Personalised medicine has finally been featured in psychiatric journals, but psychiatrists have mainly focused on the promise of using disease mechanisms to personalise treatment. Psychiatric disorders such as schizophrenia and depression are not diseases, in the medical sense, and are probably more like syndromes. Instead of spending much time and effort focusing on the mechanisms of diseases that may instead be syndromes, the author believes that psychiatrists should (1) learn more about personalising prescription via drug mechanisms, a pharmacological approach to personalised medicine; and (2) reconsider prior attempts by traditional clinical psychopharmacologists to use sophisticated clinical approaches that try to …

Retrotransposon-Based Profiling Of Mammalian Epigenomes: Dna Methylation Of Iap Ltrs In Embryonic Stem, Somatic And Cancer Cells, Arundhati Bakshi, Joomyeong Kim

© 2014 Elsevier Inc. In the current study, we have used HT-TREBS to individually analyze the DNA methylation pattern of 4799 IAP LTR retrotransposons in embryonic stem, somatic and Neuro2A cells. According to the results, half of the loci within this family show constant methylation patterns between the three cell types whereas the remaining half display variable levels of methylation. About half of the variably methylated IAP LTRs tend to be hypomethylated in ES cells, and nearly all in this group are hypomethylated in Neuro2A cells. Drosophila melanogaster has been used as a model organism for understanding muscle development. Drosophila flight and jump muscles are distinct functionally and biochemically. This distinction is due to differentially expressed genes and differentially spliced mRNA transcripts. The exact mechanism of alternative splicing in somatic muscles is not well characterized. Aret was previously shown to be a transcriptional repressor, and has been implicated in splicing regulation based upon literature analysis and preliminary work. Monitoring the distribution and subsequent effects of nanoparticle (NP) contaminants in aquatic ecosystems will be pivotal to developing regulations that minimize their environmental footprint. Regulators are in a unique position to take a proactive role in shaping how we produce and consume nanomaterials as opposed to the reactive role they have had to adopt with other contaminants. Eradication of Mycobacterium tuberculosis (MTB) infection requires daily administration of combinations of rifampin (RIF), isoniazid [isonicotinylhydrazine (INH)], pyrazinamide, and ethambutol, among other drug therapies. To facilitate and optimize MTB therapeutic selections, a mononuclear phagocyte (MP; monocyte, macrophage, and dendritic cell)-targeted drug delivery strategy was developed. Long-acting nanoformulations of RIF and an INH derivative, pentenyl-INH (INHP), were prepared, and their physicochemical properties were evaluated. This included the evaluation of MP particle uptake and retention, cell viability, and antimicrobial efficacy. Drug levels reached 6 μg/10(6) cells in human monocyte-derived macrophages (MDMs) for nanoparticle treatments compared with 0.1 μg/10(6) cells for native drugs. …

Almost fifty years after the Supreme Court revived the doctrine, substantive due process remains a puzzle. Detractors insist it is nothing more than judicial policy making. Defenders say it accords with the deepest values of the Constitution. But on all sides, the present scholarly debate suffers from an impoverished understanding of modern substantive due process's intellectual history, which has led to an impoverished understanding of the doctrine's core normative content. It is time for a revisionist turn. This Article supplies that turn by excavating the intellectual origins of modern substantive due process and relating that history to the doctrine's development. …

Glucose transport in humans is a vital process which is tightly regulated by the endocrine system. Specifically, the insulin hormone triggers a cascade of intracellular signals in target cells mediating the uptake of glucose. Insulin signaling triggers cellular relocalization of the glucose transporter protein GLUT4 to the cell surface, which is primarily responsible for regulated glucose import. Pathology associated with the disruption of this pathway can lead to metabolic disorders, such as type II diabetes mellitus, characterized by the failure of cells to appropriately uptake glucose from the blood. Nursing care occurs on a continuum that involves transferring the responsibility of a patient’s care from one nurse to another during shift change. Hand-off reports are a vital part of the communication cycle to ensure safe and effective care. Studies have shown bedside reporting increases patient and nurse satisfaction as well as the nurse-patient relationship (Agency for Healthcare Research and Quality, 2013b). Hydraulic fracturing creates large volumes of produced water, contaminated with organics, suspended solids, and dissolved minerals. This water must either be disposed of in injection wells or treated for reuse. One option is to use a combination of ultrafiltration and nanofiltration to treat this water and reuse it in future hydraulic fracturing wells. The object of this study was to test and characterize commercial nanofiltration membranes. The work done for this project will lead to future work in modifying membranes to improve the rejection of desired components.

Objective—Recent increases in the nonmedical use of pharmaceutical opioids and the adverse outcomes associated with them have stimulated a large amount of research and data collection on this public health problem. Systematic organization of the available data sources is needed to facilitate ongoing research, analysis, and evaluation. This work offers a systematic categorization of data sources regarding the nonmedical use of pharmaceutical opioids in the United States.

Objective: Named entity recognition (NER) is one of the fundamental tasks in natural language processing (NLP). In the medical domain, there have been a number of studies on NER in English clinical notes; however, very limited NER research has been done on clinical notes written in Chinese. The goal of this study is to develop corpora, methods, and systems for NER in Chinese clinical text.

Feeling concern about the suffering of others is considered a basic human response, and yet we know surprisingly little about the cultural factors that shape how people respond to the suffering of another person. To this end, we conducted 4 studies that tested the hypothesis that American expressions of sympathy focus on the negative less and positive more than German expressions of sympathy, in part because Americans want to avoid negative states more than Germans do. In Study 1, we demonstrate that American sympathy cards contain less negative and more positive content than German sympathy cards. In Study 2, we …

Recent studies have identified a cholestatic variant of nonalcoholic fatty liver disease (NAFLD) with portal inflammation and ductular reaction. Based on reports of biliary damage, as well as increased circulating free fatty acids (FFAs) in NAFLD, we hypothesized the involvement of cholangiocyte lipoapoptosis as a mechanism of cellular injury. Here, we demonstrate that the saturated FFAs palmitate and stearate induced robust and rapid cell death in cholangiocytes. Palmitate and stearate induced cholangiocyte lipoapoptosis in a concentration-dependent manner in multiple cholangiocyte-derived cell lines. The mechanism of lipoapoptosis relied on the activation of caspase 3/7 activity. There was also a significant up-regulation …

Two different Illinois newspapers have sued the University of Illinois-Springfield (UIS) and the University of Illinois at Urbana-Champaign (UIUC) for the release of documents pursuant to Illinois’ Freedom of Information Act (FOIA). These cases present somewhat new, interesting, and developing problems in the law, including, but not limited to: the public’s right to information, student privacy rights, and whether a federal law like the Federal Education Rights and Privacy Act (FERPA) can override any state’s FOIA. © 2014 Elsevier Inc. Reductive methylation of lysyl side-chain amines has been a successful tool in the advancement of high-resolution structural biology. The utility of this method has continuously gained ground as a protein chemical modification, first as a tool to aid protein crystallization and later as a probe in protein nuclear magnetic resonance (NMR) spectroscopy. As an isotope-labeling strategy for NMR studies, reductive methylation has contributed to the study of protein-protein interactions and global conformational changes. Language disorder is a deleterious, but poorly understood, symptom of schizophrenia. Burgeoning research suggests that it also occurs in depression and bipolar disorders. Building on prior research, the present study evaluated the degree to which language disorder was associated with neurocognitive deficits in a sample of patients with schizophrenia and mood disorders. Employed in this study were a standard neurocognitive battery and a behavior-based analysis of language disorder from natural speech. Speech samples were collected from two separate counterbalanced conditions administered a week apart. I stand in fundamental agreement with what Thomas Schärtl has said in his article describing recent trends in US Catholicism. I am a lifelong Catholic and a lifelong Democrat. I felt personally distressed and discouraged by the support given to Mitt Romney and the Republicans by some leading US Catholic bishops. Most of this support may have technically passed the legal test of being nonpartisan, but undeniably it functioned in a partisan manner, as did the attacks launched on President Obama in the midst of a campaign to defend religious liberty.
