Functional Analysis Of The Acetic Acid Resistance (Aar) Gene Cluster In Acetobacter Aceti Strain 1023, Elwood Mullins, T Joseph Kappock

Vinegar production requires acetic acid bacteria that produce, tolerate, and conserve high levels of acetic acid. When ethanol is depleted, aerobic acetate overoxidation to carbon dioxide ensues. The resulting diauxic growth pattern has two logarithmic growth phases, the first associated with ethanol oxidation and the second associated with acetate overoxidation. The vinegar factory isolate Acetobacter aceti strain 1023 has a long intermediate stationary phase that persists at elevated acetic acid levels. Strain 1023 conserves acetic acid despite possessing a complete set of citric acid cycle (CAC) enzymes, including succinyl-CoA:acetate CoA-transferase (SCACT), the product of the acetic acid resistance (aar …

Animals that sustain high levels of aerobic activity under hypoxic conditions (e.g., birds that fly at high altitude) face the physiological challenge of jointly optimizing blood-O2 affinity for O2 loading in the pulmonary circulation and O2 unloading in the systemic circulation. At high altitude, this challenge is especially acute for small endotherms like hummingbirds that have exceedingly high mass-specific metabolic rates. Here we report an experimental analysis of hemoglobin (Hb) function in South American hummingbirds that revealed a positive correlation between Hb-O2 affinity and native elevation. The development of hybrid PET/MRI imaging systems needs to be paralleled with the development of a hybrid intrinsic PET/MRI probes. The aim of this work was to develop and validate a novel radio-superparamagnetic nanoparticle (r-SPNP) for hybrid PET/MRI imaging. This was achieved with the synthesis of superparamagnetic iron oxide nanoparticles (SPIONs) that intrinsically incorporated 59Fe and manganese iron oxide nanoparticles (MIONs) that intrinsically incorporated 52Mn. Both [59Fe]-SPIONs and [52Mn]-MIONs were produced through thermal decomposition synthesis. This thesis project examines the history, provenance, and contemporary treatment of a rare Imperial Edition of Frederick Sander’s print collection Reichenbachia, Orchids Illustrated and Described, a high-quality orchid compendium dating to the late-nineteenth century. A local philanthropist loaned the Imperial Edition Reichenbachia, number 86 of 100 to Lewis Ginter Botanical Garden in 2011 on a long-term basis as a promised donation. Research into the origins of this collection involves several disparate historical topics, including the Victorian period of “orchid mania,” imperialist business practices, and chromolithographic printmaking. We assess climate impacts of global warming using ongoing observations and paleoclimate data. We use Earth's measured energy imbalance, paleoclimate data, and simple representations of the global carbon cycle and temperature to define emission reductions needed to stabilize climate and avoid potentially disastrous impacts on today's young people, future generations, and nature. A cumulative industrial-era limit of ∼500 GtC fossil fuel emissions and 100 GtC storage in the biosphere and soil would keep climate close to the Holocene range to which humanity and other species are adapted. Cumulative emissions of ∼1000 GtC, sometimes associated with 2°C global warming, would spur …

In this thesis, we have focused on the following two projects: (1) mechanisms of peptide hydrolysis, and (2) aggregation of biomolecules. A plethora of theoretical and computational techniques; quantum mechanics, molecular dynamics, and molecular docking have been employed to accomplish the goals of these projects. The selective hydrolytic cleavage of the extremely stable peptide bond (-(O=)C-NH-) of peptides and proteins by enzymes and their analogues is required in a wide range of biological, biotechnological and industrial applications. Florida pompano (Trachinotus carolinus) is a strong candidate for aquaculture production growth in the United States. Demonstrated through research, the species meets many of the criteria for species selection. Research is needed to complete our knowledge of the nutritional requirements at different ages in order to develop cost-effective and environmentally-friendly diets. Our goal was to examine the nutritional requirements in terms of protein and energy for market size fish. Six diets were formulated to be isoenergetic (Gross energy GE= 21 kJ/g). Lipid profiles of seven human breast milk samples obtained from milk banks and four infant formulas were compared in view of the potential food hypersensitivities of certain infants to human milk.The cholesterol (0.15-0.26 mM) content of the human samples was about 50% lower than that found in the infant formulas whereas the triglyceride (TG, 173-386 mM) contents of these products were found to be comparable.The major saturated fatty acid (SFA) and mono-unsaturated fatty acid (MUFA) were 16:0 and 18:1 respectively. In this dissertation, methods to synthesize ZnO are reported. First, zinc oxide nanoparticles were synthesized with small amounts of transition metal ions to create materials called dilute magnetic semiconductors (DMS). We employed a low temperature sol-gel method that produces ZnO nanoparticles of reproducible size and incorporates cobalt, nickel, and manganese ions into the nanoparticles. Conditions were controlled such that a range of amounts of Co, Ni, and Mn were incorporated. The incorporation was tracked by color changes in the white ZnO powder to blue for Co, green for Ni and yellow for Mn. XRD measurements showed the nanoparticles were on …

In this thesis we propose methods and metrics for human interaction with bio-inspired robot teams. We refine the concept of a stakeholder and demonstrate how a human can use stakeholders to lead a swarm as well as switch the swarm between different collective behaviors. We extend the human interaction metrics of interaction time and interaction effort presented in [1] to swarm systems and introduce the concept of interaction effort. Per capita alcohol consumption by Irish teenagers has doubled over the past three decades. There has also been a doubling of the suicide rate among young men. The aim of this study was to measure the correlation between alcohol consumption and negative mood (as measured by elements of the Beck Depression Inventory) in a sample (n = 169) of final-year secondary school male students. A questionnaire was devised to ascertain frequency, type and quantity of alcohol consumed, as well as attitudes towards drinking in general. Flood zones with 1% and 0.02% of annual flooding chance are projected in the ‎Federal Emergency Management Agency’s (FEMA) digital flood insurance rate maps ‎‎(DFIRMs) and are suited for identifying flood risk at the largest impacts. However, less ‎severe floods, which are not mapped in DFIRMs, still cause significant damage and ‎occur on a more frequent basis. This article employs an easy-to-setup GIS-based ‎solution for rapid inundation mapping of small flood events. The linear interpolation ‎technique (LITE Flood) is developed to rescale the hydraulic behavior inherent with a ‎larger flood event without performing additional hydraulic simulations. Majority of Registered Dietitians (RD) in the United States are Caucasian females. This study aims to understand the factors associated with low enrollment of minority students in the dietetics major and difficulties faced in the process of becoming an RD. This was a descriptive survey research. Three sets of survey were given to ethnic minorities who were (1) undergraduate dietetic students in Tennessee, (2) undergraduate students not majoring in dietetics at University of Memphis, and (3) dietetic interns in Tennessee. Although the risk factor for apolipoprotein E (apoE) polymorphism in Alzheimer's disease (AD) has been well described, the role that apoE plays in other neurodegenerative diseases, including Pick's disease, is not well established. To examine a possible role of apoE in Pick's disease, an immunohistochemical analysis was performed utilizing a novel site-directed antibody that is specific for an amino-terminal fragment of apoE. Application of this antibody, termed the amino-terminal apoE cleavage fragment (nApoECF) antibody, consistently labeled Pick bodies within area CA1 of the hippocampus in 4 of the 5 cases examined. Temperature is often considered a master variable controlling many chemical reactions and biological processes. Research efforts have begun address how temperature affects whole stream nutrient uptake rates, but little has attention has been paid to the influence of temperature on nutrient uptake versus concentration kinetics. In this study, we investigated 1) how ambient nutrient uptake rates and 2) nutrient uptake kinetics vary along a thermal gradient established by an altitudinal gradient in the Rocky Mountains, Colorado, USA. Poa annua (annual bluegrass; ABG) is an invasive weedy species in turfgrass. Herbicides and plant growth regulators (PGRs) are often used for ABG control, providing limited or inconsistent results. Identifying shifts in ABG populations in response to these treatments would be beneficial for understanding inconsistent control. Our research employed amplified fragment length polymorphic (AFLP) markers with the objective to determine if there are changes in genetic structure of ABG populations after multiple-year season-long control programs in three states. Zinc is an essential cofactor of several proteins. The roles of zinc in these proteins are classified as catalytic, structural or regulatory. Zinc present in structural sites is considered to be a chemically inert, static structural element. On the contrary, previous studies on a C2H2 type zinc finger model compound and the C3H type HIV-1 NCp7 C-terminal zinc knuckle have shown that zinc at these sites can undergo coordination sphere expansion under the influence of a Pt based electrophile. Climate has been inherently linked to global diversity patterns, and yet no empirical data are available to put modern climate change into a millennial-scale context. High tropical species diversity has been linked to slow rates of climate change during the Quaternary, an assumption that lacks an empirical foundation. Thus, there is the need for quantifying the velocity at which the bioclimatic space changed during the Quaternary in the tropics.
