Matrix Metalloproteinases (MMPs) are a family of more than 25 structurally-related enzymes that can degrade a variety of substances including collagen. Basal levels of these enzymes play an important role in tissue turnover under normal physiological conditions, while excessive levels of these enzymes may cause or exacerbate many diseases related to tissue loss, including arthritis, cancer, cardiovascular disease, coma, periodontal disease, stroke etc. For decades, research teams from both industrial companies and academic institutions have been trying to develop an effective matrix metalloproteinase inhibitor as a drug to treat MMP-involved diseases. Part I. Vinyl iodides with defined geometry are important chemical building blocks. Their preparation by iododesilylation of the corresponding vinyl silane is an attractive method. Kishi et al. have shown that if iododesilylation is carried out with N-iodosuccinimide (NIS) in an acetonitrile/chloroacetonitrile mixture, one obtains vinyl iodides in which the geometry of the olefin is highly retained. By carrying out iododesilylation with NIS in hexafluoroisopropanol, Zakarian et al. have shown that the olefin geometry is also retained, but to a larger degree. Bacteria use numerous small molecules for cellular signaling. These primary and secondary messengers act within the cell to relay signal transduction and regulate distinct pathways. Among these the diatomic gas molecule nitric oxide (NO) and the nucleotide cyclic-di-GMP play central role in virulence, quorum sensing, and biofilm formation. In this dissertation, we have focused on H-NOX, a heme-nitric oxide/oxygen binding protein in the biofilm-dwelling bacterium Shewanella woodyi (Sw), which mediates NO-induced biofilm dispersal by modulating the activity of a dual-functioning diguanylate cyclase/phosphodiesterase enzyme that we have named HaCE, (H-NOX-associated cyclic-di-GMP enzyme). Conjugated organic polymers have optical and electronic properties that make them candidates for applications in organic field effect transistors (OFETs) and light emitting materials. Polydiacetylenes (PDAs) are an important branch of conjugated polymers with alternating poly(ene-yne) structure. The all-carbon polymer without any side groups is carbyne, which has only alternating single and triple carbon-carbon bonds. Carbyne provides a good opportunity to investigate the inherent properties of conjugated polymers. Polydiiododiacetylene (PIDA), a simple PDA with only iodine atom substituents, can be successfully obtained by the topochemical polymerization of the monomer diiodobutadiyne. Poly(Diiododiacetylene): A Potential Precursor For New All-Carbon Materials, Daniel Joseph Resch

Poly(diiododiacetylene) (PIDA) is a polymer consisting entirely of carbon and iodine. The polymer is prepared by cocrystallizing a bis(nitrile) oxalamide host with the monomer diiodobutadiyne. These compounds are held together by a halogen bond and an ordered 1,4-topochemical polymerization occurs in the solid state. The formation of the monomer cocrystals was found to be highly solvent dependent. Acetonitrile was found to greatly improve the yield of cocrystals over solvents used in the past. Cocrystals could not be obtained from other solvents such as dimethoxyethane and acetone. THF did give some cocrystal but the yield was poor. The use of acetonitrile …

Despite the continued debate regarding the structure of temperament in young children, it is often assumed that temperament traits demonstrate temporal stability over time. This research has relied predominately on parent-report measures. The present study used an alternative approach, a laboratory-observational measure (Laboratory Temperament Assessment Battery [Lab-TAB]), to examine the structural and relative stability of temperament traits in a community sample of young children (N = 440). Using structural equation modeling (SEM), we derived a similar five-factor structure consisting of the dimensions of Positive Affect/Interest, Sociability, Dysphoria, Fear/Inhibition, and Impulsivity vs. Constraint at both age 3 and 6 years, suggesting …

The sperm acrosome reaction (AR), an essential step in mammalian fertilization, is mediated by a highly species-specific interaction of sperm surface molecules with glycan moieties on the egg. Many previous studies indicate that a subset of terminal carbohydrate residues on the mouse egg zona pellucida (ZP) trigger the AR by cross-linking or aggregating receptors on the sperm membrane. However, the exact role of those carbohydrates in AR has not been identified and the mechanism underlying the induction of the AR still needs further investigation. Effective conservation of biological diversity requires accurate assessments of species extinction risks. Both under- and over-estimating extinction risks can have serious consequences. Overlooking species that are in need of conservation management can result in extinction or expensive recovery efforts. Misidentifying species which have stable populations as threatened can misdirect limited conservation resources or unnecessarily halt economic activity and development. I used a retrospective multi-modeling framework to explore the extinction of a once abundant and wide-spread North American species, the Passenger Pigeon (Ectopistes migratorius). I simulated three different types of anthropogenic impacts: harvest, habitat loss, and nest disturbance. I …

Analog circuit design activities mainly depend on designers' expertise and their ability to produce new designs by combining basic devices, sub-circuits, and ideas from similar solutions as the source for innovation. There are very few systematic methods that can characterize similarities and differences between analog circuits while maintaining the correlation between design variables, performance, and trade-offs. Moreover, many computer-aided design tools are focused only on routine tasks, like transistor sizing and layout design. This dissertation discusses how literary writers of the Victorian age render toxicity and toxic behaviors visible in order to encourage reform, as well as offering models of healthy environmental engagement. This thesis examines line between human and non-human animals in Jonathan Swift's Gulliver's Travels and H.G. Wells's The Island of Doctor Moreau. Both Gulliver's Travels and The Island of Doctor Moreau examine this separation through the prism of language, societal views of animality, and biology. In this thesis we study the problem of modeling cross-sectional volatility structure of U.S. stock market. The thesis contains two parts. In the first part we identify a particular volatility spike phenomenon, that the cross-sectional volatility is significantly stronger at every 5 minute. An empirical spike study is conducted on individual stock level by applying Lee-Mykland jump detector. By constructing spike ratio statistics, the evolving paths of this spike phenomenon is studied during the 1993 to 2009 time period. Synalpheus shrimp species of the gambarelloides group are the only marine organisms displaying the highest level of social functioning, eusociality. Their social hierarchies are equally complex compared to the reproductive abnormalities that have been recently discovered. For instance, snapping shrimp of the genus Synalpheus were thought to be gonochoric, i.e. developing as independent sexes, until scanning electron microscopy studies revealed intersexed gonopores in several species. This project analyzed both the species composition, and accompanying reproductive structures, of Synalpheus spp. (Caridea: Alpheidae) comprised of densely aggregating communal and pair-living colonies in the Florida Keys, Florida.

Racial differences in the pathophysiology of atherothrombosis are poorly understood. We explored the function and transcriptome of platelets in healthy black (n = 70) and white (n = 84) subjects. Platelet aggregation and calcium mobilization induced by the PAR4 thrombin receptor were significantly greater in black subjects. Numerous differentially expressed RNAs were associated with both race and PAR4 reactivity, including PCTP (encoding phosphatidylcholine transfer protein), and platelets from black subjects expressed higher levels of PC-TP protein. PC-TP inhibition or depletion blocked PAR4- but not PAR1-mediated activation of platelets and megakaryocytic cell lines. miR-376c levels were differentially expressed by race and …

The organic-rich Middle Devonian Marcellus Shale of the Appalachian basin is a rapidly developing natural gas play. Stratigraphic boundaries of the Marcellus Shale in Westmoreland County, Pennsylvania were identified using geophysical logs from 10 vertical gas-producing wells in a 23 sq. km area. Gamma-ray, bulk density, and resistivity well logs were examined to assess hydrocarbon potential. Values of porosity, total organic carbon (TOC), and water saturation (SW) were derived and mapped by incorporating well-log data into Marcellus-specific formulas. Concrete has a large environmental footprint so it is desirable to find ways to efficiently design structural members. Engineers can exercise their abilities early on in the design phase of construction projects to reduce the environmental footprint by minimizing the amount of materials required. One way to achieve these results is to optimize the design of structural concrete.
