing categories was created in 2007. Understanding Internal, External, And Relational Attributions For Abusive Supervision., James P. Burton, Shannon G. Taylor, Larissa K. Barber

Despite abundant research on the consequences of perceived abusive supervision, less is known about how employees develop perceptions of supervisory abuse. Across two studies, we integrate classic and recent theoretical work on attributions to understand the causal explanations underlying employee perceptions of and reactions to abusive supervision. In the first study, we develop measures of internal, external, and relational attributions for perceived supervisor abuse. In the second study, we demonstrate that internal and external attributions are indirectly related to aggressive and citizenship behaviors through employees’ perceptions of interactional justice. Theoretical and practical implications are also discussed.

This Article explores how current terminations of undocumented immigrants’ parental rights are reminiscent of historical practices that removed early immigrant and Native American children from their parents in an attempt to cultivate an Anglo-American national identity. Today, children are separated from their families when courts terminate the rights of parents who have been, or who face, deportation. Often, biases toward undocumented parents affect determinations concerning parental fitness in a manner that, while different, reaps the same results as the removal of children from their families over a century ago. American state parentage laws have traditionally required biological or adoptive ties and no more than two parents for any one child at any one time. Biological ties were demonstrated by giving birth or sperm. The premise of this article is that an understanding of psychology and other social science disciplines can inform the effectiveness of the economic tools traditionally deployed in carrying out the functions of government, which include remedying market failures, redistributing income, and collecting tax revenue. An understanding of psychology can also lead to the development of different policy tools that better motivate desired behavior change or that are more cost-effective than traditional policy tools. What Makes Annuitization More Appealing?, John Beshears, James J. Choi, David Laibson, Brigitte C. Madrian, Stephen P. Zeldes

We conduct and analyze two large surveys of hypothetical annuitization choices. We find that allowing individuals to annuitize a fraction of their wealth increases annuitization relative to a situation where annuitization is an “all or nothing” decision. Very few respondents choose declining real payout streams over flat or increasing real payout streams of equivalent expected present value. Highlighting the effects of inflation increases demand for cost of living adjustments. Frames that highlight flexibility, control, and investment significantly reduce annuitization. A majority of respondents prefer to receive an extra “bonus” payment during one month of the year that is funded by …

A Critical Examination Of Couple Leisure And The Application Of The Core And Balance Model, Peter J. Ward, Keith W. Barney, Neil R. Lundberg, Ramon Zabriskie

This study further clarified the relationship between couple leisure involvement, couple leisure satisfaction, and marital satisfaction through examining the core and balance framework as it has been applied to the relationship between couple leisure and marital satisfaction. The sample consisted of 1,187 couples across the United States (N = 2,374). SEM analyses supported previous findings, indicating satisfaction with couple leisure involvement was far more important when predicting marital satisfaction than the amount of leisure involvement. Furthermore, while core marital leisure satisfaction was the driving force in the explanation of variance in marital satisfaction, findings suggested before that can happen there …

In this paper, we present a systematic evaluation of the effects of local clays and the manufacturing process on the performance of ceramic water filters (CWFs) impregnated with silver compounds, which are used for point-of-use water treatment in developing countries. Mineral composition, silver sorption/desorption, and strength are the important characteristics that influence effectiveness and durability of CWFs during transport and use. Laboratory tests were conducted on ceramic samples obtained from five CWF factories around the world to determine their mineral composition, silver sorption/desorption, and flexural strength. The purpose of this study was to investigate the effects of three days of foam rolling on the hamstrings range of motion in comparison with static stretching. Lower extremity injuries are prevalent in strength training and sports today. Poor flexibility has been found to increase the risk of overuse injuries and significantly affect the individual’s level of function and performance. Self myofascial release (SMR), foam rolling, is a recent modality clinically used to increase flexibility. On the other hand, there are few research studies on the technique.

Detection of explosives, explosive precursors, or other threat agents presents a number of technological challenges for optical sensing methods. Certainly detecting trace levels of threat agents against a complex background is chief among these challenges; however, the related issues of multiple target distances (from standoff to proximity) and sampling time scales (from passive mines to rapid rate of march convoy protection) for different applications make it unlikely that a single technique will be ideal for all sensing situations. In the present paper, I look at the top of the Interagency Language Roundtable (ILR) Skill Level Descriptions and critique several of their assumptions. As I do this, I speak for myself and not for the Government in general or the U.S. Department of State in particular. I also do not pretend that my conclusions are not uncontroversial. I also discuss the 2012 ACTFL proficiency standards, but note that we do not train to those standards at the Foreign Service Institute.

In response to mandates from the federal government, many academic librarians face a new challenge to become service providers for research data management. A panel at RDAP14 convened representatives from Virginia Commonwealth University, James Madison University and a collaboration of seven New England libraries to discuss their strategies in response to the regulations when faced with limited resources. A common theme was to take advantage of work already done by others, including faculty surveys and existing data management resources such as DMPTool and Data Curation and Profiles Toolkit. After the Supreme Court’s decision in Shelby County v. Holder, voting rights activists proposed a variety of legislative responses. Some proposals sought to move beyond measures that targeted voting discrimination based on race or ethnicity. They instead sought to eliminate certain problematic practices that place too great a burden on voting generally. Responses like these are universalist, because rather than seeking to protect any particular group against discrimination, they formally provide uniform protections to everyone.
