Cueing Complex Thought, Shannon Houck Jun 2013

Cueing Complex Thought, Shannon Houck

Graduate Student Theses, Dissertations, & Professional Papers

Does exposure to a complex description subsequently cue a person to be more complex? To test this question, participants read a paragraph about a specific topic. Paragraphs varied in their level of Integrative Complexity. Participants then wrote about either (1) their opinion about the topic that they read about, or (2) their opinion about a topic that is unrelated to the one that they read about. Participant responses were scored for Integrative Complexity. Contrary to expectations, reading complex paragraphs did not cue people to write more complexly, regardless of whether they were assigned to write about a topic related or …

Theses and Dissertations

The goal of this study was to explore whether Strong Kids could result in improved social and emotional competence when implemented as a school-wide universal intervention. No prior studies have examined this question. This study also evaluated whether teachers could implement Strong Kids as it was designed and whether they viewed it as socially valid. It used a non-equivalent control group design. The treatment school in the study involved 348 students and 17 teachers from a Title I school. School demographics indicated that 61% of students were Hispanic, 37% White, and 2% of other ethnicities. Approximately 82% of the students …

Dynamic Functions Of Rhoa In Tumor Cell Migration And Invasion, Kathleen L. O'Connor, Min Chen Jun 2013

Dynamic Functions Of Rhoa In Tumor Cell Migration And Invasion, Kathleen L. O'Connor, Min Chen

Markey Cancer Center Faculty Publications

RhoA is one of the more extensively studied members of the Rho family of small GTPase where it is most readily recognized for its contributions to actin-myosin contractility and stress fiber formation. Accordingly, RhoA function during cell migration has been relegated to the rear of the cell where it mediates retraction of the trailing edge. However, RhoA can also mediate membrane ruffling, lamellae formation and membrane blebbing, thus suggesting an active role in membrane protrusions at the leading edge. With the advent of fluorescence resonance energy transfer (FRET)-based Rho activity reporters, RhoA has been shown to be active at the …

Molecular and Cellular Biochemistry Faculty Publications

Background: Histone wrapping of DNA into nucleosomes almost certainly evolved in the Archaea, and predates Eukaryotes. In Eukaryotes, nucleosome positioning plays a central role in regulating gene expression and is directed by primary sequence motifs that together form a nucleosome positioning code. The experiments reported were undertaken to determine if archaeal histone assembly conforms to the nucleosome positioning code.

Results: Eukaryotic nucleosome positioning is favored and directed by phased helical repeats of AA/TT/AT/TA and CC/GG/CG/GC dinucleotides, and disfavored by longer AT-rich oligonucleotides. Deep sequencing of genomic DNA protected from micrococcal nuclease digestion by assembly into archaeal nucleosomes has established that …

Insulin Promotes Glucose Consumption Via Regulation Of Mir-99a/Mtor/Pkm2 Pathway., Wei Li, Jing Wang, Qiu-Dan Chen, Xu Qian, Qi Li, Yu Yin, Zhu-Mei Shi, Lin Wang, Jie Lin, Ling-Zhi Liu, Bing-Hua Jiang

Kimmel Cancer Center Faculty Papers

Insulin is known to regulate multiple cellular functions and is used for the treatment of diabetes. MicroRNAs have been demonstrated to be involved in many human diseases, including Type 2 diabetes. In this study, we showed that insulin decreased miR-99a expression levels, but induced glucose consumption and lactate production, and increased the expression of mTOR, HIF-1α and PKM2 in HepG2 and HL7702 cells. Forced expression of miR-99a or rapamycin treatment blocked insulin-induced PKM2 and HIF-1α expression, and glucose consumption and lactate production. Meanwhile, knockdown of HIF-1α inhibited PKM2 expression and insulin-induced glucose consumption. Taken together, these findings will reveal the …

Publications and Research

We have previously shown that treatment of androgen-sensitive LNCaP cells with the sphingosine kinase (SK) inhibitor SKi (2-(p-hydroxyanilino)-4-(p-chlorophenyl)thiazole) induces the proteasomal degradation of two N-terminal variants of SK1 (SK1a and SK1b), increases C22:0-ceramide and diadenosine 5′,5′′′-P1,P3-triphosphate (Ap3A) and reduces S1P levels, and promotes apoptosis. We have now investigated the effects of three SK inhibitors (SKi, (S)-FTY720 vinylphosphonate, and (R)-FTY720 methyl ether) on metabolite and sphingolipid levels in androgen-sensitive LNCaP and androgen-independent LNCaP-AI prostate cancer cells. The 51 kDa N-terminal variant of SK1 (SK1b) evades the proteasome in LNCaP-AI cells, and these cells do not exhibit an increase in C22:0-ceramide or …

Anti-Inflammatory Effect Of Adipose-Derived Mesenchymal Stem Cells For Flexor Tendon Healing, Cionne Nirit Manning Jun 2013

Anti-Inflammatory Effect Of Adipose-Derived Mesenchymal Stem Cells For Flexor Tendon Healing, Cionne Nirit Manning

All Theses and Dissertations (ETDs)

Despite advances in surgical techniques over the past three decades, tendon repairs remain prone to poor clinical outcomes. While previous attempts to improve flexor tendon healing have focused on the later stages of healing: i.e., proliferation and matrix synthesis), the early inflammatory phase of tendon healing remains poorly understood and its modulation during healing has not yet been studied. The overall goal of this work is therefore to determine whether flexor tendon healing can be enhanced through modulation of the inflammatory environment after surgical repair.

To meet this objective, the inflammatory response after flexor tendon injury and repair was first …

Single-Step Electrochemical Functionalization Of Double-Walled Carbon Nanotube (Dwcnt) Membranes And The Demonstration Of Ionic Rectification, Xin Zhan, Ji Wu, Zhiqiang Chen, Bruce J. Hinds

Chemistry Faculty Publications

Carbon nanotube (CNT) membranes allow the mimicking of natural ion channels for applications in drug delivery and chemical separation. Double-walled carbon nanotube membranes were simply functionalized with dye in a single step instead of the previous two-step functionalization. Non-faradic electrochemical impedance spectra indicated that the functionalized gatekeeper by single-step modification can be actuated to mimic the protein channel under bias. This functional chemistry was proven by a highly efficient ion rectification, wherein the highest experimental rectification factor of ferricyanide was up to 14.4. One-step functionalization by electrooxidation of amine provides a simple and promising functionalization chemistry for the application of …

Designing a ‘living archive’ that will enable new forms of circus performance to be realised is a complex and dynamic challenge. This paper discusses the methods and approaches used by the research team in the design of the Circus Oz Living Archive. Essential to this project has been the design of a responsive methodology that could embrace the diverse areas of knowledge and practice that have led to a design outcome that integrates the affordances of the circus with those of digital technologies. The term ‘living archive’ has been adopted as a means to articulate the dynamic nature of the …

The Fat Factory: Chewing The Fat, Mike Thompson, Danielle Arets Jun 2013

The Fat Factory: Chewing The Fat, Mike Thompson, Danielle Arets

Nordes Conference Series

In 2005, the global adult biomass hit around 287 million metric tons, 15 million metric tonnes of which being caused by an overweight global population (a body mass of 25 or greater). As the worlds population continues to soar (the UN estimates the world population will reach 9.1 billion by 2050) there will be considerably more mouths to feed, and energy needed to sustain this rate of development. Paradoxically then, fat is both a waste of resources and a valuable resource in its own right. The Fat Factory is a Critical Design Research project investigating the full, untapped potential of …

Storm System: Wearable Shelter For The Alpha Time Era, Miguel Rios Jun 2013

Storm System: Wearable Shelter For The Alpha Time Era, Miguel Rios

Nordes Conference Series

In this environmentally and demographically complex start to a post-industrial millennium, it is urgent to reflect on the transformations that occur from the interaction between individuals, the city they inhabit, its surroundings and protection conditions. STORM SYSTEM, developed by Miguel Rios Design, responds to the question of a first individual nomad clothing protection against weather adversities. Today’s population growth forces a reorganisation of space, in a variety of contexts that individuals face on large urban surfaces, as well as an interiorisation of the impacts resulting from behavioural changes. An unbearable logistic and environmental excess is therefore propagated (and vice versa), …

The extracellular matrix (ECM) is a critical tissue component, providing structural support as well as important regulatory signaling cues to govern cellular growth, metabolism, and differentiation. The study of ECM proteins, however, is hampered by the low solubility of ECM components in common solubilizing reagents. ECM proteins are often not detected during proteomics analyses using unbiased approaches due to solubility issues and relatively low abundance compared to highly abundant cytoplasmic and mitochondrial proteins. Decellularization has become a common technique for ECM protein-enrichment and is frequently used in engineering studies. Solubilizing the ECM after decellularization for further proteomic examination has not …

Another Future For Designers In America, Kate Catterall

Learn X Design Conference Series

The rationale behind mass-industrialization, now normalized, is embodied uncritically in the practice of many new designers. What if the industrial past could be explored and experienced as a foreign territory, providing a vantage point from which to critically evaluate contemporary design practices and define new paths? Young American designers are searching for alternate roles, and ways to design and live. Many experiment with models from elsewhere, places where new futures are growing from useful pre-industrial remnants. America, a country synonymous with industrialization, has no such resources upon which to build new practices, and slowing while creating value through quality is …

A 3d Interactive Model And Atlas Of The Jaw Musculature Of Alligator Mississippiensis, Casey M. Holliday, Henry P. Tsai, Rebecca J. Skiljan, Ian D. George, Sami Pathan

College of Health and Human Services

Modern imaging and dissemination methods enable morphologists to share complex, three-dimensional (3D) data in ways not previously possible. Here we present a 3D interactive model of the jaw musculature of the American Alligator (Alligator mississippiensis). Alligator and crocodylian jaw musculature is notoriously challenging to inspect and interpret because of the derived nature of the feeding apparatus. Using Iodine-contrast enhanced microCT imaging, a segmented model of jaw muscles, trigeminal nerve, brain and skull are presented as a cross-sectional atlas and 3D, interactive pdf of the rendered model. Modern 3D dissemination methods like this 3D Alligator hold great potential for morphologists to …

Two Distinct Modes Of Exocytotic Fusion Pore Expansion In Large Astrocytic Vesicles, Hong Peng, Ning Kang, Jun Xu, Patric K. Stanton, Jian Kang

NYMC Faculty Publications

Formation of the fusion pore is a central question for regulated exocytosis by which secretory cells release neurotransmitters or hormones. Here, by dynamically monitoring exocytosis of large vesicles (2-7 μM) in astrocytes with two-photon microscopy imaging, we found that the exocytotic fusion pore was generated from the SNARE-dependent fusion at a ring shape of the docked plasma-vesicular membrane and the movement of a fusion-produced membrane fragment. We observed two modes of fragment movements, 1) a shift fragment that shifted to expand the fusion pore and 2) a fall-in fragment that fell into the collapsed vesicle to expand the fusion pore. …
