Phenethylisothiocyanate Alters Site- And Promoter-Specific Histone Tail Modifications In Cancer Cells, Yi Liu, Suvobrata Chakravarty, Moul Dey

Chemistry and Biochemistry Faculty Publications

Site-specific histone modifications are important epigenetic regulators of gene expression. As deregulation of genes often results in complex disorders, corrective modulation of site-specific histone marks could be a powerful therapeutic or disease-preventive strategy. However, such modulation by dietary compounds and the resulting impact on disease risk remain relatively unexplored. Here we examined phenethylisothiocyanate (PEITC), a common dietary compound derived from cruciferous vegetables with known chemopreventive properties under experimental conditions, as a possible modulator of histone modifications in human colon cancer cells. The present study reports novel, dynamic, site-specific chemical changes to histone H3 in a gene-promoter-specific manner, associated with PEITC …

Landscape Features Impact On Soil Available Water, Corn Biomass, And Gene Expression During The Late Vegetative Stage, Stephanie Hansen, Sharon A. Clay, David E. Clay, C. Gregg Carlson, Graig Reicks, Youssef Jarachi, David Horvath

Agronomy, Horticulture and Plant Science Faculty Publications

Crop yields at summit positions of rolling landscapes often are lower than backslope yields. The differences in plant response may be the result of many different factors. We examined corn (Zea mays L.) plant productivity, gene expression, soil water, and nutrient availability in two landscape positions located in historically high (backslope) and moderate (summit and shoulder) yielding zones to gain insight into plant response differences. Growth characteristics, gene expression, and soil parameters (water and N and P content) were determined at the V12 growth stage of corn. At tassel, plant biomass, N content, 13C isotope discrimination (Δ), and …

Examining The Confluence Of Literature And Mathematics Through Art, Sofia Alavosius May 2013

Examining The Confluence Of Literature And Mathematics Through Art, Sofia Alavosius

Senior Honors Projects

Twentieth century novelist and scientist C. P. Snow noted the division of “two cultures,” namely the divide between the sciences and the humanities. Snow argued these were distinctive and separated cultures in the mid 1950s. The “two cultures” are also present during the Renaissance. This appears in the artwork and literary and mathematical texts of the period. The divide between the humanities and sciences is something that we can easily distinguish in the twenty-first century as well.

I use the discourse of C. P. Snow’s “two cultures” to work through this relationship and to explore the cultural perspectives of these …

Multicellular Phenotypic Studies Of Single Gene Variants In Myxococcus Xanthus, Jinyuan Yan

Biology - Dissertations

There are several systematic methods designed to link genes to cellular processes. These methods are derived from different hypotheses and are largely complementary to each other. This dissertation presents a systematic study of functional genetics and related phenotypes using quantitative methods. The first part of this dissertation will report the successful identification and characterization of 28 genes in the multicellular bacterium Myxococcus xanthus using three different methods: sequence homology, transcription activation and protemoics. The results from this research extended the list of M. xanthus genes involved in multicellularity, and expanded our knowledge regarding the possible molecular pathways underlying physiological and …

Thermodynamic And Structural Characterization Of The Interaction Between Wd40 Repeat Containing Protein 5 (Wdr5) And Human Set1 Family Methyltransferases, Venkat Dharmarajan May 2013

Thermodynamic And Structural Characterization Of The Interaction Between Wd40 Repeat Containing Protein 5 (Wdr5) And Human Set1 Family Methyltransferases, Venkat Dharmarajan

Biology - Dissertations

Translocations and amplifications of the mixed lineage leukemia-1 (MLL1) gene are associated with aggressive myeloid and lymphocytic leukemias in humans. MLL1 is a member of the SET1 family of histone H3 lysine 4 (H3K4) methyltransferases, which are required for transcription of genes involved in hematopoiesis and development. MLL1 associates with a sub-complex containing WDR5, RbBP5, Ash2L, and DPY-30 (WRAD), which together form the MLL1 core complex that is required for sequential mono- and dimethylation of H3K4. We previously demonstrated that WDR5 binds the conserved WDR5 interaction (Win) motif of MLL1 in vitro, an interaction that is required for the H3K4 …

Whose Streets? Our Streets!: Identity, Institutions, And Privilege In Student Activism, Christina Limpert

Cultural Foundations of Education - Dissertations & Theses

This dissertation is a qualitative ethnography of college student activism that examines student activist identity, and the role of social structures and privilege in activism. The study uses data from two years of in-depth fieldwork including over 18 months of participant observation and interviews with 13 key respondents. I took on this research project to better understand how students come to think of themselves as activists, how they speak about and experience activism, how activism--as a cultural text--tells a story about power and privilege, and to explore the role of education in the culture of activism. In this dissertation I …

Feminisms At The Door: Gender And Sex At A Women's College, Susan Chappell Overton May 2013

Feminisms At The Door: Gender And Sex At A Women's College, Susan Chappell Overton

Cultural Foundations of Education - Dissertations & Theses

This dissertation is a qualitative study of early 21st century American female college students' experiences of their gender, sexuality, and racial identities, and of institutional politics as their single-sex college transitioned to co-education. It is an ethnography that utilizes feminist theorizing to understand tensions between feminists of different generations or waves; the complexities of identities and institutions in which gender, sexuality, and race intersect (bell hooks and Kimberle Crenshaw); and the workings of dominant gender and sexual norms (Judith Butler). As the College prepared to `go coed,' the prospect of introducing male bodies into this environment generated rich discourses about …

[1] The Cosmic Ray Telescope for the Effects of Radiation (CRaTER) instrument aboard the Lunar Reconnaissance Orbiter has been measuring energetic charged particles from the galactic cosmic rays (GCRs) and solar particle events in lunar orbit since 2009. CRaTER includes three pairs of silicon detectors, separated by pieces of tissue-equivalent plastic that shield two of the three pairs from particles incident at the zenith-facing end of the telescope. Heavy-ion beams studied in previous ground-based work have been shown to be reasonable proxies for the GCRs when their energies are sufficiently high. That work, which included GCR simulations, led to predictions …

Mth1 Regulates The Interaction Between The Rgt1 Repressor And The Ssn6-Tup1 Corepressor Complex By Modulating Pka-Dependent Phosphorylation Of Rgt1, Adhiraj Roy, Yong Jae Shin, Kyu Hong Cho, Jeong-Ho Kim May 2013

Mth1 Regulates The Interaction Between The Rgt1 Repressor And The Ssn6-Tup1 Corepressor Complex By Modulating Pka-Dependent Phosphorylation Of Rgt1, Adhiraj Roy, Yong Jae Shin, Kyu Hong Cho, Jeong-Ho Kim

Biochemistry and Molecular Medicine Faculty Publications

Glucose uptake, the first, rate-limiting step of its utilization, is facilitated by glucose transporters. Expression of several glucose transporter (HXT) genes in yeast is repressed by the Rgt1 repressor, which recruits the glucose-responsive transcription factor Mth1 and the general corepressor complex Ssn6-Tup1 in the absence of glucose; however, it is derepressed when Mth1 is inactivated by glucose. Here we show that Ssn6-Tup1 interferes with the DNA-binding ability of Rgt1 in the absence of Mth1 and that the Rgt1 function abrogated by Ssn6 overexpression is restored by co-overexpression of Mth1. Thus Mth1 likely regulates Rgt1 function not by modulating …

Human Cytomegalovirus Inhibits Apoptosis By Proteasome-Mediated Degradation Of Bax At Endoplasmic Reticulum-Mitochondrion Contacts, Aiping Zhang, Richard L. Hildreth, Anamaris M. Colberg-Poley May 2013

Human Cytomegalovirus Inhibits Apoptosis By Proteasome-Mediated Degradation Of Bax At Endoplasmic Reticulum-Mitochondrion Contacts, Aiping Zhang, Richard L. Hildreth, Anamaris M. Colberg-Poley

Genomics and Precision Medicine Faculty Publications

Human cytomegalovirus (HCMV) encodes the UL37 exon 1 protein (pUL37x1), which is the potent viral mitochondrion-localized inhibitor of apoptosis (vMIA), to increase survival of infected cells. HCMV vMIA traffics from the endoplasmic reticulum (ER) to ER subdomains, which are physically linked to mitochondria known as mitochondrion-associated membranes (MAM), and to mitochondria. The antiapoptotic function of vMIA is thought to primarily result from its ability to inhibit Bax-mediated permeabilization of the outer mitochondrial membrane (OMM). Here, we establish that vMIA retargets Bax to the MAM as well as to the OMM from immediate early through late times of infection. However, MAM …

Understanding Sexual Diversity, Nate Maeys May 2013

Understanding Sexual Diversity, Nate Maeys

Undergraduate Psychology Research Methods Journal

This research assessed comfort levels toward sexual diversity. Sexual diversity encompasses anyone who identifies with a gender or sexual orientation other than one that is socially accepted for his or her gender. This research followed Ceglian and Lyons’ (2004) study which assessed people’s comfort levels towards cross-dressing men. They showed that comfort levels towards cross-dressing men could be altered. They explained their findings using the mere exposure effect which refers to the tendency that over time, people will develop a preference for something or someone they are repeatedly exposed to (Zajonc, 2001). Participants in the study filled out two surveys …

Dynamically Real-Time Anomaly Detection Algorithm With Immune Negative Selection, Lingxi Peng, Wenbin Chen, Dongqing Xie, Ying Gao, Chunlin Liang

Applied Mathematics & Information Sciences

Network anomaly detection has become the promising aspect of intrusion detection. The existing anomaly detection models depict the detection profiles with a static way, which lack good adaptability and interoperability. Furthermore, the detection rate is low, so they are difficult to be deployed the realtime detection under the high-speed network environment. In this paper, the excellent mechanisms of self-learning and adaptability in the human immune system are referred and a dynamic anomaly detection algorithm with immune negative selection, named as DADAI, is proposed. The concepts and formal definitions of antigen, antibody, and memory cells in the network security domain are …

A Computer Aided Pulmonary Nodule Detection System Using Multiple Massive Training Svms, Zhenghao Shi, Minghua Zhao, Lifeng He, Yinghui Wang, Ming Zhang, Kenji Suzuki

Applied Mathematics & Information Sciences

A computer aided pulmonary nodule detection system for chest radiography is proposed. The system consists of three models, viz., lung segmentation, lung nodule candidates detection and false positive reduction. Several innovations are offered in this system. The first one is that the detection of potential lung nodule candidates is conceived as a filtering process that searches for any region which has a spherical structure (where a potential nodule may happen to occur) in a chest radiograph by analysis eignvalues of Hessian matrix of an image. The second one is that a novel two stage classifier based method is developed to …
