The work presented in this dissertation outlines the role of stem cells in the remodeling of adipose tissue under conditions of nutrient excess. Obesity-induced variations in adipose tissue stem cell distribution were uncovered by analysis of the stromal vascular fraction isolated from mice fed a high fat diet for several weeks. Bone marrow cell transplantation was used to determine the derivation of progenitor cells found in adipose tissue; and supplementation of depleted progenitor cell populations achieved via direct cell transplantation, helped to determine the contribution of these progenitor cells to the remodeling process. We receive most information about our surrounding space and objects through the eyes. To reconstruct the 3D space and objects in the visual system from the 2D retinal images, surface representation must be a critical intermediate stage in the visual processing stream. It is hypothesized in the dissertation that the visual system represents textured surface by a border-to-interior strategy: boundary contours would be encoded first and then border-ownership assignments would be resolved. This process would solve the related problems such as figure-ground segregation, surface depth relationship, occlusion, transparency, etc. As a result, the boundary contours of the surfaces would be …

Electrical stimulation has been increasingly used by research groups to enhance and increase maturation of cells in an engineered cardiac tissue (ECT). Current methods are based on using off-the-shelf wires or electrodes to deliver a stimulus voltage to the in-vitro tissue in culture medium. A major issue with this approach is the generation of byproducts in the medium due to the voltage levels required, which are typically in the range of 5V-10V. Spinal cord injury (SCI) is a significant public health concern that leaves patients with a multitude of life-long disabilities. Major complications of SCI apart from paralysis, include deficits in bladder and bowel function. Lower urinary tract dysfunction continues to remain a top priority issue affecting quality of life for this population. The majority of visceral organs receive a dual sensory innervation from both spinal nerves as well as the vagus nerve. Skeletal muscle constitutes a highly plastic and malleable tissue that rapidly and profoundly adapts to various environmental and physiological stimuli. This response takes place by means of regulated signaling transduction which elicits changes in gene expression, biochemical, and metabolic properties. Adult muscle is composed by alignment of multinucleated, post mitotic muscle fibers formed during embryonic and neonatal development. Residing in the niche of mature muscle fibers is a small population of undifferentiated muscle progenitors termed satellite cells. Quality program evaluation investigates both the process and the outcomes of an intervention in order to accurately determine its effectiveness and impact. In the present study, the effectiveness of CHOICES, a brief, applied-theatre intervention that addresses cyberbullying and suicide was evaluated. The fidelity of implementation of the intervention was assessed to determine whether the delivery of CHOICES was consistent with the original goals of the program, as well as consistent across multiple deliveries. Investigation of the fidelity of implementation revealed that CHOICES was not implemented with high levels of fidelity. The study also determined whether exposure to CHOICES led to …

The Louisville and Jefferson County Metropolitan Sewer District (MSD) has developed a Long-Term Control Plan (LTCP) to address the combined sewer overflow (CSO) issue through a combination of gray and green infrastructure (GI) stormwater management practices in the city of Louisville. In support of this effort, the MSD installed a series of GI stormwater control measures (SCMs) to abate the CSO volumes in an urban sewershed basin. The type IIa sodium-phosphate cotransporter (Npt2a) is expressed in the apical membrane of the renal proximal tubule and is responsible for the reabsorption of the majority of the filtered load of phosphate. Parathyroid hormone (PTH) is secreted by the parathyroid gland in response to a decrease in serum calcium or an increase in serum phosphorus, and acutely induces phosphaturia through the rapid stimulation of endocytosis of Npt2a and its subsequent lysosomal degradation. Chronic PTH stimulation leads to sustained phosphaturia, but the mechanisms for the chronic regulation of Npt2a by PTH remain unclear. We hypothesize that PTH decreases Npt2a mRNA levels …

Stress is a pervasive issue among traditional college students; affecting their choice in health behaviors, and overall academic performance. What is not clear is why some students, despite the pervasiveness of stressors are able to do well in school and maintain/improve their health, while other students struggle to maintain their health and do poorly in school. Aaron Antonovsky theorized that when confronted with a stressor a person with a high sense of coherence (SOC) will be motivated to cope, believe the challenge is understood and that the necessary resources to cope with the stressor are available. The development of nanomaterials as medical diagnostic and therapeutic agents is a rapidly expanding field. A brief search of the literature revealed that roughly half of all articles related to nanoparticle-based drug delivery were published after the onset of this thesis research, over the past 4 years. A few nano-based formulations have entered into clinical use, including liposomal drug formulations, protein-linked drugs, and nanoparticle-based MRI contrast agents. My dissertation provides theoretical insights on individual level entrepreneurial identity in three inter-related essays, drawing on multiple theories and disciplines. In the first essay, I advance a dual-process model of identity development, contributing to the debates in the broad identity literature. I articulate that the intrinsic prospection of identity is another fundamental process of identity formation, besides the social construction of identity, the prevalent notion in the literature. I propose a comprehensive conceptual framework of entrepreneurial identity. Detrusor-sphincter dyssynergia after chronic spinal cord injury (SCI) leads to over-distention, high bladder pressures and vesico-ureteric reflux into the kidney, resulting in repeated infections and pyelonephritis. Given the importance/limited knowledge of SCI effects on the upper urinary tract, this study assessed expression of two proteins in the kidneys of contused (T9) male rats relative to controls that are indicative of tissue stress/damage (TGFβ, CD11b). The impact of two therapeutic rehabilitation strategies, 60 minutes of step training and general exercise, was then assessed on these protein levels relative to non-trained SCI controls, plus the effectiveness of a shorter training duration (30 …

Running economy (RE) is the amount of oxygen consumed while running at a submaximal intensity. Along with aerobic capacity (VO2max), RE is an important predictor of running performance. Little research has investigated changes in RE with restricted breathing training [i.e. controlled breath-holding (CFB)] during exercise. RE may improve ~6% amongst a novice swimming cohort through CFB training, but this has not been established in elite swimmers. The purpose was to further establish that CFB training (16 sessions of 12 x 50-m with ~15 seconds rest between each 50-m, using only ~2 breaths per 50-m) can improve RE in 25 elite …

A relationship between periodontal disease and oral squamous cell carcinoma (OSCC) has been demonstrated in recent studies. Porphyromonas gingivalis’ antigen have been found in oral cancer cells. The embryonic program “epithelial-mesenchymal transition” (EMT) is thought to trigger cancer invasion by allowing tumor cell propagation. The objective of this study was to examine the role of P. gingivalis in the expression of ZEB1, vimentin, and repression of miRNA-200 family as EMT prognostic markers in cancer cell metastasis. Dr. Braun’s students in the Polymer Electronics Lab currently have a way to measure the light intensity from their light emitting devices however it consumes an unnecessary amount of space and power. I offer to improve upon the existing transimpedance circuit that Dr. Braun currently uses, reducing the total space occupied by the circuit and the power consumption of the current setup. The transimpedance circuit measures the incidence light intensity from the polymer-based photo detector and outputs an accurate, discrete, and measurable voltage. This purpose of this project, sponsored by Boeing and advised by Dr. Arakaki and Dr. Taufik, is to build a high voltage pulse width generator that lyses algae cells to harvest the biofuel within. This project uses Emmanuel Loza's thesis on a cascaded high voltage converter with variable control to build a portable pulse width generator with independent control on the peak voltage, pulse width, and frequency. The project focuses on creating a pulse width generator capable of finding the most optimal pulsed electric field to lyse algae. Trade secret law arose to help companies protect confidential information (e.g., the Coca-Cola formula) from competitors seeking to copy their innovative efforts. But companies increasingly use trade secret law to block a wide swath of information from the scrutinizing eyes of consumers, public watchdog groups, and potential improvers. Companies can do this, in part, because trade secret law lacks clear limiting doctrines that consider the social benefits of unauthorized use. For example, trade secret law makes no allowance for the departing employee that uses proprietary information to create a substantially improved product or disclose public health risks.

There are multiple breeds of beef cattle available to utilize in breeding systems to maximize production and economics. Calving difficulty (dystocia) is a significant cost to beef production and is more prevalent in first-calf heifers. The objectives of this study were to estimate genetic parameters and breed differences for calving difficulty and birth weight as a first step towards the development of across-breed adjustment factors for calving difficulty.

Most triple-negative breast cancers (TNBCs) exhibit gene expression patterns associated with epithelial-to-mesenchymal transition (EMT), a feature that correlates with a propensity for metastatic spread. Overexpression of the EMT regulator Slug is detected in basal and mesenchymal-type TNBCs and is associated with reduced E-cadherin expression and aggressive disease. The effects of Slug depend, in part, on the interaction of its N-terminal SNAG repressor domain with the chromatin-modifying protein lysine demethylase 1 (LSD1); thus, we investigated whether tranylcypromine [also known as trans-2-phenylcyclopropylamine hydrochloride (PCPA) or Parnate], an inhibitor of LSD1 that blocks its interaction with Slug, suppresses the migration, invasion, and metastatic …

To explore a mother's feeling of attachment and the affects her working status on the attachment relationship with her child, upon ethical clearance from the institutional ethics committee, in-depth interviews of nine participants were conducted. Mothers enrolled were those who resumed the employment within the first year of post-delivery and were having a child up to two years of age. Results revealed that maternal employment itself does not enhance or deteriorate attachment with the child. It is combinations of factors that revolve around it impact on their bond. The significance of this study was to evaluate multiple underlying factors among African Americans in Shreveport, Louisiana and the influence of those factors on their relationship with HIV/AIDS. This is extremely important for two very reasons. First, in the past 30 years since the initial presentation of HIV/AIDS into society, a shift in the demographic of the infected individuals has resulted in African Americans becoming the new face of HIV/AIDS in the United States. Secondly, African Americans are only 12 to 13% of the United States population, but they represent more than 45% of all new HIV/AIDS cases reported annually. …

This study investigated the online experiences of 10 adolescents with Asperger's Disorder (AD). This study was exploratory in nature and employed a qualitative approach. Three research questions guided this study: (a) What are the positive and negative online experiences of youth with ASD?, (b) What are the perceptions of online interactions in comparison to offline interactions held by youth with ASD?, and (c) What experiences with social connectedness and cyberbullying do youth with ASD have as a result of Internet usage?
