Altered Morphology And Composition Of Zymogen Granules In Acute Pancreatitis, Amanda Flack Jan 2014

Altered Morphology And Composition Of Zymogen Granules In Acute Pancreatitis, Amanda Flack

Wayne State University Dissertations

In healthy physiology, pancreatic digestive enzymes secreted following a meal are stored as inactive zymogens within membrane bound secretory vesicles called Zymogen Granules (ZG), and activated extracellularly. In acute pancreatitis however, the digestive enzymes are prematurely activated within the cell, resulting in autodigestion of the tissue. Pancreatitis is a gastrointestinal disorder in which there are over 200,000 hospitalizations per year with a 5% mortality rate. It has been demonstrated that in acute pancreatitis the digestive enzymes are blocked from being secreted and are activated within the cell leading to acinar cell and surrounding pancreatic tissue death. Little is known about …

Dual Delivery Systems Based On Polyamine Analog Benspm As Prodrug And Gene Delivery Vectors, Yu Zhu Jan 2014

Dual Delivery Systems Based On Polyamine Analog Benspm As Prodrug And Gene Delivery Vectors, Yu Zhu

Wayne State University Dissertations

Combination drug and gene therapy shows promise in cancer treatment. However, the success of such strategy requires careful selection of the therapeutic agents, as well as development of efficient delivery vectors. BENSpm (N1, N11-bisethylnorspermine), a polyamine analogue targeting the intracellular polyamine pathway, draws our special attention because of the following reasons: (1) polyamine pathway is frequently dysregulated in cancer; (2) BENSpm exhibits multiple functions to interfere with the polyamine pathway, such as to up-regulate polyamine metabolism enzymes and down-regulate polyamine biosynthesis enzymes. Therefore BENSpm depletes all natural polyamines and leads to apoptosis and cell growth inhibition in a wide range …

Social Status, Opportunity And Repeat Victimization: The Unequal Distribution Of Safety, Zavin Nazaretian

Wayne State University Dissertations

This research examines the relationship between victimization, social status and opportunity. More specifically, the effects of social status and opportunity on repeat victimization are examined. How does social status and opportunity simultaneously effect repeat victimization? This report consists of a secondary data analysis of the 2004 and 2009 Canadian Victimization Survey with a combined sample size of 43,200 people who were interviewed by telephone. Opportunity either partially or completely mediated the effects of social status on repeat victimization; however for certain subsamples neither opportunity nor social status explained repeat victimization. Additionally, the groups whose victimization was not explained by opportunity …

High-Performance Accelerometer Based On Asymmetric Gapped Cantilevers For Physiological Acoustic Sensing, Yating Hu

Wayne State University Dissertations

Continuous or mobile monitoring of physiological sounds is expected to play important role in the emerging mobile healthcare field. Because of the miniature size, low cost, and easy installation, accelerometer is an excellent choice for continuous physiological acoustic signal monitoring. However, in order to capture the detailed information in the physiological signals for clinical diagnostic purpose, there are more demanding requirements on the sensitivity/noise performance of accelerometers. In this thesis, a unique piezoelectric accelerometer based on the asymmetric gapped cantilever which exhibits significantly improved sensitivity is extensively studied. A meso-scale prototype is developed for capturing the high quality cardio and …

Structural Basis Of Epigenetic Regulation And Protein Scaffolding In Development And Diseases, Yuanyuan Jiang Jan 2014

Structural Basis Of Epigenetic Regulation And Protein Scaffolding In Development And Diseases, Yuanyuan Jiang

Wayne State University Dissertations

Protein X-ray crystallography is a powerful approach for elucidating protein structure and function. The high-resolution data generated by X-ray allow us to visualize protein structures in a three-dimensional (3D) space, which is vital for our understanding of the protein intra- and intermolecular interactions that explain the mechanisms of various biological events. More importantly, such information can provide a structural basis for developing new methods and strategies of targeted drug discovery. In this dissertation, by using X-ray crystallography as the primary approach, we have performed the structural and functional studies of SMYD2 and NHERF1 and have determined their mechanisms of action …

Bystander Effects Due To Neutrons, And Low-Dose Hyper-Radiosensitivity To Gamma Rays In Human Cells Using Cytogenetics, Isheeta Seth

Wayne State University Dissertations

Chapter 2:

Micronuclei have been used extensively in studies as an easily-evaluated indicator of DNA damage but little is known about their association with other types of damage such as nucleoplasmic bridges and nuclear buds. Radiation-induced clastogenic events were evaluated via the cytokinesis-block micronucleus assay in two normal human lymphoblastoid cell lines exposed to neutrons or gamma radiation. Micronuclei, nucleoplasmic bridges and nuclear buds were enumerated by recording the coincident presence of these endpoints within individual cells, and the associations among these three endpoints were evaluated for all treatment conditions. The common odds ratios for micronuclei and nucleoplasmic bridges were …

Proximate And Evolutionary Insights Into The Epigenetics Of Posttraumatic Stress Disorder, Levent Sipahi Jan 2014

Proximate And Evolutionary Insights Into The Epigenetics Of Posttraumatic Stress Disorder, Levent Sipahi

Wayne State University Dissertations

Posttraumatic stress disorder (PTSD) is an important medical and social condition. Although the vast majority of individuals are exposed to traumatic events within their lifetime, a minority subsequently develop diagnosable PTSD. What underlies differential risk and resiliency in the face of trauma is an ongoing research and clinical question with implications for prevention and treatment. Recent work has revealed a putative role of epigenetic variation and modification - most notably DNA methylation - in the etiology of PTSD. That DNA methylation is stable, yet modifiable in response to lived experiences, makes it a strong candidate to mechanistically explain the ontogeny …

Role Of The Pkna And Pknb Kinases In Mycobacterium Tuberculosis, Tripti Anandan Jan 2014

Role Of The Pkna And Pknb Kinases In Mycobacterium Tuberculosis, Tripti Anandan

Wayne State University Dissertations

To respond to environmental changes, M. tuberculosis possesses eleven "eukaryotic-type" Ser/Thr protein kinases. The aim of the study described in this dissertation was to identify role of two of these kinases; PknA and PknB that are essential in M. tuberculosis. Two approaches are described to screen for potential in vivo substrates of PknA/PknB. First approach is based on proteomic search by over-expressing PknA/PknB in M. tuberculosis. Proteomic search led to identification of proteasome to be a substrate of PknA and PknB in M. tuberculosis. Furthermore, I demonstrate that the phosphorylation of PrcA and PrcB by PknA regulates processing of Pre-PrcB. …

Linking Molecular, Electrical And Anatomical Properties Of Human Epileptic Brain, Shruti Bagla Jan 2014

Linking Molecular, Electrical And Anatomical Properties Of Human Epileptic Brain, Shruti Bagla

Wayne State University Dissertations

Epilepsy is a common neurological disorder of recurrent unprovoked seizures. It affects almost 1% of the world population. Although there is a wide range of anti-epileptic drugs (AEDs) available, they only treat the seizure symptoms and do not cure the disease itself. The poor role of AEDs can be attributed to the lack of knowledge of exact mechanisms and networks that produce epileptic activities in the neocortex. At present, the best cure for epilepsy is surgical removal of electrically localized epileptic brain tissue. Surgically removed brain tissue presents an excellent opportunity to discover the molecular and cellular basis of human …

Inositol is a six-carbon cyclitol that is ubiquitous in biological systems. It is a precursor for the synthesis of numerous biologically important compounds, including inositol phosphates and phosphoinositides that are essential for cell function and viability. Inositol compounds play a role in membrane formation, gene regulation, signaling, regulation of ion channels, and membrane trafficking. Furthermore, inositol regulates hundreds of genes, including those involved in the biosynthesis of inositol and phospholipids. While transcriptional regulation of inositol biosynthesis has been extensively studied and well characterized, regulation of inositol biosynthesis at the enzymatic level has not been addressed. The current study shows that …

Flexible Mems: A Novel Technology To Fabricate Flexible Sensors And Electronics, Hongen Tu Jan 2014

Flexible Mems: A Novel Technology To Fabricate Flexible Sensors And Electronics, Hongen Tu

Wayne State University Dissertations

This dissertation presents the design and fabrication techniques used to fabricate flexible MEMS (Micro Electro Mechanical Systems) devices.

MEMS devices and CMOS(Complementary Metal-Oxide-Semiconductor) circuits are traditionally fabricated on rigid substrates with inorganic semiconductor materials such as Silicon. However, it is highly desirable that functional elements like sensors, actuators or micro fluidic components to be fabricated on flexible substrates for a wide variety of applications. Due to the fact that flexible substrate is temperature sensitive, typically only low temperature materials, such as polymers, metals, and organic semiconductor materials, can be directly fabricated on flexible substrates. A novel technology based on XeF2(xenon …

Adenosine A2b Receptor Effects On Post-Mi Remodeling And Cardiac Fibroblast Function, Enbo Zhan Jan 2014

Adenosine A2b Receptor Effects On Post-Mi Remodeling And Cardiac Fibroblast Function, Enbo Zhan

Wayne State University Dissertations

Adenosine A2B receptor (A2BR) appear to contribute to chronic inflammation. This receptor is highly expressed in macrophages and cardiac fibroblasts, cells which play key roles in inflammation and healing following myocardial infarction (MI). A2BR have been shown to induce collagen production and promote organ fibrosis, although the reports of A2BR role on MI are limited and conflicting. The results of cardiac fibroblast (CF) studies however suggest that non-selective A2BR stimulation inhibits collagen expression. The hypothesis of the present study was that deletion of A2BR reduces adverse remodeling in post-MI, …

Cytogenetic Evaluation Of Low Dose Hyper-Radiosensitivity In Human Lymphoblastoid Cell Lines Exposed To Cobalt-60 Gamma Radiation., Gnanada Sameer Joshi

Wayne State University Dissertations

The dose-effect relationships of cells exposed to ionizing radiation are frequently described by linear quadratic (LQ) models over an extended dose range. However, many mammalian cell lines, when acutely irradiated in G2 at doses below 30 cGy, show hyper-radiosensitivity (HRS) as measured by reduced clonogenic cell survival, thereby indicating greater cell lethality than is predicted by extrapolation from high-dose responses. We therefore hypothesized that the cytogenetic response in G2 cells to low doses would also be steeper than predicted by LQ extrapolation from high doses. We tested our hypothesis by exposing four normal human lymphoblastoid cell lines to 0 - …

Environmental Stressors: Pathways Of Exposure And Aquatic Invertebrate Response, Carly Jean Nowicki Jan 2014

Environmental Stressors: Pathways Of Exposure And Aquatic Invertebrate Response, Carly Jean Nowicki

Wayne State University Dissertations

The need to monitor freshwater and detect impairments prior to observable impacts is crucial to maintain species diversity and ecosystem function. Therefore, understanding the contribution of various matrices (i.e., sediment and water) to chemical exposure is critical for remediation of impacted sites.

To evaluate various matrices of chemical exposure, I focused on the organic pollutant polychlorinated biphenyls (PCBs) due to their relatively ubiquitous nature, high toxicity, and adverse effects on humans and wildlife. In complementary laboratory and field experiments, I compared the effects of aqueous versus sedimentary exposure of PCBs on invertebrates. In the laboratory, organisms exposed to PCB-contaminated sediments …

The Mechanism For Inhibition Of Vibrio Cholerae Virulence Gene Expression By Bile And Its Fatty Acid Components, Sarah Plecha Jan 2014

The Mechanism For Inhibition Of Vibrio Cholerae Virulence Gene Expression By Bile And Its Fatty Acid Components, Sarah Plecha

Wayne State University Dissertations

Vibrio cholerae is responsible for the diarrheal disease cholera by producing two major virulence factors: cholera toxin (CT) and toxin-coregulated pilus (TCP). During infection, V. cholerae downregulates its motility genes, which are actively expressed in the environment, and upregulates its virulence genes, which are inactive in the environment. Virulence gene transcription is regulated by the major transcriptional activator, ToxT. ToxT is regulated, in part, by host signals, which in turn, make ToxT active or inactive in respect to virulence gene expression. Host signals include temperature and pH as well as the chemical molecules bicarbonate and bile, which are positive and …

Cervical Remodeling/Ripening At Term And Preterm Delivery: The Same Mechanism Initiated By Different Mediators And Different Effector Cells, Juan Miguel Gonzalez Velez

Wayne State University Dissertations

Premature cervical remodeling/ripening is believed to contribute to preterm delivery (PTD), the leading cause of perinatal morbidity and mortality. Despite considerable research, the causes of term and PTD remain unclear, and there is no effective treatment for PTD. We tested the hypothesis that complement activation plays a role in cervical remodeling and PTD. We studied cervical remodeling at term.

We studied two mouse models of inflammation-induced PTD. The first model was induced by vaginal administration of lipopolysaccharide (LPS)and the second one by administration of progesterone antagonist RU486. Increased cervical C3 deposition and macrophage infiltration and increased serum C3adesArg and C5adesArg …

Characterization Of Initial Iron Binding Location And The Structure/Iron Binding Site On S.Cerevisiae Isu And On D.Melanogaster Frataxin, Andria V. Rodrigues

Wayne State University Dissertations

Iron-induced free radical damage has been implicated in the pathology of diseases of iron overload such as Friedreich's Ataxia, a genetic disorder characterized by an accumulation of iron in actively metabolizing tissues ultimately leading to cardio- and neuro- degeneration and cell death. It is caused by an inability to synthesize the mitochondrial protein, frataxin. Frataxin has been shown by numerous groups to be a part of the iron-sulfur cluster (ISC) multicomplex, where it functions in the capacity of a potential iron provider and an allosteric modulator of both the cysteine desulfurase and scaffold protein ISU. My research has been focused …
