Background: Developmental programs are implemented by regulatory interactions between Transcription Factors (TFs) and their target genes, which remain poorly understood. While recent studies have focused on regulatory cascades of TFs that govern early development, little is known about how the ultimate effectors of cell differentiation are selected and controlled. We addressed this question during late Drosophila embryogenesis, when the finely tuned expression of the TF Ovo/Shavenbaby (Svb) triggers the morphological differentiation of epidermal trichomes.

Results: We defined a sizeable set of genes downstream of Svb and used in vivo assays to delineate 14 enhancers driving their specific expression in trichome …

FOF1 ATP synthases are rotary nanomotors that couple proton translocation across biological membranes to the synthesis/hydrolysis of ATP. During catalysis, the peripheral stalk, composed of two b subunits and subunit δ in Escherichia coli, counteracts the torque generated by the rotation of the central stalk. Here we characterize individual interactions of the b subunits within the stator by use of monoclonal antibodies and nearest neighbor analyses via intersubunit disulfide bond formation. Macrophages perform a variety of functions from phagocytosis and the release of pro-inflammatory cytokines to the repair and remodeling of damaged tissues. Two phenotypically different types of macrophages have been proposed to identify their diverse roles: types M1 and M2. (Mantovani et al., 2002) The M1 macrophage refers to the cell which aggressively destroys foreign invaders and tumor cells, and releases pro-inflammatory cytokines. Antigen presentation by the M1 links innate immunity to adaptive immunity. The M2 macrophage acts as a support cell by promoting the healing of damaged cells by angiogenesis and tissue remodeling. Identity Development Of Adolescent Gay Black Males, Miles James Allen Crumley

During adolescence, self-identified gay black males may develop their identities differently than their gay white male counterparts. This may be attributed to the reconciliation of stressors when developing gay, black, and male identities within certain environmental contexts. To investigate this, twelve qualitative interviews were conducted of gay black males from which developmental themes were extracted. While many of the developmental processes are similar to their white homosexual counterparts, some differences were noted regarding racism, objectification by the white gay community, and use of the internet to develop particular identities. There are several approaches to obtaining unnatural analogues of natural products. One of these approaches is precursor-directed biosynthesis (PDB), which is the process of using natural biosynthetic machinery to transform unnatural analogues of precursor compounds to produce an unnatural analogue of a target natural product. Berberine and galanthamine are chosen as model systems for the evaluation of PDB for the production of aromatic alkaloids in plants. Several approaches to the synthesis of unnatural 4 hydroxyphenylacetaldehyde (HPAA), norcoclaurine, and tyramine analogues are explored. 3-chloro-, 3-bromo-, and 3-iodoHPAA are all synthesized in good yields via and oxidative decarboxylation of tyrosine. Purpose: This dissertation focuses on establishment of pre-clinical methods facilitating the use of PET imaging for selective sub-volume dose escalation. Specifically the problems addressed are 1.) The difficulties associated with comparing multiple PET images, 2.) The need for further validation of novel PET tracers before their implementation in dose escalation schema and 3.) The lack of concrete pre-clinical data supporting the use of PET images for guidance of selective sub-volume dose escalations. BipA is a conserved prokaryotic GTPase necessary for securing bacterial survival and successful invasion of the host. Structural and biochemical studies indicate that GTP and ppGpp compete for binding to BipA to promote differential association of BipA to either the 70S or 30S ribosomal species. Exactly how guanine nucleotide binding to BipA prompts a change in the association of this protein with the ribosome is not understood. Crystallographic models show local structural rearrangements occur near the nucleotide-binding pocket but unexpectedly the overall domain arrangement, and therefore intramolecular contacts are similar in the various guanine nucleotide bound states. The zebrafish has emerged as an important model of heart development and regeneration. While the structural characteristics of the developing and adult zebrafish ventricle have been previously studied, little attention has been paid to the nature of the interface between the compact and spongy myocardium. Here we describe how these two distinct layers are structurally and functionally integrated. We demonstrate by transmission electron microscopy that this interface is complex and composed primarily of a junctional region occupied by collagen, as well as a population of fibroblasts that form a highly complex network. In Chapter 1, the roles of genes that specify antennal identity in Drosophila melanogaster were investigated in the flour beetle Tribolium castaneum. Antenna-to-leg transformations occurred in response to RNA interference (RNAi) against homothorax, extradenticle, spineless and Distal-less. However, for homothorax/extradenticle RNAi, the extent of transformation along the proximodistal axis differed between embryogenesis and metamorphosis. In chapter 2, the metamorphic roles of the Hox genes, extradenticle, and homothorax were compared in T. castaneum. homothorax/extradenticle RNAi and Hox RNAi produced similar body wall phenotypes but different appendage phenotypes. Non-alcoholic fatty liver disease (NAFLD) is a clinicopathological entity that includes a spectrum of histological and biochemical anomalies. NAFLD begins with excessive lipid accumulation within hepatocytes (hepatic steatosis) and can transition to non-alcoholic steatohepatitis (NASH), characterized by hepatic inflammation and injury that can render the liver vulnerable to bridging fibrosis, cirrhosis, and hepatocellular carcinoma. Dietary cholesterol has been proposed as a potential mediator of the transition from steatosis to NASH. Organisms undertake actions on the basis of perceptions. Perceptions serve as the basis for what an agent takes to be the case; that is, for the agent’s beliefs. However, the idea of belief is freighted with notions of fallibility and subjectivity, since belief is often considered insofar as it is distinct from knowledge. Here, an attempt is made to link belief more closely with perception and action. This linkage is shown by considering the role belief plays in determining behavior, which is distinct from the role belief plays in language-based philosophical accounts of content and intention. that promote thriving among African American adolescents. Specifically, this study examined how religiosity, religious support, racial identity, and communalism relate to thriving. This study

Resource partitioning within the ecological niche space in which there is a high level of overlap between species can alleviate the tendency toward competitive exclusion. When competitive ability is asymmetrical due to predation or other ecological factors, it may be more effective for the less competitive species to lessen direct competition by contracting their use of local resources. Thermal barrier coatings (TBCs) are used in gas turbines to reduce the metal temperature for a given operating condition. The temperature drop in the TBC can be exploited by running the engine at a higher firing temperature leading to greater fuel efficiency and/or increased durability. Eventually TBCs spall off. One of the contributing factors to the failure in any TBC system is the surface geometry change of the interface commonly known as Bond-coat rumpling. The use of human and veterinary pharmaceuticals is increasing. Over the past decade, there has been a proliferation of research into potential environmental impacts of pharmaceuticals in the environment. A Royal Society-supported seminar brought together experts from diverse scientific fields to discuss the risks posed by pharmaceuticals to wildlife. Recent analytical advances have revealed that pharmaceuticals are entering habitats via water, sewage, manure and animal carcases, and dispersing through food chains. Pharmaceuticals are designed to alter physiology at low doses and so can be particularly potent contaminants. The near extinction of Asian vultures following exposure to diclofenac is the key …
