Deleted in breast cancer 1 (DBC1, CCAR2, KIAA1967) is a large, predominantly nuclear, multidomain protein that modulates gene expression by inhibiting several epigenetic modifiers, including the deacetylases SIRT1 and HDAC3, and the methyltransferase SUV39H1. DBC1 shares many highly conserved protein domains with its paralog cell cycle and apoptosis regulator 1 (CCAR1, CARP-1). In this study, we examined the full-length sequential and structural properties of DBC1 and CCAR1 from multiple species and correlated these properties with evolution. Our data shows that the conserved domains shared between DBC1 and CCAR1 have similar domain structures, as well as similar patterns of predicted disorder …

Tall Fescue Seed Extraction And Partial Purification Of Ergot Alkaloids, Huihua Ji, Franklin Fannin, J. Klotz, Lowell P. Bush
Kentucky Tobacco Research and Development Center Faculty Publications
Many substances in the tall fescue/endophyte association (Schedonorus arundinaceus/Epichloë coenophiala) have biological activity. Of these compounds only the ergot alkaloids are known to have significant mammalian toxicity and the predominant ergot alkaloids are ergovaline and ergovalinine. Because synthetically produced ergovaline is difficult to obtain, we developed a seed extraction and partial purification protocol for ergovaline/ergovalinine that provided a biologically active product. Tall fescue seed was ground and packed into several different sized columns for liquid extraction. Smaller particle size and increased extraction time increased efficiency of extraction. Our largest column was a 114 × 52 × …

Moral Anxiety, Mortal Terror: Considering Spielberg, Post-9/11, Jake T. Bart
Cinesthesia
In the wake of 9/11, a defining moment in the young 21st century, Spielberg’s thematic concerns undergo a marked evolution. As film historian Joseph McBride noted, “(n)o other American artist confronted the key events of the first decade of the century with such sustained and ambitious treatment” (450). Together, Spielberg’s War of the Worlds (2005), Munich (2005), and Minority Report (2002) create an informal trilogy, each exploring a different facet of American shock and anxiety in the War on Terror era.

The plant galactolipids monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG) have been linked to the anti-inflammatory and cancer benefits of a green leafy vegetable diet in humans due to their ability to regulate the levels of free radicals like nitric oxide (NO). Here, we show that DGDG contributes to plant NO as well as salicylic acid biosynthesis and is required for the induction of systemic acquired resistance (SAR). In contrast, MGDG regulates the biosynthesis of the SAR signals azelaic acid (AzA) and glycerol-3-phosphate (G3P) that function downstream of NO. Interestingly, DGDG is also required for AzA-induced SAR, but MGDG is not. Notably, …

Glutamyl-tRNA (Glu-tRNAGlu) is the common substrate for both protein translation and heme biosynthesis via the C5 pathway. Under normal conditions, an adequate supply of this aminoacyl-tRNA is available to both pathways. However, under certain circumstances, Glu-tRNAGlu can become scarce, resulting in competition between the two pathways for this aminoacyl-tRNA. In Acidithiobacillus ferrooxidans, glutamyl-tRNA synthetase 1 (GluRS1) is the main enzyme that synthesizes Glu-tRNAGlu. Previous studies have shown that GluRS1 is inactivated in vitro by hydrogen peroxide (H2O2). This raises the question as to whether H2O2 negatively affects …

New 4-Aminoquinoline Compounds To Reverse Drug Resistance In P. Falciparum Malaria, And A Survey Of Early European Antimalarial Treatments, Katherine May Liebman
Dissertations and Theses
Intermittent fevers caused by Plasmodium parasites have been known for millennia, and have caused untold human suffering. Today, millions of people are afflicted by malaria each year, and hundreds of thousands die. Historically, the most successful synthetic antimalarial drug was chloroquine, as it was safe, inexpensive, and highly efficacious. However, plasmodial resistance to chloroquine now greatly limits its utility. Previously in our laboratories it has been shown that attachment of a "reversal agent moiety" to the side chain of chloroquine can result in the restoration of activity against chloroquine-resistant strains of P. falciparum malaria. In the first part of the …
A Comparison Between High-Risk And Low-Risk Children With Autism Spectrum Disorder, Kathryn Bradbury
A Comparison Between High-Risk And Low-Risk Children With Autism Spectrum Disorder, Kathryn Bradbury
Master's Theses
Due to the elevated recurrence rates in high-risk younger siblings of children with ASD, the population is frequently used to examine early ASD symptomatology. However, the representative nature of this population compared to the general population of children with ASD is unknown. Previous research also suggests that parents of children with ASD raise concerns about the development of subsequent children earlier than first-time parents or parents of typically developing children. Whether an actual discrepancy exists in the attainment of milestones in these populations has not yet been assessed. The current study proposes to compare high- and low-risk children with ASD …

Background: We have previously characterized several antihypertensive peptides in simulated digests of cooked eggs and showed blood pressure lowering property of fried whole egg digest. However, the long-term effects of this hydrolysate and its fractions on blood pressure are not known. Therefore, the objectives of the study were to determine the effects of long term administration of fried whole egg hydrolysate and its fractions (i.e. egg white and egg yolk) on regulation of blood pressure and associated factors in cardiovascular disease such as plasma lipid profile and tissue oxidative stress.
Methods and Results: We used spontaneously hypertensive rats (SHR), an …

It was August 21, 1936, in the days when Kansas was a part of our country known as the Dust Bowl. The sun shone down on a parched and thirsty earth. The lawn, which should have been a beautiful green velvet carpet, was instead an ugly brown expanse of dead grass whose roots had long ago given up the struggle to reach life-giving moisture. Where no vegetation grew, the ground was cracked and broken. The trees, prematurely brown, swayed lazily in a hot breeze which came drifting in from the west. Birds, mostly boisterous bluejays, friendly robins, twittering sparrows, and …

I was told at lunch by one of my high school friends that Wally wanted to see me at two minutes after two on the second floor bridge. That was Wally all right, always doing something peculiar and very exact. I was on my way to the bridge at two, and as I rounded the corner of it, I saw that the "Remarkable" was already there. He was standing tall and gangly in front of the window looking down at the street and appearing in a rather desolate and serious mood.

The castle was a huge gray mass of stone, high on the hill. Once it had been the splendor and austerity of Tintagel, castle of King Arthur. Now, a bleak gray ruin, the splendor and awe-inspiring quality persisted.

The train moved into the reflection of the dawn. Paul opened his eyes and looked out at the grey-black silhouettes of telephone poles stalking by, and the slowly rounding land. For a moment in the dawn, they looked like half-remembered poles and fields moving in a dream. But then he was awake and knew he was awake and California was ahead.

Fathead minnow and zebrafish are among the most intensively studied fish species in environmental toxicogenomics. To aid the assessment and interpretation of subtle transcriptomic effects from treatment conditions of interest, better characterization and understanding are needed for natural variation in gene expression among fish individuals from lab cultures. Leveraging the transcriptomics data from a number of our toxicogenomics studies conducted over the years, we conducted a meta-analysis of nearly 600 microarrays generated from the ovary tissue of untreated, reproductively mature fathead minnow and zebrafish samples. As expected, there was considerable batch-to-batch transcriptomic variation; this "batch-effect" appeared to differentially impact subsets …

Views By St Louis County Residents Regarding The Police And Public Responses To The Shooting Of Michael Brown In Ferguson, Missouri On August 9, 2014, Tammy R. Kochel
Reports
In their own words, residents living in high crime, disadvantaged neighborhoods of St. Louis County, MO revealed how they were personally impacted by the shooting of Michael Brown in Ferguson and the police and public response to it during the first few weeks following the event last August. Three quarters of residents reported being impacted in some way. The most common negative effects were emotional, as well as the daily inconvenience brought on by closed stores and schools and traffic issues. Residents’ opinions about police also suffered. Concerns about the police response relate to a perceived overreaction and specifically, use …

The Role Of Mate Preference And Personality On Reproductive Performance In An Ex-Situ Conservation Breeding Program For The Giant Panda (Ailuropoda Melanoleuca), Meghan S. Martin
Dissertations and Theses
Successful captive-breeding and re-introduction programs must have the ability to breed a surplus of genetically suitable animals for release into the wild. Unfortunately, many individuals in captive breeding programs often do not reproduce even when they are apparently healthy and presented with genetically appropriate mates. Mate choice can affect multiple parameters of reproductive sperformance, including mating success, offspring production, survival, and fecundity. We investigated the role of mate preference and personality on the reproductive performance of male and female giant pandas (Ailuropoda melanoleuca) as measured by intromission success and litter production. We conducted these studies on giant pandas at the …

The Non-Lethal Effects Of Climate Change On The Territoriality Of Lottia Gigantea, Tracey Gunanto, Christina Chavez, Jessica Martinez, William G. Wright
Student Scholar Symposium Abstracts and Posters
The intertidal zone has been described as ground zero for global warming. Here, the owl limpet, Lottia gigantea, adapted to the cool ocean temperatures, must withstand a few hours of baking sun during day-time low tides. This hardship is predicted to increase in frequency and severity in the future as the globe warms. Our research hypothesized that heat events compromise territorial behavior of L. gigantea. All observations and experiments were performed at Inspiration Point near Newport Beach, California. We measured the natural radiant temperature of tagged limpets during day-time low tides using a field-calibrated infrared “thermogun”. We also …
Impact Of Leaflet Material On Transcatheter Aortic Valve Function, Andrea Mandragouras
Impact Of Leaflet Material On Transcatheter Aortic Valve Function, Andrea Mandragouras
Master's Theses
There has been a huge push in the medical device industry to make the current transcatheter aortic valves (TAV) smaller. This would shift some patients who are receiving transapical access site which is in the heart to the transfemoral site which is in the leg. With a decreased hospital stay, and higher endpoint safety transfemoral is typically preferred. Some patients are forced to receive the transapical approach because of the small arteries in their leg and the size of the device. The current TAVs leaflets are made from thick porcine or bovine pericardium which causes the bulkiness in the device. …
